Influence of Priming with Exogenous Selenium on Seed Vigour of Alfalfa (Medicago sativa L.)

Background: Selenium (Se) is an essential trace element to higher plants, animals and humans, but low Se levels are a global public health concern. Seed priming has become a basic strategy for the production of Se-riched agricultural products, but its application is still not clear in the production of Se-enrich alfalfa, hence this study was conducted for the production of Se-enriched alfalfa by seed priming with different concentrations and time on seed vigour. Methods: Seeds were primed with 0, 0.5, 1.0, 2.0, 4.0 and 8.0 mmol L-1 of sodium selenite solution for 0, 3, 6, 9 and 12 h at 20°C and their germination percentage, germination index, seedling vigour index and mean germination time were analyzed. Result: Seed vigour of alfalfa was improved by priming with low selenium (Se) concentration (0.5 and 1.0 mmol L-1), but was inhibited by high Se concentration ( greater than 2.0 mmol L-1). Hence, it must be necessary to carefully select appropriate concentration and time for the application of Se priming in alfalfa seeds. The optimal manner of Se priming in alfalfa seeds might be at 1.0 mmol L-1 concentration for 9 h.

Background: Black gram is the important pulses crop, grown throughout the country; gives low seed yield mainly due to poor management and low soil fertility. Development of suitable technology is essential to enhance productivity. Seed priming is one such effective technology which enhance rapid and uniform emergence to achieve high vigour, leading to better stand establishment and yield. The current research aimed to study micronutrient seed priming and rhizobium coating on seed germination, seedling vigour, crop growth and seed yield in black gram.Methods: Blackgram seeds were primed with 10 different micronutrient solutions with different concentrations and evaluated for its quality parameters to find out suitable seed priming technique. Seeds from two best priming treatments were coated with rhizobium @ 30g/kg of seed and evaluated for its production potential and resultant seed quality under field condition.Result: Seed priming with 0.5% ZnSO4 at 1/3rd seed to solution ratio for 3 hours recorded higher germination (96%) and vigour index (3811) than control which recorded 84% seed germination and 2528 of vigour index under laboratory experiment. Field experiments revealed that, seeds primed with 0.5% ZnSO4 and coated with rhizobium @ 30g per kg of seeds improved seed yield by 13.2% over control.

Seed priming has proved to be an effective method in imparting stress tolerance to plants using natural and/or synthetic compounds to treat the seeds before germination. The present study was designed to investigate the physiological mechanism of seed priming with ZnSO4 (osmopriming) and distilled water (hydropriming) on sugar beet genotypes (‘Shokofa’, ‘Sina’, ‘Paya’, ‘Turbata’, and ‘Aria’) germination indices, seedling growth parameters, and biochemical properties under salinity stress (0, 2, 5, and 12 dS/m NaCl). A significant reduction in germination percentage (33.23%), germination rate (77.2%), chlorophyll a, b, and total contents (43.9, 31.9, and 39.9%, respectively) while, a significant increase in radical, plumule, and seedling length (57.1, 44.4, and 51.2%, respectively), seedling vigour index (48.9%), superoxide dismutase activity (61.3%), proline (54.0%) and sugar (56.3%) contents were achieved at 12 dS/m NaCl in compared to the control treatment. Seed hydropriming and osmopriming caused significant improvements in photosynthetic pigments, antioxidant enzyme activity, and proline content reflected in high germination percentage and rate as well as seedling vigour index and reduced mean germination time under salinity. ‘Paya’ and ‘Aria’ genotypes had a superiority according to the germination percentage and seedling vigour index, respectively. The hydropriming of ‘Paya’ genotype resulted in the highest germination percentage (95%) under high level of salinity (12 dS/m) which 11.84% increase compared to the control treatment. Hydropriming of ‘Sina’ seeds showed the highest chlorophyll a and total, and carotenoids under non-stress conditions (22.89, 31.65, and 2116.6 µg/g FW). Also, hydropriming by increases chlorophyll b content led to the modulation of the negative effects of high salinity stress (12 dS/m). In conclusion, different seed priming treatments in sugar beet seeds improved the salinity tolerance by physiological characteristics nonetheless hydropriming was the most effective treatment to get higher germination indices in ‘Paya’ and ‘Aria’ genotypes.

            The study was conducted to standardize the methodology of humid priming for improving the seed germination and vigour of okra seeds and to compare the efficacy of different enhancement techniques such as hydro priming, spin priming, seed coating and the combination of ‘humid priming + seed coating’ in improving the seed vigour characteristics of okra (Abelmoschus esculentus L.). In order to standardize the humid priming technique, okra seeds were subjected to humid priming for four different duration’s viz., 2, 4, 6 and 8 h, shade dried to original moisture content and subjected to evaluation of seed germination   under shade net conditions in raised beds. The results revealed that 4 h of humid priming was statistically significant to the rest of the durations, with respect seed germination and seedling growth. With respect to anatomical potential, the radicle length (3.39mm), cotyledon length (6.69mm) and cotyledon width (6.30mm), recorded by humid priming (4 h) was 62.2 %, 6.4 % and 2.6 % higher than control seeds. The second experiment conducted to compare the efficacy of hydro priming, spin priming, polymer coating and combination of humid priming + seed coating. The study revealed that seeds subjected to combination of humid priming (4 h) + seed coating (3g kg-1) recorded highest physiological potential in terms of germination percentage (87 %), seedling length (20.2 cm), dry matter production (421.3 mg), vigour index (1765), root volume (3.5 cm3) and root sheath (2.560 g) while the control recorded only 70 %, 15.1 cm, 328.7 mg, 1055, 2.0 cm3 and 1.532 g, respectively.

The effects of seed priming with Fe-Zn-NA chelate (FeSO4+ZnSO4+niacinamide) and the longevity of the beneficial priming effects were investigated for aged seeds of hybrid rice cv. Lingyouhuazhan (LYHZ) and cv. Longxiangyou 130 (LXY130). As compared with untreated and hydroprimed seeds, Fe-Zn-NA chelate priming significantly enhanced germination energy (GE), germination percentage (GP), germination index (GI), vigour index (VI) and normal seedling rate (NSR), and increased seedling shoot height (SH) and seedling dry weight (DW) in both cultivars. After priming with Fe-Zn-NA chelates, the GP of LYHZ was 96.0%, which was 12.7 and 12.0% higher than hydroprimed and non-primed seeds, respectively. Similarly, the GP of LXY130 was 89.5%, significantly higher than the hydroprimed and non-primed seeds (by 11.5 and 10.0%, respectively). When stored at 25°C, the benefits of Fe-Zn-NA chelate priming on seed vigour and viability was maintained for three months; while after six months storage, the GP of LYHZ and LXY130 was only 48.0 and 34.0%, respectively, which was significantly lower than the non-primed seeds (84.0 and 71.1%). When stored at 15 or 5°C for six months, the germination potential of Fe-Zn-NA chelate-primed seeds was still high. The GP of LYHZ and LXY130 seeds was 94.4% and 77.8%, respectively, after storage at 15°C, and 97.8% and 85.6% after storage at 5°C. In conclusion, the Fe-Zn-NA chelate priming was an effective method to improve the vigour and viability of hybrid rice aged seeds; however, the duration of beneficial priming effects needs to be determined according to the storage temperature.

Seed priming is a pre-sowing treatment that enhances germination performance and stress tolerance of germinating seeds. The effect of seed priming with proline on germination and seedling growth of mungbean (Vigna radiata L.) under salt stress was investigated. The experiment carried out in completely randomized design with three replications in May 2018 at the Research Laboratory of the School of Agriculture and Rural Development, Bangladesh Open University, Gazipur, Bangladesh. Salt stress at 5 dSm-1 decreased seed germination percentage, plumule length, radicle length, plumule fresh weight, radicle fresh weight and seed vigour index compared to control. Seed priming with proline increased germination percentage (53.84%), plumule length, radicle length, plumule fresh weight, radicle fresh weight and seed vigour index under salt stress. The results suggested that seed priming with proline could effectively alleviate the inhibitory effects of salt stress on seed germination and seedling growth of mungbean. South Asian J. AbstractThe stimulatory role of 3-methyl-2H-furo[2,3-c]pyran-2-one, a smoke-derived butenolide, on germination and post-germination events is well documented. Previous studies have involved germinating seeds in the continuous presence of the compound. However, commercial growers cannot exploit the potential of the butenolide for large-scale production of crops due to limited availability and environmental constraints. The present study was undertaken to assess the potential of the butenolide as a priming agent of tomato (Solanum esculentum Mill.) seeds. Flow cytometry data revealed that butenolide-primed seeds had a higher percentage of nuclei at the 4C stage than water-primed seeds. Emergence of the radicle was much faster in the primed seeds. After 36 h of imbibition, a higher percentage of the butenolide-primed seeds (22%) exhibited radicle emergence compared to the water-primed seeds (12%). While butenolide-primed seeds initially germinated more rapidly than either water-primed or unprimed seeds in a 48-h period, water-imbibed seeds reached a similar germination level as the butenolide-primed seeds by 60 h of incubation. The butenolide-primed seeds produced significantly (P ≤ 0.05) more vigorous seedlings than water-primed seeds or seeds in the continuous presence of either water or butenolide. A gradual decrease in the seedling vigour index was recorded for both water and butenolide-primed seeds with increased seed storage at room temperature. Nevertheless, the vigour index was significantly greater in the butenolide-primed seeds than the water-primed seeds. Vigour indices were significantly (P ≤ 0.05) higher for the butenolide-primed seeds under various stress conditions (salinity, osmoticum or temperature) compared to control or water-primed seeds. Results of the present study suggest that the butenolide is a good seed-priming agent. Additionally, primed seeds retained the promotive effect for a considerable time. This was also the case for tomato seeds under various simulated field stress conditions.

UDK: 582.661.51:631.53.027]:547.913 In order to investigate the effects of seed priming with silver fir and oregano essential oils on certain important seedling characteristic and seed vigour of interesting endemic and horticultural species Silene sendtneri, an experiment was conducted based on randomized completely design with three replications. Traits such as germination rate, water content, dry weight, vigour index and photosynthetic pigments were analysed. Results revealed that the seedlings obtained with priming seeds showed increased growth, water content, vigour and photosynthetic pigment contents but decreased germination rate and dry weight compared with that obtained with non-primed seeds. Analysis of seed priming effects had demonstrated even germination rate is smaller the seedling vigour is slightly higher especially with all oregano oil treatments. We suggest that oregano oil has a potential as a priming agent for improvement of seedling synchrony, although at lower rate of germination.

The continuous rise in global population demands more food production under limited land resources accompanied by climate change. More oversupply of quality seeds to meet ever-growing food demand is a highly challenging task. Seeds with low vigour level limit the growth and yield of crops at varied field conditions. Hence, an experiment was conducted to develop a technique for the fast and homogeneous growth of kabuli chickpea seeds. The priming treatments evaluated in the present study enhanced the various seed quality parameters of kabuli chickpea significantly when compared to control. Hydro-priming for 4 hours (at 20°C) and air drying for 72 hours showed better results compared to rest of the treatments both in fresh seeds which recorded higher first count (66.38%), germination (92.50%) root length (11.85 cm), shoot length (6.16), mean seedling length (18.01), dry seedling weight (747 mg), seedling vigour index-I (1665), higher speed of emergence (0.3992) and old seeds with higher first count (61.88%), germination (74.13%) root length (8.15 cm), shoot length (5.85 cm), mean seedling length (14.00 cm), seedling dry weight (611 mg), seedling vigour index-I (1056), higher speed of emergence (0.3814). The aged seeds have recorded 29 % increase in germination percentage in contrast to 8 percent increases in fresh seeds. Kabuli chickpea seeds could be subjected to hydropriming, an affordable, alternative, and eco-friendly technique for improving seed and seedling vigour of kabuli chickpea.

Seedling vigour is a key critical component to produce well-established canola (Brassica napus) seedlings under less than ideal conditions in western Canada. Our objective was to determine if seed lots from late October (fall) or early spring (April) seeding dates improve seed vigour relative to mid-May (May) seeding dates. We compared seed size, seed maturity, germination percentage, and seedling emergence under controlled-environment conditions from six seed lots obtained from fall-sown (1997, 1998), April-sown (1998, 1999) and May-sown canola (1998, 1999) cv. Quest. On average, 80% of fall-derived seed for both years was larger than 1.7 mm compared to only 32% for May-derived canola seed. April-derived seed was intermediate. Fall and April derived seed were considered 40% mature compared to 16% for May-derived seed. In germination tests at 8°C, May derived seed germinated and emerged slower with a lower total germination percentage compared to falland April-derived seed lots. Priming reduced differences in emergence between the seed lots. Field studies at Scott, SK, sown in the fall of 1999 and 2000, and in early May of 2000 demonstrated that seed derived from fall- and April-sown canola produced higher plant densities, higher biomass at bolting, and higher seed yield than seed derived from May-sown canola. Yield was 10 to 45% less from seed derived from May-sown canola than seed derived from fall- and April-sown canola. This study established the impact of seeding date on seed quality and vigour, which in turn affected emergence, seedling vigour and yield. Also, seed vigour slowly declined within 1 yr, primarily with seed derived from the May-sown canola. Key words: Canola, Brassica napus, vigour (seed), seeding (fall), emergence, yield (quality)

The current experiment was conducted to evaluate the Performance of Organic amendments and botanical treatments on seed quality parameters of French bean under stimulated environmental conditions. An experiment was conducted in 2021 at postgraduate seed testing laboratory, Department of Genetics and Plant Breeding Naini Agriculture Institute, Sam Higginbottom University of Agriculture Technology and Sciences, Prayagraj. The French bean Cultivar Arka Bold seeds were used for priming. Organic treatments are designated as Panchagavya 2%, 4%,6%,8%, Agniasthra 2%,4%,6%,8% and Botanicals treatments are Tulasi leaf extract 5%, Aloevera leaf extract 5%, Vinca rosea leaf extract 5% and also did hydroprming treatment using distilled water respectively. Seeds were soaked in organic, botanicals and in distilled water for duration of 6 hours followed by shade drying. The results were obtained and it was found that all the treatments showed significant effect on quality parameters. The highest was found in panchgavya of all doses followed by tulasi leaf extract found to be best in Germination percentage, Speed of Germination, Root length, Shoot length, Seedling length, Seed vigour index-I, Seed vigour Index-II compared to control or untreated seeds which indicates the effect of organics and botanicals on physiological processes of the seeds. This study revealed that priming with organic amendments and botanicals can be effective in enhancing the seed quality parameters of french bean and they are low cost, effective and nature friendly and toxic free.
