Abstract The fish paste product, fish balls ‘tsumire’, is a traditional type of Japanese food made from minced fish as well as imitation crab, kamaboko and hanpen. Although tsumire is known as a high-protein and low-fat food, there is a lack of scientific evidence on its health benefits. Hence, we aimed to investigate the effects of tsumire intake on organ weight and biomarker levels in Sprague–Dawley rats for 84 d as a preliminary study. Six-week-old male Sprague–Dawley rats were divided into two groups: group I, fed normal diets, and group II, fed normal diets with 5 % dried tsumire. Throughout the administration period, we monitored their body weight and food intake; at the end of this period, we measured their organ weight and analysed their blood biochemistry. No significant differences were observed with respect to body weight, food intake, organ weight and many biochemical parameters between the two groups. It was found that inorganic phosphorus and glucose levels were higher in group II rats than in group I rats. On the other hand, sodium, calcium, amylase and cholinesterase levels were significantly lower in group II than in group I. Interestingly, we found that the levels of aspartate aminotransferase, alanine transaminase, lactate dehydrogenase and leucine aminopeptidase in group II were significantly lower than in group I, and that other liver function parameters of group II tended to be lower than in group I. In conclusion, we consider that the Japanese traditional food, ‘tsumire,’ may be effective as a functional food for human health management worldwide.

ABSTRACT Background: Infertility may give an impact on psychosocial. In Indonesia, the habit of consuming young dates is often done by couples to increase fertility. FSH is one of the factors that play a role in folliculogenesis. The lack of scientific evidence of young dates consumption effects on female reproduction has led to this study. This study aims to determine the levels of FSH hormone and the picture of ovarian tissue in mice after administration of young dates. Method: This study used an experimental design using 28 female Sprague-Dawley rats which were randomly divided into 4 groups. Group I to III was given young dates in successive doses: 17 mg, 34 mg, 68 mg per 200 grams of body weight and group IV was given distilled water. The treatment was carried out for 28 days. FSH levels were examined before treatment during the proestrus phase which was known from microscopic examination of rat vaginal swabs. The treatment begins during the proestrus phase. Result: The mean baseline and post-treatment FSH levels were 0.08 and 0.09 respectively Conclusion: There is no significant increase of FSH levels and FSH receptors as the effect of giving young dates (Phoenix Dactylifera) to female Sprague-Dawley rats Keywords: Dates, FSH, FSH receptors, Rats   ABSTRAK Latar Belakang: Infertilitas dapat memberikan dampak psikososial. Metode: Penelitian ini menggunakan desain ekperimental menggunakan tikus Sprague dawney betina sebanyak 28 ekor yang dibagi secara acak menjadi 4 kelompok. Kelompok I-III diberikan kurma muda dosis berturut -turut 17 mg, 34 mg, 68 mg per 200 gram BB dan kelompok IV diberikan aquadest. Perlakuan dilakukan selama 28 hari. Kadar hormon FSH diperiksa sebelum perlakuan saat fase proestrus yang diketahui dari pemeriksaan mikroskopis swab vagina tikus. Perlakuan dimulai saat fase proestrus. Hasil: Rerata kadar FSH baseline dan paska perlakuan adalah 0,08 dan 0,09 Kesimpulan: Tidak terdapat peningkatan yang signifikan kadar FSH dan reseptor FSH terhadap efek pemberian buah kurma muda (Phoenix Dactylifera) pada uterus tikus Spague Dawney Kata kunci : kurma, FSH, Reseptor FSH, tikus

BACKGROUND: Consumption of foods and drinks high in energy, fat, and/or sugar beyond the recommended quantities can cause obesity, which triggers the incidence of brain nerve cell death related to oxidative stress, high levels of tumor necrosis factor (TNF)-α and triglycerides, and low high-density lipoprotein (HDL) levels. Progressive nerve cell death causes decreasing cognitive performance. This study aims to prove that an American Institute of Nutrition committee in 1993 (AIN-93M) diet modified with high-fat-high-fructose (HFHF) can decrease the number of hippocampal neurons. A decrease in the number of hippocampal neurons indicates progressive nerve cell death.METHODS: An experimental study using a post-test control group design was carried out using male Sprague Dawley rats. Samples were selected using simple random sampling to divide them into two groups, Group I was AIN-93M-modified HFHF diet (n=14) and Group II was AIN-93M standard (n=16). The number of visible neurons was measured in the hippocampus area of Sprague Dawley rats’ brains, stained with haemotoxylin and eosin (H&E) and scanned under 400x magnification. Neurons were counted in 10 visual fields using the "Cell_Count" application.RESULTS: The data were analysed by Pearson’s correlation test using SPSS. The results show that rats in Group I had a greater weight gain and fewer neurons than those in the Group II (p=0.023, r=-0.413).CONCLUSION: The consumption of foods high in fat and fructose can cause an increase in nerve cell death, as shown by the decrease in the number of hippocampal neurons.KEYWORDS: brain nerve cells, high fat, high fructose, increased body weight

Introduction: Xiang Sha Yang Wei Wan (XSYWW) is a Chinese traditional medicine that is used for gastrointestinal disorders, specifically gastric ulcer in many countries of South-East Asia. The aim of the study was to evaluate the potential effects of XSYWW on ethanol-induced gastric ulcer in rats by means of histological Study. On a similar basis of treatment, ranitidine, a conventional medication was used as gold standard. Methods: Fifty five male Sprague-Dawley rats (250-300 gm) were divided into four groups. Group I (ethanol treated group) was the control group and gastric ulcers were induced by administering 100% ethanol (1 ml/200 g). Group II (Pre-treatment group) was divided into two subgroups; they were orally fed with 1.0 gm/kg and 2.0 gm/kg respectively of XSYWW solution. Thirty minutes later they were administered with absolute ethanol as in group I. Group III, was given an oral dose of 2gm/kg of XSYWW solution after one hour of ethanol administration. Group IV was given an oral dose of 200mg/kg ranitidine solution after one hour of ethanol administration. Five rats from groups I, III and IV were sacrificed on day 1, 2 and 3 while the animals of group II were sacrificed one hour after ethanol administration. Results: Histological study of the stomachs from ethanol treated rats showed multiple ulcers of various depths that reached the muscularis and the serosa. Conclusion: Pre or post-treated rats with XSYWW showed that XSYWW has protective effect against ethanol-induced gastric mucosal lesion. However, there was a faster and more complete healing process in the ranitidine treated group when compared to the XSYWW treated subjects.

We investigated the effect of repetitive postnatal (2–7 days) intracerebroventricular administration of neuropeptide Y (NPY) on food intake and body weight gain in the 3- to 120-day-old Sprague-Dawley rats. NPY caused a 32% transient increase in body weight gain with elevated circulating insulin concentrations within 24 h. This early intervention led to the persistence of hyperinsulinemia and relative hyperleptinemia with euglycemia in the 120-day-old female alone. This perturbation was associated with 50% suppression in adult female hypothalamic NPY concentrations and a 50–85% decline in NPY immunoreactivity in the paraventricular and arcuate nuclei. This change was paralleled by a ∼20% decline in food intake and body weight gain at 60 and 120 days. However, when exogenous NPY was stereotaxically reinjected into the paraventricular nucleus of the ∼120-day-old adult females who were pretreated with NPY postnatally, an increase in food intake and body weight gain was noted, attesting to no disruption in the NPY end-organ responsivity. We conclude that postnatal intracerebroventricular NPY has long-lasting effects that predetermine the resultant adult phenotype in a sex-specific manner.

Traditional herbal medicines have been used for centuries in Asian countries. However, recent studies have led to increasing concerns about the safety and toxicity of herbal prescriptions. Bojungikgi-tang (BJIGT), a herbal decoction, has been used in Korea to improve physical strength. To establish the safety information, BJIGT water extract was evaluated in a 4-week repeated-dose oral toxicity test in Crl:CD Sprague Dawley rats. BJIGT was orally administered in daily doses of 0, 500, 1000, and 2000 mg/kg/day for 4 weeks via oral gavage in male and female rats. We examined the mortality, clinical signs, body weight change, food intake, organ weights, hematology, serum biochemistry, and urinalysis parameters. No significant changes were observed in mortality, clinical sings, body weight, food intake, organ weights, hematology, serum biochemistry, and urinalysis parameters between the control group and the BJIGT-treated groups in the rats of both sexes. The results indicate that BJIGT did not induce toxic effects at a dose level up to 2000 mg/kg in rats. Thus, this concentration is considered the nonobservable effect dose in rats and is appropriate for a 13-week subchronic toxicity study.

ABSTRACTObjectives: The aim of this study was to investigate the effects of low levels of zinc intake on the rat mandible and maxilla during growth and to compare these results with those of zinc-containing rats.Methods: The study was carried out on 14 Sprague-Dawley rats. The rats were randomly divided into two groups. Group I rats were fed with a Zn-deficient diet, and Group II rats with a Zn-containing diet. At the end of the fourth week on the experimental diet, all the rats were killed and blood samples were taken. Serum Zn levels were measured by atomic absorption spectrophotometry. Then, the s ulls and mandibles were freed from soft tissues and measurements were made on the dry skulls, the mandibles, and teeth in both of the two groups.Results: The zinc-deficient group showed a significantly lower value in dry skull, mandible, and teeth measurements when compared with those of the Group II.Conclusions: Changes in zinc intake might exert an effect on the growth of craniofacial structures. A low-zinc diet during adolescence might slow bone and teeth growth and enhance the risk of oral, periodontal, and orthodontic problems in later years. (Eur J Dent 2009;3:10-15)

Ethane in alveolar expirate may have its source in organs other than the lung and be transported to the lung for elimination. We determined ethane production rates in rats (group I) ventilated with hydrocarbon-free air (HFA) before and after exsanguination. To determine whether the lung is the source of increased ethane production during exposure to 100% O2, we measured ethane in the expirate of nine exsanguinated, Sprague-Dawley rats (group II) mechanically ventilated with HFA and then with 100% O2. In all nine animals, ethane elimination rates on 100% O2 increased compared with HFA values. In five of the nine rats, HFA ventilation was reinstated after O2 (group III). In all five, ethane elimination fell with HFA ventilation compared with the value on 100%. Six rats with circulation intact were ventilated with HFA and then 100% O2 (group IV). Ethane production rate for group IV animals breathing HFA was not significantly different from the exsanguinated animals in group II while ventilated with HFA. The mean increase in ethane production for the group II animals was not significantly different from the group IV animals. Lung slices from four other rats (group V) were incubated in saline at 37 degrees C with FeCl2 (10 mg) added to enhance free radical formation. Paired lung samples from the same rat were incubated with either HFA or 100% O2. Headspace gas was analyzed chromatographically for ethane at 120 min. Mean ethane in the O2 samples was higher than for HFA. Rat lung tissue is the main source of increased ethane production during 100% O2 exposure.

Abstract: Polycyclic aromatic hydrocarbons (PAH) is one of the largest group of carcinogen in the environment.  An agent that has antigenotoxic and anticarcinogen potency is needed to prevent DNA damage and carcinogenesis. Soursop leaves have a chemopreventive action. This study was aimed to assess the effectiveness of ethanolic extract of soursop leaves in prevention of dysplasia of upper surface of tongue in rats. This was a true laboratory experimental study with the post test-only control group design, using 24 male Sprague Dawley rats divided into six groups. The upper surfaces of tongues of group I-III were induced by DMBA topically three times a week for 16 weeks; group II and III were induced by DMBA added soursop leaves ethanolic extract of 100 and 200 mg/kg body weight daily for 18 weeks; group IV was given soursop leaves ethanolic extract 200 mg/kg body weight; group V was given DMSO 1%; and group VI was untreated. After 18th week, rats were terminated and tongue necropsies in longitudinal anteroposterior direction were conducted. The clinical changes of the upper surfaces of the tongues were observed and the histological changes were observed by using HE staining to confirm signs of dysplasia. The Kruskal Wallis test showed differences between groups, and the Mann-Withney test showed not significantly decrease of mild dysplasia of group II and group III compared to group I (p>0.05). In conclusion, ethanolic extract of soursop leaves was not effective in prevention of dysplasia of the upper surface of rat’s tongue.Keywords: dysplasia; soursop leave extract; dorsal part of tongue; DMBA  Abstrak: Senyawa golongan PAH merupakan salah satu kelompok karsinogen terbesar di lingkungan. Sebagai upaya untuk mencegah kerusakan DNA serta karsinogenesis, diperlukan suatu agen yang berpotensi antigenotoksik sekaligus antikarsinogenik. Daun sirsak terbukti mempunyai aksi kemopreventif. Penelitian ini bertujuan untuk mengkaji efektivitas ekstrak etanol daun sirsak dalam mencegah displasia pada dorsum lingue tikus. Jenis penelitian ialah true experimental laboratory dengan the post test-only control group design yang menggunakan 24 ekor tikus galur Sprague Dawley jantan, dibagi dalam enam kelompok. Dorsum lingue tikus kelompok I - III diinduksi DMBA secara topikal tiga kali seminggu selama 16 minggu; kelompok II dan III selain diinduksi DMBA, juga diberi ekstrak etanol daun sirsak 100 dan 200 mg/kg berat badan setiap hari selama 18 minggu; kelompok IV diberi ekstrak etanol 200 mg/kg BB; kelompok V diberi DMSO 1%; dan kelompok VI tidak diberi perlakuan. Terminasi dan nekropsi lidah tikus longitudinal anteroposterior dilaksanakan setelah minggu ke 18. Perubahan pada dorsum lingue diamati secara klinis dan perubahan histologik dilihat dengan pewarnaan HE untuk mengonfirmasi tanda displasia. Uji Kruskal Wallis menunjukkan perbedaan antar kelompok, sedangkan uji Mann Whitney menunjukkan penurunan tidak bermakna displasia ringan pada kelompok II dan III dibanding kelompok I. Simpulan penelitian ini ialah ekstrak etanol daun sirsak tidak efektif dalam mencegah displasia epitel dorsum lingue tikus.Kata kunci: displasia; ekstrak daun sirsak; dorsum lingue; DMBA

The objective of this study was to analyze the ratio of LDL/HDL level of Sprague Dawley rats induced by high fat ration after given of sea shrimp carapace (Penaeus monodon F.). The animals for this study were twenty adult male rats divided into four groups, i.e. group I as the control was fed with basal ration containing normal fat for 3 months, group II was fed ration containing high fat for 3 months, group III was fed ration containing high fat and given sea shrimp carapace 180 mg per kg body weight per day orally in 2 ml aquadest for 3 months, group IV was fed ration containing high fat for 3 months and after 1 month given sea shrimp carapace 180 mg per kg body weight per day orally in 2 ml aquadest for 2 months. Each group consisted of five animals. After 90 days, the rats were necropsied and the blood were collected to analyzes ratio of LDL/HDL level. LDL level and HDL level were measured using colorimetric enzymatic method and Roche/Hitachi cobas c systems automatically calculate.  The difference between treatments was statistical analysis by Anava, and continued by Lowest Significant Difference Test to locate the difference. The result showed that high fat diet increased ratio of LDL/HDL level and Sea shrimp carapace decreased ratio of LDL/HDL level.   Keywords: Sea shrimp carapace, Sprague Dawley rats, ratio of LDL/HDL level
