Presence of Aspartate Aminotransferase in Peri-Implant Crevicular Fluid With and Without Mucositis

The aim of this study was to assess the presence of aspartate aminotransferase (AST) in peri-implant crevicular fluid, with or without clinical signs of mucositis, to determine its predictive diagnostic value, sensitivity, and specificity. The AST levels were determined (at a threshold of 1200 µIU/mL) for 60 clinically successful implants in 25 patients with or without peri-implant mucositis. Samples were taken prior (AST1) to peri-implant probing with a manual constant-pressure probe (0.2 N) and 15 minutes after probing (AST2). Clinical assessments included radiographic determination of preexisting bone loss, probing, and the evaluation of mucositis, plaque, and bleeding upon probing. Analysis was performed at both the level of the implant and the patient as a unit. We detected a significant difference between AST1 and AST2 at both levels. A significant difference was observed at AST1 between implants that bled upon probing and those that did not. However, when we considered the patient as a unit, there were no significant differences. The plaque index was not significant at either level. AST1 had high specificity and positive predictive diagnostic value (80%) for bleeding upon probing. Probing induces a greater release of AST from inflamed tissues compared with healthy tissues in situ but not at the systemic level. At the implant level, the implant position could be responsible for this difference. Aspartate aminotransferase was a reliable predictor of patients with mucositis.

BackgroundThis study aimed to evaluate the diagnostic value of telomerase activity (TA) for bladder cancer (BC) by meta-analysis.MethodsWe conducted a systematic search of studies published on PubMed, Embase, and Web of Science up to June 1, 2019. We used Stata 15 and Review Manager 5.3 for calculations and statistical analysis.ResultsTo evaluate the diagnostic value of TA for BC, we performed a meta-analysis on 22 studies, with a total of 2,867 individuals, including sensitivity, specificity, positive and negative likelihood ratio (PLR, NLR), diagnostic odds ratio (DOR), and 95% confidence intervals (CIs). The pooled parameters were calculated from all studies, and we found a sensitivity of 0.79 (95% CI: 0.72–0.84), a specificity of 0.91 (95% CI: 0.87–0.94), a PLR of 8.91 (95% CI: 5.91–13.43), an NLR of 0.24 (95% CI: 0.15–0.37), a DOR of 37.90 (95% CI: 23.32–61.59), and an AUC of 0.92 (95% CI: 0.90–0.94). We also conducted a subgroup analysis based on the different stages and grades of BC. Results from the subgroup analysis showed that there was no significant difference in TA in either high and low stages of BC, but that low-grade tumors had a lower TA than high-grade tumours.ConclusionsTA can be used as a potential biomarker for the diagnosis of bladder cancer with its high specificity. Rigorous and high-quality prospective studies are required to verify our conclusion.

Abstract Objectives This study was designed to evaluate the role of anti-CD74 antibodies in diagnosis of axial spondyloarthritis (axSpA) and their relationship to disease duration and disease activity. Methods Fifty patients with axSpA, 15 patients with RA and 15 healthy subjects were included in the study. Clinical examination and laboratory tests were done. The ESR, CRP level and ASDAS were measured as markers of the disease activity. Quantitative determination of human CD74 IgG antibodies was done. Results The mean age of the patients was 38.22 (S.D.12.20) years. The level of CD74 autoantibodies was significantly higher in axSpA in comparison to control groups. Most patients with positive articular and extra-articular manifestations were positive for CD74 autoantibodies. In patients with inactive disease, 33.3% were positive for CD74 autoantibodies, as were 83% with active disease. High percentages of patients with early and late axSPA were CD74 autoantibody positive. The majority of patients with positive disease activity in early and late axSpA were CD74 autoantibody positive. CD74 autoantibodies had 80% sensitivity vs both control groups with 87% specificity vs the healthy control group and 80% vs the RA control group in the diagnosis of axSpA. Conclusions The frequency of positive anti-CD74 IgG antibodies was as high in patients with early axSpA as in those with late axSpA, with no significant differences. There was a significant difference in the frequency of positive anti-CD74 IgG antibodies between patients with positive and negative disease activity. Based on the sensitivity and specificity of anti-CD74 IgG, this is a promising diagnostic tool to support the clinical diagnosis of axSpA.

Abstract Abstract 2826 Poster Board II-802 To investigate whether genetic risk factors affect the outcome of relapsed/refractory myeloma patients undergoing bortezomib therapy, we evaluated the clinical features of 85 patients treated with bortezomib and correlated with the high-risk cytogenetic abnormalities as detected by interphase cytoplasmic fluorescence in situ hybridization (cIg-FISH). Forty (47%) patients had an objective response to bortezomib with median progression free (PFS) and overall survivals (OS) of 9.4 and 12.6 months, respectively. cIg-FISH determination of del(13q), del(17p), del(1p21), t(4;14), and amp. (1q21)(CKS1B) was done on 79, 77, 77, 78, and 80 cases and the frequency of their detection was 38%, 22%, 26%, 18%, and 39%, respectively. There was no statistically significant difference in terms of response or duration of response to bortezomib for patients with or without any of the cytogenetic abnormalities. PFS was also comparable among patients with and without the aforementioned genetic markers. However, patients with 1q21(CKS1B) amplification had significantly shorter OS than those without such abnormality (5.3 vs. 24.6 months, p=0.0005). Multi-variate analysis adjusting for all 5 high-risk genetic factors confirmed that 1q21(CKS1B) amplification is an independent risk factor for OS (p=0.008). The determination of myeloid and lymphoid lineage is essential for the diagnosis and therapy of acute leukemia. Immunophenotyping is the gold standard to determine the lineage of acute leukemia, but it is still constrained and relatively expensive. Mie Map RBC in the ADVIA 2120i is a parameter that can give additional information about myeloid and lymphoid lineage but has never been studied before. It is expected that Mie Map RBC can be used to differentiate the lineage of acute myeloid and lymphoid leukemia if immunophenotyping is not present. This study aimed to analyze the diagnostic value of Mie Map RBC with ADVIA 2120i towards immunophenotyping in determining myeloid and lymphoid lineage in acute leukemia. Child and adult patients diagnosed with acute leukemia (n=30) that had peripheral blood smear and bone marrow aspiration with blasts > 20% were examined using ADVIA 2120i. The Mie Map RBC lineage results were compared to the lineage of immunophenotyping. The sensitivity and specificity of the Mie Map RBC myeloid series are respectively 60.00%, 93.33%. The sensitivity and specificity of the Mie Map RBC lymphoid series are respectively 93.33% and 60.00%. The diagnostic accuracy value of Mie Map RBC is 76.67%. The determination of acute leukemia myeloid series lineage has high specificity. If there is no population outside the matrix of Mie Map RBC, it highly suggests myeloid series. On the other hand, the determination of acute leukemia lymphoid series lineage has a relatively low specificity meaning that the population outside the matrix of Mie Map RBC does not always suggest a lymphoid lineage

This article describes the application and performance of an inexpensive, simple and portable device for colorimetric quantitative determination of drugs in pharmaceutical preparations. The sensor is a light detector resistor (LDR) incorporated into a black PTFE cell and coupled to a low-cost multimeter (Ohmmeter). Quantitative studies were performed with captopril/p-chloranil/H2O2 and methyldopa/ammonium molybdate systems. Calibration curves were obtained by plotting the electrical resistance of the LDR against the concentration of the colored species in the ranges 1.84 × 10-4 to 1.29 × 10-3mol L-1 and 5.04 × 10-4 to 2.52 × 10-3 mol L-1 for captopril/p-chloranil/H2O2 and methyldopa/ammonium molybdate systems, respectively, exhibiting good coefficients of determination. Statistical analysis of the results obtained showed no significant difference between the proposed methodologies and the official reported methods, as evidenced by the t-test and variance ratio at a 95% confidence level. The results of this study demonstrate the applicability of the instrument for simple, accurate, precise, fast,in situ and low-cost colorimetric analysis of drugs in pharmaceutical products.

Abstract The current AOAC procedure for semiautomated determination of niacin specifies the use of externally generated cyanogen bromide. Because of the safety concerns in handling this material, we investigated the use of an alternative system of generating cyanogen chloride in situ, using chloramine-T and potassium cyanide. Recovery studies conducted on 9 different food products yielded average recoveries of 101 %. A repeatability study resulted in a measured coefficient of variation of 2.9%. The AOAC niacin method was compared with this semiautomated method; 115 paired analyses on 8 different food types over 6 separate analytical replications indicated no significant difference by a paired t-test at the 95% confidence level.

Background Determination of the extent of involvement or pathological staging is one of the requirements for adequate evaluation of bladder cancer specimens. Therefore, the differentiation between MP and MM is essential for proper treatment and avoiding over or under staging. Aim The present study aimed at evaluation of diagnostic value of smoothelin and vimentin expression both singly and in combination for differentiation between MM and MP. Methods This study was carried out on 59 cystectomy specimens of primary bladder carcinoma and eleven cystoscopic biopsies for non-neoplastic bladder lesions (cystitis). Results Histologically, MM was identified clearly in 40 cases and MP was identified in all 70 examined cases. The cases were immunostained for smoothelin and vimentin. Intensity of smoothelin expression showed significant difference (P = 0.001) between MM and MP with 97.5% sensitivity and 95% specificity and the percentage of smoothelin expression was significantly higher in MP compared to MM (P = 0.001) with 95.7% sensitivity and 85% specificity (using 65% as cut-off point). Vimentin was negative in MP and showed positive expression in 32 cases (80%) of MM with a statistical significant difference (P = 0.001) providing 80% sensitivity and 100% specificity. Combined moderate to strong smoothelin and negative vimentin offered 100% sensitivity and 100% specificity towards the identification of MP. Conclusions Differentiation of MM from MP can be made based on histopathological criteria, which are unfortunately overlapping in many cases. Moderate to strong smoothelin expression with negative vimentin could be very helpful procedure in difficult and overlapping cases with a high diagnostic validity.

Three visible spectrophotometric methods (A–C) were developed for the analysis of someβ-blocker drugs, namely atenolol (ATE) and timolol (TIM) based on their reactivity with bromine, generated in situ by the action of hydrochloric acid on bromate–bromide mixture. The determination of residual bromine is based on its ability to bleach the indigo carmine dye and measuring the absorbance at 610 nm (method A). Methods B and C involve treating the unreacted bromine with a measured excess of iron (II), the remaining iron (II) is complexed with 1,10-phenanthroline (method B) or with 2,2'-bipyridyl (method C) and measuring the increase in absorbance at 510 and 522 nm, respectively. In all the methods, the amount of bromine reacted corresponding to the drug content. Regression analysis of Beer's plot showed good correlation in the concentration ranges of 0.4–16.4, 0.8–10.4 and 0.4–12.8 µg/ml using methods A–C, respectively, for ATE and 14–38, 12–32 and 14–38 µg/ml using methods A–C, respectively, for TIM. The optimum reaction conditions and other analytical parameters are evaluated. No interference was observed from the additives and the applicability of the methods was tested by analyzing the pharmaceutical preparations containing the investigated drugs. Statistical comparison of the results with those of official methods shows excellent agreement and indicates no significant difference in precision.

Concrete strength represents by far the most critical property of concrete. It represents the mechanical properties of concrete. On-site evaluation of concrete strength remains the fundamental challenge in the condition assessment of existing infrastructure. Although standard laboratory methods can be typically used but most of these testing methods are costly and time-consuming. Among the in-situ methods, the “twist-off ” method with very slight damage is genuinely a convenient, fast and also low-cost technique that provides accurate results for engineers. In this study, the twist-offmethod has been used for the assessment of in-situ strength of the 30 concrete structures in Qazvin in Iran. The results showed structures studied had a strength of 45 to 600 kg/cm2 and the average is about 200 kg/cm2. The observed variation is very high, as well as a significant difference between the compressive strength of the columns, and the floors of the buildings that all indicate non-standard concrete mixing and inadequate control over construction. However, according to the past experience and results of the samples, some recommendations in this regard have been suggested.

Introduction: Diagnosis determination of chronic periapical abscess can be achieved by subjective, objective, and other examination such as radiographic and microbiological examination based on clinical finding and differences intensity at the periapical area with healthy bone suspected to be chronic abscess periapical. A microbiological examination performed to obtain the predominant bacteria that involved to determine an adequate medicament to succeed endodontic treatment. This study was aimed to compare intensity abscess lesion segmentation image reconstruction and surrounding bone for determine the diagnosis of periapical abscess more accurately and to determine the characteristic of bacteria colony from a periapical abscess tooth. Methods: The study sampling method was consecutive sampling (6 female) consisted of inclusion criteria patient with a periapical lesion in anterior maxillary that diagnosed based on clinical signs and symptoms. CBCT 3D scan of the maxillofacial region taken from of patients was measured image intensity periapical lesion and surrounding bone using semi-auto segmentation with ITK-SNAP 3.6.0. Samples from the root canals taken using three sequential sterile paper points. The samples were plated, and microorganisms were then isolated and identified by the colony characteristics. Results: There was a significant difference between the mean content of lesion intensity of (n = 6,256.710 ± 81.930 and 636.022 ± 79.981 lesion boundary. The two-tailed p-value = 0.0050, this difference was considered to be very statistically significant. The mean of group lesion minus group surrounding bone equals to 163.715. 95% confidence interval of this difference from 75.542 to 251.888. Conclusions: Demineralisation of periapical bone can be determined by voxel intensity value in CBCT 3D radiograph using semi-auto segmentation ITK SNAP, could be recommended as a quantitative interpretation for detection periapical lesion and chronic periapical abscesses is a poly-microbial disease. Keywords: Chronic periapical abscess, endodontic, microbiology, radiograph CBCT
