The aim of this study was to investigate the use of protein-phenolic based coating made from fermented rice bran on cherry tomatoes (Lycopersicum esculentum). Tests were performed with glycerol 3% (v/v), glycerol with protein-phenolic rice bran extract (5%), glycerol with protein-phenolic extract after 96 hours of fermentation (5%), and a control (without coating). The coated cherry tomatoes were kept at room temperature for 28 days. Mass loss, pH and acidity, total soluble solids, and carotenoids were determined every 96 hours. The coating made from the biomass extract reduced the carotenoid and acidity levels in the fruits studied by 17 and 21.1%, respectively, compared to the control. The coating proved an efficient barrier to water vapor with mass loss of 57% less than the control suggesting that it can be used as an alternative for vegetable tissue conservation.

The plum is a fruit prized for its nutraceutical properties because of its high content of fiber and sorbitol, which aid in digestion; furthermore, it is characterized as containing antioxidant pigments and an antiseptic action with anthocyanins. These fruits are classified as climacteric and continue the process of respiration and ripening after harvesting; because of this, it is necessary to harvest early so they can withstand transport, which often causes the fruits to not reach the consumption maturity required by consumers. The effect of ethylene on the ripening of plum fruits was evaluated, for which a completely randomized design with 10 treatments was used, which resulted from the combination of two factors plus a control without applications. The first factor was the ethylene dose (100; 1,000 and 2,000 mg L-1) and the second factor was the length of exposure to the ethylene (5, 10 and 15 minutes). The fruits were stored at room temperature for 9 days. In the fruits treated with ethylene, a significant increase in the respiration index and total soluble solids was observed. Additionally, greater total titratable acids and firmness values were obtained with the control treatment than with the ethylene; similarly, the ethylene application increased the fresh mass loss in the plum fruits. Overall, differences between the fruits treated with ethylene and the control were observed, but not between the doses, indicating that the lowest ethylene dose (100 mg L-1) can obtain the desired uniformity in plum fruits.

Fast ripening of fruits is one of the main hindrances to peach preservation. Even though the use of cold storage has extended commercialization periods, fruit quality is affected during storage. This study aimed at evaluating the influence of application of salicylic acid (SA) during cold storage, followed by commercialization simulation, regarding physical and chemical characteristics of ‘BRS Kampai’ peaches, which were harvested in the 2017 crop. The experiment was conducted in a completely randomized factorial design (4×3: SA concentrations × storage days), with four replications of ten fruits each. SA was applied at the following concentrations: 0, 2, 4 and 6 mM. Afterwards, fruits were submitted to cold storage at 1±0.5ºC and 80-85% relative humidity. Analyses were carried out on harvest day, on the 10th cold storage day, followed by a day at room temperature (20ºC) and on the 20th cold storage day, followed by a day at room temperature. Fruits were evaluated in terms of their mass loss, epidermis color, chromatic hue, pulp firmness, soluble solids (SS), pH and titratable acidity (TA). Both application of SA and cold storage contributed to decrease mass loss and TA, besides increasing the SS/TA relation. Fruits which were stored for 20 days and kept at room temperature for a day exhibited the highest values of SS and the lowest values of both brightness and pulp firmness. SA was efficient to maintain the quality of ‘BRS Kampai’ peaches submitted to cold storage for 10 and 20 days and an extra day of commercialization simulation.

The market demand for corn starch and cassava continues to increase because of their use in edible applications, their biodegradable nature, and other appealing properties. As a result, there is a need to identify alternative starch sources, for example, the seeds of S. burchelli, with the potential to be modified for use in post-harvest applications. Therefore, this study aimed to develop and characterize edible films based on the starch phosphates of the seeds of S. burchelli, with the specific aim to apply these starches to cherry tomatoes for post-harvest conservation. After extraction, the starch was phosphorylated with sodium tripolyphosphate (STP) in different concentrations and times according to a 2 x 2 factorial design with additional treatment (native starch). After modification, the starch phosphates were selected for the preparation of edible films using glycerol as a plasticizer, in proportions of 5, 10, 15 and 20% for each selected starch. The films were measured for thickness, permeability to water vapor and solubility in water. According to their permeability values, 4 films were selected for application in the coverage of cherry tomatoes. The conservation of cherry tomatoes with and without coverage was studied over 8 evaluation times (up to 21 days) at 10±2 °C and 80±5% relative humidity. The weight loss, soluble solids, titratable acidity, maturation index, and firmness were measured every 3 days during storage. The starch phosphates showed a phosphorus content within that established by standards, such that the resulting films are acceptable for use in food for human consumption. The edible films presented with an acceptable appearance and without the development of cracks. The concentration of glycerol and the type of starch influenced the characteristics of the films, increasing the permeability and reducing the water solubility of the various edible films. The best result obtained regarding the conservation of cherry tomatoes was observed for fruits with edible coatings of greater permeability to water vapor; such fruits exhibited only a slight decline in firmness during storage compared with the control. It can be concluded that the modification performed on the starch and the glycerol concentration positively influenced the properties of the edible films, allowing for optimal use in post-harvest applications. Similarly, the edible coatings conferred better appearance to the fruits, maintaining their maturation index and firmness during the experimental period and providing better post-harvest storage conditions for the cherry tomatoes.

In order to observe room temperature hydrated specimens in an electron microscope, the following conditions should be satisfied: The specimen should be surrounded by water vapor as close as possible to the equilibrium vapor pressure corresponding to the temperature of the specimen.The specimen grid should be inserted, focused and photo graphed in the shortest possible time in order to minimize dehydration.The full area of the specimen grid should be visible in order to minimize the number of changes of specimen required.There should be no pressure gradient across the grid so that specimens can be straddled across holes.Leakage of water vapor to the column should be minimized.

In cotton seeds the radicle has 12% moisture content which makes it possible to prepare freeze-fracture replicas without fixation or cryoprotection. For this study we have examined replicas of unfixed radicle tissue fractured at room temperature to obtain data on organelle and membrane structure.Excised radicles from seeds of cotton (Gossyplum hirsutum L. M-8) were fractured at room temperature along the longitudinal axis. The fracture was initiated by spliting the basal end of the excised radicle with a razor. This procedure produced a fracture through the tissue along an unknown fracture plane. The warm fractured radicle halves were placed on a thin film of 100% glycerol on a flat brass cap with fracture surface up. The cap was rapidly plunged into liquid nitrogen and transferred to a freeze- etch unit. The sample was etched for 3 min at -95°C to remove any condensed water vapor and then cooled to -150°C for platinum/carbon evaporation.
