<p>Black cumin (Nigella sativa, L.) is a plant with a lot of advantages. Many research have been conducted on this plant and most of them use nonpolar, such as petroleum ether and chloroform, as a solvent in extraction process. In this research, extraction of black cumin using ethanol was applied.Black cumin extraction started with maceration using ethanol p.a. and partition using chloroform (1:1). The antioxidant activity of ethanol fraction of black cumin was examined by DPPH method. To identify the composition, phytochemical screening, including test for terpenoid, flavonoid, saponin, tannin and alkaloid, was also conducted.The result showed that the scavenging ability on DPPH method was 22.483% with IC50 2743.59. Phytochemical screening on ethanol fraction showed the presence of flavonoid, tannin and alkaloid. The study concluded that polar fraction of black cumin has small antioxidant activity.</p>

Tecoma stans (L.) Juss. Ex Kunth is a species belonging to the Bignoniaceae family, popularly known as yellowing, yellow bell, and garden little ipe. Studies with T. stans seeds are scarce and it have potential in the search for natural compounds with biological activities. This study aimed to evaluate the toxicity and antioxidant activity of the ether extract (EE) and fractions from T. stans seeds. The EE was obtained in Soxhlet apparatus with petroleum ether and fractions were obtained by hydrolysis and esterification reactions. Phytochemical screening evaluated the presence of steroids, triterpenoids, alkaloids, coumarins and anthraquinones in EE. The toxicity was evaluated by the A. salina lethality test and antioxidant activity by DPPH method. It was observed the presence of steroids, triterpenoids and alkaloids in EE, which had no toxicity to A. salina (LC50 > 1000 µg/mL). The fractions of seeds exhibited toxicity on A. salina. All samples showed antioxidant activity with EC50 between 0.9 and 1.1 µg/mL. These results indicated potential applications for T. stans seeds.

Objective: To assess the antioxidant activity from another part of the plant, in this study, leaf extracts in n-hexane were fractionated.Methods: Ten fractions were obtained and tested in vitro for antioxidant activity using two methods, 1,1-diphenyl-2-picrylhydrazyl (DPPH) and ferricreducing antioxidant power (FRAP), to identify the most active fraction.Results: The IC50 of the most active fraction was 36.24 μg/mL using the DPPH method, and the EC50 was 39.54 μg/mL using the FRAP method. Themost active fraction was also shown to contain terpenoids.Conclusion: The most active fraction of an n-hexane extract of the leaves of Gacinia bancana Miq., which was tested by both DPPH and FRAP methodshad antioxidant activities with IC50 and EC50 values of 36.2482 μg/mL and 39.5442 μg/mL, respectively. Phytochemical screening showed that activefraction contains terpenoids.

Chlorogenic acid has been proved to have cardiovascular protection, antibacterial, antiviral, hemostatic, and hypolipidemia effects. Modern scientific research on the bioactivity of chlorogenic acid has been extended to the fields of food, medicine, health care and daily-use chemical industry. The aim of this research was to optimize the extraction conditions for chlorogenic acid from Eucommia ulmoides (Eucommiaceae) leaves. The significant variables were screened and optimized by a combination of Plackett-Burman test and Box-Behnken design. Optimum extraction parameters with ethanol concentration of 50%, solvent pH value of 3, and particle size of 60 mesh were determined according to variance analysis and contour plots. Under these conditions, the yield of chlorogenic acid was up to 4.36 mg/g, which was basically consistent with the theoretical prediction value of 4.50 mg/g. This study also proved the potential antioxidant activity of E. ulmoides leaves. The optimal extract of E. ulmoides leaves rich in chlorogenic acid showed the highest antioxidant activity in the FRAP method, which was 219.8 μM Trolox equivalents (TE) per g extract weight (EW) (μM TE/g EW). The DPPH method gave a similar value (168 μM TE/g EW) to the ABTS method (152 μM TE/g EW). The established extraction process was efficient in the recovery of chlorogenic acid from E. ulmoides leaves, encouraging its valorization as a cheap and sustainable alternative for the isolation of chlorogenic acid.

Abstract The study aimed to evaluate the antimicrobial activity, antioxidant, toxicity and phytochemical screening of the Red Propolis Alagoas. Antimicrobial activity was evaluated by disk diffusion method. Determination of antioxidant activity was performed using the DPPH assay (1.1-diphenyl-2-picrylhydrazyl), FTC (ferric thiocyanate) and determination of phenolic compounds by Follin method. Toxicity was performed by the method of Artemia salina and cytotoxicity by MTT method. The phytochemical screening for the detection of allelochemicals was performed. The ethanol extract of propolis of Alagoas showed significant results for antimicrobial activity, and inhibitory activity for Staphylococcus aureus and Candida krusei. The antioxidant activity of the FTC method was 80% to 108.3% hydrogen peroxide kidnapping, the DPPH method showed an EC50 3.97 mg/mL, the content of total phenolic compounds was determined by calibration curve gallic acid, resulting from 0.0005 mg/100 g of gallic acid equivalent. The extract was non-toxic by A. salina method. The propolis extract showed high activity with a higher percentage than 75% inhibition of tumor cells OVCAR-8, SF-295 and HCT116. Chemical constituents were observed as flavonones, xanthones, flavonols, and Chalcones Auronas, Catechins and leucoanthocyanidins. It is concluded that the extract can be tested is considered a potential source of bioactive metabolites.

Abstrcact Jamu Selokarang is one of the traditional drinks originating from an area, namely Madura, the ingredients used in the manufacture of Jamu Selokarang are turmeric, ginger, ginger, betel leaf, soursop leaves, cloves, nutmeg, coriander, temu ireng and ginger. The ingredients used in the manufacture of herbal medicine selokarang contain a lot of antioxidants and essential oils. Testing is needed to determine the antioxidant activity contained in the herbal medicine selokarang. This study aims to determine the effect of substitution of jamu selokarang on antioxidant compounds, total phenols, flavonoids, ash content and water content. This study used a simple completely randomized design method (simple CRD) with a single factor, namely the black cumin formulation of 0%, 2%, 4%, 6% and 8% which were included in jamu selokarang. The research was repeated 3 times. The parameters observed were: water content, ash content, antioxidants, total phenols, and flavonoids. The results showed that the substitution had a significant effect on water content, ash content and antioxidant activity, while the total phenol and flavonoid analysis had no significant effect. Treatment 0% black cumin had water content of 2.68%; Ash content of 7,90%; DPPH anti-oxidant for 49.08%; antioxidant FRAP 26.25 μgAAE / mL; total phenol was 2.05 μgGAE / mg, and total flavonoids was 0.91 μgQAE / mg.

The oil obtained from the seeds of Nigella sativa L. (N. sativa), also known as black cumin, is frequently used in the Mediterranean area for its anti-inflammatory, anti-oxidant, and anti-cancer activities. The aim of the present study was to evaluate the antioxidant and anti-inflammatory properties of the oil extracted from seeds of a N. sativa cultivar produced in the Marche region of Italy, and to determine if the thymoquinone content, antioxidant properties, and biological activity would decay during storage. Cytotoxicity and anti-inflammatory properties of N. sativa oil were tested in an in vitro model of low-grade inflammation in Simpson–Golabi–Behmel syndrome human pre-adipocytes. The fresh extracted oil (FEO) contained 33% more thymoquinone than stored extracted oil (SEO), demonstrating that storage affects its overall quality. In addition, the thymoquinone content in the N. sativa oil from the Marche region cultivar was higher compared with other N. sativa oils produced in the Middle East and in other Mediterranean regions. Pro-inflammatory cytokines (e.g., Interleukin (IL)-1alpha, IL-1beta, IL-6) were differently modulated by fresh and stored extracts from N. sativa oils: FEO, containing more thymoquinone reduced IL-6 levels significantly, while SEO inhibited IL-1beta and had a higher antioxidant activity. Total antioxidant activity, reported as µM of Trolox, was 11.273 ± 0.935 and 6.103 ± 0.446 for SEO and FEO (p = 1.255 × 10−7), respectively, while mean values of 9.895 ± 0.817 (SEO) and 4.727 ± 0.324 (FEO) were obtained with the 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) assay (p = 2.891 × 10−14). In conclusion, the oil capacity to counteract proinflammatory cytokine production does not strictly depend on the thymoquinone content, but also on other antioxidant components of the oil.

Antioxidant are compounds that are able to inactivate the development of the oxidation reactions of the molecules or neutralize free radicals. Mahogany bark (Swietenia mahagoni Jacq.) has properties as a medicinal plant. Through the phytochemical screening, the mahogany bark (Swietenia mahagoni Jacq.) contains antioxidant compounds such as alkaloids, saponins and phenolics. The aim of this study was to determine the presence of antioxsidant activity in the extract of mahogany bark (Swietenia mahagoni Jacq.). using the DPPH method with a concentration of 250 mg / L, 200 mg / L, 150 mg / L and 100 mg / L and Vitamin C p.a as a positive control. Each sample made three repetitions of the test. The test uses a UV-Vis Spectrophotometer. The result of the study showed that mahogany bark (Swietenia mahagoni Jacq.) showed antioxidant activity with the percentage of 21,50% at the concentration at 250 mg / LKeywords : Swietenia mahagoni Jacq., Antioxidant, Ethanol, DPPHABSTRAKAntioksidan ialah senyawa yang mampu menginaktivasi berkembangnya reaksi oksidasi molekul lain atau menetralisir radikal bebas. Mengandung senyawa antioksidan seperti alkaloid, saponin dan fenolik. Penelitian ini bertujuan untuk mengetahui adanya aktivitas antioksidan didalam ekstrak tanaman Kulit Batang Switenia mahagoni Jacq. menggunakan metode DPPH dengan konsentrasi 250 mg/L, 200 mg/L, 150 mg/L dan 100 mg/L dan Vitamin C p.a sebagai kontrol positif. Masing-masing sampel dibuat tiga kali pengulangan uji. Pengujian menggunakan alat Spektrofotometer UV-Vis. Hasil penelitian menunjukkan bahwa ekstrak Kulit Batang Swietenia mahagoni Jacq. memiliki aktivitas antioksidan dengan presentase sebesar 21,50% pada konsentrasi 250 mg/L Kata Kunci : Swietenia mahagoni Jacq., Antioksidan, Etanol, DPPH

The extraction process from leaves and twigs of gambier (Uncaria gambier Roxb)plant was conducted mechanically by using traditional hydraulic press, conventional screw press, and modified twin-screw press. The leaves and twigs of gambier plant was obtained from traditional farmer in Limapuluh Kota, West Sumatera, Indonesia. The water, ash and catechin contents of the gambier extracts were analyzed based on SNI 01-3391-2000 using spectrophotometry and thermogravimetry method. Antioxidant activity analysis of the extracts was also performed by 1,1-diphenyl-2-picrylhidrazyl (DPPH) method and compared tovitamin C as a standard antioxidant. The analysis results showed that the extracts contain no ash. Meanwhile, the catechin and water contents of the extracts were approximately 50 and 13% thus the extracts were classified as quality gambier 2. Other than that, analysis result of catechin and epicatechin contents of the extracts using HPLC and compared to the reference materials showed that catechins contents of the extracts using traditional hydraulic press, conventionalscrew press, and modified twin-screw press give catechin content in about 94.296-95.030%. However, epicatechin was detected in a trace amount. The antioxidant activity of the extracts were 2.5 times stronger than reference. The IC50 value of 4.37-4.52 µg/mL and was categorized as active antioxidant.

Abstract Thymoquinone (TQ) is a natural compound occurred in black cumin ( Nigella sativa L.), which possesses potent antioxidant activity without having any phenolic hydroxyl group believed to be responsible for antioxidant activity of a molecule. In the present study, computational calculation based on Density Functional Theory (DFT) have been executed to assess systematically the worth of antioxidant behavior of this compound. Geometrical characteristics, HOMO-LUMO and MEP surface have been studied. Thermochemical parameters correlated to the leading antioxidant mechanisms such as HAT, SETPT and SPLET have been studied in gas and water media. In addition, the changes of thermochemical parameters such as ∆G and ∆H have been computed for HA from TQ to hydroxyl radical in gas and water phases to investigate its free radical scavenging potency. The low and comparable values of BDE, PDE, IP, PA and ETE suggest the antioxidant activity. The ∆G and ∆H also convey apposite thermodynamic evidence in favor of antiradical capability of TQ. The attack of the free radical takes place preferentially at 3CH position of the molecule.

SKRINING FITOKIMIA DAN UJI AKTIVITAS ANTIOKSIDAN EKSTRAK BIJI ADAS (Foeniculum vulgare) MENGGUNAKAN METODE DPPH ABSTRAK Telah dilakukan penelitian tentang skrining fitokimia dan uji aktivitas antioksidan ekstrak biji adas. Tujuan dari penelitian ini yaitu melakukan skrining fitokimia untuk mengetahui kandungan senyawa alkaloid, triterpenoid dan steroid, flavonoid, tannin, saponin dan menetukan aktivitas antioksidan dari ekstrak biji adas. Skrining Fitokimia untuk alkaloid ditentukan dengan menggunakan pereaksi Mayer, Dregendorff, Wagner. Triterpenoid dan steroid ditentukan menggunakan Mg bubuk. Senyawa tannin ditentukan dengan larutan FeCl3 1%. Senyawa saponin ditentukan menggunakan aquades. Ekstrak biji adas diperoleh dengan metode soxhletasi menggunakan pelarut petroleum eter dan uji aktivitas antioksidan menggunakan metode               1-1-difenil-2-pikrihidrazil (DPPH).  Hasil penelitian menunjukan kandungan senyawa kimia biji adas positive mengandung senyawa falvonoid, tannin dan saponin, sedangkan untuk senyawa alkaloid, triterpenoid dan steroid memberikan hasil yang negatif. Aktivitas antioksidan dengan 1000; 2000; 3000; 4000; 5000 ppm,  memberikan hasil berturut-turut yaitu 48.99 %, 33.92 %, 5.93 %, 21.23 %, 6.40 %. Aktivitas antioksidan biji adas tertinggi terdapat pada konsentrasi    1000 ppm dengan hasil 48.99 %. Biji adas memiliki persen aktivitas antioksidan yang baik, sehingga dapat didigunakan sebagai salah satu sumber antioksidan alami.   Kata kunci : Antioksidan, Biji adas, DPPH phytochemical screening and antioxidant activity testing of fennel seed extract (foeniculum vulgare) Using the dpph method Abstract Has done research on phytochemical screening and testing fennel seed extract antioxidant activity. The purpose of this study is to determine the phytochemical screening compounds content of alkaloids, triterpenoids and steroids, flavonoids, tannins, saponins and determine the antioxidant activity of extracts of fennel seeds. Phytochemical screening for alkaloids was determined using reagent Mayer, Dregendorff, Wagner. Triterpenoids and steroids were determined using Mg powder. Tannin compounds determined with 1% FeCl3 solution and saponin determined using distilled water. Fennel seed extract obtained by the method soxhletasi using petroleum ether solvent and antioxidant activity assay using method 1-1-diphenyl-2-pikrihidrazil (DPPH). Research has been done showing the chemical content of fennel seeds contain compounds falvonoid positive , tannins and saponins . Antioxidant activity with 1000 ppm , 2000 ppm,    3000 ppm , 4000 ppm , 5000 ppm , respectively yield is 48.99 % , 33.92 % , 5.93 % , 21.23 %, 6.40 %. The highest antioxidant activity of fennel seeds contained 1000 ppm with 48.99 % result. Fennel seeds have a good percent antioxidant activity , so it can didigunakan as a source of natural antioxidants. Keywords : Antioxidants, Fennel seeds, DPPH
