AbstractThe aim of this study was to evaluate the effects of 780-nm laser therapy on the expression of genes related to muscle healing.Sixty-three rats were distributed into three groups: 1) injured control group (CG), 2) injured treatment group at 10 J/cmAt day 7, G10 presented a smaller necrosis area compared to CG and G50. Fourteen days post-surgery, G10 and G50 presented a smaller amount of inflammatory cells and a better tissue organization compared to CG. On day 21, G10 and G50 showed better muscle structure than the control. A significantly decreased COX-2 expression was observed in groups G10 and G50 at day 7 compared to the control animals. No difference was found among the experimental groups after 14 days, but G50 presented statistically higher COX-2 down-regulation at day 21. VEGF expression decreased in the first period analyzed in both treatment groups, increased after 14 days in G10, and increased after 21 days in G50. Both irradiated groups had a higher MyoD expression in all the evaluated periods and myogenin levels increased after 14 days in both treatment groups and in G10 after 21 days.780-nm laser therapy had positive effects during muscle regeneration through the gene expression modulation related to the inflammatory process and the new muscle fibers formation, in both fluencies used in the present study, but the fluence of 10 J/cm

Abstract Background: Melatonin (MT) is important for plant growth and development; however, it is not known whether MT is involved in apple adventitious root (AR) development. In this study, we treated Malus prunifolia (MP) at four different stages of AR development, and analysed the level of the endogenous hormones MT, auxin (IAA), zeatin-riboside (ZR), gibberellins (GA1+3) and abscisic acid (ABA) in all four treatment groups and in the untreated control group. The expression of genes related to MT, IAA biosynthesis, transport and signal transduction, the cell cycle, and root development was quantified by RT-qPCR, and the function of MdWOX11 was analysed in transgenic apple plants.Results: The promotion of AR development by MT was dependent on the stage of AR induction between 0–2 d in apple rootstocks. MT-treatment increased the level of IAA and crosstalk existed between MT and IAA during AR formation. The expression of MdWOX11 was induced by MT treatment and positively regulated AR formation in apple. Furthermore, transgenic lines that overexpressed MdWOX11 lines produced more ARs than ‘GL3’. Phenotypic analysis indicated that MdWOX11 overexpression lines were more sensitive to exogenous MT treatment than ‘GL3’, suggesting that MdWOX11 regulates AR formation in response to MT in apple rootstock.Conclusions: MT promotes AR formation mainly during the AR induction stage by inducing IAA levels and upregulating MdWOX11.

BACKGROUND There is no in vivo evidence for the effectiveness of adjuvant intrastromal and combination of intrastromal and intracameral voriconazole (VCZ) for treating Aspergillus fumigatus keratitis. This study aimed to compare the efficacy of both agents against it.  METHODS A randomized, masked, controlled experimental study was conducted on 11 albino New Zealand white rabbits in which moderate fungal keratitis was induced by inoculating spores of A. fumigatus to the cornea. The rabbits were allocated into 3 groups: 50 μg/0.1 ml intrastromal VCZ injection, 50 μg/0.1 ml intrastromal VCZ and intracameral VCZ injections, and topical VCZ (control). The treatment was given 5 days after inoculation. Epithelial defect, infiltrate size, corneal ulcer depth, and hypopyon were evaluated clinically. Histopathological and mycological examinations were also done 14 days after treatment.  RESULTS All rabbits in the adjuvant treatment groups demonstrated a tendency of a better clinical response with decreasing size of epithelial defect (p = 0.679) and infiltrate (p = 0.755) than in the control group. Direct microscopy, corneal culture, and chop corneal tissue culture were still positive in most of the rabbits from all groups. Histopathological examination showed an increase of inflammatory cells after treatment in all groups, especially in rabbits which were inoculated with A. fumigatus spores in both eyes.  CONCLUSIONS An adjuvant combination of intrastromal and intracameral VCZ showed a tendency of better clinical response for A. fumigatus-induced moderate fungal keratitis in rabbits.

Biomaterials and bone grafts, with the ability of stimulating tissue growth and bone consolidation, have been emerging as very promising strategies to treat bone fractures. Despite its well-known positive effects of biosilicate (BS) on osteogenesis, its use as bone grafts in critical situations such as bone defects of high dimensions or in non-consolidated fractures may not be sufficient to stimulate tissue repair. Consequently, several approaches have been explored to improve the bioactivity of BS. A promising strategy to reach this aim is the inclusion of an organic part, such as collagen, in order to mimic bone structure. Thus, the present study investigated the biological effects of marine spongin (SPG)-enriched BS composites on the process of healing, using a critical experimental model of cranial bone defect in rats. Histopathological and immunohistochemistry analyzes were performed after two and six weeks of implantation to investigate the effects of the material on bone repair (supplemental material-graphical abstract). Histological analysis demonstrated that for both BS and BS/SPG, similar findings were observed, with signs of material degradation, the presence of granulation tissue along the defect area and newly formed bone into the area of the defect. Additionally, histomorphometry showed that the control group presented higher values for Ob.S/BS (%) and for N.Ob/T.Ar (mm2) (six weeks post-surgery) compared to BS/SPG and higher values of N.Ob/T.Ar (mm2) compared to BS (two weeks post-surgery). Moreover, BS showed higher values for OV/TV (%) compared to BS/SPG (six weeks post-surgery). Also, VEGF immunohistochemistry was increased for BS (two weeks post-surgery) and for BS/SPG (six weeks) compared to CG. TGFb immunostaining was higher for BS compared to CG. The results of this study demonstrated that the BS and BS/SPG scaffolds were biocompatible and able to support bone formation in a critical bone defect in rats. Moreover, an increased VEGF immunostaining was observed in BS/SPG.

Background Indoleamine 2,3-dioxygenase (IDO) catalyzes the rate-limiting step in the catabolism of tryptophan along the kynurenine pathway. In tumors, increased IDO activity inhibits T cell and natural killer cell proliferation and induces apoptosis. We have previously reported that IDO expression in lymphoma cells and serum concentration of kynurenine in diffuse large B-cell lymphoma are useful prognostic factors. Preclinical studies in rodents have demonstrated that IDO inhibitors, such as 1-methyl-D-tryptophan (D-1MT), are therapeutically beneficial, especially when combined with different types of cytotoxic chemotherapeutic agents. We investigated the therapeutic potential of IDO inhibitor D-1MT with cyclophosphamide (CY) by using an IDO-positive B-cell lymphoma model mouse. Materials and Methods To establish tumors, 1x106 A20 cells (BALB/c B-cell lymphoma cell line) were subcutaneously injected into the lower back of naïve syngeneic BALB/c mice. Seven days later, 20 BALB/c mice were treated with D-1MT (Sigma-Aldrich, Saint Louis, MO, USA) and/or CY. The mice were fed with D-1MT-supplemented water (5mg/mL), which was replaced every two to three days. CY was administered at 80 mg/kg i.p. on day 10. To examine the effect of D-1MT on A20 tumors and tumor-draining lymph nodes (TDLNs), mice were sacrificed on day 28. Serum concentration of kynurenine and tryptophan were measured by high-performance liquid chromatography. CD4+CD25+Foxp3+ regulatory T cells (Tregs) were counted by flow cytometry with a FACSCantoII flow cytometer (BD Biosciences, San Jose, CA, USA), and data were analyzed with FACSDiva 6.1 software (BD Biosciences). Quantitative real-time reverse transcription-polymerase chain reaction (RT-PCR) was performed with specific primer/probe sets that amplify IDO1, Foxp3, IFN-γ, cyclooxygenase-2 (COX-2), and GAPDH genes (TaqMan Gene Expression Assays; Applied Biosystems, Foster City, CA). Results The mean body and spleen weights of the mice did not differ significantly among the groups. The mean drinking water intakes (mL/day/mouse) of the D-1MT, CY, and D-1MT+CY groups were significantly less than that of the control group (p< 0.05). All mice implanted with A20 cells developed tumors that became palpable after day 7. On day 28, the tumor volume in the D-1MT+CY group was significantly less than that in the control group (p< 0.05), and there were no significant differences among the other treatment groups (Figure 1). There were no significant differences in the serum kynurenine-to-tryptophan ratio and the IDO1 expression level in the tumors among any treatment groups. The expression levels of IFN-γ and COX-2 mRNA in TDLNs were found to be significantly up-regulated in the CY and D-1MT+CY groups. By quantitative RT-PCR, the expression levels of IFN-γ and COX-2 mRNA in TDLNs were found to be significantly up-regulated in the CY and D-1MT+CY groups. The Foxp3 expression level and number of Tregs in TDLNs in the CY and D-1MT+CY groups were significantly higher than in the control group on day 28. The number of Tregs in TDLNs in the D-1MT+CY group was significantly lower than those in CY groups on day 17 (p< 0.05) (Figure 2). Conclusion Our results suggest that D-1MT in combination with CY is an effective treatment for IDO-positive lymphoma in a model mouse by reducing Tregs and breaking tumor tolerance. This antitumor effect might be induced by the suppression of Tregs by D-1MT. Our findings suggest that inhibition of IDO might offer a promising treatment strategy for lymphoma. Abstract Background Yan Hou Qing (YHQ) is a Chinese medicinal formula designed to alleviate sore throat symptoms, but underlying mechanism of YHQ treatment for pharyngitis is poorly defined up to now. Methods In this study, the modulation of YHQ on pharyngitis is investigated in ammonia-induced acute pharyngitis rat models. After treatment with YHQ or dexamethasone respectively for five consecutive days, all rats were sacrificed for biomolecular and histopathologic study. Protein expressions of MAPKs, NF-κB, COX-2 and 5-LOX in pharyngitis tissue were evaluated by western blot analysis and the levels of TNF-α, IL-6, prostaglandin (PG) E2, leukotrienes (LT)-B4 and LT-D4 in pharyngeal tissue were measured via ELISA assay. Evans blue (EB) dye exudation test was performed parallelly to assess the integrity of pharyngeal tissue. Results Compared with normal control group, EB dye exudation, and inflammatory cytokines in the model group were significantly increased, and the pharynx tissue was obviously infiltrated by inflammatory cells. YHQ treatment improved the inflammatory infiltrate in pharyngeal tissue, and reduced EB dye exudation in AP rat models. The up-regulated TNF-α and IL-6 in pharyngeal tissue of AP were significantly reduced by YHQ through inhibition of phosphorylation of p38, Erk and NF-κB. YHQ treatment also reversed the increased level of PGE2 through down-regulation of COX-2. Conclusions YHQ formula attenuated the pharyngitis related symptoms via suppression of COX-2 and phosphorylation of p38, Erk and NF-κB (p65).

Abstract: Aspirin is an anti-inflammatory drug which can cause side effect such as damage of the gastric mucous. Soursop leaf is usually used for medical treatment because it contains flavonoid compound which has the antioxidant and anti-inflamatory activity and may protect gastric mucous from the side effects of aspirin. This study aimed to reveal the histopathological features of the gaster of wistar rats administered with soursop leaf extract after induced with aspirin. This was an experimental study using 20 Wistar rats. Rats were divided into negative control group (A) and treatment groups. Treatment groups were divided into; rats induced with aspirin 30mg for 10 days (B); rats administered with soursop leaf extract 80mg before induced with aspirin 30mg for 10 days (C); rats induced with aspirin 30mg for 10 days and administered with soursop leaf extract 80mg for the next 3 days (D); and rats induced with aspirin 30mg for 10 days and not treated for the next 3 days (E). Groups A, B and C were terminated on 11th day, meanwhile groups D and E were terminated on 14th day. The results showed normal histological features in group A. Group B showed acute gastritic features such as many PMN inflammatory cells in the mucous to serous layers, submucous edema, and capillary dilatation. Groups C and D showed many PMN inflammatory cells in the mucous to submucous layers. Group E showed decreased PMN inflammatory cells in mucous to submucous layers. Conclusion: Administration of soursop leaf extract could not decrease the acute gastritic signs such as inflammatory cells, edema and capillary dilatation in the gaster of Wistar rats induced with aspirin.Keywords: aspirin, soursop leaves, gaster. Abstrak: Aspirin merupakan obat anti inflamasi yang bisa menyebabkan efek samping gangguan mukosa lambung. Daun sirsak sering digunakan sebagai obat tradisional yang berkhasiat karena mengandung senyawa flavonoid yang berkhasiat sebagai antioksidan dan antiinflamasi yang mungkin dapat melindungi lambung dari efek samping aspirin. Penelitian ini bertujuan untuk mengetahui gambaran histopatologik lambung tikus Wistar yang diberikan ekstrak daun sirsak setelah induksi aspirin. Jenis penelitian ini ialah eksperimental yang menggunakan 20 ekor tikus Wistar. Hewan uji dibagi dalam kelompok kontrol negatif (A) dan kelompok perlakuan. Kelompok perlakuan dibagi atas kelompok tikus yang diberi aspirin 30mg selama 10 hari (B); tikus yang diberi ekstrak daun sirsak 80mg sebelum induksi aspirin 30mg selama 10 hari (C), tikus yang diberi aspirin 30mg selama 10 hari dan diberikan ekstrak daun sirsak 80mg selama 3 hari berikutnya (D); dan tikus yang diberi aspirin 30mg selama 10 hari dan tidak diberi perlakuan selama 3 hari berikutnya (E). Kelompok A, B dan C diterminasi pada hari ke-11, kelompok D dan E diterminasi pada hari ke-14. Hasil penelitian menunjukkan gambaran histologik lambung normal pada kelompok A. Kelompok B menunjukkan gambaran histopatologik gastritis akut yakni baanyak sel-sel radang PMN pada lapisan mukosa sampai serosa, edema submukosa, dan pelebaran pembuluh darah kapiler. Kelompok C dan D menunjukkan banyak sel-sel radang PMN pada lapisan mukosa sampai submukosa. Kelompok perlakuan E menunjukkan sel-sel radang PMN yang lebih sedikit pada lapisan mukosa sampai submukosa. Simpulan: Pemberian ekstak daun sirsak tidak dapat mengurangi tanda-tanda gastritis akut berupa sel-sel radang, edema, dan pelebaran pembuluh darah kapiler pada lambung tikus wistar yang diinduksi aspirin. Abstrak: Gastritis akut merupakan proses inflamasi yang bersifat akut dan biasanya terjadi pada bagian permukaan mukosa lambung. Penyakit ini biasanya disebabkan karena banyak faktor salah satunya yaitu penggunaan asam mefenamat dalam dosis yang berlebihan. Pada penelitian yang telah dilakukan sebelumnya didapatkan bahwa lengkuas dapat mengurangi terjadinya radang akut. Tujuan penelitian ini adalah untuk mengetahui gambaran histopatologi lambung tikus wistar yang diberikan lengkuas setelah diinduksi dengan asam mefenamat. Penelitian ini menggunakan metode eksperimental dengan sampel 11 ekor tikus wistar yang dibagi dalam 4 kelompok perlakuan. Kelompok A tidak diberikan perlakuan. Kelompok B diberikan asam mefenamat 10 mg selama 7 hari. Kelompok C diberikan asam mefenamat 10 mg selama 7 hari kemudian diberikan perasan lengkuas selama 7 hari. Kelompok D diberikan asam mefenamat 10 mg kemudian tidak diberikan perlakuan selama 7 hari. Hasil penelitian ini menunjukkan bahwa gambaran mikroskopik lambung tikus wistar pada kelompok C terdapat sel-sel radang PMN yang lebih sedikit dibandingkan dengan kelompok B dan D. Simpulan: lengkuas dapat mengurangi sel-sel radang PMN pada mukosa lambung yang diberikan dengan asam mefenamat. Kata kunci: Lengkuas, Asam Mefenamat, Gastritis akut.   Abstract : acute gastritis is an acute inflammatory process normally occurring in the mucosal lining of the stomach. This disease is caused by a variety of factors, one of which is the use of mefenamat acid in large doses. Previous research has revealed that galangal can be used to reduce acute inflammation. The objective of this research was to reveal the histopathological pictures of stomach of wistar rats that treatment with galangal after induce by mefenamat acid. This study is an experimental, employing 11 wistar rats which were assigned for four treatment groups. Group A was the control group in which the rats received no treatment. Rats in group B were administered mefenamat acid 10 mg for 7 days. In group C, the rats were also administered mefenamat acid 10 mg for 7 days and then received galangal distillation for 7 days. In group D, rats were administered mefenamat acid 10 mg for 7 days and then for 7 days without receiving anymore mefenamat acid or galangal distillation. Results showed that microscopic pictures of stomach of wistar rats in group C were presented with PMN inflammatory cells fewer than rats in group B and D. Conclusion: galangal can reduce PMN inflammatory cells in mucosal lining of the stomach exposed to mefenamat acid. Key words: galangal, mefenamat acid,  acute gastritis.

AbstractObjective:The aim of this study was to evaluate the effects of low level laser therapy (LLLT) on the degenerative process in the articular cartilage after an anterior cruciate ligament transection (ACLT) model in rats.Methods:Eighty male rats (Wistar) were divided into four groups: 1.) intact control group (CG), 2.) injured control group (ICG), 3.) injured laser-treated group at 10 J/cmResults:Initial signs of tissue degradation could be observed 5 weeks post-ACLT, evidenced by the decrease of proteoglycan concentration and increase in cartilage thickness of the ICG. After 8 weeks post-surgery, analysis showed a progression of the degenerative processes in the ICG revealed by the increased cellularity and higher TNF-α, IL1-β and MMP-13 immunoexpression. LLLT was able to modulate some of the aspects relating to the degradative process, such as biomodulation of the number of chondrocyte proliferation, prevention of proteoglycan loss, and decrease of MMP-13 immunoexpression.Conclusion:This study showed that the 685-nm laser irradiation, especially at 10 J/cm

BACKGROUND: Crohn's disease accompanied by nonspecific or idiopathic ulcerative proctocolitis corresponds to a condition called intestinal inflammatory disease. The immunoexpression of cyclooxygenase 2 (COX-2) in Crohn's disease becomes more marked with progression of the disease and the presence of wild-type p53 suppresses the transcription of COX-2. AIMS: To investigate the immunoexpression of cyclooxygenase 1 (COX-1), COX-2 and p53 in Crohn's ileocolitis and to correlated this expression with clinical and histopathological parameters. METHODS: Forty-five cases of Crohn's disease, 16 cases of actinic colitis (diseased-control group) and 11 cases without a history of intestinal disease (normal control group) were studied. Hematoxylin-eosin-stained sections were submitted to histopathological analysis and the immunohistochemical expression of COX-1, COX-2 and p53 was evaluated by the streptavidin-biotin-peroxidase method. RESULTS: Sixty percent of the Crohn's disease patients were women and 40% were men, with 75.5% whites and 25.5% non-whites. The disease involved the terminal ileum in 44.5% of cases, ileum in 33.3%, colon in 20% and duodenum-ileum in 2.2%. A significant association was observed between COX-2 immunoreactivity and age <40 years. Histopathological analysis of Crohn's disease samples showed mild or moderate crypt distortion (57.8% and 35.6% of cases), atrophy (6.6%), mild, moderate and marked chronic inflammation (46.7%, 26.7% and 20%), acute inflammatory activity (93.3%), ulceration (24.4%), mucin depletion (37.8%), Paneth's cells (24.4%), intraepithelial lymphocytes (93.3%), and subepithelial collagen (6.7%). In the CD group, COX-1 immunoreactivity in epithelial and inflammatory cells was observed in 26.7% and 22.2% of cases, respectively. COX-2 immunoreactivity was detected in epithelial cells in 68.9% of cases and in inflammatory cells in 46.7%. A marginal difference in COX-2 reactivity was observed between epithelial and inflammatory cells in association with acute inflammatory activity and increase in intraepithelial lymphocytes. Comparison of the date among the threes groups (Crohn's disease, actinic colitis and normal controls) showed a higher proportion of cases presenting COX-2 immunoreactivity in inflammatory cells in the Crohn's disease group. No p53 reactivity was observed in all cases. CONCLUSIONS: COX-2 immunoexpression is high in Crohn's disease, which suggest a possible role of the protein in the pathogenesis of the inflammation. The absence of epithelial dysplasia in all Crohn's disease samples was correlated with the lack of expression of p53.

Pain is the most common complaints in everyday practice. Pain is a clinical manifestation caused by stimuli due to potential tissue damage. Low level laser therapy (LLLT) has been used in reducing pain. One of indicator tissue healing is activation of transforming growth factor -b1(TGF-b1). Tissue healing will eliminate the pain. Do LLLT stimulate TGF-b1 expression? The objective of this study was to investigate the expression of TGF-b1 in low level laser therapy. Thirty male Wistar rats, body weight 250 – 300 gram, 3 month old, were divided into 3 groups, negative control (n=10), positive control (n=10) and treatment groups (n=10). The positive control and treatment groups were injected with Complete’s Freund Adjuvant (CFA) 100 mL at left footpad. The treatment group treated with low level laser therapy Lasermed type 2100 wavelength 905 nm, out power 25-500 mW, dose 1 joule/cm2;5 times with interval 24 hours on left footpad. Samples were taken from the skin of footpad and immunohistochemical examination with monoclonal antibody anti- TGF-b1. Calculations carried out on brown reaction in macrophage cell. Data were analyzed using the Games Howell with SPSS 17. There were significant differences in the expression of TGF-b1 in treatment group (15.8±3.1) compared with the positive control group (3.9±1.1) and negative control group (4.6±2.1). There were no significant differences between expression TGF-b1 in positive control group and negative control group. In conclusion, low level laser therapy increased the expression of TGF-b1.
