Seed quality may be affected by several factors, including permeability, color, and lignin content in the seed coat. This study aimed at evaluating influence of lignin content in the tegument of seed samples of six different soybean cultivars, in which half of each sample was inoculated with the fungus Aspergillus flavus, on the physical and physiological quality, and on the seed health, during 180 days storage period, under cold chamber with controlled conditions of temperature and RH. For that, at each interval of 60 days, samples were removed, and the physiological quality of these seeds was assessed by means of moisture and lignin contents; and by tests of seed health, germination, and electrical conductivity. The moisture content of seeds remained constant during all storage period. In the seed health test, it was found that inoculation was efficient, once the minimum incidence of the fungus in the inoculated seeds was 85%. In the germination test, there was a trend of reduction on percentage germination with the increase in storage period. However, there was an increase on electrical conductivity of seeds assessed. It was concluded that there is no interference of the lignin content in the seed coat on the resistance to infection by the fungus Aspergillus flavus, even after seed storage for a period of 180 days.

Seed coat tear is frequently occurs in some soybean cultivars. The impact of seed coat tear on seed physiology and sanitation is uncertain. Our objective was to analyze the physiological and pathological effect of tear on soybean (Glycine max) seed coat. The cultivars NS-8338-IPRO (with high incidence of tear) and NS-6906-IPRO (with low incidence of tear) were used. A tetrazolium test was used to assess the physiological quality of the seed coat before and after storage. The sanitary quality was assessed through the "Blotter Test". For each storage period, we evaluated seeds with 0% coat tear, up to 10%, and above 10%. The experiment was performed in a completely randomized design using the methodologies proposed by RAS. The coat tear on soybean seed positively contributed to the moisture damage affecting the inner part of the seed. Incidences of Fusarium sp. and Aspergillus sp. were frequently observed in torn seeds and in seeds without tear (around 9-10%) but did not interfere with seed quality. The appearance of coat tear on soybean seed is increased by moisture damage and do not serve as a gateway for the fungi to cause damage during seed emergence.

Abstract: The aim of this study was to evaluate changes in the physiological quality of soybean seeds throughout a storage period in two environments, and to evaluate how these changes are related to metabolites of the phenylpropanoid pathway. A completely randomized experimental design was used in a 4 × 8 factorial arrangement (storage periods × cultivars) with four replications. We used seeds from cultivars with contrasts in lignin content, color, and presence of anthocyanin in the seed coat, and isoflavone content in the seed. The seeds were kept for six months under cold and dry storage and in a non-controlled environment. Germination, first germination count, seedling emergence, and tetrazolium testing were performed on the seeds every two months. The physiological quality of the seeds declines during the storage period, with higher rates of decrease in the non-controlled environment. The metabolites of the phenylpropanoid pathway that were studied, especially lignin, affect seed storage potential. Cultivars with higher lignin content show greater storage potential, especially in a non-controlled environment.

The aim of this study was to evaluate the influence of pod wall permeability on the physiological quality of soybean seed. The cultivars studied were Sant'Ana, FT-2, FT-10, Bossier, Davis and the breeding line F 84-7-30, with a black seed coat. Pods were collected from plants at the R4, R5, R6, R7 and R8 development stages, which composed the treatments in regard to time of harvest. The parameters of permeability and the lignin content of the pods and the seeds within the pods were evaluated. The seeds were collected just after full maturity (R8), and the following tests were performed: germination, electrical conductivity, and tetrazolium, which determined seed viability and vigor. A randomized complete block design in a split-plot in time arrangement was used, with four replications per treatment. The soybean genotypes (six) composed the plots, and the split-plots consisted of the development stages (R4, R5, R6, R7 and R8). In seed evaluation, the same design was used, reducing the number of treatments to three in the split-plots (R6, R7 and R8). Pod permeability varied with the genotype and stage of development; this affected seed vigor, but not the viability of newly-harvested seeds. The pod lignin content did not show any influence on pod permeability.

Breeding soybean for high seed quality is an important approach for developing cultivars for tropical regions, and the lignin content in the seed coat is one of the screening parameters for this trait. Considering that many breeding lines are evaluated in each growing season using the presently recommended method for lignin determination, a long period is required for the evaluation of the whole breeding program. This time limitation may influence lignin content assessment, if lignin is degraded during storage. This research reported was designed to determine whether lignin was degraded in the seed coat of soybean seed cultivars stored for one year in a controlled environment (10°C temperature and 50% air relative humidity). Seeds of 12 selected soybean cultivars that had a range in seed coat lignin content were evaluated. Seeds were hand harvested just after physiological maturity and evaluated for seed coat lignin content at harvest and after one year of storage in a cold room (10°C and 50% RH). The lignin content in seed coats differed significantly among cultivars in both analyses, but for both results the sequence of cultivar classification and the lignin content values of each cultivar did not change. A regression analysis of lignin content at harvest and after one year of storage indicated a direct relationship between both lignin determinations suggesting no differences between the lignin content of each cultivar due to prolonged storage (r² = 0.98***). This indicates that the lignin determination in the soybean seed coat can be performed over a long time period without any bias due to change in its content.

Increased productivity of cocoa needs high quality of cocoa seeds which are generally provided by certified seed gardens located far from smallholders farm, where seed delivery takes long time and may reduce physiological quality of seeds. One effort to maintain the seed quality is by treatment the seeds with polyethylene glycol6000 (PEG 6000). This study was aimed to study the interaction of PEG 6000 concentration and storage period on cocoa seed quality, and to determine the best concentration of PEG 6000 and storage period to maintain cocoa seed quality. The research was conducted in seed storage room, seed laboratory and green houseof PPPPTK, Cianjur. The experimental design used was a factorial completely randomized design and each combination treatment repeated three times. The first factor was concentration of PEG 6000 of 0%, 20%, 40%, and 60%, and the second factor was the storage period of 3 weeks, 6 weeks, 9 weeks, and 12 weeks. The results showed that concentration of PEG 60% with three weeks storage period was able to minimize the number of moldy seeds. The concentration of PEG 20%, 40%, and 60% were able to prevent seed germination in storage until six weeks. Concentration of PEG 6000 20% was able to maintain moisture content, seed germination, and germination rate.Key words:cocoa seed, storage, recalcitrant, PEG

The electrical conductivity test measures the electrolytes that leach out of seeds when they are immersed in water and this leakage is an indication of seed vigor. The level of standardization reached by the procedures of this test is such that the test is recommended for pea seeds and suggested for other large seeded legumes, including soybean [Glycine max (L.) Merrill]. This study was conducted to contribute to the standardization of this test for soybean seeds by verifying whether the seed storage temperature influences the composition of the leachate from soaked seeds solution. Two soybean seed lots of distinct physiological potential were stored in moisture-proof containers either at constant temperatures of 10ºC and 20ºC or at the temperature of 20ºC during the first seven months of storage followed by a change to 10ºC for the rest of the storage time (nine months). The chemical composition of the soaked water was evaluated every three months from January to October 1998. The highest amount of leakage was observed for potassium, followed by calcium and magnesium, iron and sodium regardless of temperature and storage period. The amount of electrolytes in the soaked water increased as the period of time and the temperature of storage increased. On the other hand the amount of leakage decrease along the time for those seeds stored at 10ºC or transferred from the temperature of 20 to that of 10ºC. The temperature at which soybean seeds remain during storage may affect the amount of electrolytes in the soaked water and consequently the results of the electrical conductivity test.

In consequence of several studies and speculations concerning the issue of RR transgenic soybean after the application of glyphosate, additional scientific investigations became necessary to clarify the actual viability of the product use when applied in different developmental stages of the soybean crop. Therefore, this study was aimed to evaluate the physiological quality as well as seed health quality of RR soybean subjected to application of the herbicide glyphosate in different phonological stages of the transgenic soybean, cultivar CD 219RR. For this, an experiment with a complete block experimental design with treatments randomly distributed within the block, with four replications, was carried out. The assessed treatments were foliar sprayings of glyphosate in three increasing dosages [0 (control); 1,440 g ha-1; and 2,880 g ha-1] of acid equivalent, applied in two crop developmental stages: vegetative (V6) and reproductive (R2). The variables assessed were: germination; first count of germination; fresh and dry mass of seedlings, lengths of seedling and root; vigor and viability by the tetrazolium test; and seed health quality. Glyphosate application may adversely affect physiological quality of RR soybean seeds, when applied in dosages varying from 1,440 to 2,880 g acid equivalent per hectare at the stages V6 and R2.

This study aimed to assess the effect of genetic diversity on physiological quality of soybean seeds stored in cold chamber and under environmental conditions. Ten cultivars were assessed in a randomized factorial design (2x10). Factor 1 corresponded to two storage conditions and factor 2 to ten soybean cultivars, with four replications. The evaluated variables were total germination (G%), first count of germination (F%), percentage of abnormal seedlings (AS%), germination speed index (GSI), water content (WC), electrical conductivity (EC), dry matter of seedlings (DMS) and length of seedlings (LS). Data underwent ANOVA, followed by Scott Knott test, as well as multivariate analysis of genetic diversity. The results showed a higher physiological quality for seeds under cold storage. Half of the cultivars (FPS Júpiter, FPS Urano, FPS Antares, FPS Netuno and CD 250) presented high germination rates and seed vigor, being thus indicated as high-standard materials for further breeding programs. Besides that, storage environment had influence on the clustering of soybean cultivars. Moreover, cultivars had genetic dissimilarity for almost all assessed traits as G%, GSI, F%, AS%, EC, DMS and LS.

Differences in levels of lignin in the plant between conventional and transgenic cultivars RR has been reported by several authors, however, there are few studies evaluating the influence of spraying of glyphosate on the lignin in the plant and RR soybean seeds. The aim of this study was to evaluate the physiological quality of RR transgenic soybean seeds and the lignin contents of plants sprayed with the herbicide glyphosate. The assays were conducted both in greenhouse and field in the municipality of Lavras, MG, in the agricultural year 2007/08. The experiment was arranged in a splitplot design with four replicates, considering the treatments hand weeding and herbicide glyphosate as plots, and five RR soybean cultivars (BRS 245 RR, BRS 247 RR, Valiosa RR, Silvânia RR and Baliza RR) as splitplots. In the greenhouse, the cultivars tested were BRS 245 RR and Valiosa RR in a randomized block design with four replicates. The sprayings were carried out at stages V3, V7 and early R5 (3L/ha). The 1000 seed weight, mechanical injury, germination and germination velocity index, emergence velocity index, accelerated aging, electrical conductivity and water soaking seed test, lignin content in the seed coat, in the stem and legumes were determined. The spraying of glyphosate herbicide, in greenhouse and field, did not alter the physiological quality of seeds and the lignin contents in the plant.

<p>ABSTRACT<br />Information of seed quality during storage can be determined through the actual storage and storability vigor estimation. This study aimed at comparing effective accelerated aging method<br />between physical and chemical, and studying the seed deterioration during storage in ambient (T =28-30 0C, RH=75-78%) and AC (T =18-20 0C, RH =51-60%) condition with three levels of initial moisture content (8-10%, 10-12%, and 12-14%) for 20 weeks. The final objective of this research<br />was to develop model for storability vigor of papaya seed. Two experiments, accelerated aging and seed storage were conducted at Seed Laboratory, Department of Agronomy and Horticulture, Bogor Agricultural University from October 2015 to May 2016. A completely randomized design with nested factors and four replications was applied to both experiments. The results showed that physical accelerated aging using IPB 77-1 MMM machine was more effective than chemical accelerated aging using IPB 77-1 MM machine for papaya seed. The viability of seed stored in AC condition remained high until the end of the storage period, whereas it declined at 16 week storage period in the ambient condition. The viability of seed with initial moisture content of 12-14% declined faster than that of initial moisture content of 8-10% after 18 week storage periode. The model used to estimate the storability vigor of papaya seed accurately was the equation y = a + b expcx where y : storability vigor estimation, x : aging time and a,b,c : constant value. Penelitian ini bertujuan untuk membandingkan metode<br />pengusangan cepat yang efektif antara fisik dengan kimia serta mempelajari pola penurunan viabilitas benih selama penyimpanan aktual pada kondisi simpan kamar (suhu =28-30 0C, RH =75-78%) dan AC (suhu =18-20 0C, RH =51-60%) dengan tiga tingkat kadar air awal (8-10%, 10-12%, dan 12-14%) selama 20 minggu. Tujuan akhirnya adalah membangun model vigor daya simpan benih pepaya. Penelitian pengusangan cepat dan penyimpanan dilakukan pada bulan Oktober 2015 sampai Mei 2016 di Laboratorium Benih, Departemen Agronomi dan Hortikultura, Institut Pertanian<br />Bogor. Kedua penelitian menggunakan rancangan acak lengkap tersarang dengan empat ulangan. Hasil penelitian menunjukkan bahwa pengusangan cepat secara fisik dengan alat IPB 77-1 MMM lebih efektif daripada pengusangan kimia dengan alat IPB 77-1 MM untuk benih pepaya. Viabilitas benih yang disimpan pada kondisi AC tetap tinggi hingga akhir periode simpan, sedangkan pada kondisi kamar penurunan viabilitas dimulai pada periode simpan 16 minggu. Benih yang disimpan dengan tingkat KA awal sebesar 12-14% lebih cepat mengalami penurunan viabilitas mulai periode simpan 18 minggu dibandingkan dengan benih dengan KA awal 8-10%. Hasil penelitian juga menunjukkan terdapat korelasi yang erat antara pola kemunduran benih pada pengusangan cepat dan penyimpanan aktual, sehingga model pendugaan vigor daya simpan (y) berdasarkan waktu pengusangan (x) dapat disusun dengan persamaan y = a + b expcx.
