Intestinal mucus protects epithelial and immune cells from the gut resident microorganisms, and provides growth-promoting factors as mucus-derived O-glycans for beneficial bacteria. A lack of intestinal protective mucus results in changes in the commensal microflora composition, mucosal immune system reprogramming, and inflammation. Previous work has shown that fucose, the terminal glycan chain component of the intestinal glycoprotein Mucin2, and fucoidan polysaccharides have an anti-inflammatory effect in some mouse models of colitis. This study evaluates the effect of fucose on reproductive performance in heterozygous mutant Muc2 female mice. We found that even though Muc2+/− females are physiologically indistinguishable from C57Bl/6 mice, they have a significantly reduced reproductive performance upon dietary fucose supplementation. Metagenomic analysis reveals that the otherwise healthy wild-type siblings of Muc2−/− animals have reduced numbers of some of the intestinal commensal bacterial species, compared to C57BL/6 mice. We propose that the changes in beneficial microflora affect the immune status in Muc2+/− mice, which causes implantation impairment. In accordance with this hypothesis, we find that macrophage polarization during pregnancy is impaired in Muc2+/− females upon addition of fucose. Metabolic profiling of peritoneal macrophages from Muc2+/− females reveals their predisposition towards anaerobic glycolysis in favor of oxidative phosphorylation, compared to C57BL/6-derived cells. In vitro experiments on phagocytosis activity and mitochondrial respiration suggest that fucose affects oxidative phosphorylation in a genotype-specific manner, which might interfere with implantation depending on the initial status of macrophages. This hypothesis is further confirmed in BALB/c female mice, where fucose caused pregnancy loss and opposed implantation-associated M2 macrophage polarization. Taken together, these data suggest that intestinal microflora affects host immunity and pregnancy outcome. At the same time, dietary fucose might act as a differential regulator of macrophage polarization during implantation, depending on the immune status of the host.

Recent reviews highlight the importance of the liver in the coordination of nutrient fluxes in support of pregnancy and lactation (e.g. Drackley et al., 2001). Mobilisation of body fat reserves in the late dry period and early lactation leads to an increase in uptake of non-esterified fatty acids (NEFA) by the liver. Their metabolic fate is either oxidation or esterification into triacylglycerides (TAG) that are either exported in very low density lipoproteins (VLDL) or accumulated within liver cells. Recent evidence indicates that TAG accumulation impairs ureagenic and gluconeogenic capacity of the liver, with consequent reductions in feed intake and milk yield, increased incidence of disease and decreased reproductive performance (Overton and Piepenbrink, 1999).LiFTTM (NuTec Ltd.) is a proprietary mixture of B-group vitamins and methyl group donors (rumen protected choline, niacin, vitamin B12, biotin, folic acid and thiamine) designed to reduce the accumulation of TAG in the liver and accelerate VLDL export. The objective of this experiment was to evaluate the effect of LiFT on milk yield and composition and concentrations of metabolites in blood.

Lactation is an energy-demanding physiological process for the maternal organism and life-giving for the offspring. Likewise, exercise is an energy-demanding process. This review addresses the Compatibility of exercise during lactation. Human studies suggest no detrimental effect of exercise during lactation on milk composition and volume, infant growth and development, or maternal health. Studies also demonstrate improved cardiovascular fitness in lactating, exercising women and suggest a quicker return to pre-pregnancy body weight and a more positive sense of well-being, compared to sedentary controls. Findings from rodent studies, although of questionable value for humans, have generally shown no detrimental influence of exercise during pregnancy and lactation on pup growth and development. To date, findings suggest that exercise and lactation are compatible activities. Key words: maternal health, infant growth, breast milk composition, breast milk volume, cardiovascular fitness

The major alteration in the milk composition due to mastitis includes presence of somatic cells (neutrophills, monocytes, macrophages and mammary epithelial cells), bactericidal and bacteriostatic compounds and certain enzymes. Antibiotic residue violation and presence of foodborn pathogens with/without their toxins is another threat of immense importance. These changes in the milk account for the deterioration of the milk quality, risk of health hazards and also its acceptance in the international market. Of all these components, milk somatic cells are the single most widely used indicator of the udder health and mastitis. Somatic cell count (SCC) is used as a system for measurement of milk quality internationally. Strict legislation for monitoring the minimum SCC level is followed to ensure healthy milk in most of the developed countries. This paper introduces the concept of ‘Clean White Revolution’ and reviews the importance of SCC as a measure to ensure hygienic milk production in India. It also emphasizes upon a need to strengthen the existing legislations concerned.
