Respiratory problems in cats have a multifactorial character. Therapy without the detection of pathogen is often ineffective. Our study was therefore focused on the detection of important feline respiratory bacterial pathogens such as Mycoplasma felis, Chlamydia felis and Bordetella bronchiseptica and viral pathogens such as Felid alphaherpesvirus-1 and feline calicivirus. The goal of this study was to map the occurrence of these pathogens in cat populations in the Czech Republic with the aim of introducing rapid and highly sensitive methods into routine diagnostics and to provide consulting services to animal health professionals based on the acquired data. A total of 218 cats were investigated in the study: 69 were outdoor and 149 were indoor cats. Three groups of animals were compared: up to one year of age (60 cats), one to three years of age (68 cats) and more than three years of age (90 cats). Samples were taken from conjunctiva and/or the oropharynx. Samples originated from cats with various forms of respiratory disease or from healthy cats from different parts of the Czech Republic. Real-Time RT-PCR, multiplex Real-Time PCR, nested PCR and sequencing analyses were performed. Outdoor cats were infected more often (84 detected pathogens in 69 cats) than indoor cats (110 detected infections in 149 cats). More than one pathogen was detected in a total of 38 cats, and six cats were infected with more than two pathogens. The difference was statistically significant in the case of co-infections, but not for mono-infections (P < 0.05). Kittens and young adults up to the age of one year were the most common reservoirs of respiratory infections (only 19 cats out of 60 were negative and positive cats often harboured coinfections). The difference in age groups were not statistically significant (P > 0.05). Concerning the site of the sampling, feline calicivirus, M. felis and B. bronchiseptica were detected more often from oropharynx than from conjunctival swabs. M. felis was slightly more common in clinically diseased animals (39.6%) than in healthy ones (26.1%). The obtained results reveal the frequency of individual pathogens and their co-infections in cats kept on the territory of the Czech Republic, data which can be used to make the treatment of respiratory infections and breeding measures more effective. Therefore, the diagnostic methods are now available to veterinary surgeons with the possibility of consultation and discussion of the results.

Unemployment affects individuals socially, personally, and economically. The impact of being jobless can be long-lasting. Five different generations participate in the workplace today. As countries throughout the world went into lockdown to combat the spread of Covid-19, unemployment numbers rose rapidly. This study aimed to examine the effects of unemployment in three-generation groups in V4 and Austria in the pre-Covid-19 era and during Covid-19. Descriptive statistics were used to present the collected data. OECD data were used for the analysis. Based on the data collected, unemployment decreases with age. The group aged 15-24 shows significantly higher unemployment than the other two groups. A gender difference in unemployment was confirmed only in the Czech Republic and Slovakia. Unemployment has increased during the Covid-19 pandemic. The unemployment gap for females before and during Covid-19 was not confirmed. The T-Test confirmed the difference in unemployment before and during the crisis in the age categories 15-24 and 25-54. In Hungary, Poland, and Slovakia, the number of people with a duration of employment of up to one year differs in all age categories. In the Czech Republic, there is a significant difference only between the youngest group and the other two. In all countries, the largest number of people with the employment of up to one year is in the age group 25-54. In none of the examined countries was a gender unemployment gap proved before Covid-19.

Outbreaks of the three capripox virus species, namely lumpy skin disease virus, sheeppox virus, and goatpox virus, severely affect animal health and both national and international economies. Therefore, the World Organization for Animal Health (OIE) classified them as notifiable diseases. Until now, discrimination of capripox virus species was possible by using different conventional PCR protocols. However, more sophisticated probe-based real-time qPCR systems addressing this issue are, to our knowledge, still missing. In the present study, we developed several duplex qPCR assays consisting of different types of fluorescence-labelled probes that are highly sensitive and show a high analytical specificity. Finally, our assays were combined with already published diagnostic methods to a diagnostic workflow that enables time-saving, reliable, and robust detection, differentiation, and characterization of capripox virus isolates.

Abstract The companies engaged in brown coal mining are looking for ways of managing the mining process as efficiently as possible. The principal mining technology used for brown coal mining in our country, but also in Germany and Poland, are wheel excavators. The evolving GNSS technology has enabled designing and realisation of systems for determining the spatial position of the excavator wheel. The visualisation of the wheel's spatial position and tracking of its real-time motion is performed in the Czech Republic by the program Mine Model developed by the company KVASoftware. One of the most important tasks of mine surveyors is the calculation of the volumes of the extracted masses. The described system performs this task in real-time. This article describes an application that is used to automate volume calculations.

Aim: To evaluate the occurrence of melanoma in the period 1996–2017 in East Bohemia region in the Czech Republic. Method: We studied the incidence of melanoma and the age of diagnosis (adjusted calculation) and the parameters such as histology, body site of lesions, the length of the duration of lesions in 2810 patients. Results and conclusion: No change in the occurrence of melanoma and in age of melanoma during this period was found. The difference between men and women was not confirmed in histology, but the difference between men and women was confirmed in the body site of lesion and in the length of duration of lesion. No relation between the length of duration of lesions from which melanoma had originated and its histology was confirmed. The relation was confirmed between histology and body site of melanoma. The relation between the body site and the length of duration of previous lesions was confirmed also. The increasing occurrence of melanoma on the trunk according to the duration of the previous lesions was confirmed.

The Common Agricultural Policy has been implemented in order to guarantee the appropriate life quality for farmers and to preserve the European heritage. Costs of its realization amounted to 40% of the EU budget. The EU has not established the same conditions for all member states. The aim of the paper is to assess the influence of agricultural subsidies and the structure of production on the incomes of agricultural holdings and their comparison with the largest producers in the EU with similar production structure. The shift-share analysis is used. Different amount of subsidies according to the type of farming together with increasing subsidy rate may influence the type of farming. Therefore, it may cause a paradox that the structure of subsidies according to the type of farming will stimulate products that are currently suppressed. The difference in subsidies in comparison with the largest producers with a similar structure of agricultural production is significant for the Czech Republic and it is possible to compare it to the increase of the SAPS by 75%.

Abstract In polygynous mammals, females are expected to bias maternal investment in favor of male calves. The mother should invest more in males to enhance their reproductive success in adulthood, or the males require greater investment as they are bigger and stronger than females. In this study, we used nursing duration to compare the difference in the amount of maternal investment provided by females. We compared differences according to sex of the offspring and the influence of calves’ identification by sniffing, using captive giraffes (Giraffa camelopardalis) as a model. Since a high intensity of allonursing (nursing of a nonfilial calf) was reported in giraffes, we also focused on the difference between filial and nonfilial calves. We observed 22 nursing females and 47 suckling calves in four zoological gardens in the Czech Republic from 2007 to 2011. Nursing duration was longer for male calves than for female calves and for calves sniffed by the nursing female regardless whether the calves were filial or nonfilial. We conclude that male calves are more demanding for the amount of investment received and they are more successful in this effort than female calves. Since females provided investment in the same way to filial and nonfilial calves, selection for higher demand for investment by male than by female offspring should be important for their future development. Our results also demonstrate the importance of sniffing for the identification of the suckling calf by the female.

A survey to estimate the prevalence of verotoxigenic E. coli (VTEC) or enterohaemorrhagic E. coli (EHEC) in rectal swabs from healthy dairy cattle aged three weeks, three months and one year was conducted in three herds from the Czech Republic. Screening for the presence of the stx1, stx2 and eaeA genes in faecal swab cultures was performed by PCR, and in positive samples, isolated colonies were examined. Immunomagnetic separation was used for the isolation of the VTEC serogroup O157 from samples. VTEC were detected in animals from all three herds under study. In the group of 3-week-old calves, VTEC were only detected in samples collected in the summer months. However, in the other age-groups, VTEC were detected in both the summer and winter months. EHEC shedding was observed in 30 to 100% of the total samples collected from cattle aged three months and one year in the summer months, and in 30 to 60% of samples taken in the winter months. EHEC strains of serogroup O157 were detected in two herds. The range of verotoxins shed by VTEC isolates of serogroup O157 differed between herds. Besides serogroup O157, additional EHEC belonging to the antigen groups O26, O103, O128 and O153 have been identified, and in some of them, no somatic antigen was detected.

Lamb growth performance traits in relation to parental breeding values (BVs) for these traits were evaluated in a purebred Suffolk sheep population in the Czech Republic. The research lasted over 8 years and included 24 886 lambs. Four relevant parental BVs were observed: BV predicted for lamb live weight direct effect (BVLW-DE), BV predicted for lamb live weight maternal effect (BVLW-ME), BV predicted for lamb musculus longissimus lumborum et thoracis depth (BV-MLLT), and BV predicted for lamb backfat thickness (BV-BT). The lamb live weight (LW; kg), musculus longissimus lumborum et thoracis depth (MLLT; mm), and backfat thickness (BT; mm) were assessed at 100 days of age. A dataset was created using the most current parental BVs for each year (2007–2014) and subsequent growth traits of their lambs in the next season (2008–2015). Linear regressions showed an increased tendency when one point in dam BVs was associated with an increase in lamb LW (0.393 kg; P < 0.01 in BVLW-DE and 0.090 kg; P < 0.05 in BVLW-ME), MLLT (0.340 mm; P < 0.01 in BV-MLLT), or BT (0.243; P < 0.01 mm in BV-BT). Lower (but significant – P < 0.01) values on linear regression were detected for sire BVs, when 0.135 kg of LW, 0.217 mm of MLLT, and 0.214 mm of BT corresponded to 1-point increases of BVLW-DE, BV-MLLT, or BV-BT. This was confirmed by ANOVA evaluation, especially for LW and MLLT traits. Maximal differences (P < 0.05) in lamb LW were 1.84 kg or 0.88 kg regarding to dam or sire BVLW-DE groups. Similarly, the difference (P < 0.05) in lamb MLLT reached 0.82 mm in dam BV-MLLT, while 0.57 mm was detected in sire BV-MLLT groups. These results have practical implications for the objectives of selection schemes used in the Suffolk sheep population in the Czech Republic.
