Development of data reduction function for hyperspectral imaging

Algorithms have been developed for controlling the calibration and the measurement cycles of hyperspectral equipment. Special calibration and preprocessing methods were necessary to obtain suitable signal level and acceptable repeatability of the measurements. Therefore, the effect of the noise of NIR sensor was decreased, the signal level was enhanced and stability was ensured simultaneously. In order to investigate the properties of the number of objects suitable for statistical analysis, the enormous size of acquired hypercube (gigabytes per object) should be reduced by vector-to-scalar mathematical operators in real-time to extract the desired features. The algorithm developed was able to calculate the score of operators during scanning and the matrices were displayed as pseudo-images to show the distribution of the properties on the surface. The operators had to be determined by analysis of a sample set in preliminary experiments. Stored carrot was chosen as a model sample for investigation of the detection of moisture loss by hyperspectral properties. Determination of the proper operator on different tissues could help to analyze and model drying process and to control storage. Hyperspectral data of different carrot cultivars were tested under different storage conditions. Using improved measurement method the spectral parameter of the suitable operator described quite well the moisture loss of the different carrot tissues.

Background:A growing part of the root is one of the most active sinks for sucrose coming from source leaves through the phloem. In the root, sucrose is unloaded from conducting bundles and is distributed among the surrounding cells. To be involved in the metabolism, sucrose should disintegrate into hexoses by means of degrading enzymes.Aims:The aim of this research was to explore the possibility of the involvement of one such enzymes, invertase, in phloem unloading as well as distribution of its activity in the functionally different tissues of the plant root tips.Method:To estimate the enzyme activities in root tissues, we applied two techniques: the histochemical method using nitro blue tetrazolium. The localization of phloem unloading was studied with carboxyfluorescein, a fluorescent marker for symplastic transport.Results:Invertase activity was not detected in the apical part of the meristem. It appeared only between the basal part of this zone and the beginning of the elongation zone. There is the root phloem unloading in that area. Invertase activity increased with increasing the distance from the root tip and reached the highest values in the region of cell transition to elongation and in the elongation zone. The activities of the enzyme varied in different tissues of the same zone and sometimes in the neighboring cells of the same tissue. Biochemical determination of invertase activity was made in the maize root segments coincident to the zones of meristem, cell elongation and differentiation. The results of both methods of determination of invertase activity were in agreement.Conclusion:It was concluded that phloem unloading correlated with invertase activity, possibly because of the activation of invertase by unloaded sucrose. Invertase is one of the factors involved in the processes preparing the cells for their transition to elongation because the concentration of osmotically active hexoses increases after cleavage of sucrose, that stimulates water entry into the cells, which is necessary for elongation growth.

The hydrodynamic characteristics of the planing hulls in particular at the planing regime are completely different from the conventional hull forms and the determination of these characteristics is more complicated. In the present study, calm water hydrodynamic characteristics of planing hulls are investigated using a hybrid method. The hybrid method combines the dynamic trim and sinkage from the Zarnick approach with the Savitsky method in order to calculate the total resistance of the planing hull. Since the obtained dynamic trim and sinkage values by using the original Zarnick approach are not in good agreement with experimental data, an improvement is applied to the hybrid method using a reduction function proposed by Garme. The numerical results obtained by the hybrid and improved hybrid method are compared with each other and available experimental data. The results indicate that the improved hybrid method gives better results compared to the hybrid method, especially for the dynamic trim and resistance. Although the results have some discrepancies with experimental data in terms of resistance, trim and sinkage, the improved hybrid method becomes appealing particularly for the preliminary design stage of the planing hulls.

A laboratory prototype for hyperspectral imaging in ultra-violet (UV) region from 225 to 400 nm was developed and used to rapidly characterize active pharmaceutical ingredients (API) in tablets. The APIs are ibuprofen (IBU), acetylsalicylic acid (ASA) and paracetamol (PAR). Two sample sets were used for a comparison purpose. Sample set one comprises tablets of 100% API and sample set two consists of commercially available painkiller tablets. Reference measurements were performed on the pure APIs in liquid solutions (transmission) and in solid phase (reflection) using a commercial UV spectrometer. The spectroscopic part of the prototype is based on a pushbroom imager that contains a spectrograph and charge-coupled device (CCD) camera. The tablets were scanned on a conveyor belt that is positioned inside a tunnel made of polytetrafluoroethylene (PTFE) in order to increase the homogeneity of illumination at the sample position. Principal component analysis (PCA) was used to differentiate the hyperspectral data of the drug samples. The first two PCs are sufficient to completely separate all samples. The rugged design of the prototype opens new possibilities for further development of this technique towards real large-scale application.

Under the condition of nine kinds of storage, In this thesis, through the determination of acid value of soybean oil, to search for the best storage condition, used to prevent the soybean oil acid value rise, through the analysis of the acid value data, We find the best storage condition, is 0°C, 6% moisture content, and PVC plastic wrap packaging.
