Background : Streptococcus mutans is the most cariogenic microorganism in the oral cavity. Streptococcus mutans has the ability to use dietary carbohydrates, such as sucrose, to synthesize extracellular polysaccharides (glucan and fructan) through glucosyltransferase and fructosyltransferase. Extracellular polysaccharide has a role in promoting the adhesion of bacteria. Chlorhexidine  is the gold standard as antiplaque agent but chlorhexidine is not fully able to inhibit the adhesion of bacteria because it has an effect that can increase gtfC and gtfD expression on S. mutans in planktonic form. Temulawak extract (Curcuma xanthorrhiza Roxb.) contains of curcumin, xanthorrhizol, saponin, flavonoid and tannin that have an effect to inhibit activity and secretion of glucosyltransferase and fructosyltransferase. Purpose: The aim of this study is to find out the ability of temulawak extract (Curcuma xanthorrhiza Roxb.) to inhibit adhesion of Streptococcus mutans.  Method: This study was designed as an experimental laboratory study with post test only control group design using Streptococcus mutans. Temulawak was extracted using maceration method. The number of bacteria that can perform adhesion to the tooth surface can be determined by performing calculations using formulas plate count method. Result: Temulawak extract (Curcuma xanthorrhiza Roxb.) at concentrations of 25%, 37.5%, 50% and chlorhexidine can inhibit bacterial adhesion of Streptococcus mutans. Conclusion: Temulawak extract at concentration of 50% and 37.5% has the best ability to inhibit the adhesion of bacteria among other treatment groups.

Background: Dental caries is one of the most common infectious diseases and often occurs in the community caused by bacteria. Attached bacteria in the tooth surface for a long time will form a biofilm and will lead to demineralization characterized by damage in the structure of the tooth enamel. The bacteria that cause dental caries and can form biofilms is Streptococcus mutans. The bacteria inside biofilms are more resistant to antibacterial agents. Flavonoids in mangosteen pericarp extract can be a cleaner alternative for the anti-biofilm cavity that has properties against Streptococcus mutans. Purpose: To determine the activity of flavonoids in mangosteen pericarp extract at a certain concentration against Streptococcus mutans bacteria. Methods: This study was a laboratory experimental study with a post-test only control group design. Streptococcus mutans were diluted according to the Mc Farland dilution standard 106 in Tryptic Soy Broth (TSB) medium and put in a flexible U-bottom microtiter plate. Then it was incubated for 5x24 hours and checked using crystal violet simple staining to see the formation of biofilms. Flavonoid extract of mangosteen pericarp performed serial dilution in a concentration of 100%, 50%, 25%, 12.5%, 6.25%, 3.125%, 1.56%, and 0.78% was added, and the incubation process were conducted for 1x24 hours. OD (Optical Density) readings were done with a wavelength of 595 nm. Results: There was a significant difference between the test groups and the positive control group. The concentration of 100% had the anti-biofilm activity and showed the value of the highest percentage of inhibition, whilst the concentration of 0.78% showed a minimum biofilm inhibition concentration. The results were demonstrated by a statistical analysis test. Conclusion: Flavonoid extract of mangosteen pericarp at a certain concentration has anti-biofilm activity against Streptococcus mutans biofilm.

Abstrak: Minyak cengkeh berguna sebagai antibakteri alami. Minyak esensial dari cengkeh mempunyai fungsi anestetik dan antimikrobial. Zat yang terkandung dalam cengkeh yang bernama eugenol dapat membunuh bakteri termasuk bakteri yang resisten terhadap antibiotika, salah satunya adalah bakteri Streptococcus mutans. Bakteri ini merupakan mikroorganisme penyebab utama terjadinya karies. Tujuan dari penelitian ini yaitu untuk mengetahui efek antibakteri ekstrak cengkeh terhadap bakteri Streptococcus mutans secara in vitro. Penelitian ini merupakan penelitian eksperimental menggunakan post test only control group design. Penelitian ini menggunakan bahan coba ekstrak cengkeh dengan konsentrasi 40%, 60%, dan 80%, Ciprofloxacin, aquades dengan pengulangan sebanyak lima kali. Data dikumpulkan dan dianalisis dengan one-way ANOVA dan post-hoc uji LSD ( = 0,05). Berdasarkan hasil uji statistik penelitian uji efek antibakteri ekstrak cengkeh terhadap bakteri streptococcus mutans secara in vitro, dapat disimpulkaan bahwa ekstrak cengkeh memiliki efek antibakteri dalam menghambat pertumbuhan bakteri Streptococcus mutans secara in vitro. Hasil uji lanjut post-hoc uji LSD menunjukan daya hambat ekstrak cengkeh 40%, 60%, 80%, lebih kecil (p<0,05) dalam menghambat Streptococcus mutans secara in vitro dibandingkan Ciprofloxacin. Kata Kunci: Ekstrak cengkeh, Streptococcus mutans.   Abstract: Clove oil is useful as a natural antibacterial agent, essential oil of clove has anesthetic and antimicrobial effect. Substances contained in clove called eugenol can kill bacteria including antibiotic resistant bacteria, one of which is the bacteria Streptococcus mutans. This bacteria is a major cause for caries. The purpose of this study was to mengetahui clove extrack antibacterial effects againts Streptococcus mutans bacteria in vitro. This study is an experimental study using a post test only control group design. This research try using clove extract with a concentration of 40%, 60%, and 80%, Ciprofloxacin, aquades repetition five times. Data collected and analyzed by one-way ANOVA and post-hoc LSD test (α = 0.05). Based on the results of the statistical test to test the effects of anti-bacterial research clove extracts against Streptococcus mutans bacteria in vitro, can disimpulkaan that clove extracts have antibacterial effects in inhibiting the growth of Streptococcus mutans bacteria in vitro.further test result post-hoc LSD test shoved its inhibitory clove extract 40%, 60%, 80% smaller (p<0,05)in hibiting Streptococcus mutans in vitro compared Ciprofloxacin. <p><strong>Abstract</strong></p><p><strong>Background : </strong>Malaria is a disease caused by the <em>Plasmodium</em> parasite and is transmitted by the <em>Anopheles</em> mosquito and is still a health problem in Indonesia due to high mortality and morbidity. One form of a severe complication of malaria in addition to cerebral malaria is a function failure of the spleen. Today, the management of malaria is increasingly limited due to resistance. Therefore, further development is needed to find new innovations in malaria treatment.</p><p><strong>Purpose : </strong>The purpose of this study was to determine the effect of temulawak rhizome extract (<em>Curcuma xanthorhizza</em> Roxb.) On the level of necrosis in the spleen tissue of male BALB / c mice (<em>Mus musculus</em> L.) inoculated with <em>Plasmodium berghei</em> ANKA.</p><p><strong>Methods :</strong>Experimental research used a post-test only control group design that used five groups of mice. One group of mice was left normal while the other four groups were inoculated with <em>Plasmodium berghei</em> ANKA, positive control groups were given aquades and three treatment groups treated with temulawak extract (<em>Curcuma xanthorrhiza</em> Roxb.) With a dose of 150 mg / KgBB, 100 mg / KgBB, and 50 mg / KgBB for four day. On the fifth day an observation of the level of necrosis in the spleen organ of mice to determine the level of necrosis by histopathological examination using a light microscope.</p><p><strong>Conclusion and Result : </strong>The results of this study indicate that the administration of ginger rhizome extract (<em>Curcuma xanthorriza</em> Roxb.) Has an influence on the level of necrosis of male mice (<em>Mus musculus</em> L.) BALB / c inoculated with <em>Plasmodium berghei</em> ANKA α = 0,002 (ρ&lt;0,05), where the administration of temulawak extract can increase necrosis levels compared to the control group . Objective: Marshmallow is a soft candy loved by all levels of society, especially children. Sucrose contained in marshmallow increases the growth of Streptococcus mutans and plaque. Betel chew as Indonesian traditional plants add in marshmallow was believed to decrease S. mutans and plaque. The aim of this study was to evaluate the effect of chewing marshmallow containing betel chew in reducing S. mutans and plaque index in children. Methods: This research was a quasi-experimental with pre-test and post-test control group design. It was conducted in the Laboratory of Agricultural Chemistry, Laboratory of Agricultural Technology Department, and Islamic School of Al-Amalul Khair. Thirty students were divided into two groups. Group A was chewing marshmallow without betel chew and Group B was chewing marshmallow containing betel chew. Results: The results showed that chewing marshmallow without betel chew increased the growth of S. mutans and plaque index significantly. Chewing marshmallow without betel chew inhibited the growth of S. mutans and dental plaque formation. Conclusion: Chewing marshmallow containing betel chew reduces S. mutans and plaque index on children.

Abstract: Streptococcus mutans is the most common bacterium that causes dental caries due to its ability to ferment carbohydrates into acid resulting in the decreased pH on the tooth surface. Prevention of dental caries could be achieved by inhibiting the growth of cariogenic bacteria. Various efforts to control and prevent the cariogenic bacteria include the usage of herbal ingredients; one of them is gedi leaves (Abelmoschus manihot L.). These gedi leaves contain bioactive compounds such as flavonoids, alkaloids, steroids, and saponins. This study was aimed to prove that gedi leaf extract had inhibitory effect on the growth of S.mutans and to obtain the minimum inhibitory concentration (MIC) of this extract on the growth of S. mutans. This was a true experimental design with a randomized pretest-posttest control group design. Gedi leaf extract was obtained by maceration method in 96% ethanol. The results showed that gedi leaf extract had an antibacterial effect on the growth of S. mutans. We used turbidimetry, UV-Vis spectrophotometer, and two times of treatment to obtain the MIC of gedi leaf extract on Streptococcus mutans which was 6.25%. Conclusion: Gedi leaf extract could inhibit the growth of S. mutans with a MIC of 6.25%.Keywords: dental caries, gedi leaf extract (Abelmoschus manihot L.), Streptococcus mutans Abstrak: Streptococcus mutans merupakan bakteri yang paling banyak menyebabkan karies gigi karena bakteri ini berkemampuan memfermentasi karbohidrat menjadi asam yang berakibat turunnya pH pada permukaan gigi. Berbagai upaya dilakukan untuk mengen-dalikan dan mencegah bakteri kariogenik, antara lain dengan menggunakan bahan herbal; salah satunya yaitu tanaman gedi (Abelmoschus manihot L.). Daun gedi mengandung senyawa bioaktif antara lain flavonoid, alkaloid, steroid, dan saponin. Penelitian ini bertujuan untuk membuktikan efek inhibisi ekstrak daun gedi terhadap pertumbuhan S. mutans dan mendapatkan konsentrasi hambat minimum (KHM) ekstrak daun gedi terhadap pertumbuhan bakteri S. mutans. Jenis penelitian ini ialah eksperimental murni dengan randomized pretest-posttest control group design. Ekstrak daun gedi dibuat dengan metode maserasi dengan menggunakan etanol 96%. Hasil penelitian menunjukkan bahwa ekstrak daun gedi (Abelmoschus manihot L.) memiliki efek antibakteri dalam menghambat pertumbuhan bakteri Streptococcus mutans. Dengan menggunakan metode turbidimetri dan spektrofotometer UV-Vis dalam 2 (dua) kali perlakuan maka diperoleh KHM ekstrak daun gedi (Abelmoschus manihot L.) terhadap bakteri Streptococcus mutans terdapat pada konsentrasi 6,25%. Abstract: In addition to daily consumption, avocado is also used to treat oral diseases. Scientific studies showed that avocado seed contained flavonoids, tannins, and alkaloid which were expected to inhibit bacterial growth. This study was aimed to obtain the bacterial inhibitory effect of avocado seed (Persea americana Mill.) extract on Streptococcus mutans as the primary cause of dental caries. This was a true experimental study with the post test only control group design. This study used a modified Kirby-Bauer method with paper disk. The positive control was erythromycin and the negative control was aquadest. Avocado seeds were extracted by using maceration method with 96% ethanol. The Streptococcus mutans bacteria were obtained from pure stock of Microbiology Laboratory of Pharmacy Study Program University of Sam Ratulangi Manado. The results showed that the mean diameter of inhibition zone of avocado seed extract was 21.8 mm which was classified as very strong inhibition. Conclusion: Persea Americana Mill. seed extract had a very strong inhibitory effect on the growth of Streptococcus mutans.Keywords: avocado seed (Persea americana Mill.), Streptococcus mutans, inhibition effectAbstrak: Selain menjadi bahan konsumsi masyarakat yang lezat, ternyata alpukat telah lama dipercaya dapat mengobati penyakit di dalam rongga mulut. Di dalam buah alpukat terdapat biji yang terbukti melalui penelitian ilmiah mengandung flavonoid, alkaloid, dan tannin yang mampu menghambat pertumbuhan bakteri. Penelitian ini bertujuan untuk mengetahui daya hambat ekstrak biji buah alpukat (Persea americana Mill.) terhadap pertumbuhan Streptococcus mutans. Jenis penelitian ialah eksperimental murni dengan post test only control group design. Metode yang digunakan ialah modifikasi Kirby-Bauer dengan paper disk. Kontrol positif menggunakan antibiotik eritromisin dan kontrol negatif menggunakan akuades. Biji buah alpukat diekstraksi dengan metode maserasi menggunakan pelarut etanol 96%. Bakteri Streptococcus mutans diambil dari stok bakteri murni Laboratorium Mikrobiologi Program Studi Farmasi Fakultas MIPA Universitas Sam Ratulangi Manado. Hasil penelitian menunjukkan diameter rerata zona hambat dari ekstrak biji buah alpukat yang terbentuk ialah 21,8 mm dan digolongkan sebagai zona hambat sangat kuat. Simpulan: Ekstrak biji buah alpukat memiliki daya hambat sangat kuat terhadap pertumbuhan Streptococcus mutans.Kata kunci: biji buah alpukat (Persea americana Mill.), Streptococcus mutans, daya hambat

Background: Streptococcus mutans (S.mutans) is a gram-positive bacteria in oral cavity which produced acid as the common cause of dental caries. In order to prevent the growth or kill Streptococcus mutans, antibacterial is required. Red grape (Vitis vinifera) is suspected to contain antibacterial.Method: An experimental study using the post test only control group design is carried out to Streptococcus mutans with tube dilution method and streaking on the Brain Heart Infusion. A range of concentration from 40%, 50%, 60%, 70%, 80%, 90%, 98%. The control group are group of bacteria and red grape juice control.Result: The analysis of Minimum Inhibitor Bactericidal (MIC) is show a difference absorbent value as the given concentration (Anova, p<0,05). The correlation test show is strong reciprocal association between concentration of res grape juice and absorbent value (r=-0,978; p< 0,05). The analysis of Minimum Bactericidal Concentration (MBC) is shows difference of the number of colonies as given concentration (Anova, p<0,05). The correlation reveals a very strong reciprocal association between concentration of red grape juice and the number of Streptococcus mutans colonies growth (r=-0,925; p< 0,05).Conclusion: The conclusion is supernatant of red grape juice has effect the growth as antibacterial agent on Streptococcus mutans in vitro, with MIC at concentration 60% and MBC at concentration 80%

Papaya leaves contain several active substances that are thought to inhibit bacterial adhesion. This study aims to determine the inhibitory power of papaya leaf extraction and papaya leaf infusion to the adhesion of Streptococcus mutans bacteria to neutrophils and differences in inhibition in various concentrations. The method used is an in vitro experimental laboratory with a post-test-only control group design. The sample consisted of several concentrations of papaya leaf extraction and papaya leaf infusion (control, 25%, 50%, 75%, and 100%). The treatment group was added with neutrophil isolates and incubated for 3 hours, then exposed to Streptococcus mutans for 2.5 hours. Knowing the adhesion index can be done by calculating the average number of Streptococcus mutans attached to 100 neutrophils. The results showed successively with rounding in control, 25%, 50%, 75%, 100% papaya leaf infusion, adhesion of 9237, 8929, 7436, 3870, 3162 bacteria to neutrophils occurred. The minimum concentration of 25% papaya leaf extraction and 50% papaya leaf infusion can inhibit the adhesion of Streptococcus mutans to neutrophils. There is a difference in the ability between papaya leaf extraction and papaya leaf infusion in inhibiting the adhesion of Streptococcus mutans to neutrophils as follows: papaya leaf extraction 25% to papaya leaf infusion 25% obtained sig value ≥0.001* with a mean square of -16.38, papaya leaf extraction 50% to Papaya leaf infusion 50% got a sig value of ≥0.001* with a mean square of -42.91, 75% papaya leaf extraction to 75% papaya leaf infusion got a sig value of ≥0.001* with a mean square of -28.85 and 100% papaya leaf extraction to 100% papaya leaf infusion obtained a sig value of ≥0.001* with a mean square of -26.11.

Streptococcus mutans (S. mutans) merupakan bakteri utama penyebab karies gigi yang mampu mendemineralisasi email, menginvasi dentin yang dapat berlanjut menjadi inflamasi pulpa (pulpitis). Streptococcus mutans menginvasi inang diawali dengan melakukan adhesi (perlekatan). Proses ini merupakan langkah awal dalam proses inflamasi. Oleh karena itu, adhesi Streptococcus. mutans pada sel inang perlu dicegah agar tidak terjadi infeksi oleh bakteri. Neutrofil merupakan sel pertahanan pertama yang datang pada proses inflamasi. Salah satu upaya untuk mencegah terjadinya adhesi tersebut bisa menggunakan bahan alami yang bersifat anti-inflamasi, salah satunya yaitu daun wungu. Penelitian ini bertujuan untuk  mengetahui pengaruh pemberian ekstrak daun wungu (EDW) terhadap adhesi  bakteri  Streptococcus. mutans pada neutrofil dan perbedaan indeks adhesi dalam berbagai konsentrasi dengan eksperimental laboratoris menggunakan the post test only control group design. Sampel terbagi menjadi 5 kelompok (klp): klp I/kontrol (tanpa inkubasi EDW), klp II (EDW 3,125%), klp III (EDW 6,25%), klp IV (EDW 12,5%), dan klp V (EDW 25%). Isolat neutrofil diinkubasi dengan EDW selama 3 jam, kemudian dipapar S. mutans selama 4 jam. Indeks adhesi dihitung berapa rata-rata jumlah S. mutans  yang menempel pada 100 neutrofil. Simpulan penelitian menunjukkan bahwa ekstrak etanol daun wungu berpotensi menurunkan indeks adhesi Streptococcus mutans. Kata kunci: adhesi, S. mutans, neutrophil, daun wungu

Abstract: Dental caries is still a health problem in Indonesia. There are several factors that play some important roles in the occurence of caries, as follows: microorganism, host, food, and time. Streptococcus mutans is one of the microorganisms that cause caries. Squid ink contains melanin which has an active compound to inhibit microbial activity. This study was aimed to determine the inhibitory effect of squid ink extract (Loligo sp) on the growth of Streptococcus mutans. This was a laboratory experimental study, with a post test only control group design. The results showed that the mean diameter of the inhibitory zones of the squid ink extract (Loligo sp) was 10.50 mm which was categorized as strong inhibition (Davis and Stout criteria). In conclusion, the squid ink extract (Loligo sp) had a strong inhibitory effect on the growth of Streptococcus mutans bacteria.Keywords: squid ink extract (Loligo sp), Streptococcus mutans, zone of inhibitionAbstrak: Karies gigi merupakan salah satu masalah kesehatan gigi dan mulut di Indonesia. Penelitian ini bertujuan untuk mengetahui daya hambat ekstrak tinta cumi-cumi (Loligo sp) terhadap pertumbuhan bakteri Streptococcus mutans. Jenis penelitian ialah eksperimental laboratorik, dengan post test only control group design. Hasil penelitian menunjukkan diameter rerata zona hambat dari ekstrak tinta cumi-cumi (Loligo sp) sebesar 10,50 mm dan digolongkan dalam kategori kuat (kriteria Davis dan Stout).
