Seed propagation preserves the population genetic variability and helps selecting desirable features. This study evaluated the in vitro germination of Hamatocactus setispinus in six different culture media, 1- MS basal medium full strength; 2- half-strength MS basal medium; 3- 1.0 g L-1 of Peter’s CalMag® 15-05-15 formulation; 4- 0.5 g L-1 of Peter’s CalMag® 15-05-15 formulation; 5- MS basal medium supplemented with 10% coconut water and; 6- water and agar, with and without activated charcoal, and the speed of germination index, the mean germination time and the germination rate, root length, shoot length and the number of roots were evaluate. The seedlings with superior development obtained from in vitro germination were acclimatized in two substrates: Biomix® Floreira; Biomix® Floreira + sand. Seedling survival, shoot length, shoot diameter, root length, root number, shoot fresh matter weight, root fresh matter weight, shoot dry matter weight and root dry matter weight were evaluated. Peter’s 1.0 g L-1 medium without activated charcoal led to the best results for root length (11.36 mm) and root number (3.84). There was 100% of seedling survival. Acclimatization substrates did not differ among themselves and, therefore, they did not affect seedling growth.

Optimal axillary shoot proliferation was obtained from stem explants of a clone of Eriostemon myoporoides DC. on Murashige and Skoog (MS) basal medium containing 0.1 mg BA/liter, and of Eriostemon `Stardust' on MS medium containing 0.5 mg BA/liter. Overall average number of shoots and shoot lengths for all treatments was greater for E. `Stardust' (22.4 shoots and 12.1-mm shoot length) than for E. myoporoides (4.5 shoots and 8.3-mm shoot length). Maximum percent rooting of E. myoporoides (42%) and E. `Stardust' (95%) was obtained on MS medium supplemented with 1.0 mg K-IBA/liter for E. myoporoides and 0.1 mg NAA/liter for E. `Stardust'. Overall average percent rooting and root lengths were greater for E. `Stardust' (42% rooting and 11.0-mm root length) than for E. myoporoides (27% rooting and 2.3-mm root length). For E. `Stardust', reducing sucrose in the rooting medium from 50 to 25 g·liter-1 significantly decreased overall average percent rooting to 1670 and root length to 6.8 mm. Plantlets of both clones were acclimatized in the greenhouse and transferred successfully to soil, although survival was <7070. The use of foliar fertilizers and coconut water is potential; foliar fertilizer is the alternative medium and coconut water is the plant growth regulator (PGR) in the banana tissue culture. This study aimed to examine the ability of foliar fertilizer to substitute Murashige and Skoog (MS) media and coconut water as the main source of cytokine. There are two factors in this research. The first factor is the media, which consist of three levels, i.e. fully MS, ½ MS + ½ foliar fertilizer and fully foliar fertilizer. The second factor is the coconut water with four levels of concentration i.e. 50, 100, 150 and 200 ml l<sup>-1</sup>. The results show that fully formulated foliar fertilizer had not been able to substitute or even compete with a fully MS media, but conversely foliar fertilizer was used to substitute ½ MS media. This can be seen from the parameters of shoot length, leaf length, root number and root length with the highest yield that was found in MS media (8.7, 4.66, 3.33 and 3.23 cm). The 50-100 ml l<sup>-1 </sup>concentrations of coconut water showed the best results by giving a significant effect on the number of roots and root length. The acclimatization of plantlets in this study was nearly 100%, indicated by the formation of a complete organ.

The effect of glutamine, biotin and adenosine diphosphate (ADP) on growth and cultivars micropropagation of three ornamental species was investigated. The addition of 10 ? 100 mg/l glutamine in culture media significantly increased rate of multiplication in Cordyline fruticosa and 100 mg/l glutamine showed the same effect for Gerbera jamesonii. Addition of glutamine did not show any effect on shoot length, root number and length in all the three species. Addition of 1 ? 3 mg/l biotin increased shoot length of Gerbera jamesonii but inhibited shoot length of Cordyline fruticosa and decreased root length of Chrysanthemum hybridum and Gerbera jamesonii. Addition of 5.0 mg/l ADP significantly increased multiplication rate of Cordyline fruticosa and 1.0 mg/l ADP showed similar effect for Gerbera jamesonii.Plant Tissue Cult. & Biotech. 26(1): 97-104, 2016 (June)

Climate change is emerging phenomena and causing frequent drought which lead to scaricity of water, which ultimately nagetively affecting wheat (Triticumaestivum L.) yield all around the world. The aim of this study was to explore the potential deought tolerant wheat genotypes for candidate genes exploration. This study was conducted during the year 2014-2015 at Plant Physiology Division, Nuclear Institute of Agriculture (NIA) Tandojam. The six wheat genotypes (cv. MT-1/13, MT-2/13, MT-3/13, MT-4/13 Chakwal-86 and Khirman) were investigated for their response at germination and seedling stage under different water stress treatments (0, -0.5, -0.75 and -1.0 MPa) in controlled conditions. The results of experiments with reference to genotypes revealed that genotype Chakwal-86 shows maximum seed germination (82.58 %), while the genotype Khirman shows maximum shoot length  (7.23 cm), root length  (15.1 cm), shoot fresh wt. (5.85 g 10-1shoots), root fresh wt.  (3.45 g 10-1roots), shoot dry wt. (1.33 g 10-1shoots), root dry wt. (0.69 g 10-1roots). Among the genotypes tested Khirman and MT-4/13 are the tolerant genotypes had the potential to perform better under drought conditions, whereas  MT-4/13 and Chakwal-86 were moderate tolerant under water stress conditions. Moreover, the genotypes i.e. MT-1/13 and MT-2/13 are the sensitive genotypes under drought environment. It is concluded from present in-vitro studies that osmotic stress significantly reduced the seed germination shoot/root length fresh and dry weight in all six wheat genotypes. The maximum reduction was found at higher osmotic stress induced by PEG-6000 (-1.0 MPa) significantly.

The production of seedlings occurred in polyethylene plastic trays, where they were placed 2/seeds paprika cells of Yolo Wonder cultivar. After the emergence of seedlings was thinning. The experimental design was completely randomized design, containing 5 treatments with 4 replications. The treatments were T1-trays remained out of the "floating", receiving water irrigation 2 times a day; T2-trays of floating, remained out more irrigation the same solution of the "floating" 3 times a week, using 500 ml of solution per Pan; T3-were pans 12:00 are in "floating" and 12:00 am out; T4-trays remained throughout the period of the test in the "floating"; T5-trays remained throughout the period in the "floating", except in the last week. The solution was composed of soluble fertilizer, calcium nitrate, potassium chloride, Triple Superphosphate and Urea, and was changed every Ten days. The parameters analyzed were height of the plant; Root length; Fresh matter weight of shoot; Weight of fresh root matter; Dry matter weight of shoot; Root dry matter weight; Diameter of the stem. All parameters analyzed were submitted to analysis of variance and averages compared by Tukey test to 1% in the case of significance. CONDICIONAMENTO DE MUDAS DE PIMENTÃO (CAPSICUM ANNUM L.), INFLUENCIADAS PELO SISTEMA HIDROPÔNICO “FLOATING”           2 RESUMO   Objetivou-se com esta pesquisa avaliar o desenvolvimento de mudas de pimentão submetidas a diferentes condicionamentos nutricionais, sobre influência do sistema “Floating” visando, uma nova alternativa de produção, para propiciar aos produtores maior segurança em produzir sustentavelmente, para com isso garantir a produção de alimentos saudáveis e geração de renda. A produção de mudas ocorreu em bandejas de polietileno plástico, onde foram colocadas 2 sementes/célula de pimentão da cultivar Yolo Wonder. Após a emergência das plântulas, foi feito o desbaste. O delineamento experimental foi Inteiramente Casualizado, contendo 5 tratamentos com 4 repetições. Os tratamentos foram: T1- As bandejas permaneceram sempre fora do “floating”, recebendo irrigação de água 2 vezes ao dia; T2 – As bandejas permaneceram fora do floating, mais irrigação da mesma solução do “floating” 3 vezes por semana, usando 500 ml da solução por bandeja; T3 – As bandejas ficaram 24 h em “floating” e 24 h fora; T4 - As bandejas permaneceram todo o período do ensaio no "floating"; T5 - As bandejas permaneceram todo o período no "floating” ", exceto na última semana. A solução foi composta pelos fertilizantes solúveis, Nitrato de Cálcio, Cloreto de potássio, Superfosfato Triplo e Ureia, e foi trocada a cada Dez dias. Os parâmetros analisados foram: Altura da planta; Comprimento da raiz; Peso da matéria fresca da parte aérea; Peso da matéria fresca da raiz; Peso da matéria seca da parte aérea; Peso da matéria seca da raiz; Diâmetro do caule. Todos os parâmetros analisados foram submetidos à análise de variância e as médias comparadas pelo teste de Tukey a 1% no caso de significância. O tratamento que se mostrou mais viável foi o T4, pois este apresentou maiores médias para as variáveis altura da planta, diâmetro caulinar, massa fresca da parte aérea e massa seca da parte aérea.   Palavras-chave: Solução Nutritiva, Alternativa de Produção, Segurança produtiva.

The Medicilândia municipality is the main dried cocoa beans producer in Pará state, however, researches are being carried out in relation to the seedlings production. The study objective was to study the cacao seminal seedlings growth under a drip irrigation system, receiving different nitrogen doses with fixed doses of phosphorus (4,5 g) and potassium (3 g) through fertirrigation. The experiment was realized under greenhouse conditions with four treatments and five repetitions in a design of randomized blocks, being each composed by seven plants. Thus, T1 = check, without fertilization; T2 = 2 g of N + P and K; T3 = 7 g of N + P and K; T4 = 12 g of N + P and K. The treatments were scientific from the 31st to the 156th day after sowing, where the values of height, collar diameter and leaves number were obtained fortnightly. On the 156th day, the plants were harvested and obtained the root length, fresh matter and aerial part dry matter and root and the nutritional contents from the leaf and root plant tissue. The plants obtain results with the highest values of height, collar diameter and leaves number in the T2 treatment.

The objective of this work was to evaluate the pH, chemical composition, minerals, vitamins, and in vitro rumen fermentation characteristics of silage prepared with lucerne, sweet maize stalk (MS), and their mixtures. Freshly chopped lucerne and MS were combined in ratios of 100:0 (M0, control), 80:20 (M20), 60:40 (M40), 40:60 (M60), 20:80 (M80), and 0:100 (M100) on a fresh matter basis. Each treatment was prepared in triplicate, and a total of eighteen silos were fermented for 65 days. After 65 days of fermentation, the pH values in M0, M20, M40, M60, M80, and M100 silages were 5.47, 4.84, 4.23, 4.13, 3.79, and 3.61, respectively. As the MS proportion in the mixtures increased, silage K, Ca, P, Na, Fe, and Cu concentrations linearly decreased (p < 0.001) and so did vitamins B5 and K1 and α-tocopherol. In vitro rumen dry matter and organic matter degradability, pH, ammonia, total volatile fatty acid, and gas production linearly decreased (p < 0.01), while neutral detergent fiber concentration linearly increased (p < 0.001), with increasing proportion of MS. The in vitro dry matter and organic matter degradability rapidly decreased when the MS percentage was ≥60%. In conclusion, the M40 silage is the most suitable for livestock utilization in local forage production considering the balance of silage pH, nutritional quality, and in vitro ruminal fermentation characteristics.

Salinity is a major problem affecting crop production all over the world. Excessive soil salinity can reduce the productivity of many agricultural crops including many vegetables and spices. Onion is one of the most important spices in the Asiatic region which is now in high demand. The experiment was conducted to observe in vitro regeneration of onion (Allium cepa L.) under salt stress condition from September 2016 to July 2017. The experiment was conducted as two factorial (genotype and treatment) Completely Randomized Design (CRD) with 3 replications for each treatment. Shoot tip segments of three genotypes namely Faridpuri, Taherpuri and Pusa red (Indian) were cultured in MS (Murashige and skoog, 1962) media supplemented with 25, 50, 75 and 100 mM NaCl. The genotype Faridpuri gave maximum salt tolerance upto 100 mM salinity level with 10.60 cm shoot length and 1.94 cm root length having the highest relative shoot and root growth. Pusa red was found to be salinity sensitive genotype which showing lowest shoot length of 7.03 cm and root length of 0.96 cm at 100 mM NaCl treatment. However, Taherpuri was tolerant up to 100 mM salinity level with 8.14 cm shoot length and 1.25 cm root length. Both the highest fresh weight of root (54.77 mg) and dry weight of root (41.36 mg) was from the genotype Faridpuri with 25 mM NaCl treatment. However, a convenient in vitro regeneration protocol of onion genotypes under different salinity level has been developed and the genotype Faridpuri can be used for further investigation in field condition to evaluate its performance at various salinity levels.

Bacterial wilt (BW) disease caused by Ralstonia solanacearum species complex is a devastating plant disease that inflicts heavy losses to the large number of economic host plants it infects. In this study, the potential of dried powder of the arid-land medicinal shrub Rhazya stricta to control BW of tomato was explored. Both, in vitro and in planta studies were conducted, using different concentrations of dried powder of plant parts, and applied (surface mulched or mixed) to infested soil at 0, 10, and 20 days before transplanting (DBT). Aqueous extract of leaves (16% w/v) was found to be as effective as streptomycin (100 ppm) in inhibiting the in vitro growth of R. solanacearum. As evident from the scanning electron micrograph, 16% aqueous extract of leaves produced severe morphological changes, such as rupture of the bacterial cell walls. Results from the greenhouse experiments demonstrated that the higher powder dose (succulent shoot), namely, 30 g/kg of soil mixed with infested soil 20 DBT, was found to be the most effective in controlling BW. It increased root length (cm), shoot length (cm), and plant fresh biomass (g) by 55, 42, and 40%, respectively, over control plants. Mixing of plant powder with the artificially infested (35 ml of 108 CFU/ml per kilogram of soil) pot soil was better than surface mulching. The 30 g/kg of soil dose mixed with soil increased root length (cm), shoot length (cm), and plant fresh biomass (g) of treated plants by 67, 36, and 46%, respectively, over control plants. A 37% decrease in disease severity over the control was observed with drench application of 30 g of powder per kilogram of soil applied once at 20 DBT. Our results indicated that the dried powder (30 g/kg of soil) of leaves or succulent shoots of R. stricta, thoroughly mixed with soil, 20 DBT, could act as an effective control method against BW.

The first experiment involving different explants and concentrations of 2,4-D and kinetin showed highly significant differences for length and weight of callus formed except interaction of callus weight. Leaf explant appeared to be best of all for callus length and weight when 1.0 mg/L 2,4-D + 0.25 mg/L kinetin concentration was used. Similarly, different explants versus different concentrations of BAP/GA3/IAA showed significant differences for shoot length and leaf number per plantlet and also for root length. However, interaction term confirmed node and node/internode explants produced better results in shoot length and number of leaves per plantlet when concentrations 1 .0 mg/L BAP + 0.1 mg/L GA3 and 1.0 mg/L BAP + 0.2 mg/L GA3, 1.0 mg/L BAP + 0.4 mg/L GA3, respectively, were used. Similarly, internode explants produced better results for root length after 21 days plantlet-1 when concentration of 1.0 mg/L IAA + 0.25 mg/L GA3 was used. Shoot tip explants also produced better results in root length after 28 days plantlet-1 when concentrations 1.0 mg/L IAA + 0.25 mg/L GA3 were used.
