The effects of ethanol (0.75 g/kg body weight) on the activities of selected enzymes in sera of healthy young adults: 1. Intermediate-term effects.

Abstract We report the intermediate-term effects of three consecutive evenings of moderate ethanol ingestion (0.75 g/kg body weight each evening) on activity values for alkaline phosphatase, gamma-glutamyltransferase, creatine kinase, aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase in sera of nine apparently healthy young adults. We define "intermediate-term" effects as those occurring between 10 h and 100 h after completion of the ethanol consumption schedule. The most pronounced changes in enzyme activity for the group of volunteers were: gamma-glutamyltransferase, +25% at 60 h after ethanol ingestion; alanine aminotransferase, +12% at 60 h after ethanol; and aspartate aminotransferase,--12% at 60 h after ethanol. All three enzymes exhibited similar time courses, i.e., mean peak activity changes were observed at 60 h, and all three mean enzyme activity values returned to near baseline by 100 h. The possible explanations for the observed changes and the clinical significance are discussed.

Abstract We measured individual temporal changes in gamma-glutamyltransferase activity in serum after four consecutive ethanol challenges in each of eight apparently healthy volunteers. Each challenge consisted of ingesting 0.75 g of ethanol per kilogram of body weight on two evenings, followed by five days of abstention during which the activity was assessed at regular intervals. A nine-day abstention period preceded the study, and initial baseline activity was determined. The highest values for the group mean activity were +12%, +11%, +22%, and +18% greater than baseline after the first, second, third, and fourth ethanol challenges, respectively. We noted marked intra-individual and interindividual variation in the enzymic response; peak values ranged from +1% in one subject to +57% in another. The possible causes for these biological variations are discussed.

Introduction/Aim. By analyzing activity of some of the enzymes normally present in the saliva and the level of malondialdehyde in gingivitis, it is possible to estimate the functional condition of parodontium, and the examined parameters can be considered as biochemical markers of its functional condition. The aim of this paper was to examine activity of alanine aminotransferase, aspartate aminotransferase, gamma glutamyl transferase, lactate dehydrogenase and the level of malondialdehyde in the saliva of children affected with gingivitis, as well as the values of the mentioned parameters in relation to the level of the inflammation of gingiva. Methods. The research included 120 children at the age of 12.2 with permanent dentition. L?e and Silness gingival index was used to estimate the condition of gingiva, based on which the children were classified into four groups: the children with healthy gingiva (the control groups), the children with mild, moderate and severe inflammation of gingiva (the study group). Enzymes of the saliva were determined by the use of original tests and measured by the autoanalyser (Bio Systems A25, Spain). A modified method with tiobarbituric acid was used to determine malondialdehyde in nonstimulated mixed saliva. Results. The results of the examined enzyme activity and the level of malondialdehyde in the saliva of the study groups showed statistically considerably higher values for the level of malondialdehyde (p < 0.001), for the activity of aspartate aminotransferase and gamma glutamyl transferase (p < 0.01), as well as for alanine aminotransferase (p < 0.05) in comparison with the control group, whereas the activity of lactate dehydrogenase did not show a statistically significant increase. In relation to the level of the inflammation of gingiva, the results of the examination of the enzyme activity in the study groups showed statistically significantly higher values in the group with severe inflammation in comparison with those with mild, as well as the moderate inflammatory, except for the gamma glutamyl transferase, and in the group with moderate inflammation compared to that with the mild one, except for alanine aminotransferase. The results of the examination of the level of malondialdehyde in the saliva of the study groups did not show a statistically significantly increase in relation to the level of the inflammation of gingiva. Conclusion. There is a higher level of alanine aminotransferase, aspartate aminotransferase, gamma glutamyl transferase and lactate dehydrogenase enzyme activity together with the higher level of malondialdehyde in the saliva of children with gingivitis in comparison with the activity of the same enzymes and the level of malondialdehyde in the saliva of children without gingivitis. The activity of the examined enzymes in the saliva of children with gingivitis increases in relation to the intensity of the pathological process, whereas the level of malondialdehyde shows no significant difference in relation to the level of the inflammation of gingiva.

Metabolic changes in the body of dogs with atopic dermatitis and the causes of its occurrence still remain poorly understood. This study confirms the need for biochemical blood tests for the differential diagnosis of skin diseases in animals. The aim of the research. To identify metabolic changes in dogs with atopic dermatitis. Methods. Serum samples from five ill animals and five intact animals were tested. The diagnosis of the nosological form of skin lesions was established based on the results of anamnesis and clinical manifestations of the disease. Additionally, biochemical studies of the serum of sick animals were performed. Blood of dogs (n=5) was taken from the anterior subcutaneous vein of the forearm or the lateral subcutaneous vein of the tibia into blood tubes. Results. In the study of mineral metabolism, it was found that the concentration of inorganic phosphorus in the serum of sick animals is reduced by only 0.01 mmol/l, but the concentration of total calcium by 10.0 %. The concentration of cholesterol increases 1.3 times and exceeds the reference values by 0.16 mmol/l. In the blood of sick animals, the activity of alanine aminotransferase is significantly reduced by 21.0 % and aspartate aminotransferase – by 20.0 % compared to intact animals. The enzyme gamma-glutamyltransferase probably increases by 1.51 times (P<0.05) in sick dogs, but does not exceed the reference values. Conclusions. It was found that in atopic dermatitis in the serum of dogs decreases the activity of alanine aminotransferase by 21.0 % and aspartate aminotransferase – by 20.0 %, while the activity of gamma-glutamyltransferase increases 1.51 times. The results show the toxic effect of an allergic agent on animals, especially on the liver, which confirms the previous diagnosis – atopic dermatitis.

Background: Liver plays a major role in detoxification and excretion of many endogenous and exogenous compounds. Any injury may lead to severe liver damage and impairment of liver function. Harbal plants such as Terminalia chebula (Haritaki) may have free radical scavenging activity thereby can be used for the prevention and treatment of liver damage.Objective: To observe the effect of Terminalia chebula on paracetamol induced changes of serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in Wister albino rats.Methods: This experimental study was carried out in the Department of Physiology, Dhaka Medical College, Dhaka from January to December’ 2013. Total 44 rats with age 90 to 120 days, weighing between 150 to 200 gm were selected. After acclimatization for 14 days, they were divided into base line control (BC, n=11), paracetamol treated control (PC, n=11),Terminalia chebula pretreated and paracetamol treated (TCP-PCT n=11) and paracetamol pretreated and Terminalia chebula treated group (PCP-TCT, n=11). All groups received basal diet for 21 consecutive days. In addition to basal diet, rats of BC received propylene glycol (2ml/kg body weight, orally) and PC received single dose of paracetamol suspension (750mg/kg body weight, orally) on 21st day. Rats of TCP-PCT received Terminalia chebula extract (200 mg/kg body weight, orally) for 21 consecutive days and paracetamol suspension (750mg/kg body weight, orally) on 21st day. Again, rats of PCP-TCT received paracetamol suspension (750mg/kg body weight, orally) on the 1st day and Terminalia chebula extract (200 mg/kg body weight orally) for 21 consecutive days. All rats were sacrificed on 22nd day and then blood samples were collected. For assessment of liver function serum AST and ALT levels were estimated by using standard laboratory kits. The statistical analysis was done by one way ANOVA and post hoc Bonferroni test as applicable.Results: The mean serum AST and ALT levels were significantly (p<0.001) higher in PC in comparison to those of BC. Serum AST and ALT levels of all experimental groups were significantly (P<0.001) lower than PC group. Conclusion: From the results of this study, it may be concluded that Terminalia chebula may have some hepatoprotective effects in paracetamol induced liver damage in rats.Bangladesh Soc Physiol. 2015, June; 10(1): 1-5

Abstract We examined the measurement of serum aspartate and alanine aminotransferase activities with a short-interval enzyme-activity analyzer. Double-beam spectrophotometry was used to elucidate the source of errors in such measurements. For both enzymes the principal error sources are: (a) the presence of endogenous substrate for lactate dehydrogenase, and (b) reaction of 2-oxoglutarate with serum glutamate dehydrogenase and ammonium ions added to the reaction mixture in admixture with the secondary enzymes of the coupled reactions. In the case of serum aspartate aminotransferase, a less important source of error is the reaction of serum alanine aminotransferase with endogenous substrate. Use of 2-oxoglutarate as a reaction initiator in conventional methods causes errors. Suitable blank reagent mixtures are described that permit accurate, rapid measurement of these activities by double-beam spectrophotometry in a short-interval enzyme-activity analyzer.

In this study the effect of diazepam on some blood chemistry values was studied in male rats. Intraperitoneal administration of diazepam at a dose of 0.6mg/kg body weight for 60 days didn't induce significant changes in serum glucose, urea, cholesterol, aspartate aminotransferase (AST), alanine aminotransferase (ALT), total bilirubin, creatinine and alkaline phosphatase levels. Findings of this study give further sound for the safety of benzodiazepines.

Objectives: Our goal was to contribute to the production of reference values ​​of plasma or serum biochemical markers by determining the reference values ​​of gamma-glutamyltransferase (GGT), aspartate aminotransferase (ASAT) and alanine aminotransferase (ALAT) in young Congolese presumed to be healthy. Methods: 250 young Congolese presumed to be healthy (125 boys and 125 girls) aged 15 to 25 participated in the study. They were selected according to anamnestic and clinico-biological criteria. Samples were taken on a tube containing EDTA and the resulting plasma was stored at -20 ° C. The KENZA MAX spectrophotometer was used to analyze GGT, ASAT and ALAT. The median and the 2.5-97.5 percentiles were used to set the reference limits for each enzyme. The benchmarks determined were compared with those reported by other Africans, Europeans, Indians and Americans. Results: The established reference values ​​were: GGT 12.15-61.85 IU/L for boys and 7-51.95 IU / l for girls (p˂0.0001); ASAT 21.60-94.85 IU/L for boys and 17-84.85 IU/L for girls (p = 0.0003); ALAT 8.30-74.40 IU/L for boys and 8-53.85 IU/L for girls (p˂0.0001). In addition, the comparison between our values ​​and those of other populations showed significant differences. Conclusion: Our results underline the importance of establishing reference values ​​for plasma enzymes specific to the Congolese population. The use of the values ​​established in the ’other populations could induce errors of judgment by excess or by default. The aim of the study was to investigate the effect of toxocariasis invasion on protein-synthesizing function and functional state of the liver of dogs. For experimental studies, 12 dogs aged from two to four months were used and two groups of six animals were formed in each: control and experimental. Puppies of the experimental group were experimentally infected with the pathogen toxocariasis at a dose of 5,000 invasive eggs of T. canis per kg of body weight. The control puppies were clinically healthy. Protein-synthesizing function of the liver of dogs in experimental toxocariasis was studied by the level of total protein and its fractions. It was found that on the 25th day of the experiment the level of total protein in the blood of the experimental group of dogs was 56.9 ± 1.47 g/l, while in the control group – 63.8 ± 2.92 g/l. The lowest level of the studied indicator was on the 30th day of the experiment in the blood of the experimental group of dogs, where it decreased by 9.6 %. The results of a study of protein fractions in the blood of dogs infested with toxocara showed that the percentage of albumin is likely to decrease. However, in the blood of infected dogs in this period of research there was a probable increase in globulin levels. The study of the effect of toxocariasis invasion on the activity of enzymes in blood serum is a test for the morphological and functional state of tissues and organs. Based on the studies, it was found that with the development of toxocariasis invasion in dogs there is an increase in alanine aminotransferase in their serum. On the 25th and 30th day of the experiment, the activity of the enzyme in the serum of infected dogs was the highest, where compared with the control group of dogs, the activity of ALT increased by 51.2 and 59.1 %, respectively. Similar changes in activity were observed in the determination of aspartate aminotransferase, where, respectively, on the 15th, 20th and 25th day of the experiment, the activity of AST in the serum of infected dogs increased by 19.3, 27.5 and 39.1 %.

An outbreak of natural poisoning by Cestrum laevigatum was reported among cattle in the Agreste region of Paraíba, which affected six out of 20 animals. Four animals were found dead and two presented clinical signs, including, dyspnea, drooling, jugular vein engorgement, muscle tremors and ataxia, which subsequently progressed to recumbence and death. An experimental poisoning was performed in two bovines who were administered single doses of 35 g kg-1 and 50 g kg-1 body weight (BW), respectively, of fresh leaves and fruits. The animal that received 35 g kg-1 BW had mild clinical signs, consisting of apathy, salivation and reduced ruminal movements with recovery 30 hours after the onset of initial signs. The animal that received 50 g kg-1 BW showed apathy, dry stool, drooling, hyperexcitability, head pressing, opisthotonos, nystagmus, miosis, jugular and episcleral vessel engorgement, ruminal atony, muscle tremors, ataxia, falling, seizures, and sternal recumbence, followed by lateral recumbence, with death occurring 21 hours and 37 minutes after ingestion. The enzyme activities of aspartate aminotransferase and gamma-glutamyltransferase in the serum increased significantly 18 hours after the administration of the plant. The primary gross lesions in the natural and experimental cases were enlarged livers, with rounded edges and accentuation of the lobular pattern on the capsular and cut surfaces. Histopathology revealed diffuse centrilobular coagulative necrosis with hemorrhages and congestion, and the presence of degenerated hepatocytes in the midzonal regions. Based on the epidemiological and clinicopathological data, we concluded that C. laevigatum was responsible for an acute hepatotoxic disease among cattle in the Agreste region of Paraíba.
