Shi-Zhen-An-Shen decoction (SZASD), a Chinese herbal medicine that is a liquor extracted from plants by boiling, has been reported to be effective in treating schizophrenia. However, the mechanism is unclear. Abnormal demyelination has been implicated in schizophrenia. The aim of this study was to investigate the effect of SZASD on myelin in demyelinated mice exhibiting schizophrenia-like behaviors. Sixty male C57BL/6 mice were randomly divided into six groups ( n = 10 per group): (1) control group, (2) cuprizone (CPZ, a copper chelator that induced demyelination, 0.2% w / w )+saline, (3) CPZ+low-dose SZASD (8.65 g·kg-1·d-1), (4) CPZ+medium-dose SZASD (17.29 g·kg-1·d-1), (5) CPZ+high-dose SZASD (25.94 g·kg-1·d-1), and (6) CPZ+quetiapine (QTP, an atypical antipsychotic that served as a positive treatment control, 10 mg·kg-1·d-1). Mice in groups 2-6 were treated with CPZ added to rodent chow for six weeks to induce demyelination. During the last two weeks, these mice were given an oral gavage of sterile saline, SZASD, or quetiapine. Behavioral tests and brain analyses were conducted after the last treatment. The brain expression of myelin basic protein (MBP) and neuregulin-1 (NRG-1) was assessed using immunohistochemistry and Western blots. CPZ induced significant schizophrenia-like behaviors in the mice, including reduced nest-building activity and sensory gating deficits. Hyperlocomotor activity was accompanied by significant reductions in MBP expression in the corpus callosum, hippocampus, and cerebral cortex. However, both QTP and SZASD significantly reversed the schizophrenia-like behaviors and demyelination in CPZ-fed mice. The QTP and medium-dose SZASD resulted in better therapeutic effects compared to the low and high SZASD doses. Reduced NRG-1 expression was observed in CPZ-fed mice compared with controls, but neither QTP nor SZASD showed significant influence on NRG-1 expression in the hippocampus. Together, SZASD showed a therapeutic effect on demyelinated mice, and the improvement of demyelination might not be through the NRG-1 pathway.

Abstract Background Our previous study indicated a therapeutic effect of Shi-Zhen-An-Shen-Tang (SZAST), a Chinese herb formula, on schizophrenia, but the related mechanism is unknown(citation). A large body of evidence suggests the important role of white matter of the brain in the pathophysiology of schizophrenia. This study was designed to evaluate the effect of SZAST on schizophrenia with demyelinated mice. Methods Male C57BL/6 mice were given mixed cuprizone (CPZ, a copper chelator, 0.2 %, w/w) rodent chow for six successive weeks to induce demyelination. During the last two weeks, mice were given an oral gavage of saline, or SZAST of three different doses (a low dose of 5.5g·kg-1·d-1, a medium dose of 8.24g·kg-1·d-1, or a high dose of 10.98 g·kg-1·d-1), or quetiapine, respectively. Behavioral tests were conducted after the last treatment. Meanwhile, the expression of myelin basic protein (MBP) and neuregulin-1(NRG1) in the brain was tested by immunohistochemistry staining or Western Blot. Results Mice exposed to CPZ for six weeks showed obvious schizophrenia-like behaviors, including lower nest-building activity, sensory gating activity, and higher locomotor activity. CPZ-fed mice also displayed a lower myelin density in the corpus callosum, hippocampus, and cerebral cortex and a reduction of MBP and NRG1 protein in the hippocampus compared with controls. Both quetiapine and SZAST significantly alleviated the abnormal schizophrenia-like behaviors and the impairment of myelin sheath in CPZ-fed mice, however, SZAST with medium dose showed better neuroprotective effect than the low dose or the high dose of SZAST. Furthermore, the expression of NRG1protein in the hippocampus was slightly, but not significantly increased in all SZAST-treated and quetiapine-treated groups. Discussion These results indicate that the neuroprotective effect of SZAST in demyelinated mice might partially relate to remyelination in the hippocampus in CPZ-fed mice.

Objective. To investigate the effects of topical and subconjunctival tigecycline on the prevention of corneal neovascularization.Materials and Methods. Following chemical burn, thirty-two rats were treated daily with topical instillation of 1 mg/mL tigecycline (group 1) or subconjunctival instillation of 1 mg/mL tigecycline (group 3) for 7 days. Control rats received topical (group 2) or subconjunctival (group 4) 0.9% saline. Digital photographs of the cornea were taken on the eighth day after treatment and analyzed to determine the percentage area of the cornea covered by neovascularization. Corneal sections were analyzed histopathologically.Results. The median percentages of corneal neovascularization in groups 1 and 3 were 48% (95% confidence interval (CI), 44.2–55.8%) and 33.5% (95% CI, 26.6–39.2%), respectively. The median percentages of corneal neovascularization of groups 1 and 3 were significantly lower than that of the control group (P=0.03andP<0.001, resp.). Histologic examination of samples from groups 1 and 3 showed lower vascularity than that of control groups.Conclusion. Topical and subconjunctival administration of tigecycline seems to be showing promising therapeutic effects on the prevention of corneal neovascularization. Furthermore, subconjunctival administration of tigecycline is more potent than topical administration in the inhibition of corneal neovascularization.

Background.Blood pressure variability (BPV) independent of average blood pressure is related to cardiovascular damage. Meanwhile, BPV is also associated with measures of endothelial injury. Decoction, a traditional used form of Traditional Chinese Medicine (TCM), is inconvenient to prepare, carry, and store. Dispensing granules is now developing as an alternative to decoction, but the evidence supporting its clinical efficacy the same as decoction remains unclear.Objective.To examine the therapeutic effects on mean blood pressure (MBP), blood pressure variability, and endothelial function by giving Bushen Qinggan Formula, a compound Chinese Herbal Medicine and also to evaluate the difference in efficacy between decoction and granule.Methods.A total of 150 patients with hypertension were enrolled and randomly assigned to receive the placebo, Bushen Qinggan decoction, or Bushen Qinggan granule in addition to the standard medications (amlodipine-5 mg/d) for the treatment of essential hypertension (EH). The outcome was the reduction in the MBP and BPV and also included changes in the endothelial markers including endothelin-1 (ET-1) and nitric oxide (NO) after 8 weeks of treatment.Results.Compared with the control group, the Bushen Qinggan decoction and granule groups had significant improvement (P<0.01) in BPV and endothelial founction. The level of BPV and endothelial function between decoction and granule group had no significant difference (P>0.05).Conclusion.Based on the standard treatment, Bushen Qinggan Formula further improved BPV and endothelial function. The efficacy of Bushen Qinggan decoction and granule is similar in improving BPV and endothelial function. However, no significant antihypertensive effects could be demonstrated.

5-year results of combined treatment of 160 patients with respectable rectal cancer (TNM: IIab - IIIa-b) are analyzed. In 40 patients (study group)) neoadjuvant (72 h before surgery), endovascular oil Chemoembolization of the Rectal Arteries (RACHEL procedure) with a Lipiodol and 5-Fluorouraci was used. The results were compared with surgical treatment (control group 1, n=40) and preoperative radiotherapy methods (control group 2: 5 х 5 Gr, up to a Total Focal Dose of 25 Grandcontrolgroup3: High Dose radiotherapy with a Single Focal Dose of 13 Gr with program Endovascular Radio modification Metronidazole, for 40 patients). The preoperative RACHEL procedure in treatment of patients with resectable rectal cancer was effective with low local recurrence (2,6%) rate, at 5-year overall (89,7%) and disease -free survival (84,6%) and can compete with known preoperative radiotherapy in combined treatment of rectal cancer.

This study explores the antiskin cancer effect of formulated agar (FA) from Laminaria digitata on dimethyl benzanthracene- (DMBA-) induced skin cancer mice. The agar was extracted and formulated (emulgel), and FA was biochemically characterized. The in vitro cytotoxicity of FA was tested using NTT 3T3 mice fibroblast cells. The mice were divided into 5 groups: group 1 served as control mice, group 2 mice were considered as DMBA-induced cancer control, group 3 mice were FA pretreated (low dose) + DMBA-induced mice, group 4 mice were FA pretreated (high dose) + DMBA-induced mice, and group 5 were positive control + DMBA-induced mice. The behaviour and biochemical markers of cancer were significantly decreased in group 2 (DMBA-induced) mice, which were brought to near normalcy by FA pretreated mice (groups 3 and 4). The levels of p53 and keratin were significantly elevated in group 2 mice and these levels were decreased in 3 and 4 mice as well. The histopathological examination of DMBA-induced mice was shown degenerated cervical patches in the skin, cirrhosis in liver, oedema in the renal tissue, and swollen and damage in cardiac tissue, which were reduced for the mice applied with FA. This confirms that FA pretreatment offered potential antiskin cancer property.

<p>Several kind of heavy metals that present in the environment are considered as toxic substance to human and animal. Lead (Pb) is one of  heavy metals that increase in use for the last decade. Lead toxicity to human had wide influence in medical aspect, from nerve problem, bone metabolics disturbance until liver and renal failure. This experiment tried to found out the effect of cronic oral lead consupmtion to lead plasma rate in mice. Design of this experiment is true research experiment to test wheater there is an effect of oral lead consumption to acumulation of   lead in blood plasma of mice. The dose devided to two kinds, medium dose (50 ppm) and high dose (100 ppm). The results show increase of lead accumulation in blood plasma of the mice compared to control group.</p> <br />

Objectives: This study was undertaken to reveal therapeutic effects and the preliminary mechanism of Chinese medicine formula Qianlie Tongli decoction (QTD) in chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS). Methods: A total of 50 male C57BL/6 mice were randomly divided into five groups. All groups except the control group were injected subcutaneously T2 peptide emulsion, which induced the CP/CPPS model. After the induction of CP/CPPS, the model group was given 0.9% NaCl by oral gavage while low-dose, medium-dose, and high-dose groups were treated with Chinese medicine formula. Micturition habits and pain behavior of mice were analyzed for each group. Hematoxylin and eosin staining were used to investigate prostate inflammation. The serum level of tumor necrosis factor-&alpha; (TNF-&alpha;) was measured by enzyme-linked immunosorbent assay (ELISA) kit. Key findings: Chinese medicine formula significantly reduced the number of urine spots and improved pain response frequency in the medium-dose and high-dose group. The high-dose group showed reduced considerably inflammatory lesion and inflammatory cell infiltration than the low-dose and medium-dose groups. Serum levels of TNF-&alpha; in the high-dose group were significantly reduced compared with the model group. Conclusions: The results demonstrated the therapeutic effects of Qianlie Tongli decoction in CP/CPPS mice by analyzing clinically relevant symptoms (urinary tract system, pelvic pain, and prostate inflammation), and preliminary explored the inflammatory-related treatment mechanisms by measuring TNF-&alpha;.

Background : Heart disease is the leading cause of death for both men and women, but heart disease is preventable and controllable. Ganoderma lucidum is widely used as traditional medicine for centuries particularly in China, Japan, and Korea. Previous study showed antioxidative activity of polysaccharide peptide (PsP) from Genoderma lucidum.Objective : This study was aimed to evaluate anti-inflammatory and antioxidant effect of polysaccharide peptide (PsP) from Ganoderma lucidum in atherosclerotic rats.Methods : The atherosclerotic rats were randomly divided into four groups (5 rats each group) : atherosclerotic model with high-fat diet, low dose PsP treated group (50 mg/kgBW), medium dose PsP treated group (150 mg/kgBW), high dose PsP treated group (300 mg/kgBW), with normal mice used as a control group. Parameters measured were the level of MDA, SOD, IL - 6 , IL - 10, hsCRP, TNF - ?, lipid profile and foam cell.Results : After PsP therapy for 5 weeks, the levels of MDA (p=0.01), hsCRP (p=0.018) in rats model of atherosclerosis decrease significantly. PsSP can reduce levels of IL - 6 (p=0.933) and increase levels of SOD (p=0.28) descriptively at PsP doses 150 mg/kgBW. While the levels of TNF-? (p=0.894) and IL-10 (p=0.98) was not affected by administration of PsP. PsP improve the lipid profile by increasing HDL (p=0.002) and lowering total cholesterol (p=0.04). The formation of foam cells (p=0.024) as a marker of atherogenesis significantly decreased by administration of PsP .Conclusion : PSP can be useful to reduce inflammatory processes and oxidative stress to prevent the process of atherogenesis.

Trichinellosis caused by the gastrointestinal nematode Trichinella spiralis occurs in humans, domestic animals and wild animals. This heavy ubiquitous parasitic disease. The muscle phase of the parasite of difficult to controlling. Homeopathic drugs such as Cina have anthelmintic and immunostimulation properties. The purpose of this study was assess the protective efficiency of homeopathic drug such as Cina C6cH and Roncoleukin ® (recombinant interleukin-2 (rIL-2) immunostimulation) against experimental trichinosis, that can affect the muscle phase of the parasite T. spiralis in mice. An assay was carried out on 30 white outbred mice. These were divided into tree groups of ten mice each. The group 1 were treated with the Roncoleukin ® (100 IU/per mouse in 0,2 ml sterile saline intramuscularly, twice with 48 hours intervals); the group 2 - desoldering dissolved in water Cina C6cH (under the scheme). The group 3 received 0.2 ml of sterile 0.9% NaCl. After a 21 day regimen, the groups were inoculated with a dose of 80 ± 5 units of Trihinella spiralis larva per mouse. After 90 days of incubation, the mice were euthanized and dissected for evaluation. The maximum protection was obtained in mice immunized with Roncoleukin ® (80,61%; 869,5±16,6 larva/per animal). The protective effect in the group immunized by Cina C6cH was of 36,67 ( (2840,5±183,3 larva/per animal)%. In control group invasion on muscle phase T.spiralis was 4485±430,6 larva/per animal/ In conclusion, the use of homeopathic drugs also reduces in the number of infective Trichinella spiralis larvae in mice and has a synergistic effect on the treatment.

Purpose: To investigate the hepatoprotective role of ganoderic acid A (GAA) on liver cancer induced by diethylnitrosamine (DEN) via Nrf-2/HO-1/NF-κB signal pathway in mice. Methods: Sixty male C57BL/6J mice were randomly divided into 4 groups: (1) control group, (2) DEN (25 mg/kg) group, (3) GAA (20 mg/kg) + DEN group, (4) GAA (40 mg/kg) + DEN group. The protective effect of GAA on liver was evaluated by determining malondialdehyde (MDA), superoxide dismutase (SOD), inflammatory cytokines including interleukin-6 (IL-6), interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), and the expression of heme oxygenase-1 (HO-1), nuclear factor erythroid- 2-related factor-2 (Nrf-2), IκBα, p-IκBα, p65, p-p65, glyceraldehyde-3-phosphate dehydrogenase (GAPDH) in serum. Results: The results demonstrate that GAA treatment significantly suppressed the generation of MDA, proinflammatory cytokines, and restored the activity of SOD in the serum of DEN-induced liver cancer in mice. Western blots analysis revealed that GAA significantly restored Nrf-2/HO-1/NF-κB signal pathwayrelated protein levels in DEN-induced mice liver cancer model. Conclusion: This research reveals the anticancer activity of GAA in liver tissue, and suggests that GAA counters DEN-induced liver  cancer through Nrf-2/HO-1/NF-κB signal pathway.
