The Development of Antibiotics Based on Nanostructured Manganese Oxide; Understanding Mechanism from Fundamental Aspects to Application

The emergence of bacterial resistance to currently available antibiotics emphasized the urgent need for new antibacterial agents. Nanotechnology-based approaches are substantially contributing to the development of effective and better-formulated antibiotics. Here, we report the synthesis of stable manganese oxide nanostructures (MnO NS) by a facile, one-step, microwave-assisted method. Asprepared MnO NS were thoroughly characterized by atomic force microscopy (AFM), field emission scanning electron microscopy (FESEM), dynamic light scattering (DLS), UV-Visible spectroscopy and X-ray powder diffraction (XRD). UV-Visible spectra give a sharp absorption peak at a maximum wavelength of 430 nm showed surface plasmon resonance (SPR). X-ray diffraction (XRD) profile demonstrated pure phase and crystalline nature of nanostructures. Morphological investigations by a scanning electron microscope showed good dispersity with spherical particles possessing a size range between 10–100 nm. Atomic force microscope data exhibited that the average size of MnO NS can be controlled between 25 nm to 150 nm by a three-fold increment in the amount of stabilizer (o-phenylenediamine). Antimicrobial activity of MnO NS on both gram-positive (Bacillus subtilis) and gram-negative (Escherichia coli) bacterial strains showed that prepared nanostructures were effective against microorganisms. Further, this antibacterial activity was found to be dependent on nanoparticles (NPs) size and bacterial species. These were more effective against Bacillus subtilis (B. subtilis) as compared to Escherichia coli (E. coli). Considering the results together, this study paves the way for the formulation of similar nanostructures as effective antibiotics to kill other pathogens by a more biocompatible platform. This is the first report to synthesize the MnO NS by green approach and its antibacterial application.

Two bacterial species, Gram-negative Escherichia coli and Gram-positive Bacillus subtilis, were exposed to different auxins to examine possible effects of these substances on bacterial stress tolerance. Bacterial resistance to UV irradiation, heat shock, and streptomycin was assessed with and without previous exposure to the following auxins: indole-3-acetic acid (IAA), indole-3-butyric acid (IBA), and 1-naphthalene acetic acid (NAA). Escherichia coli and B. subtilis cultures pretreated with any of the 3 auxins survived UV irradiation better than the untreated cultures. Also, B. subtilis cultures pretreated with IBA or NAA survived prolonged heat exposure better than the untreated cultures, while IAA pretreatment had no effect on heat shock survival. In contrast, auxin pretreatment rendered E. coli more sensitive to heat shock. Escherichia coli cultures pretreated with auxins were also more sensitive to streptomycin, while auxin pretreatment had no effect on sensitivity of B. subtilis to streptomycin. These results show that auxins may either enhance or reduce bacterial tolerance to different stressors, depending on the bacterial species and the type and level of the stress. Auxins usually had similar effects on the same bacterial species in cases when the same type and level of stress were applied.

Anatase TiO2 thin films are deposited on K9 glass samples at different substrate temperatures by radio frequency magnetron sputtering. N ion implantation is performed in the as-deposited TiO2 thin films at ion fluences of 5 × 1016, 1 × 1017, and 5 × 1017 ions/cm2. X-ray diffraction, atomic force microscope, X-ray photoelectron spectroscopy (XPS), and UV–visible spectrophotometer are used to characterize the films. With increasing N ion fluences, the absorption edges of anatase TiO2 films shift to longer wavelengths and the absorbance increases in the visible light region. XPS results show that the red shift of TiO2 films is due to the formation of N–Ti–O compounds. As a result, photoactivity is enhanced with increasing N ion fluence.

Self-patterning presents an appealing alternative to lithography for the production of arrays of nanoscale ferroelectric capacitors for use in high density non-volatile memory devices. Recently a self-patterning method, based on the use of the instability of ultrathin films during hightemperature treatments, was used to fabricate nanosized ferroelectrics. This paper reports the use of the method for the preparation of PZT nanoislands on different single crystalline substrates - SrTiO3, MgO and LaAlO3. Moreover, a multi-step deposition procedure in order to control lateral the dimension of the crystals was introduced. The nanostructures obtained were studied by atomic force microscopy, scanning electron microscopy and X-ray diffraction.

The toxicity of the first ten MEIC chemicals to Escherichia coli and Bacillus subtilis was examined. Nine of the chemicals were toxic to the bacteria, with the minimal inhibitory concentration (MIC) ranging from 10-3 to 4.4M. The sensitivities of both organisms were similar, but the effect on E. coli was often bactericidal, while it was bacteriostatic for B. subtilis. Digoxin was not detectably toxic to either bacterial species. Amitriptyline and FeSO4 were relatively less toxic to the bacteria than to human cells. For seven chemicals, a highly significant linear regression was established between log MIC in bacteria and log of blood concentration, giving lethal and moderate/mild toxicity in humans, as well as with toxicity to human lymphocytes.

Two new bis-Schiff bases containing a piperazine ring, N,N‘-bis(4-chlorobenzylidene)- and N,N‘-bis(4-cyanobenzylidene)-1,4-bis(3-aminopropyl)piperazine, were prepared by the reaction of N,N‘-bis(3-aminopropyl)piperazine with 4-chloro- and 4-cyanobenzaldehyde, respectively. The dichloro compound was fully identified by X-ray crystallography and it exhibited good antibacterial activity against Escherichia coli, Staphylococcus aureus and Bacillus subtilis.

The block single-crystals of taurine were obtained, and its structure was determined by single-crystal X-ray diffraction. The single-crystal X-ray analysis of taurine reveals that the crystal belongs monoclinic, space group P2(1)/c with a = 0.52824(10) nm, b = 1.1647(8) nm, c = 0.79236(13) nm, ß = 94.0850(10). The magnesium complex with taurine has been synthesized in ethanol. The antibacterial assay of the Mg (II) complex was measureed using a modified version of the 2-fold serial dilution method. The results show that the complex shows considerable antibacterial activity against escherichia coli, bacillus subtilis and staphylococcus white.

Telah dilakukan penelitian tentang kemampuan aktivitas anti bakteri untuk bakteri Staphylococcus aureus (gram positif) dan Escherichia coli (gram negatif) dengan menggunakan nanokomposit nata de coco/TiO2, nata de coco/Ag, dan nata de coco/TiO2/Ag dengan variasi konsentrasi Ag 0,5 M; 0,6 M; 0,7 M; 0,8 M dan 0,9 M  menggunakan metode reduksi kimia. Nanopartikel tersebut dikarakterisasi menggunakan X-Ray Diffractometry (XRD), scanning electron microscopy-energy dispersive X-ray spectroscopy  (SEM-EDS) dan anti bakteri. Untuk uji aktivitas antibakteri menggunakan metode kertas cakram dan dilakukan sebanyak dua kali ulangan untuk tiap-tiap sampel dan bakteri yang diuji. Hasil penelitian menunjukan, aktivitas anti bakteri nanokomposit yang paling baik dalam menghambat pertumbuhan bakteri adalah nanokomposit Nata de coco/TiO2/Ag mampu menghambat pertumbuhan bakteri Escherichiacoli dan Staphylococcusaureus, namun nanokomposit tersebut lebih efektif menghambat pertumbuhan bakteri Escherichiacoli.ABSRACT Research on the ability of anti-bacterial activity for Staphylococcus aureus (gram positive) and Escherichia coli (gram negative) bacteria using nata de coco / nanocomposites TiO2, nata de coco / Ag, and nata de coco / TiO2 / Ag with variations of Ag 0,5 M; 0.6 M; 0.7 M; 0.8 M and 0.9 M using the chemical reduction method. Nanoparticles were characterized using X-Ray Diffractometry (XRD), scanning electron microscopy-energy dispersive X-ray spctroscopy  (SEM-EDS) and anti-bacterial actvity. Test the antibacterial activity using the paper disc method and repeated two times for each sample and bacteria tested. The results showed that the good anti-bacterial activity of nanocomposites in inhibiting bacterial growth was nanocomposite nata de coco /TiO2/Ag  able to inhibit the growth of Escherichia coli and S. aureus, but the nanocomposite is more effective in inhibiting the growth of Escherichia  coli bacteria.

Insertion of metal-oxide nanoparticles to polymers stipulate the modification of physical properties of polymers over and above the accomplishment of new features in the polymer matrix. In the current study, an attempt was made to disperse the CuO nanoparticles in the polyvinyl alcohol and hydroxypropyl methylcellulose (HPMC) blend to investigate the structural, mechanical and optical properties of the nanocomposite. Blend was prepared in different ratios using PVA and HPMC, viz. 25:50, 50:50 and 75:25 wt%. The CuO nanoparticles were added to the 75:25 PVA:HPMC blend in different percentage like 0.5,1 and 1.5%. The polymer with and without CuO incorporation were subjected to X-ray diffraction, Fourier transform infrared spectroscopy, scanning electron microscopy, UV-visible spectral analyses and mechanical strength, etc. The results revealed that the incorporation of the CuO nanoparticles enhanced the structural and mechanical properties of the polymer by forming successful nanocomposite.

Aiming to improve the nanotribological response of Si-based materials we implanted silicon wafers with different fluences of iron ions (up to 2x1017 cm-2). Implantation was followed by annealing treatments at temperatures from 550°C to 1000°C. The implanted surfaces were analyzed by scanning electron microscopy (SEM), X-ray diffraction (XRD), atomic force microscopy (AFM) and wettability tests. Then, samples were submitted to AFM-based nanowear tests. We observe an increase of both hidrophobicity and and wear resistance of the implanted silicon, indicating that ion implantation of Si can be a route to be deeper explored in what concerns tribomechanical improvement of Si.
