ABSTRACT – HCTN 06The Sea urchin Tripneustes gratilla (Linnaeus, 1758) is a famous species of invertebrate member of the phylum Echinodermata of marine animals living in shallow tropical water. In Viet Nam, it is usually called by the name of yellow sea urchin. This species is mentioned much about its economic and commerce properties. Recently, many researchers have been paying attention to sea urchin because of its positive effect on human health. In terms of medicine and nutrition, the egg of yellow sea urchin was rich in Polyunsaturated Fatty Acids (PUFAs) and essential amino acids. In this research, we focused on determining the total lipid contents, the components of lipid classes and the compositions and contents of fatty acids, amino acids from the body and the egg of the yellow sea urchin T. gratilla (Linnaeus, 1758) collected in Hon Tam, Nha Trang, Khanh Hoa, Viet Nam in 2016. Although the yellow sea urchin T. gratilla has low percentage of total lipid in fresh weigh basis, they are full of essential lipid ingredients such as Wax and Hydrocabon (H + W), Triacylglycerol (TG), Monodiacylglycerol MDAG, Free Fatty Acid (FFA), sterol (ST), and Polar Lipid (PL). In both egg and body samples palmitic acid (16:0) was the most abundant fatty acid with the proportion of about 25%. In addition, arachidonic acid (20:4n-6) is dominent Omega-6 fatty acid in the Monounsaturated Fatty Acid group (MUFA). Furthermore, the research result also showed the very high percentages of omega-3 fatty acids in the egg and body of T. gratilla, with the total contents of 13.97% and 20.67% respectively, especially the presence of eicosapentaenoic fatty acid (C20:5n-3, EPA), a very valuable fatty acid. By the HPLC method, the research also found 17 amino acids in the egg and body of T. gratilla with the existence of 7 essential amino acids.Keywords: Tripneustes gratilla, lipid classes, yellow sea urchin, echinodermata, fatty acid content.

This study studied the content and composition of the total lipid, lipid classes and fatty acids in 13 brown seaweed Sargassum species collected from Con Dao and Van Phong, Vietnam. The total lipid has a low content and varies among species from 0.10–1.70% of the fresh weight. From 13 species, seven lipid classes including polar lipid (Pol), free fatty acids (FFA), sterol (ST), hydrocarbon and wax (HW), triacylglycerol (TG), diacylglycerol (DG), and monoalkydiacylglycerol (MADG). Using the GC-FID technique, we have identified 29 fatty acids classified into 3 groups of saturated fatty acid, monounsaturated fatty acids, polyunsaturated fatty acids with an average content of 44.93%, 24.57% and 27.44%, respectively. Among those, many value fatty acids have been detected with high content such as C18:3n-3, C20:4n-6, 20:5n-3, and 22:6n-3. The lipid of 13 brown seaweed Sargassum species also fully contains omega-3,6,9 fatty acids with the content of 9.28%, 16.28% and 16.63%, respectively.

The fatty acid profiles of Euphausia superba, the Antarctic prymnesiophyte, Phaeocystis pouchetii, and a temperate diatom, Phaeodactylum tricornutum were analysed and compared. The lipid content, lipid class, fatty acid and sterol composition of E. superba fed on each cultured phytoplankton and a mixed diet of both species, were determined. No significant difference was found between total lipid levels of E. superba reared on each of these different diets. Phaeocystis pouchetii, although deficient in a number of the essential fatty acids, is apparently an adequate food source for E. superba. The proportion of polyenoic fatty acids varied within lipid classes although there was no significant difference between levels of the long chain polyunsaturate 20:5(n-3) in the total lipid of E. superba fed on these diets. This acid was found to be less than 1% of the total lipid in Phaeocystis pouchetii compared to 37% in Phaeodactylum tricornutum. This suggests that krill may possess the ability to convert exogenous shorter chain fatty acids to 20:5(n-3) and 22:6(n-3). Significant differences were detected in the isomeric ratio of 16:1(n-7c)/16:0 between krill fed the diatom compared to the prymnesiophyte diet. Significant differences were also detected in several shorter chain fatty acids and between fatty acids within their lipid classes. Such differences may have the potential to be used as biochemical signatures to provide information on food sources and possible feeding grounds of E. superba. Phaeocystis pouchetii in a very late stationary phase, although not used in this feeding study, was found to contain 11% of 22:6(n-3) for which there are few substantive sources in natural algal populations.

The effects of hypoglycin A on the metabolism of L-leucine-C14, L-alanine-C14, and L-glutamic-acid-C14 by rat liver slices have been investigated. Hypoglycin exerted markedly inhibitory effects on the conversion of leucine-C14 to fatty acid, cholesterol, and CO2. Conversion of alanine-C14 and glutamic acid-C14 to fatty acids was also inhibited by hypoglycin. No effects of hypoglycin on the conversion of C14-amino acids into protein or glycogen were demonstrated.

Abstract Sewage sludge was subjected to hydrothermal fueling (HTF) (330 °C for 40 min), obtaining hydrochar at 13.5 MJ kg−1. The higher heating value (HHV) of the raw sludge was related to its fatty acid content. The results showed that although the higher heating value (HHV) of the raw sludge was related to its fatty acid content, with the intensification of HTF, the increase in aliphatic/cyclic amino acids determined the production of HHV in the hydrochar. In order to increase the content of fatty acids and amino acids, the sludge was fermented. However, the Bacteroidetes consumed the organic matter too early, which was detrimental to the production of HHV. Therefore, appropriate sludge fermentation is recommended to restrict excessive Bacteroidetes proliferation, decompose lipids to saturated fatty acids, and convert proteins to aliphatic/cyclic amino acids to increase the efficiency of converting sludge to fuel.

Dolabella auricularia are found in the waters of Indo - Pacific and has active compound in health, which until now is still limited information about nutritional content from sea hare. The aim of this research were to determine morphometric and chemical characteristic D. auricularia which includes the proximate, amino acids, fatty acids and minerals. The composition of fatty acid were measured by gas chromatography (GC), amino acids were measured by high performanced liquid chromatography (HPLC), and mineral was measured by atomic absorption spectrophotometer (AAS). The sea hare contained 9 essential amino acids and 6 non essential amino acids. The highest essential amino acid was arginine (1.61%) while the highest non essential amino acids was glycine (3.02%). Sea hare contained 26 fatty acids such as saturated fatty acids 5.33%, monounsaturated fatty acids 2.11% and polyunsaturated fatty acids 4.10%. The high mineral was calcium 68100 mg/kg.

Fatty acid compositions were determined for total lipid (17.5% of the milk and > 95% triglycerides), 2-monoglyceride obtained by enzymatic hydrolysis of isolated triglyceride, and isolated phospholipid (~1% of total lipids). The total lipid fatty acids of the milk had a composition similar to fin whale depot fat but were enriched in hexadecanoic acid and polyunsaturated fatty acids at the expense of monoethylenic acids; correspondingly the iodine value of 136 (methyl esters) was higher than the normal range (105–120) of North Atlantic fin whale blubber oils. Over 80% of the fatty acids in the 2-position of the triglycerides were accounted for by relatively short chain fatty acids, especially hexadecanoic (54.6%), tetradecanoic (13.7%), and hexadecenoic (11.2%), so that the ester iodine value was only 48. The milk phospholipids had a fatty acid composition basically similar to that of liver phospholipids (methyl ester iodine value 120) with somewhat more polyunsaturated fatty acids and accordingly an iodine value of 144 for methyl esters.

The processing carried out on coffee beans such as fermentation and roasting can affect the contents of amino and fatty acids of coffee beans. This study aimed to determine the amount of amino acid and fatty acid content in Luwak coffee bean during processing. The amino acids of coffee were analyzed using HPLC, while fatty acids of coffee were analyzed using GC-MS. The results showed a change in total amino acid content in raw coffee bean (3.04%), green bean coffee (6.93%), and roasted coffee (6.83%). The total fatty acid of raw coffee bean (1199.86 mg/100 g), green bean coffee (3147.56 mg/100 g), and roasted coffee (6282.4 mg/100 g) also experienced significant changes

Spirochete MA-2, which is anaerobic, ferments glucose, forming acetate as a major product. The spirochete also ferments (but does not utilize as growth substrates) small amounts of l -leucine, l -isoleucine, and l -valine, forming the branched-chain fatty acids isovalerate, 2-methylbutyrate, and isobutyrate, respectively, as end products. Energy generated through the fermentation of these amino acids is utilized to prolong cell survival under conditions of growth substrate starvation. A branched-chain fatty acid kinase and two acetate kinase isozymes were resolved from spirochete MA-2 cell extracts. Kinase activity was followed by measuring the formation of acyl phosphate from fatty acid and ATP. The branched-chain fatty acid kinase was active with isobutyrate, 2-methylbutyrate, isovalerate, butyrate, valerate, or propionate as a substrate but not with acetate as a substrate. The acetate kinase isozymes were active with acetate and propionate as substrates but not with longer-chain fatty acids as substrates. The acetate kinase isozymes and the branched-chain fatty acid kinase differed in nucleoside triphosphate and cation specificities. Each acetate kinase isozyme had an apparent molecular weight of approximately 125,000, whereas the branched-chain fatty acid kinase had a molecular weight of approximately 76,000. These results show that spirochete MA-2 synthesizes a branched-chain fatty acid kinase specific for leucine, isoleucine, and valine fermentation. It is likely that a phosphate branched-chain amino acids is also synthesized by spirochete MA-2. Thus, in spirochete MA-2, physiological mechanisms have evolved which serve specifically to generate maintenance energy from branched-chain amino acids.

Water buffalo (Bubalus bubalis) conservation in Serbia is under an in situ program, but additional efforts are needed to ensure the development of this animal’s genetic resources biodiversity. This research aims to describe challenges and possible strategies for sustainable water buffalo milk production. In this study, the physicochemical characteristics of buffalo milk and buffalo dairy products (cheese, butter, and kajmak) were determined. Furthermore, amino and fatty acids composition and the related health lipid indices (atherogenic and thrombogenic) were assessed. The findings support the fact that buffalo milk is a reliable source of high-quality nutrients (dry matter: 16.10%, fat: 6.02%, protein: 4.61%). Leucine, lysine, and valine content were found to be high in buffalo milk and cheese. A substantial quantity of non-essential glutamic and aspartic amino acids was observed in milk, as well as glutamic acid and tyrosine in cheese. It was established that milk protein of buffalo cheese had a favorable proportion of essential and non-essential amino acids (61.76%/38.24%). The results revealed significant differences (p < 0.05) in fatty acid profiles among the three dairy products for saturated short-chain, n-3, and n-6 fatty acids. Conversely, no significant difference (p < 0.05) was observed in monounsaturated fatty acids content. Kajmak showed the most favorable anti-atherogenic and anti-thrombogenic properties due to lower saturated and higher polyunsaturated fatty acid content. These results confirmed that buffalo milk could be successfully used in producing high-quality traditional dairy products with added value and beneficial characteristics from the aspect of a healthy diet. Furthermore, it could actively contribute to the promotion of sustainable production of buffaloes and strengthen the agricultural production of rural areas and their heritage.

From a population of cells of Pseudomonas aeruginosa sensitive to 33 parts per million (ppm) of quaternary ammonium compound (QAC) and to 0.1 mg/ml of chloramphenicol (CAP), pure strains resistant to 750 ppm and 2.5 mg/ml of these antibacterial substances were isolated. Lipids from the sensitive and resistant cells grown under various conditions were extracted and characterized. Increased lipid biosynthesis was investigated as a possible factor in the resistance of this bacterium to CAP and QAC.Resistant cells grown in medium containing 207 ppm QAC at 25C averaged 77% more total lipid than sensitive cells. Resistant cells grown in the absence of QAC at 25C retained their resistance and averaged 27% more total lipid than sensitive cells. Cells grown at 32C in the presence of QAC contained 8% more total lipid than sensitive cells but less than cells grown at 25C under the same conditions. There was no increased lipid production by QAC-resistant cells grown at 37C in a Fermacell fermentor. Cells grown in CAP-containing medium (2.5 mg/ml) at 25C developed 28% more total lipid than sensitive cells grown in antibiotic-free medium.Gas–liquid chromatograms of the phospholipid, free fatty acid, and triglyceride fractions of the various cell types indicated that the same fatty acids were present. There was a 10% increase of the C18:1 fatty acid in both the phospholipid and free fatty acid fractions of the free lipids of cells grown in a Fermacell fermentor in the presence of CAP or QAC. However, there was no compensatory decrease in any single fatty acid, but rather a general decrease.Sensitive P. aeruginosa contained 14% free lipid and 3% to 6% bound lipid. The major phospholipid was phosphatidyl ethanolamine, which constituted 59% of the total phospholipid fraction of the sensitive cells. Analysis of fatty acids revealed that C16 amounted to 34%, C18:1 19%, and C16:1 and C18 each about 5% of the total present. These four fatty acids accounted for 63% of the free lipid present in the sensitive cells.
