AbstractThe study investigated whether dietary methionine (Met) affects egg weight and antioxidant status through regulating gene expression of ovalbumin (OVAL), nuclear factor erythroid 2 like 2 (Nrf2) and haem oxygenase 1 (HO-1) in laying duck breeders. Longyan duck breeders (n540, 19 weeks) were randomly assigned to six treatments with six replicates of fifteen birds each. Breeders were fed diets with six Met levels (2·00, 2·75, 3·50, 4·25, 5·00 and 5·75 g/kg) for 24 weeks. The egg weight (g), egg mass (g/d), feed conversion ratio, hatchability, 1-d duckling weight, albumen weight, albumen proportion andOVALmRNA level improved with dietary Met levels, whereas yolk proportion decreased (P<0·05). The weight of total large yellow follicles increased linearly (P<0·001) and quadratically (P<0·05) with dietary Met concentration, and their weight relative to ovarian weight showed a linear (P<0·05) effect. Dietary Met level had a linear (P<0·05) and quadratic (P<0·001) effect on the gene expression of glutathione peroxidase (GPX1),HO-1andNrf2, and quadratically (P<0·05) increased contents of GPX and total antioxidant capacity (T-AOC) in liver of duck breeders. In addition, maternal dietary Met enhanced gene expression ofGPX1,HO-1andNrf2, increased contents of GPX and T-AOC and reduced carbonylated protein in the brains of hatchlings. Overall, dietary Met concentration affected egg weight and albumen weight in laying duck breeders, which was partly due to gene expression ofOVALin oviduct magnum. A diet containing 4·0 g Met/kg would achieve optimal hepaticGPX1andNrf2expression, maximise the activity of GPX and minimise lipid peroxidation.

Context Organic and herbal additives in feed may lead to more healthy animal products. For this study, we hypothesised that an organic form of selenium and/or a herbal additive (oregano, Origanum vulgare L.) may improve performance of laying hens. Aims The study was designed to determine the effects of selenium source (SS, sodium selenite; or selenium yeast, SY), oregano essential oil (OEO) and a combination of Se and OEO on performance, egg quality and blood parameters of laying hens. Methods In total, 216 Lohmann LSL-Lite laying hens, 66 weeks of age, were assigned to feed on one of six diets: control group, receiving basal diet (BD); BD + 0.3 mg SS/kg; BD + 0.3 mg SY/kg; BD + 250 mg OEO/kg; BD + 250 mg OEO + 0.3 mg SS/kg; BD + 250 mg OEO + 0.3 mg SY/kg. For each treatment, there were six replicates of six hens each for 12 weeks. Key results Feed conversion ratio was higher (P &lt; 0.05) with the OEO diet than in the control. The SY diet decreased egg production and the OEO diet decreased egg mass (P &lt; 0.05), although this effect was reversed with the SY + OEO diet. This would illustrate a synergistic effect of OEO with SY. Highest and lowest yolk colour values were found with the SY and SS diets, respectively (P &lt; 0.05). Serum uric acid was lower with diets containing Se than the control diet (P &lt; 0.05). Glutathione peroxidase activity was highest with SY and SY + OEO diets and lowest with the OEO diet (P &lt; 0.05). Birds fed the SS + OEO diet showed the highest total antioxidant capacity and those fed SY showed the lowest total antioxidant capacity (P &lt; 0.05). Conclusions Based on the results of the current study, it can be concluded that SY diet have increasing effect on egg weight in laying hens. In addition, novel synergistic effects between OEO and SY diets on improving egg production, egg mass and feed conversion ratio have been found. In terms of oxidative status, the detected synergistic effects between OEO and SS diets on total antioxidant capacity and between OEO and SY diets on glutathione peroxidase (GPx) have not been reported before, that appreciate more clarifying investigations in future. Although, dietary supplemental SY individually and also with OEO increased GPx, adding SS to diet together with OEO showed more improving effect on GPx compared to the separate usage. Implications Dietary supplemental SY can improve egg weight in laying hens. In addition, adding SY to diet of laying hens individually and with OEO have beneficial effects on oxidative status of bird in terms of GPs activity, which in turn can ameliorate the unfavourable impressions of probable environmentally oxidative stress on productive performance.

The supplementation of a phytogenic feed additive (PFA) characterised by menthol and anethole was evaluated at three levels on broiler growth performance, nutrient digestibility, biochemical parameters, total antioxidant capacity of plasma, breast and thigh meat as well as on the relative gene expression of immune-related biomarkers. A total of 225 1-day-old male Cobb-500 were assigned into three treatments with five replicates of 15 chicks each. Wheat-soybean meal basal diets were formulated according to a three-phase (i.e. starter, grower and finisher) feeding program. Dietary treatments were: no PFA, PFA at 100 mg/kg diet and PFA at 150 mg/kg diet. Feed and water were available ad libitum. Performance parameters were monitored weekly and all other biological responses were determined at 42 days of broiler age. Increasing PFA level increased (P = 0.044) bodyweight gain at finisher period, decreased quadratically (P = 0.035) overall feed intake, and quadratically improved (P = 0.024) overall feed conversion ratio. Moreover, increasing PFA level increased plasma total antioxidant capacity linearly (P = 0.001) whereas linearly decreased (P = 0.005) triglyceride concentration. Thigh meat cholesterol decreased linearly (P = 0.016) with increasing PFA level. Expression of pro-inflammatory cytokine IL-2 in caecal tonsils increased quadratically (P = 0.046) with increasing PFA level. In conclusion, PFA inclusion at 100 mg/kg diet affected positively performance whereas a stronger improvement mainly in plasma total antioxidant capacity and triglyceride as well as in meat cholesterol was noted for the 150 mg/kg diet level. Inclusion of PFA resulted in increasing pro-inflammatory biomarker IL-2 at local caecal level.

There are close links among hyperglycaemia, oxidative stress and diabetic complications. Glutamine (GLN) is an amino acid with immunomodulatory properties. The present study investigated the effect of dietary GLN on oxidative stress-relative gene expressions and tissue oxidative damage in diabetes. There were one normal control (NC) and two diabetic groups in the present study. Diabetes was induced by an intraperitoneal injection of nicotinamide followed by streptozotocin (STZ). Rats in the NC group were fed a regular chow diet. In the two diabetic groups, one group (diabetes mellitus, DM) was fed a common semi-purified diet while the other group received a diet in which part of the casein was replaced by GLN (DM-GLN). GLN provided 25 % of total amino acid N. The experimental groups were fed the respective diets for 8 weeks, and then the rats were killed for further analysis. The results showed that blood thioredoxin-interacting protein (Txnip) mRNA expression in the diabetic groups was higher than that in the NC group. Compared with the DM group, the DM-GLN group had lower glutamine fructose-6-phosphate transaminase 1, a receptor of advanced glycation end products, and Txnip gene expressions in blood mononuclear cells. The total antioxidant capacity was lower and antioxidant enzyme activities were altered by the diabetic condition. GLN supplementation increased antioxidant capacity and normalised antioxidant enzyme activities. Also, the renal nitrotyrosine level and Txnip mRNA expression were lower when GLN was administered. These results suggest that dietary GLN supplementation decreases oxidative stress-related gene expression, increases the antioxidant potential and may consequently attenuate renal oxidative damage in rats with STZ-induced diabetes.

AbstractThis study aimed to evaluate the effect of in ovo feeding (IOF) of vitamin C at embryonic age 11 (E11) on post-hatch performance, immune status and DNA methylation-related gene expression in broiler chickens. A total of 240 Arbor Acres breeder eggs (63 (sem 0·5) g) were randomly divided into two groups: normal saline and vitamin C (VC) groups. After incubation, newly hatched chicks from each group were randomly divided into six replicates with ten chicks per replicate. Hatchability, average daily feed intake (D21–42 and D1–42), and average daily gain and feed conversion ratio (D1–21) were improved by vitamin C treatment (P < 0·05). IOF of vitamin C increased vitamin C content (D1), total antioxidant capacity (D42), IgA (D1), IgM (D1 and D21), stimulation index for T lymphocyte (D35) and lysozyme activity (D21) in plasma (P < 0·05). On D21, vitamin C increased the splenic expression of IL-4 and DNMT1 and decreased IL-1β, Tet2, Tet3 and Gadd45β expression (P < 0·05). On D42, vitamin C increased the splenic expression of IL-4 and DNMT3A and decreased IFN-γ, Tet3, MBD4 and TDG expression (P < 0·05). In conclusion, the vitamin C via in ovo injection can be absorbed by broiler’s embryo and IOF of vitamin C at E11 improves the post-hatch performance and immune status and, to some extent, the antioxidant capacity of broiler chickens. The expression of enzyme-related DNA methylation and demethylation indicates that the level of DNA methylation may increase in spleen in the VC group and whether the fluctuating expression of pro- and anti-inflammatory cytokines is related to DNA methylation change remained to be further investigated.

Hepatocyte nuclear factor-3 (HNF-3) belongs to a large family of forkhead transcription factors and is made up of three members (HNF-3alpha, -3beta and -3gamma). It has been shown that HNF-3 regulates a number of metabolically important genes. However, the mechanisms underlying this regulation of HNF-3 activity by hormones and nutrition have not yet been well elucidated. In attempting to explore the regulation of gene expression of HNF-3 members by physiological status, we analyzed the effects of insulin, dexamethasone and protein malnutrition on the hepatic mRNA level of each member. Male Wistar rats were fed on a 12% casein diet, 12% gluten diet (deficient in lysine and threonine) or a protein-free diet for 1 week. The protein-free diet and gluten diet caused a 3. 7-fold increase in HNF-3g mRNA in the liver and did not affect the mRNA level of either HNF-3alpha or HNF-3beta. Daily administration of dexamethasone caused the mRNA levels of HNF-3alpha and HNF-3beta to increase (2.3- and 1.4-fold, respectively), but had no effect on the HNF-3gamma mRNA level. In diabetic rats that had been injected with streptozotocin, an elevation of the hepatic mRNA levels of HNF-3beta and HNF-3gamma was observed (1.6-and 1.9-fold, respectively). Insulin replacement in the diabetic rats decreased both mRNA levels in a dose-dependent manner. HNF-3alpha mRNA was not affected by insulin status. These results show that the genes of the three members of the HNF-3 family respond differently to hormonal and nutritional factors suggesting that the activities of HNF-3 members are regulated, at least in part, by the levels of their gene expression.

This study was conducted to know the production performance of laying hen by using different graded level of linseed. At 56 weeks of age, a total of 160 laying hens were raised up to 63 weeks of age. All hens were fed a basal corn soyabean meal diet. The experiment was allowed 4 dietary treatments (T0, no addition of whole linseed in the basal diet; T1,  addition of 5% whole linseed; T2, addition of 10% whole linseed and T3, addition of 15% whole linseed) with 5 replications and per replication contains eight hens. Egg production, feed intake, egg weight of each treatment was recorded. Egg samples were collected and analyzed for egg quality. The results showed that egg production and egg mass (kg) exhibited for all dietary treatment groups were unaffected (P>0.05) by feeding linseed to the laying hens. Feed Conversion Ratio (FCR) was found significant difference within the four dietary group (P<0.05). Egg quality aspects such as yolk weight, egg weight, HU and yolk index were not differed significantly (P>0.05) among treatments.  A trend for lower yolk weight was observed in birds fed 10% linseed, where as the highest yolk weight percentage was recorded in control diet. There was no statistical difference among the different diets for yolk index. The results of the present trial suggested that feeding linseed to the laying hens has no detrimental effects on egg production and egg quality. This study aimed to demonstrate the effects of dietary glutathione (GSH) on growth, intestinal antioxidant capacity, histology, gene expression, and microbiota in juvenile triploid rainbow trout (Oncorhynchus mykiss). Different diets (G0-control, G100, G200, G400, and G800) containing graded levels of GSH (0, 100, 200, 400, and 800mgkg−1) were fed to triplicate groups of 30 fish (initial mean weight 4.12±0.04g) for 56days. G400 had significantly improved weight gain and feed conversion rate. Based on the broken-line regression analysis, the optimum dietary GSH level was 447.06mgkg−1. Catalase and superoxide dismutase activities were significantly higher in G200–G800. G200 had significantly lower malondialdehyde content. The height of the intestinal muscular layer in G400 was significantly higher than that of the control group. Intestinal PepT1 and SLC1A5 gene expression was significantly increased, and the highest was observed in G400. TNF-α, IL-1β, IL-2, and IL-8 expression were significantly decreased than that of G0. Next-generation sequencing of the 16S rDNA showed a significant difference in alpha diversity whereas no differences in beta diversity. On the genus level, LefSe analysis of indicator OTUs showed Ilumatobacter, Peptoniphilus, Limnobacter, Mizugakiibacter, Chelatococcus, Stella, Filimonas, and Streptosporangium were associated with the treatment diet, whereas Arcobacter, Ferrovibrio, Buchnera, Chitinophaga, Stenotrophobacter, Solimonadaceae, Polycyclovorans, Rhodococcus, Ramlibacter, and Azohydromonas were associated with the control diet. In summary, feeding juvenile triploid O. mykiss 200–800mgkg−1 GSH improved growth and intestinal health.

The objective of this study was to examine the effects of diets supplemented with corn silk meal (CSM) and non-starch polysaccharide (NSP) enzyme on growth performance, blood biochemistry, immunological response, and growth-related gene expression in broiler chickens. A total of 270 broiler chickens were divided into six experimental groups: (1) basal diets (BD) as control; (2) BD supplemented with 0.5 g/kg feed NSP enzyme; the other four groups are CSM diets as following; (3) and (4) fed diet contain 40 and 80 kg/ton of CSM; (5) and (6) fed diet contain 40 and 80 kg/ton CSM and supplemented with 0.5 g/kg NSP enzyme. Body weight gain (BWG), feed conversion ratio (FCR), protein retention and fiber digestibility were synergistically improved (p < 0.05) when fed CSM supplemented with NSP enzyme. Moreover, a synergistic decrease (p < 0.05) in the serum glucose and total cholesterol were found. Immune organ weights and Newcastle disease virus titers were increased with CSM diets. Interestingly, the relative mRNAs of the growth hormone receptor (GHR) and insulin growth factor (IGF) were increased (p < 0.05) with the CSM and NSP enzyme: the relative mRNA expressions of cholecystokinin (CCK) and leptin were decreased by feeding CSM diets with the NSP enzyme. It could be concluded that the dietary inclusion of CSM with the NSP enzyme might improve growth performance, modify plasma lipids, and enhance immune response in broilers.

The experiment was conducted to evaluate the production performance and egg quality of laying hens supplemented with phytobiotics and magnetized drinking water. This experiment used 2700 gauss level in Magnetized Drinking Water (MDW). The phytobiotics had two form: the non-encapsulated phytobiotics (PTO) and encapsulated phytobiotics (EPTO). There were untreated water (control) and the treated water (MDW, PTO, PTO+MDW, EPTO, EPTO+MDW). The treatments were presented by 0.6% for each treatment. The result showed highly significant (p<0.01) improvement on feed conversion ratio (FCR) but no significant effect on other production performance variables including feed intake, hen day production (HDP), egg mass, and income over feed cost (IOFC). The results indicated a highly significant improvement (p<0.01) on egg weight and yolk cholesterol. There were no significant effects on shape index (SI), shell weight, shell thickness, Haugh unit (HU), albumen height, yolk weight, yolk index (YI), and yolk color of egg quality variables. It was concluded that the encapsulated form (EPTO and EPTO+MDW) had the best improvement on FCR and yolk cholesterol, on another hand the non-encapsulated form (PTO and PTO+MDW) increased the egg weight of laying hens
