Potyviruses are the largest genus of plant viruses that cause significant losses over a wide range of crops. In this paper, the presence of potyvirus in different plant crops in the provinces of Ocaña and Pamplona located in the north and south of the Department of Norte de Santander (Colombia) was evaluated with RT-PCR analysis using universal oligonucleotides specific to the region that encodes the NIB protein. The results indicate the presence of several potyvirus in Pamplona in economically important crops such as corn (Zea mays), tomato (Solanum lycopersicum), potato (Solanum tuberosum) and zucchini (Cucurbita pepo). In Ocaña, potyvirus was found in bean (Phaseolus vulgaris), corn and pumpkin (Cucurbita maxima). In corn, one of the most important crops, the presence of the Sugarcane mosaic virus (SCMV) was confirmed with nucleotide sequencing. This is the first report of this virus in the department. The presence of several potyviruses in different crops in Norte de Santander indicate an alarming phytosanitary condition that must be addressed with priority to establish detection and control systems that maximize production, ensure agricultural sustainability, and propose certification schemes and improvement programs to reduce economic losses.

Food safety is very important in the food industry as most pathogenic bacteria can cause food-borne diseases and negatively affect public health. In the milk industry, contamination with Salmonella has always been a challenge, but the risks have dramatically increased as almost all bacteria now show resistance to a wide range of commercial antibiotics. This study aimed to isolate a bacteriophage to be used as a bactericidal agent against Salmonella in milk and dairy products. Here, phage ZCSE6 has been isolated from raw milk sample sand molecularly and chemically characterized. At different multiplicities of infection (MOIs) of 0.1, 0.01, and 0.001, the phage–Salmonella interaction was studied for 6 h at 37 °C and 24 h at 8 °C. In addition, ZCSE6 was tested against Salmonella contamination in milk to examine its lytic activity for 3 h at 37 °C. The results showed that ZCSE6 has a small genome size (<48.5 kbp) and belongs to the Siphovirus family. Phage ZCSE6 revealed a high thermal and pH stability at various conditions that mimic milk manufacturing and supply chain conditions. It also demonstrated a significant reduction in Salmonella concentration in media at various MOIs, with higher bacterial eradication at higher MOI. Moreover, it significantly reduced Salmonella growth (MOI 1) in milk, manifesting a 1000-fold decrease in bacteria concentration following 3 h incubation at 37 °C. The results highlighted the strong ability of ZCSE6 to kill Salmonella and control its growth in milk. Thus, ZCSE6 is recommended as a biocontrol agent in milk to limit bacterial growth and increase the milk shelf-life.

The northern root-knot nematode, Meloidogyne hapla, is a biotrophic parasite that infects many crops and causes severe economic losses worldwide. Rapid and accurate detection of M. hapla is crucial for disease forecasting and control. We developed a recombinase polymerase amplification combined with a lateral flow dipstick (RPA-LFD) assay for rapid detection of M. hapla. The primers and a probe were designed based on the effector gene 16D10 sequence and were highly specific to M. hapla. The RPA reaction was performed at a wide range of temperatures from 25 to 45°C within 5 to 25 min, and the amplicon was visualized directly on the LFD within 5 min. The detection limits of the RPA-LFD assay were 10-3 female and 10-2 J2/0.5 g of soil, which was 10 times more sensitive than the conventional PCR assay. In addition, the RPA-LFD assay can detect M. hapla from infested plant roots and soil samples, and the entire detection process can be completed within 1.5 h. These results indicate that the RPA-LFD assay is a simple, rapid, specific, sensitive, and visual method that can be used for rapid detection of M. hapla in the field and in resource-limited conditions.

Drought stress remains one of the most detrimental environmental cues affecting plant growth and survival. In this work, the DNA methylome changes in mulberry leaves under drought stress (EG) and control (CK) and their impact on gene regulation were investigated by MethylRAD sequencing. The results show 138,464 (37.37%) and 56,241 (28.81%) methylation at the CG and CWG sites (W = A or T), respectively, in the mulberry genome between drought stress and control. The distribution of the methylome was prevalent in the intergenic, exonic, intronic and downstream regions of the mulberry plant genome. In addition, we discovered 170 DMGs (129 in CG sites and 41 in CWG sites) and 581 DMS (413 in CG sites and 168 in CWG sites). Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis indicates that phenylpropanoid biosynthesis, spliceosome, amino acid biosynthesis, carbon metabolism, RNA transport, plant hormone, signal transduction pathways, and quorum sensing play a crucial role in mulberry response to drought stress. Furthermore, the qRT-PCR analysis indicates that the selected 23 genes enriched in the KEGG pathways are differentially expressed, and 86.96% of the genes share downregulated methylation and 13.04% share upregulation methylation status, indicating the complex link between DNA methylation and gene regulation. This study serves as fundamentals in discovering the epigenomic status and the pathways that will significantly enhance mulberry breeding for adaptation to a wide range of environments.

Survey on macro fungi was made in Gajni forest, Sherpur, Bangladesh which is located in between 24°18' and 25°18' north latitudes and in between 89°53' and 90°91' east longitudes. It is bounded by Meghalaya state of India on the north, Mymensingh and Jamalpur districts on the south with a wide range of ecosystem. The survey was conducted on July to December, 2018 to identify and preserve wood-rot causal macro fungi for future industrial utilization. Morphology of basidiocarp and characteristics of basidiospore were recorded. A total of 20 samples were collected and identified to 12 species belonging 7 families. Dominant species was Ganoderma species. The identified four species were from Ganodermataceae family and these were G. applanatum, G. lucidum, G. tropicum and G. lobetum. Other dominant genus was Russula. Other recorded genera were Hebeloma, Boletus, Phlebopus and Entoloma. Among them the highest frequency (85.72%) was recorded for G. applanatum and lowest frequency (7.14%) was recorded for Phlebopus marginatus. Similarly highest density (20.25%) was recorded for Agaricus sp. followed by G. lucidum (15.85%). The lowest density was (2.14%) was recorded for Phlebopus marginatus. Collected specimens were preserved in Sher-e-Bangla Agricultural University Herberium of Macro fungi (SHMF) for further study.

&lt;p&gt;The characterization of the basement architecture of the Pyrenean Axial Zone, backbone of the chain, is crucial to understand its geodynamic evolution and the interplay between tectonism and magmatism. In this work, a new gravity-constrained cross section was built along the Central Pyrenees, between two of the largest Pyrenean Late Variscan granitic complexes, La Maladeta and Andorra-Mont Louis granites, to infer the geometry at depth of the basement host rocks. This cross section is ca. 65 km long and extends from the Mesozoic B&amp;#243;ixols basin in the South to the Late Variscan Bassi&amp;#232;s granite to the North, close to the northern end of the Axial Zone. It is based on available geological maps, previous published works and new geological field data; together with newly acquired gravimetric stations (1141), to improve the existent spatial resolution of the gravity data from the databases of the Spanish and Catalan Geological Surveys, and density values from 65 rock samples covering all different lithologies in the cross section. Thus, its geometry at depth is constrained by means of an integrated 2.5D gravity/structural/petrophysical modelling.&lt;/p&gt;&lt;p&gt;The La Maladeta and Andorra-Mont Louis granites appear aligned in a WNW-ESE direction and both lie within the same Alpine basement unit, the Orri thrust sheet. They are separated about 40 km by the WNW-ESE-oriented Llavors&amp;#237; syncline, formed by Devonian and Silurian rocks and limited to the north and south by south vergent thrusts. This syncline is located between two large Cambro-Ordovician anticlinorium structures, the La Pallaresa and Orri massifs to the north and south respectively, formed by a monotonous alternation of shales and sandstones with some intercalations of limestones and conglomerates affected by very low to medium grade of metamorphism. Most structures show southern vergence along the cross section, and its southern part is characterized by the occurrence of Triassic evaporites, a significant detachment level decoupling deformation between the Paleozoic basement and the Mesozoic-Cenozoic cover rocks.&lt;/p&gt;&lt;p&gt;The observed residual anomaly along the cross section shows a relative maximum, coinciding with the southern edge of the Axial Zone (Nogueras Zone) and southern half of the Orri massif, followed to the north by a relative large minimum. This gravity minimum in the core of the Axial Zone coincides with the northern half of the Orri massif, the Llavors&amp;#237; syncline and southern half of the La Pallaresa massif and must be related at depth with rocks of lower density with respect to rocks located to the North and South. Two possible solutions have been postulated to explain the presence of lower density rocks: (i) the presence of Triassic evaporites at depth as a continuation to the North of the Triassic evaporites outcropping in the Rialp window located to the South and/or (ii) the presence of buried granitic bodies equivalent to the adjacent La Maladeta and Andorra-Mont Louis granites.&lt;/p&gt;

Abstract Background: Pasteurella multocida is one of the important pathogens that infect rabbits, causing major economic losses in commercial rabbit farming. In this study, 205 P. multocida isolates recovered from lungs of dead rabbits with respiratory disease were defined by capsular serogroups, lipopolysaccharide (LPS) genotypes and multi-locus sequence types, screened virulence factors and antimicrobial susceptibility. Results: The 205 isolates were assigned into 2 capsular types, A and D, and 2 LPS genotypes, L3 and L6. When combining capsular types with LPS genotypes, 4 serotypes were detected. A:L3 (51.22%, 105/205) was the most predominant serotype, followed by A:L6 (24.88%, 51/205), D:L6 (19.02%, 39/205) and D:L3 (4.88%, 10/205). The 205 isolates were grouped into 3 sequence types, ST10, ST11 and ST12. ST12 (56.10%, 115/205) was the most prevalent sequence type, followed by ST10 (24.88%, 51/205) and ST11 (19.02%, 39/205). In the 205 isolates, virulence associated genes ptfA , fur , hgbB , ompA , ompH and oma87 were positive in the PCR screening, whereas the toxA and tbpA genes were negative. Notably, the 156 capsular serogroup A isolates carried the pmHAS gene. All the 205 isolates were susceptible to most of the used antibiotics, except for streptomycin, gentamycin, kanamycin and ceftriaxone, and the resistance rates of which were 27.80%, 15.61%, 9.27% and 2.44%, respectively. Conclusions: This study, for the first time, described the prevalence and characteristics of P. multocida causing respiratory disease in rabbits in Fujian Province, which might be useful for tracking the epidemic strains and development of efficient vaccines and methods to prevent and control the pathogen.

This chapter re-examines the powerful nomadic-equestrian societies in the North and South American grasslands and makes the case for a distinctive imperial formation, the kinetic empire. Kinetic empires were shape-shifting, action-based regimes that turned mobility into an imperial strategy and revolved around mobile activities: long-distance raiding, seasonal expansions, transnational diplomatic missions, semi-permanent trade fairs, recurring political assemblies, and control over shifting nodes. Their governing systems were light and flexible, and they rose and stayed in power by capitalizing on their superior capacity to access and connect political and economic centers around them. The Comanche and Lakota Indians created the Western Hemisphere’s most prominent examples of kinetic empires in the North American Great Plains. In South America the Araucanians frustrated European colonizing efforts, commanded a large and expanding territory, and pulled other Native groups on their orbit, but lacked the Comanches’ and Lakotas’ strong collective identities and political unity.

Cryptosporidiosis is a cosmopolitan parasitosis that affects a wide range of hosts including birds. As information concerning Cryptosporidium in birds is limited, the present study examined the prevalence and genotypes of Cryptosporidium in Java sparrows in Beijing and Shangqiu, northern China. Three hundred and fifty fecal samples were collected from Java sparrows (Lonchura oryzivora, 225 white Java sparrows and 125 gray Java sparrows) in Beijing and Shangqiu in October 2015, and the samples were examined by PCR amplification of the small subunit ribosomal RNA (SSU rRNA) gene. The overall Cryptosporidium prevalence is 13.42% (47/350), with 16.44% (37/225) in white Java sparrows and 8.00% (10/125) in gray Java sparrows. Cryptosporidium prevalence was 9.82% (16/163) in Java sparrows from Beijing and 16.58% (31/187) in Java sparrows from Shangqiu. The prevalence of Cryptosporidium in females and males was 40.63% (26/64) and 7.34% (21/286), respectively. The Cryptosporidium prevalence in Java sparrows of different ages varied from 10.47% to 16.33%. Sequence analysis of the SSU rRNA gene revealed that all the samples represented C. baileyi. This is the first report of Cryptosporidium in gray Java sparrows in China, which extend the host range for C. baileyi. These results provide baseline information for further studies of molecular epidemiology and control of Cryptosporidium infection in poultry in China.
