The article reflects the scientific fate of the main genre model – the “questionnaire of the speech genre” of T. V. Shmeleva. The author discusses the reasons for the high demand for the “questionnaire” in genre studies: the parameters of the “questionnaire” affect the most important aspects, the “pain points” of the genre; the formal simplicity and consistency of the “questionnaire”. Along with the advantages and achievements (the article gives their brief description), the author points to a few critical issues concerning the “questionnaire”: the lack of items of the “questionnaire” (the juxtaposition of primary and secondary speech genres, direct and indirect speech genres, speech and rhetorical genres, cultural characteristics of speech genres, etc. are not presented in it) and its crucial points which are not commonly recognized, i.e. the ranking points of the “questionnaire”. In relation to modern genre studies, the article deals with the items that either should be or can be, or have already been added to the “questionnaire”, as well as the theoretical justification of their relations, which, according to the author of the article, will allow to clarify some other items of the “questionnaire”. The emphasis is on those points that are less spelled out in the “canonical” “questionnaire”, but that should be used for a more adequate study of genres (especially new ones), and are already actually used. The discussion focuses on the following points: typological, including the stadial-typological and historical-etymological, features of speech genres; determination of the place of the genre in the structural-hierarchical typology; cultural, linguistic and linguocultural characteristics of speech genres; characteristics of the communicative concept, through which the genre is presented and interpreted, and its components; the ability of the genre to form its variants; corpus and quantitative characteristics of speech genre; online genres.

The article defines the methodological problems of constructing a model of the speech genre and suggests the way of solving them. The existing models of Text – Text components – Genre type are recognized as the main tool for the study and description of speech genres. It is argued that the speech genre description methodology in modern linguistics should be focused on the ideas of systematic construction. The principles of text analysis in the aspect of speech genre studies are determined and some errors of scientifically employed approaches are identified. The elimination of these errors will allow genre studies to shift to a new methodological principle of constructing a genre model that has a diagnostic potential. The current problematic issues of genre studies are formulated. As a productive technology for describing a speech genre, we propose a universal method of functional modelling, which will enable designing genres and their modules on the basis of regularity, obligation, and oppositional character. In order to solve the problem of genre description in the attributive aspect, that is, to determine its units, it is productive to analyze the genre applying the so-called communicative semiotic model, as well as the method of transformational-and-oppositional analysis. The proposed principle of genre description is postulated as a method of objective modelling. Thus, within the framework of this study, the analysis of the theoretical problems of modern genre studies is carried out, the objectives of speech genres investigation are formulated and the approaches to their achieving are presented in an overview.

The article touches upon current problems of speech genres functioning on the Internet and fits into the context of modern research in the field of virtual genre studies. The article discusses the speech genre of the movie announcement and describes the means of its language embodiment. The research is focused on the analysis of clichéd constructions and is based on 250 Indian film announcements written in Russian and posted on Indiankino.net. The author offers a descriptive model of the movie announcement speech genre which includes the following parameters: the communicative goal and type of speech genre, the image of the author and the addressee, the image of the future and the image of the past, the composition of movie announcement, the type of dictum content, means of its linguistic embodiment, non-verbal means. There is a special focus on the use of clichéd constructions in all compositional parts of the analyzed texts (name of the film, exposition, plot of the film, denouement). The research has shown that clichés function in every compositional element. The author singles out the main functions of clichéd constructions in the speech genre of the movie announcement: persuading function, advertising function, as well as text-building and genre-forming functions.

Thiols play vital and irreplaceable roles in the biological system. Abnormality of thiol levels has been linked with various diseases and biological disorders. Thiols are known to distribute unevenly and change dynamically in the biological system. Methods that can determine thiols’ concentration and distribution in live cells are in high demand. In the last two decades, fluorescent probes have emerged as a powerful tool for achieving that goal for the simplicity, high sensitivity, and capability of visualizing the analytes in live cells in a non-invasive way. They also enable the determination of intracellular distribution and dynamitic movement of thiols in the intact native environments. This review focuses on some of the major strategies/mechanisms being used for detecting GSH, Cys/Hcy, and other thiols in live cells via fluorescent probes, and how they are applied at the cellular and subcellular levels. The sensing mechanisms (for GSH and Cys/Hcy) and bio-applications of the probes are illustrated followed by a summary of probes for selectively detecting cellular and subcellular thiols.

The sikness rate of children living in the territories of the city with various levels of anthropogenic load is studied. The informativity of the multifactor dispersed analysis for determination of quantitative characteristics of the effect of environmental factors on the population sickness rate is noted. The effect of six inassociated factors (dust, sulphur dioxide etc.) on the sickness rate of children aged 4 to 7 is studied by this method.

Clustered regularly interspaced short palindromic repeats (CRISPR) and CRISPR-associated protein (Cas) system facilitates targeted genome editing in organisms. Despite high demand of this system, finding a reliable tool for the determination of specific target sites in large genomic data remained challenging. Here, we report SSFinder, a python script to perform high throughput detection of specific target sites in large nucleotide datasets. The SSFinder is a user-friendly tool, compatible with Windows, Mac OS, and Linux operating systems, and freely available online.

The following paper deals with the application of methodology of pedagogical situations cognitive modeling, which is considered by the authors as a process consisting of six consecutive and interconnected stages. The first stage is a formulation of the purpose and the corresponding tasks. The second stage provides collecting, systematization and analysis of a pedagogical situation with the subsequent allocation of the major factors influencing development of the situation and determination of interrelation between them, i.e. creation of a cognitive map. At the third stage a focused count is created as a result of accounting of the cause and effect chains reflecting the system of interaction between the educational process subjects and allowing to form a pedagogical theory on the basis of basic person study categories: consciousness, thinking, knowledge, understanding, etc. The fourth stage assumes combination of the cognitive map and the focused count in a uniform cognitive model of the studied pedagogical situation. The fifth stage is focused on a real pedagogical situation cognitive model adequacy check i.e. on its verification. The last sixth stage allows to define possible options of a pedagogical situation development by a cognitive model, to find ways and mechanisms of a situation impact.

The article dealt with the assessment of the quality of hydraulic oil and determination of the mode of wear of the friction surfaces of Baljer & Zembrod manipulating lines through the information traces in the oils by applying tribotechnical diagnostics. We presented the assessment of the level of degradation of the oils. In addition, we presented the mode of wear of the friction surfaces washed in oil through evaluation of the qualitative and quantitative characteristics of the particles found in the oil. In detail, we focused on the application of suitable multivariate statistical methods on the data matrix. The article also presents predictive models that can sort oils into groups based on the assessment of quality of the oil and the state of the friction couples. The models can be used in research and in solving practical tasks in tribotechnical diagnostics of hydraulic fluids in woodworking equipment. Our results showed that the manipulation lines were greatly thermically stressed due to inadequate oil and machine maintenance. By correlative integration of all methods used, we could determine the real mode of the wear of the tribologic nodes of the machine. The experiment enabled the early detection of an undesirable process in the tribological node and implementation of corrective measures before the machine would break down.

The article deals with the problem of the relationship between two linguistic theories: a speech genre theory and a speech act theory. The research is based on the genre of a contact excursion. From the point of view of genre studies a contact excursion is a speech genre and it can be described using the “speech genre questionnaire” proposed by T. V. Shmeleva, which includes the communicative goal of the genre, the concept of the author, the concept of the addressee, the dictum content, the image of the communicative past, the image of the communicative future, and the language embodiment. From the point of view of functional and structural classification, a contact excursion is a complex genre or a hypergenre, consisting of a number of simple genres. Within the framework of the actional or the activity approach, which is based on the speech act theory, a contact excursion can be considered as a communicative event and described using speech strategies and tactics which represent one or more speech actions, which traces to the concept of a speech act. The notion of the communicative style serves as the basis to reconcile and combine both theories.

The article presents the results of studies of changes in the vitamin composition of pumpkin, grown in the Kuban region. Standard and modern methods of physical and chemical analysis were used in the research. It is shown that the nutritional value of this raw material allows to maintain its nutritional value for a long time. Differential determination of carotenoid pigments contained in pumpkin fruits was carried out. The quantitative characteristics of carotenoids were established and the predominant carotenoid pigments for the studied pumpkin varieties were specified.
