Effect of Biofilm of Aeromonas Hydrophila Oral Vaccine on Growth Performance and Histopathological Changes in Various Tissues of Striped Catfish, Pangasianodon Hypophthalmus (Sauvage 1878)

Biofilm of Aeromonas hydrophila oral vaccine was evaluated on growth and histopathological features of striped catfish, Pangasianodon hypophthalmus. Fish were fed with feed incorporating biofilm (BF) or free cell (FC) of A. hydrophila vaccine at 1010 cells/fish/day for 20 days and basal feed for 60 days. From 30th day onwards, till end of the trial, the mean weight gain (g/fish) was significantly higher (P less than 0.05) in BF treated group than both FC and control group. At the end of the study, histopathological changes in gills, liver and kidney were evaluated using haematoxylin eosin technique. The histopathological findings, in BF fed P. hypophthalmus, showed normal architechture of the gills, liver and kidney. However, fishes fed on FC and control group, resulted several histopathological abnormalities in all the organs.

An experiment was conducted to investigate the counteracting effects of spirulina in Long Evans rats exposed to oral potentiated sulfonamide administration. 20 rats were randomly assigned into four equal groups (A, B, C and D) and were fed with standard broiler pellet (25g/rat/day) throughout the experimental period of 60 days. Rats of Group A were fed only with pellet without any experimental diet and were defined as control. Rats  of Group B were treated with potentiated sulfonamide @ 96 mg/rat/day orally whereas Group C was treated with potentiated sulfonamide @ 96 mg/rat/day plus spirulina (Spirulina maxima) @ 50 mg/rat/day orally (low dose spirulina). In Group D, potentiated sulfonamide and spirulina (Spirulina maxima) were given through feed @ 96 mg/rat/day and @ 100 mg/rat/day (high dose spirulina) respectively. Hematological parameters (TEC, Hb and absolute count of lymphocyte, neutrophil and eosinophil) and hispathological profile of liver and kidney were recorded. The investigation revealed that the oral administration of sulfonamide significantly (p<0.01) decreased the TEC (5.93±0.24) value, number of lymphocyte (581.76±3.70) and neutrophil (581.76±3.70) compared to other treated groups and control group. On the other hand significant (p<0.01) increase (422.86±2.34) in eosinophil population has been found in rats fed on sulfonamide irrespective of spirulina supplementation on the final day of experiment compared to other treated group and control group. From this experiment it is evidenced that spirulina has a potential counteracting effect against sulfonamide. These experiments were performed to determine the effects of dietary &beta;-glucan on stress responses of striped catfish (Pangasianodon hypophthalmus). Fish were fed for nine weeks with a diet containing 0 (control), 0.5% (G1), 1% (G2) and 2% (G3 group) &beta;-glucan. Subsequently, stress responses were studied by evaluating serum cortisol and glucose levels following a constant 24 h cold shock (from 28 &deg;C to 15 &deg;C). Serum cortisol and glucose concentrations were measured after cold treatments of varying durations (prior to, and after one, 12 and 24 h of cold shock stress, respectively). No differences in serum cortisol and glucose levels were found between control and &beta;-glucan-treated fish. However, the mortality rate was significantly lowered in cold challenged fish fed appropriate doses of &beta;-glucan (in G1 and G2 vs. G3 and control group). The results of the present study demonstrate that a proper administration&beta;--glucan in the diet could ameliorate the detrimental effects of a severe stress resulting in a reduction in fish mortality. &nbsp;

The aim of this study to conduct the effects of fimbrial and lipopolysacchride (LPS) immunization is on the pathohistological changes in rabbits, Fifteen rabbits of both sexes (Weight 1500-2000 gm) divided into three groups (5 animals of each group). The first group was immunized by 1ml (200µg /animal) of fimbrial subcutaneously the second group gave 1 ml ( 200 µg /animal) LPS while the third group was left as negative control group that injected 1 ml phosphate buffer control subcutaneously. First and second groups recived the same dose after two weeks give as booster dose. All animals challenged after 5 weeks of immunization by5X107CFU/ml Proteus vulgaris intra peritoneally .After 7 days from challenge all the animals, sacrificed for histopathological examination . The results showed that the fimbrial group had a severe infiltrations of mononucleart cells in liver and kidney ,but there was no clear histopathological changes observed in the spleen compared with lipopolysaccharide and control group . Also this group showed a slight mononuclear cells infiltration in lamina properia of intestine ;while the lipopolysaccharide group showed hypertrophy of epithelial cells with a mild mononuclear cells infiltration in lamina properia as compared with the control group that showed presence of mononuclear cells aggregation in the lamina properia .Our conclusion That the defense of animals against Proteus vulgaris more efficient in fimbrial antigen than lipopolysaccharide antigen by decrease the pathological effects of this bacteria.

Objective was to establish efficiency of sodium nucleospermat in correcting thrombocytopenia after chemoradiotherapy in oncological patients. Methods and materials. The study included data on 32 patients that had undergone combined treatment from January till May 2016. After detecting thrombocytopenia patients were randomized into two groups (16 patients in each): treated group, where patients received sodium nucleospermat, and control group, where sodium nucleospermat was not used. Thrombocyte level control was done on 5th, 10th and 15th day after treatment was over. Results and discussion. All 16 patients showed positive dynamics in increasing thrombocyte level after Sodium nucleospermat injection course was finished. This was proven by first (5th day) blood analysis. On average thrombocyte level after sodium nucleospermat treatment has risen to normal, at 161х109/1. Only 3 patients from this group had to pause radiotherapy for 5 days. Control group patients, which did not receive sodium nucleospermat, showed evidence of thrombocyte level recovery by 10th day only. On average thrombocyte level increase was insignificant, and median number was 111*109/l. Low thrombocyte level was main reason to pause radiotherapy for 11 (69%) patients in control group. Conclusion. Sodium nucleospermat allowed raising thrombocyte level to the lower normal range, which surpassed by 40%-50% in control group patients. Use of sodium nucleospermat did not show any cases of allergic reactions, toxicity or complications in oncological patients.

PURPOSE: Aloe vera is a tropical plant popularly known in Brazil as babosa. We have investigated the effect of aqueous extract of Aloe vera on the biodistribution of Na99mTcO4 and laboratorial parameters in Wistar rats. METHODS: Twelve animals were divided into treated and control groups. In the treated group, Aloe vera was given by gavage (5mg/mL/day) during 10 days. The control group received sorbitol by the same way and period. One hour after the last dose, we injected 0.1mL of Na99mTcO4 by orbital plexus. After 60 min, all the animals were killed. Samples were harvested from the brain, liver, heart, muscle, pancreas, stomach, femur, kidneys, blood, testis and thyroid and the percentage of radioactivity (%ATI/g) was determined. Biochemical dosages were performed. RESULTS: There was a significant increase of %ATI/g in blood, femur, kidneys, liver, stomach, testis and thyroid and also in blood levels of AST and ALT. A significant decrease in levels of glucose, cholesterol, triglycerides, creatinine and urea occurred. The statistical analyses were performed by Mann-Whitney test and T-Student test (p<0.05). CONCLUSION: The aqueous extract of Aloe vera facilitated the uptake of Na99mTcO4 in organs of rats and it was responsible to a high increase of levels of AST and ALT.

The aim of this study was to investigate the dose-related effects of fenitrothion (FNT) on the liver and kidney. The study was conducted on 8-week-old male Wistar rats that were divided into four groups (three experimental groups and one control group) and were treated orally with different doses (25, 50, 100 mg/kg) of FNT for 28 consecutive days. After treatment, the rats were anesthetized with ether and liver and kidney samples were taken for histological studies. The results showed that the histopathological changes in the liver were mainly represented by parenchymatous degeneration of hepatocytes with mild necrosis, leukocytic infiltration in the portal area, severe congestion, and hemorrhage. These changes were dose dependent. Marked tubular dilation, hydropic degeneration in tubular epithelium, moderate congestion, and hemorrhage in the cortical and medulla part of the kidney were recorded. Histopathologic examination of the liver and kidney indicated a significant injury only in rats receiving 100 mg/kg FNT.

The metabolic response to nutrient ingestion and the rate of digestion and absorption of nutrient molecules in bowel physiology plays an important role in the metabolic control of some human chronic non-infectious diseases. This experimentally-controlled designed nutritional study which lasted eight weeks aimed to determine the effects of proportional high-protein/low-carbohydrate (HP/LC) formulated diet on glycemic tolerance, glycemic control, body weight, organ weight and organ morphometry in healthy and diabetic adult male Wistar rats. Twenty-four male Wistar rats purchased from a disease-free stock were randomly categorized into four groups (n = 6, each) after two weeks acclimatization period in raised stainless steel cages with 6 mm2mesh floor and replaceable numbered blotters papers placed under each cage in a well-ventilated animal house. Animal groups include: Healthy control group (HC), Healthy treated group (HT), Diabetic control group (DC) and Diabetic treated group (DT. The animals were fed according to the experimental design with water ad libitumfor eight weeks. Diabetes was inducted with freshly prepared alloxan monohydrate solution (150 mg/kg bw, intraperitoneally). Body weights and fasting blood sugar concentrations were measured twice weekly, while oral glucose tolerance test was conducted on the last day of the eighth-week study and subsequently followed by organs extraction after anesthesia for weight and gross assessment. Proportional high-protein/low-carbohydrate formulated diet caused significant reduction in mean body weight of treated diabetic (DT: 22.6%; P= .001) and healthy (HT: 5.8%; P= .007) rats while the control animals on control diet recorded significant (P< .05) increase in body weight gain (DC: 12.4%; HC: 11.2%). Glycemic tolerance and control improved significantly in diabetic treated rats over that of the healthy treated rats. Gross morphometry of the extracted organs (kidneys, liver, heart, lungs, spleen and testes) revealed sustained normal morphological features without any visible lesion. In conclusion, consumption of proportional high-protein/low-carbohydrate formulated diet enhanced body weight reduction and sustained normal organ morphological features with good glycemic tolerance and control in experimental rats, suggesting its dietary potentiality, safety and suitability to ameliorate obesity-related diabetes.

Background: The present study was planned to assess the biochemical and micro-minerals profile in blood and other tissues of buffaloes environmentally exposed to heavy metals. Methods: Tissues (liver and kidney) and blood samples (n=50) were collected from local abattoir. Based on the level of heavy metals, animals were classified as exposed and control was found to have heavy metals in normal range. Blood and tissue sample from both groups were analyzed for micro-minerals, antioxidant status, metabolic profile and expression of metallothionein-2 (MT-2).Result: Exposed group was found to have significantly (p less than 0.05) higher level of arsenic and chromium as compared to the control group. Level of Copper (Cu) and zinc (Zn) were observed to be significantly (p less than 0.05) higher in exposed animals as compared to control but their concentrations were below the permissible limit in both the groups. Cobalt (Co) and iron (Fe) level were normal in all tissues but Fe level was lower than permissible limit in blood. Malonldialdehyde (MDA), the activities of superoxide dismutase (SOD) and catalase (CAT) was found to be significantly increased (p less than 0.05) in tissues and blood of exposed group. The exposed buffaloes were found to have significantly (p less than 0.05) increased glucose, alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), creatine kinase (CK), alkaline phosphatase (ALP), urea and creatinine level as compared to control group. Fold change expression of metallothionein (MT-2), had maximum in liver, followed by kidney and blood as compared to control group. The study concluded that heavy metals exposure and low concentration of micro-minerals in buffaloes could result in oxidative damage and alterations in the expression of metallothionein.

This study examines the effect of dexamethasone (Dex), a phospholipase A2 inhibitor, on the reversal of 1-kidney, 1-clip (1K,1C) hypertension and the synthesis of phospholipase A2-dependent products. Male Sprague-Dawley 1K,1C hypertensive rats [blood pressure (BP) greater than 190 mmHg] were allocated to three groups: two groups were given daily oral doses of Dex (0.142 mg/kg in water) for 72 h, whereas the third group was given water only (controls). One of the Dex-treated groups was then sham unclipped (n = 9), while the other Dex-treated group (n = 8) and the control group (n = 8) were unclipped. Dex attenuated the BP fall in the unclipped (223 +/- 8-148 +/- 9 mmHg) compared with the control unclipped (226 +/- 9-114 +/- 5 mmHg) animals (P less than 0.005). Aortic 6-ketoprostaglandin F1 alpha (6-keto-PGF1 alpha) was reduced in unclipped Dex-treated rats (13.4 +/- 1.2 ng/mg) compared with unclipped control rats (16.3 +/- 1.4 ng/mg; P less than 0.05) but was higher than in the sham-unclipped Dex group (11.5 +/- 1.2 ng/mg; P less than 0.05). Serum thromboxane B2 (TxB2) in the unclipped Dex-treated group was lower than in the unclipped control rats (P less than 0.05) but higher than in sham-unclipped rats (P less than 0.05). Dex significantly increased urinary prostaglandin E2 (PGE2) excretion, whereas urinary 6-keto-PGF1 alpha was unaltered. After unclipping, both urinary PGE2 and 6-keto-PGF1 alpha increased significantly, although there was no obvious difference between Dex-treated and control animals. These findings demonstrate opposite effects of Dex on renal compared with extrarenal prostanoid synthesis and support the hypothesis that attenuation of aortic 6-keto-PGF1 alpha synthesis may be responsible for the smaller fall in BP after unclipping in Dex-treated rats.

Background: Dermatologic toxicities are the most common side effects associated with the targeted drugs epidermal growth factor receptor inhibitors (EGFRIs), in which xerosis commonly complicated by pruritus severely disturbs the quality of life. The study has observed the curative effect of Zizao Yangrong granules (ZYG) from Chishui Xuanzhu in the treatment of EGFRIs-related xerosis and pruritus, as well as evaluating the safety of the prescription. Methods: Patients (n = 68) who had xerosis after using EGFRIs were enrolled and then randomly divided into the treatment group and control group, respectively, receiving ZYG and placebo granules combined with vitamin E ointment. The intervention lasted 4 weeks. Changes in xerosis and pruritus were observed, and blood routine examination as well as liver and kidney function are observed as safety indexes. The water content of skin and qualify of life were observed. Results: A total of 66 out of 68 patients finished the study with 34 patients in each group. The effective rates of xerosis among the treatment group and control group were 84.8% and 69.7% after 2 weeks’ treatment ( P < .05), while they were 84.8% and 75.8% after 4 weeks’ treatment ( P < .05). The patients in the experimental group had better quality of life than that in the control group ( P = .045). Conclusion: ZYG can effectively improve the skin dryness associated with EGFRIs, and significantly improve the quality of life of patients with good safety; however, larger randomized controlled trials are needed to verify these findings.
