Kasturi (Mangifera casturi) is a typical plant of South Kalimantan which has only been used for consumption because of its distinctive taste and aroma. Mangifera indica, which is in the same genus as Mangifera casturi, shows antioxidant, antibacterial, antifungal, and antidiarrheal activity, so it is suspected that this plant has the same activity. Crude methanol extract from the meat of Casturi mango was fractionated using n-hexane, ethyl acetate, and methanol-water. Testing for antibacterial activity and minimum inhibitory concentration (MIC) were carried out using the disk diffusion method (Kirby-Bauer). The positive control used was Chloramphenicol because it has an antibacterial range that is rather broad and the negative control used was 10% DMSO. The fraction that had the greatest antibacterial activity was the ethyl acetate fraction; with the formation of a clear zone of 7.98mm against Pseudomonas aeruginosa and 7.52mm against Bacillus cereus, with a minimum inhibitory concentration of 2mg/mL. The next test was a cell leakage analysis which revealed an increase in absorbance at MIC 1 and MIC 2. The Gas Chromatography – Mass Spectrometry (GC-MS) results from the ethyl acetate fraction revealed the presence of phenolic, flavonoid, and glycoide content, with the highest compound concentration of 5-Hydroxymethylfurfural. Keywords: Antibacterial, Extract, Ethyl acetate, fraction, Mangifera casturi Abstrak Kasturi (Mangifera casturi) merupakan tumbuhan khas Kalimantan Selatan yang selama ini hanya dimanfaatkan untuk dikonsumsi karena rasa dan aromanya yang khas. Mangifera indica yang masih satu genus dengan Mangifera casturi menunjukkan aktivitas antioksidan, antibakteri, antijamur, dan antidiare sehingga diduga tanaman ini memiliki aktivitas yang sama. Ekstrak kasar metanol daging buah mangga kasturi difraksinasi dengan menggunakan n-heksan, etil asetat, dan metanol-air. Pengujian aktivitas antibakteri dan kadar hambat minimum dilakukan dengan menggunakan metode difusi cakram (Kirby- Bauer). Kontrol positif yang digunakan adalah Chloramfenikol karena memiliki spektrum antibakteri yang cukup luas dan kontrol negatif yang digunakan adalah DMSO 10%. Fraksi yang memiliki aktivitas antibakteri terbesar pada penelitian ini adalah fraksi etil asetat dengan terbentuknya zona bening sebesar 7,98mm terhadap Pseudomonas aeruginosa dan 7,52mm terhadap Bacillus cereus dengan kadar hambat minimum 2mg/mL. Uji selanjutnya adalah uji kebocoran sel yang menyatakan adanya peningkatan absorbansi pada KHM 1 dan KHM 2. Hasil pemeriksaan Gas Chromatography – Mass Spectrometry (GCMS) dari fraksi etil asetat menyatakan adanya kandungan fenolik, flavonoid, dan glikosida dengan senyawa tertinggi yaitu 5-Hydroxymethylfurfural. Cissus trifoliata (L.) L belongs to the Vitaceae family and is an important medicinal plant used in Mexico for the management of infectious diseases and tumors. The present study aimed to evaluate the metabolic profile of the stems of C. trifoliata and to correlate the results with their antibacterial and cytotoxic activities. The hexane extract was analyzed using gas chromatography coupled with mass spectrometry (GC-MS) and the CHCl3-MeOH and aqueous extracts by ultraperformance liquid chromatography quadrupole time of fly mass spectrometry (UPLC-QTOF-MS). The antibacterial activity was determined by broth microdilution and the cytotoxicity was evaluated using MTS cell proliferation assay. Forty-six metabolites were putatively identified from the three extracts. Overall, terpenes, flavonoids and stilbenes characterize the metabolic profile. No antibacterial activity was found in any extract against the fifteen bacteria strains tested (MIC >500 µg/mL). However, high cytotoxic activity (IC50 ≤ 30 µg/mL) was found in the hexane and aqueous extracts against hepatocarcinoma and breast cancer cells (Hep3B, HepG2 and MCF7). This is the first report of the bioactive compounds of C. trifoliata stems and their antibacterial and cytotoxic properties. The metabolic profile rich in anticancer compounds correlate with the cytotoxic activity of the extracts from the stems of C. trifoliata. This study shows the antitumor effects of this plant used in the traditional medicine and justifies further research of its anticancer activity.

Mikania guaco Bonpl. methanol extract showed an interesting activity against Trypanosoma cruzi with an IC50 of 4 μg/mL. Bioassay-guided fractionation of the methanol extract resulted in the isolation of a new sesquiterpene lactone, 2α-acetoxy, 15-acetylartemisiifolin (1), which was shown to be active (IC50 = 25 μM) against the amastigote form of T. cruzi,. The chemical structure of 1 was elucidated using 2-D NMR techniques and mass spectrometry.

Bioassay guided fractionation of the MeOH extract of the plant Gutierrezia sarothrae (Asteraceae) using an assay for antiplasmodial activity against the drug-resistant Dd2 strain of Plasmodium falciparum led to the isolation of the two new diterpenes 3α-angeloyloxy-15-hydroxylabda-7,13-dien-16,15-olid-18-oic acid (1) and 3α-angeloyloxy-15-methoxylabda-7,13-dien-16,15-olid-18-oic acid (2). The structures of 1 and 2 were elucidated by interpretation of ID and 2D NMR spectroscopic data, mass spectrometry, and comparison with the data of related compounds reported in the literature. Compound 1 exhibited moderate antiplasmodial activity with an IC50 values of 10.4 ± 4.3 μM.

ABSTRACTTropodithietic acid (TDA) is an antibacterial compound produced by somePhaeobacterandRuegeriaspp. of theRoseobacterclade. TDA production is studied in marine broth or agar since antibacterial activity in other media is not observed. The purpose of this study was to determine how TDA production is influenced by substrate components. High concentrations of ferric citrate, as present in marine broth, or other iron sources were required for production of antibacterially active TDA. However, when supernatants of noninhibitory, low-iron cultures ofPhaeobacter inhibenswere acidified, antibacterial activity was detected in a bioassay. The absence of TDA in nonacidified cultures and the presence of TDA in acidified cultures were verified by liquid chromatography–high-resolution mass spectrometry. A noninhibitory TDA analog (pre-TDA) was produced byP. inhibens,Ruegeria mobilisF1926, andPhaeobactersp. strain 27-4 under low-iron concentrations and was instantaneously converted to TDA when pH was lowered. Production of TDA in the presence of Fe3+coincides with formation of a dark brown substance, which could be precipitated by acid addition. From this brown pigment TDA could be liberated slowly with aqueous ammonia, and both direct-infusion mass spectrometry and elemental analysis indicated a [FeIII(TDA)2]xcomplex. The pigment could also be produced by precipitation of pure TDA with FeCl3. Our results raise questions about how biologically active TDA is produced in natural marine settings where iron is typically limited and whether the affinity of TDA to iron points to a physiological or ecological function of TDA other than as an antibacterial compound.

Essential oil extracted from fresh aerial parts ofAthamanta siculaL. (syn.Tingara sicula) was analysed by gas phase chomatography coupled to mass spectrometry (GC-MS). The main constituents were: germacrene B (88.5%) and apiol (4.9%). Comparing with the tested bacteria, the growth ofEscherichia coliandKlebsiella pneumoniaestrains was more inhibited by the essential oil ofA. sicula.
