Effect of tree age and portions of Cassia fistula L. on variation of total extractives and its reactivity towards formaldehyde

O trabalho teve por objetivo: verificar a influência da idade da árvore de candeia sobre os elementos anatômicos, teores de extrativos totais, lignina, cinzas e densidade básica, relacionado a duas condições de relevo (1.000 m e 1.100 m de altitude), em Aiuruoca, Minas Gerais. Em cada área de estudo foram amostradas 15 árvores distribuídas em 3 classes diamétricas, das quais foram retirados discos da base da árvore para determinações da idade, características anatômicas e densidade básica. Os fustes foram reduzidos na forma de cavacos onde foram retiradas amostras aleatórias de material para análises químicas. As características anatômicas das madeiras das duas áreas foram semelhantes, com variações na quantidade de vasos e tamanho de pontoações. As células de óleo apresentaram-se nas células do parênquima radial. A freqüência de vasos foi menor em árvores mais velhas. A densidade básica variou de 0,74 g/cm3 na área com 1.000 m e 0,68 g/cm3 para a área de 1.100 m de altitude, sendo maior em árvores mais velhas. Os teores de extrativos totais, lignina e cinzas, diferiram estatisticamente entre as madeiras das duas áreas estudadas. As árvores mais jovens apresentaram maior teor de material inorgânico; o teor de extrativos totais e de lignina, não foi influenciado pela idade.Palavras-chave:Caracterização anatômica; composição química; densidade básica; Eremanthus erythropappus.                                                                                                                                                AbstractInfluence of altitude and stand age on basic density, anatomical, and chemical characteristics of candeia wood - Eremanthus erythropappus. This work aimed to study the influence of tree age and altitude on some anatomical elements characteristics (basic density, total extractives, lignin, and leached ashes) of candeia wood. The sampled trees were collected in the Aiuruoca County, Minas Gersis State, Brazil, in two different altitudes (1,000 and 1,100m above sea level). From each altitude level 15 trees, enclosed in 3 diametric classes, were cut. Disks were taken from the trunk base of each tree in order to determine the age, the anatomical characteristics, and the basic density. Random samples from the trees trunks were collected for the chemical analyses. The wood density varied from 0.74 to 0.68 g/cm3 in the trees collected at 1,000 and 1,100m of altitude respectively, being higher in the older trees. The wood anatomical characteristics of both sites were similar, with some variations in the amount of vases and pit sizes. Oily cells were observed in the radial parenchyma tissue. The vases frequency was lower in the older trees. The content of total extractives, amount of lignin, and leached ashes were statistically different in the two studied sites. The youngest trees presented higher leached ashes content; total extractives and lignin were not influenced by tree age.Keywords:Anatomical characterization; chemical components; basic density; Eremanthus erythropappus.

During the first 20 years of a Pinus radiata tree rotation, tree growth and pasture yield were assessed under a range of tree spacings at Invermay and Akatore, two coastal sites in Eastern Otago. Pasture yield in association with trees thinned to 100 stems per hectare (sph) was comparable to that from open pasture up to a tree age of 12 years. By the 19th year, however, pasture production declined to 63% of open pasture yield at Invermay and to 42% at Akatore. At 200 and 400 sph at Akatore, pasture yield was similar to that from open pasture at tree age 12 years but declined to 27% and 0% of open pasture yield respectively by year 20. At both Invermay and Akatore, the ryegrass and clover content of open pasture was relatively constant throughout the term of the trial. However, both the ryegrass and clover content of pasture beneath trees began to decline by tree age 12 years with a very rapid decline at Akatore in the number of pasture species at 200 sph by the 19th year. No pasture remained at 400 sph, after 19 years. Livestock carrying capacity with sheep on tree treatments at Invermay decreased from 100% of open pasture at year 6 to 60% by year 10. At Akatore, livestock carrying capacity averaged over the 20-year life of the trial was 4.1 stock units per hectare with a maximum of 8.1 stock units at a tree age of 8 years. Tree growth at both sites was similar, averaging between 1 and 1.1 m/year in height over 20 years, with trees at Invermay at 100 sph averaging 9% greater height and diameter growth than at Akatore. Increasing tree stocking from 100 to 200 to 400 sph at Akatore, resulted in increased tree height, but decreased diameter at breast height. A comparison of the East Otago trees with those in a similar trial at Tikitere (Rotorua) 900 km further north indicated that the southern trees were about 6 years later in their growth pattern by tree age 20 years. On both sites, soil pH tended to be lower in the presence of trees and was significantly lower than in open pasture by year 20. The results and comparisons with the Tikitere data suggest that, in an integrated agroforestry regime, there will be livestock grazing under the trees further into the tree rotation in Otago than in North Island sites. However, slower tree growth would result in a longer rotation time to harvest. Current recommendations to farmers are to plant trees on the less productive areas of the farm and adopt a tree stocking rate which fully utilises the site. Trees planted from agroforestry practices can become valuable resources in meeting the wood requirements of many nations. Gliricidia sepium is an exotic species introduced to the agricultural sector in Malaysia mainly for providing shade for cocoa and coffee plantations. This study investigates its wood physical properties (specific gravity and moisture content) and fibre morphology (length, lumen diameter and cell wall thickness) of G. sepium at three intervals according to age groups ( three, five and seven years of ages). Specific gravity (0.72) was significantly higher at seven years ofage as compared to five (0.41) and three (0.35) years age group with a mean of 0.43 (p<0.05). Mean moisture content was 58.3% with no significant difference existing between the tree age groups. Fibre diameter (22.4 mm) was significantly lower (p<0.05) for the trees which were three years of age when compared to five and seven years age groups (26.6 mm and 24. 7 mm), respectively. Means of fibre length, lumen diameter and cell wall thickness were 0.83 mm, 18.3 mm, and 6.2 mm, respectively, with no significant differences detected between trees in all age groups. Further calculation on the coefficient of suppleness and runkel ratio suggest that wood from G.sepium may have the potential for insulation board manufacturing and paper making. However, future studies should experiment the utilisation of this species for these products to determine its full potential.

Filipendula ulmaria (L.) Maxim. (Meadowsweet) is known in traditional medicine as anti-inflammatory, wound-healing,astringent and antibacterial remedy. However recent studies show that it also has neurotropic activity. In Russia meadowsweet flowers are used as crude herbal drugs (temporary pharmacopoeial monograph 42-1777-87), also leafs and herb are used in the traditional medicine. Objective of the study was to carry out comparative investigation of composition and content of major biologically active compounds (BAC) in Filipendula ulmaria herb, flowers and leafs by thin-layer chromatography, differential spectrophotometry with aluminum chloride reagent (total flavonoids in terms of rutoside), gravimetry (total extractives, extracted by water), permanganatometric titration (total tannins in terms of tannin). Rutoside, tannin, gallic acid and salicylic acid were identified in Filipendula ulmaria herb, flowers and leafs by TLC. Also we analyzed content of substances extracted by water, flavonoids and tannins. Total extractives, extracted by water in F. ulmaria herb is 13.12±0.10%, in leafs – 13.98±0.37%, in flowers – 18.09±0.17%. Total tannins in F. ulmaria herb is 11.87±0.47%, in leafs – 12.06±0.18%, in flowers – 12.26±0.29%. Total flavonoids in F. ulmaria herb 4.34±0.17%, in leafs – 6.98±0.23%, in flowers – 11.75±0.57%. The obtained data will be used for development of a pharmacopoeial monograph project "Filipendula ulmaria (L.) Maxim., herba" for inclusion in the State Pharmacopoeia of the Russian Federation.

Temporal studies were conducted from mid- to late-harvest season of `Ruby Red' grapefruit (Citrus paradisi Macf.) to evaluate the effect of on- and off-tree storage, fruit size, and juice vesicle position on the development of granulation. Juice vesicle fresh and dry masses were highest at the stem and stylar positions of the fruit section and were not affected significantly by time of harvest or by storage. Juice vesicles isolated from each position were subjectively evaluated for the presence of granulation. Granulation was highest in stylar juice vesicles obtained from large fruit (≈600 g) that were harvested late in the season (March and May) and stored in air at 21 °C for 60 days. Large fruit harvested in March and May and examined immediately, and fruit harvested in January and stored for 60 days had low granulation scores. Thus, fruit remaining on the tree until May are less susceptible to the disorder than those harvested in March and held in storage until May. Levels of alcohol-insoluble solids (AIS), largely composed of pectins and other cell wall materials, were significantly higher in juice vesicles that were granulated. The results suggest that storage itself was not responsible for the marked accumulation of AIS in granulated juice vesicles. Rather, some interaction of fruit size with maturation, as well as other factors such as tree age and rootstock, likely contributed to the development of granulation.

Aim: This study was focused on the formulation of the multi-unit extended-release peroral delivery device of lamotrigine for better management of epilepsy. Background: The single-unit extended-release peroral preparations often suffer from all-or-none effect. A significant number of multi-unit delivery systems have been reported as a solution to this problem. But most of them are found to be composed of synthetic, semi-synthetic or their combination having physiological toxicity as well as negative environmental impact. Therefore, fabrication and formulation of multi-unit extended-release peroral preparations with natural, non-toxic, biodegradable polymers employing green manufacturing processes are being appreciated worldwide. Objective: Lamotrigine-loaded extended-release multi-unit beads have been fabricated with the incorporation of a natural polysaccharide Cassia fistula seed gum in calcium-cross-linked alginate matrix employing a simple green process and 23 full factorial design. Methods: The total polymer concentration, polymer ratio and [CaCl2] were considered as independent formulation variables with two different levels of each for the experiment-design. The extended-release beads were then prepared by the ionotropic gelation method using calcium chloride as the crosslinkerions provider. The beads were then evaluated for drug encapsulation efficiency and drug release. ANOVA of all the dependent variables such as DEE, cumulative % drug release at 2h, 5h, 12h, rate constant and dissolution similarity factor (f2) was done by 23 full factorial design using Design-Expert software along with numerical optimization of the independent variables in order to meet USP-reference release profile. Results: The optimized batch showed excellent outcomes with DEE of 84.7 ± 2.7 (%), CPR2h of 8.41± 2.96 (%), CPR5h of 36.8± 4.7 (%), CPR12h of 87.3 ± 3.64 (%) and f2 of 65.9. Conclusion: This approach of the development of multi-unit oral devices utilizing natural polysaccharides might be inspiring towards the world-wide effort for green manufacturing of sustained-release drug products by the QbD route.

Abstract Objective This study was aimed at evaluating the toxicity and the antioxidant property of Cassia fistula stem bark extracts in zebrafish. Materials and Methods Crude aqueous and ethanol extracts of C. fistula stem bark were obtained following a standard solvent-based extraction method. The toxicity of these extracts on zebrafish embryonic development was determined and the LC50 values were calculated. Finally, the antioxidant property of C. fistula stem bark extracts was determined in arsenic-induced oxidative stress models of zebrafish. Results The aqueous extract of C. fistula stem bark showed a slightly larger LC50 value (213.6 ppm) compared with the ethanol extract (LC50 = 63.5 ppm), suggesting a lower toxicity of the aqueous extract. A significant reduction of reactive oxygen species (ROS) signal was observed in arsenic-exposed embryos treated with the aqueous extract, but not the ethanol extract, indicating that the antioxidant activity is present only in the aqueous extract of C. fistula stem bark. Conclusion Identification of antioxidants from natural sources is desirable because of increasing safety concerns associated with synthetic antioxidants. This study demonstrated that aqueous extract from C. fistula stem bark possesses antioxidant properties, which can be further characterized for mechanism of action and potential applications.

Pecan is native to the United States. The US is the world’s largest pecan producer with an average yearly production of 250 to 300 million pounds; 80 percent of the world’s supply. Georgia, New Mexico, Texas, Arizona, Oklahoma, California, Louisiana, and Florida are the major US pecan producing states. Pecan trees frequently suffer from spring freeze at bud break and bloom as the buds are quite sensitive to freeze damage. This leads to poor flower and nut production. This review focuses on the impact of spring freeze during bud differentiation and flower development. Spring freeze kills the primary terminal buds, the pecan tree has a second chance for growth and flowering through secondary buds. Unfortunately, secondary buds have less bloom potential than primary buds and nut yield is reduced. Spring freeze damage depends on severity of the freeze, bud growth stage, cultivar type and tree age, tree height and tree vigor. This review discusses the impact of temperature on structure and function of male and female reproductive organs. It also summarizes carbohydrate relations as another factor that may play an important role in spring growth and transition of primary and secondary buds to flowers.
