Abstract A major objective of the North American Monsoon Experiment (NAME) was to quantify microphysical processes within convection occurring near the steep topography of northwestern Mexico. A previous study compared examples of isolated convection using polarimetric radar data and noted a dependence on mixed-phase processes via drop freezing and subsequent riming growth along the coastal plain and western slopes, with an even greater role of melting ice in rainfall production over the highest terrain. Despite the higher frequency of these isolated cells compared to organized convective systems, the latter were responsible for 75% of rainfall. Therefore, this study seeks to evaluate the role of mesoscale organization on microphysical processes and describes the evolution of these systems as a function of topography. Similar to isolated convection, both warm-rain and ice-based processes played important roles in producing intense rainfall in organized convection. Although similarities existed between cell types, organized convection was typically deeper and contained greater ice mass, which melted and contributed to the development of outflow boundaries. As convection organized along the slopes, these boundaries spread over the lower terrain, converging with diurnally driven upslope flow, thus allowing for the generation of new convection and propagation toward the coast. Once over lower elevations, additional warm-cloud depth contributed to intense rainfall and allowed for continued ice production. This, along with the development of rear inflow in the trailing stratiform region, led to further development of convective outflow, similar to organized systems in the tropics and midlatitudes.

Abstract To address questions regarding microphysical processes occurring in the core North American monsoon region, data from NCAR’s S-band polarimetric Doppler radar (S-Pol) deployed during the North American Monsoon Experiment (NAME) in the summer of 2004, were used to investigate the location, size, and type of hydrometeors in convection. A cell identification and tracking algorithm was applied to this data over 100 h of microphysical scans, characterized by increased temporal and vertical resolution, to locate and track individual convective elements. Only isolated cells over land were included for this study to investigate potential elevation-dependent trends in microphysical processes in this region. Examples of intense, isolated convection over all elevations revealed deep cells and polarimetric signatures comparable to other studies of tropical and midlatitude convection. A case over the low terrain highlighted deep, isolated convection with precipitation-sized ice extending to 15 km. In addition, the presence of differential reflectivity ZDR columns in these cells indicated the lofting of supercooled water above the melting level, and an enhanced linear depolarization ratio LDR “cap” above the column implied subsequent freezing to produce graupel. Similar features were also observed in an isolated cell over the western slopes, highlighting the combined roles of collision–coalescence and melting precipitation-sized ice for producing intense rainfall over the lower elevations. Despite previous observations of weaker and shallower cells with less precipitation ice over the Sierra Madre Occidental (SMO), case studies and general statistics using polarimetric data reveal the potential for accretional processes to also play an important role in producing intense rainfall over these higher elevations. For these isolated SMO cells, reduced warm-cloud depths, increased ice mass observed just above the melting level, and a narrower distribution of drop sizes suggest a reduced role of warm-rain processes compared to intense cells over the lower terrain. A potential relationship between microphysical processes and degree of organization is also hypothesized and will be the focus of a future study.

Abstract. Using data for a common period (1976–2005) for a set of 31 sites located in south-east Australia, variations in frequency and magnitude of intense rainfall events across durations from 6 min to 72 h were assessed. This study was driven by a need to clarify how variations in climate might affect intense rainfall and the potential for flooding. Sub-daily durations are of particular interest for urban applications. Worldwide, few such observation-based studies exist, which is mainly due to limitations in data. Analysis of seasonality in frequency and magnitude of events revealed considerable variation across the set of sites, implying different dominating rainfall-producing mechanisms and/or interactions with local topography. Both these factors are relevant when assessing the potential effects of climate variations on intense rainfall events. The set of sites was therefore split into groups ("north cluster" and "south cluster") according to the characteristics of intense rainfall events. There is a strong polarisation in the nature of changes found for the north cluster and south cluster. While sites in the north cluster typically exhibit decrease in frequency of events, particularly in autumn and at durations of 1 h and longer; sites in the south cluster experience an increase in frequency of events, particularly for summer and sub-hourly durations. Non-stationarity found in historical records has the potential to significantly affect design rainfall estimates. An assessment of quantile estimates derived using a standard regionalisation technique and periods representative of record lengths available for practical applications show that such estimates may not be representative of long-term conditions, so alternative approaches need to be considered, particularly where short records are concerned. Additional rainfall information, in particular radar data, could be used for an in-depth spatial analysis of intense rainfall events.

<p>Three consecutive extratropical cyclones named Daniel, Elsa, and Fabien affected the northwest of the Iberian Peninsula during December 2019. In this region is located the Miño-Limia-Sil Hydrographic Demarcation (MLSHD), which includes part of Galicia, in Spain and the north of Portugal. The water resources of the MLSHD are of great importance for the socio-economic framework of both countries, particularly for the agricultural and livestock activities, tourism, and the production of electrical energy from renewable sources like the eolic and the hydroelectric. In this study was analysed the synoptic characteristics of these extratropical cyclones, particularly during the life cycle close to the Iberian Peninsula, when the greatest damages associated with strong winds and intense rainfall occurred. The storm Daniel was formed from a secondary low located to the west and close to the Iberian Peninsula during the afternoon of December 15. Nevertheless, Elsa was formed in the Gulf of Mexico and Fabian in the north Atlantic Ocean, then both crossed the north Atlantic Ocean to finally affect with intense rainfall that caused floods in the MLSHD from 18 to 21 December 2019. The moisture supplies from the tropical north Atlantic Ocean, revealed by the integrated water vapour transport favoured the intensification of all these systems. The consecutive impact of these systems provided great amounts of rainfall to the MLSHD, causing positive anomalies of the total accumulated rainfall for this month. An assessment of drought conditions through the SPI and the SPEI on time scales of 1, 3, 6, and 12 months exposed the role of these systems on drought busting in the MLSHD. Therefore, despite the negative impacts, these systems favoured a recovery of the hydrological conditions of the Demarcation. Abstract Three methodologies for correcting the radar reflectivity factor (ZH) in the presence of partial beam blockage are implemented, compared, and evaluated using a polarimetric radar dataset from the North American Monsoon Experiment (NAME) in northwestern Mexico. One methodology uses simulated interactions between radar beams and digital terrain maps, while the other two invoke the self-consistency of polarimetric radar measurands in rainfall, and the relative insensitivity of a specific differential phase to beam blockage. While the different methodologies often agree to within 1–2 dB, significant disagreements can occur in regions of sharp azimuthal gradients in beam blockage patterns, and in areas where the terrain-caused radar clutter map is complex. These disagreements may be mitigated by the use of additional radar data to develop the polarimetric correction techniques, by a more sophisticated terrain-beam interaction model, or by a higher-resolution digital terrain map. Intercomparisons between ground radar data and Tropical Rainfall Measuring Mission satellite overpasses suggest that all of the methodologies can correct mean ZH to within the expected uncertainty of such intercomparisons (1–1.5 dB). The polarimetric correction methods showed good results even in severely blocked regions (>10 dB reduction). The results suggest the possibility that all of the techniques may be valid approaches to correcting partial beam blockage, and within that context relative advantages and disadvantages of each technique are discussed. However, none of the techniques can correct radar data when weak echoes are reduced to noise by strong blocks, thus leading to biases in corrected ZH and rainfall climatologies.

The role of calcium in modulating cellular replication and differentiation has been described for various cell types. In the present study, the effects of Ca++ on the growth and differentiation of cultured rat esophageal epithelial cells was investigated.Epithelial cells were isolated from esophagi taken from 8 week-old male CDF rats by the enzymatic dissociation method of Kaighn. The cells were cultured in PFMR-4 medium supplemented with 0.25 mg/ml dialyzed fetal bovine serum, 5 ng/ml epidermal growth factor, 10-6 M hydrocortisone 10-6 M phosphoethanolamine, 10-6 M ethanolamine, 5 pg/ml insulin, 5 ng/ml transferrin, 10 ng/ml cholera toxin and 50 ng/ml garamycin at 36.5°C in a humidified atmosphere of 3% CO2 in air. At weekly intervals, the cells were subcultured with a solution containing 1% polyvinylpyrrolidone, 0.01% EGTA, and 0.05% trypsin. After various passages, the replication rate of the cells in PFMR-4 medium containing from 10-6 M to 10-3 M Ca++ was determined using a clonal growth assay.

Protein kinase C (PKC) isozymes, when activated, are translocated to particulate membrane fractions for transport to the apical membrane surface in a variety of cell types. Evidence of PKC translocation was demonstrated in human megakaryoblastic leukemic cells, and in cardiac myocytes and fibroblasts, using FTTC immunofluorescent antibody labeling techniques. Recently, we reported immunogold localizations of PKC subtypes I and II in toad urinary bladder epithelia, following 60 min stimulation with Mezerein (MZ), a PKC activator, or antidiuretic hormone (ADH). Localization of isozyme subtypes I and n was carried out in separate grids using specific monoclonal antibodies with subsequent labeling with 20nm protein A-gold probes. Each PKC subtype was found to be distributed singularly and in discrete isolated patches in the cytosol as well as in the apical membrane domains. To determine if the PKC isozymes co-localized within the cell, a double immunogold labeling technique using single grids was utilized.

SummaryThe variability of the platelet GP Ia/IIa density has been associated with the 807 C/T polymorphism (Phe 224) of the GP Ia gene in American Caucasian population. We have investigated the genotype and allelic frequencies of this polymorphism in Spanish Caucasians. The T allele was found in 35% of the 284 blood donors analyzed. We confirmed in 159 healthy subjects a significant association between the 807 C/T polymorphism and the platelet GP Ia density. The T allele correlated with high number of GP Ia molecules on platelet surface. In addition, we observed a similar association of this polymorphism with the expression of this protein in other blood cell types. The platelet responsiveness to collagen was determined by “in vitro” analysis of the platelet activation and aggregation response. We found no significant differences in these functional platelet parameters according to the 807 C/T genotype. Finally, results from 3 case/control studies involving 302 consecutive patients (101 with coronary heart disease, 104 with cerebrovascular disease and 97 with deep venous thrombosis) determined that the 807 C/T polymorphism of the GP Ia gene does not represent a risk factor for arterial or venous thrombosis.

Maintenance of the individual patterns of gene expression in different cell types is required for the differentiation and development of multicellular organisms. Expression of many genes is controlled by Polycomb (PcG) and Trithorax (TrxG) group proteins that act through association with chromatin. PcG/TrxG are assembled on the DNA sequences termed PREs (Polycomb Response Elements), the activity of which can be modulated and switched from repression to activation. In this study, we analyzed the influence of transcriptional read-through on PRE activity switch mediated by the yeast activator GAL4. We show that a transcription terminator inserted between the promoter and PRE doesnt prevent switching of PRE activity from repression to activation. We demonstrate that, independently of PRE orientation, high levels of transcription fail to dislodge PcG/TrxG proteins from PRE in the absence of a terminator. Thus, transcription is not the main factor required for PRE activity switch.

In reading archaeological texts, we expect to be engaged in a characteristically archaeological discourse, with a specific and recognisable structure and vocabulary. In evaluating the published work of 19th Century antiquarians, we will inevitably look for points of contact between their academic language and our own; success or failure in the identification of such points of contact may prompt us to recognise a nascent archaeology in some writings, while dismissing others as naïve or absurd. With this point in mind, this paper discusses the written and material legacies of three 19th Century antiquarians in the north of Scotland who worked on a particular monument type, the broch. The paper explores the degree to which each has been admitted as an influence on the development of the broch as a type. It then proceeds to compare this established typology with the author's experiences, in the field, of the sites it describes. In doing so, the paper addresses wider issues concerning the role of earlier forms of archaeological discourse in the development of present day archaeological classifications of, and of the problems of reconciling such classifications with our experiences of material culture.

One of the results of the educational reform of the 1860s was the formation of the regular personnel of village teachers. In Vilna educational district the goal was not to invite teachers from central Russia, but to train them on the spot by establishing special seminaries. Trained teachers were supposed to perform the role of «cultural brokers» – the intermediaries between local peasants and the outside world, between the culture of Russian intelligentsia and the culture of the Belarusian people. The article examines how officials and teachers of Vilna educational district saw the role of rural teachers as «cultural brokers» in the context of the linguistic and cultural diversity of the North-Western Provinces. According to them, the graduates of the pedagogical seminaries had to remain within the peasant estate and to keep in touch with their folk «roots». The special «mission» of the village teachers was in promoting the ideas of «Russian elements» and historical proximity to Russia among Belarusian peasants.
