Background: Enterococcus. faecalis (E. faecalis) and Fusobacterium nucleatum (F. nucleatum) are the most common bacteria found in infected tooth root canal. Most of these bacteria often cause failure in endodontic treatments. Pluchea indica Less leaf is a species of plants that has several chemical properties. It consists of flavonoids, tannins, polyphenols, and essensial oils which have been reported as antibacterial agents. Because of its benefits, the extract of Pluchea indica Less leaves may be potentially developed as one of root canal sterilization dressing. Purpose: This study aimed to determine antibacterial activity of Pluchea indica Less leaves extract against E. faecalis and F. nucleatum bacteria. Method: Dilution method was conducted first to show Minimum Inhibitory Concentration (MIC) of the extract against E. faecalis and F. nucleatum. The antibacterial activity test on Pluchea indica Less leaves extract was performed on E. faecalis and F. nucleatum bacteria using agar diffusion method. The Pluchea indica Less leaves extract used for antibacterial activity test was at a concentrations of 100%, 50%, 25%, 12.5%, and 6.25%. Thirty-five petridiscs were used and divided into five groups based on the extract concentration. Result: The results showed strong and moderate antibacterial effects of the Pluchea indica Less leaves extract on E. faecalis at the concentrations of 100% and 50%, while on F. nucleatum only at the concentration of 100% with moderate effect. Conclusion: Pluchea indica Less leaves extract has antibacterial activity against E. faecalis and F. nucleatum bacteria with strong-moderate effect.

Because of its benefits, the extract of Pluchea indica Less leaves can be potentially developed as one of  sterilization dressing in root canal teeth. <strong>Purpose: </strong>The aim of this study was to determine biofilm formation inhibition of Pluchea indica Less leaves extract against Enterococcus faecalis and Fusobacterium nucleatum. <strong>Materials and Methods: </strong>The dilution method was done first to show the Minimum Inhibitory Concentration (MIC) of the extract. The inhibition biolfilm formation was tested using microtitter plate assay by measuring the bacterial biofilm Optical Density (OD) from ELISA reader’s results and using autoagregation assay to show the inhibition of adherance bacteria.  The Pluchea indica Less leaves extract concentration used for inhibition of biofilm formation were 100%, 50%, 25%, 12,5%, and 6,25%. <strong>Results:</strong> The result of biofilm formation inhibition showed that Pluchea indica Less leaves extract were able to inhibit Enterococcus faecalis and Fusobacterium nucleatum’ biofilm formation with strong moderate effect. Crude methanol extract of 9 Vietnamese plants were in vitro screened for their antibacterial activity against three common freshwater fish pathogens including Aeromonas hydrophila, Edwardsiella ictaluri, and Streptococcus agalactiae. Agar disc diffusion method was used to evaluate the antibacterial activity, then solvent extract was performed for the extracts which exhibited the strongest and a broad-spectrum antibacterial activity. Minimal inhibitory concentration (MIC) was conducted for effective plant extracts using broth dilution method. The results indicated that most of the plant extracts exhibited antibacterial propeties to at least one tested bacterium. Headache tree (Premna corymbosa), bushwillows (Combretum quadrangulare) and Celandine spider flower (Cleome chelidonii) showed a broad-spectrum antibacterial activity. The largest inhibitory zones of 35 mm and 21 mm were observed  for the extract of Premna corymbosa against E. ictaluri and S. agalactiae, respectively. E. ictaluri was found to be the most susceptible for all of the extracts while A. hydrophila was the most resistant. The MIC of effective plant extracts against tested bacteria ranged between 0.39 mg/mL and 3.125 mg/mL. The result can be considered for further investigation of the development of an alternative therapy against bacterial infection in aquaculture.

Microbial resistance to antibiotics has become a problem plaguing the world. Currently, interest has been focused on exploring antimicrobial properties of plants and herbs. This work aim to evaluate the antibacterial activity of garlic (Allium sativum) bulbs and ginger (Zingiber officinale) rhizome on Brucella abortus isolates. Some concentrations of garlic and ginger extracts were tested for their antibacterial activity against B. abortus isolate brought from Central Veterinary Research Laboratory (CVRL), Soba, using well diffusion method. Moreover, minimum inhibitory concentration (MIC) and minimum lethal concentration (MLC) of garlic and ginger were tested using broth dilution method. Sensitivity pattern of the conventional antibacterial against common pathogenic bacteria was tested using disc diffusion method. Aqueous extract of ginger produced dose-dependent increase in the zone of inhibition at a concentration of 15% and higher, whereas the garlic extract produced inhibition zone at a concentration of 5% and higher, i.e. B. abortus isolate showed relatively high sensitivity toward garlic extract than ginger which required a more concentrated extract to kill or inhibit B. abortus isolate that brought from (CVRL), Soba, Khartoum, Sudan. Further studies are needed to find out the efficacy, safety, and kinetic data of their active ingredients.

Aim and Objectives: To compare and evaluate the antibacterial efficacy of various nanoparticles incorporated in orthodontic primer with that of conventional antimicrobial agents at different concentrations on Streptococcus mutans ( S. mutans) strain. Materials and Methods: Transbond XT Primer was mixed with 2.5% and 5% benzalkonium chloride (BAC), 0.2% and 2.5% chlorhexidine, 1% and 3% titanium dioxide (TiO2) nanoparticles, 0.2% and 0.5% nanohydroxyapatite, and 0.2% and 0.5% silica-doped nanohydroxyapatite powders. Antibacterial activity against S. mutans for all the materials was evaluated by the disk diffusion method for periods of 48 (T1) and 72 (T2) hours. Results: There was a significant increase in the antimicrobial activity of the orthodontic primer modified by the addition of antibacterial agents. The highest zone of inhibition against S. mutans was observed for silica-doped nanohydroxyapatite of 0.5% (11.03 mm) among all the nanoparticles, which was similar to the conventional antibacterial agents used in our study. Conclusions: • Among all the groups, BAC at 5% concentration showed the highest antimicrobial activity, and the least activity was exhibited by 1% TiO2 nanoparticles. • Silica-doped nanohydroxyapatite at 0.5% expressed the greatest antibacterial activity among all the nanoparticles. • All the materials showed sustained antibacterial activity even after 72 hours.

Aim: Cough due to Klebsiella pneumoniae and Streptococcus pneumoniae is currently managed by conventional antibiotics and herbal extracts in Uganda. However, much as these herbal extracts are extensively used, their antibacterial activity is not known. This study aimed at determining the antibacterial activity of the selected locally prepared herbal cough extracts against two bacterial strains i.e. Klebsiella pneumoniae (ATCC 700603), and Streptococcus pneumoniae (ATCC 49619). Methods: The herbal cough extracts were screened for antibacterial activity using Agar-well diffusion method for determining zone of inhibition, macro broth dilution method for Minimum Inhibitory Concentration (MIC) determination and Streak plate method for Minimum Bactericidal Concentration (MBC). Results: In vitro evaluation of antibacterial activity of the 5 brands of herbal cough extracts against K. pneumoniae and S. pneumoniae   revealed that all extracts possessed significant antimicrobial effects against all microorganisms tested (p < 0.05). However, MM04 (35.6±0.0) mm and MM03 (33.6±1.5) mm had maximum zones of inhibition as compared to other herbal extracts against K. pneumoniae and S. pneumoniae respectively. Average MIC results for extracts against K. pneumoniae   indicated that MM01 had the highest MIC (2.5000 mg/ml) while MM03 had the least MIC (0.0625 mg/ml). Average MIC results for extracts against S. pneumoniae showed MM01 had the highest MIC (2.0000 mg/ml) while MM03 3 had the least MIC (0.0438 mg/ml). Average MBC results for extracts against K. pneumoniae indicated that MM01 had the highest MBC (4.000 mg/ml) while MM03 had the least MBC (0.030 mg/ml). Average MBC results for extracts against S. pneumoniae showed MM01 had the highest MBC (4.000 mg/ml) while MM03 had the least MBC (0.033 mg/ml). Conclusion: The results obtained in present study were revealed that locally prepared herbal extracts had significant antibacterial activity. Hence they can be used as promising alternatives of antibiotics used against Respiratory Tract Infections due to K. pneumoniae and S. pneumoniae.

Honey is one of the natural treatments with antibacterial effects such as hydrogen peroxide, low pH and short water activity that may inhibit bacterial growth. This study aimed to determine the activity of honey as antibacterial to bacteria Escherichia coli. Antibacterial activity test was done in vitro through diffusion method by using disk disc in MHA media then measuring the diameter of a clear zone that inhibition zone form growth of Escherichia coli bacteria and scaled it millimetre unit. This research was done in the laboratory of the Faculty of Medicine Universitas Kristen Indonesia. The concentrations used are 10%, 30%, 50%, 70%, 100% by using sterile equates as honey diluents. The control used was chloramphenicol, the results obtained at a concentration of 10% with a zone of 7.6 mm inhibition of honey had been able to inhibit the growth of Escherichia coli bacteria, and the optimum concentration was 100% with a zone of 24.5 mm. This test showed that natural honey could inhibit bacteria, which conclude that honey might be used as alternative medicine in society. Keywords: Antibacterial effect, Escherichia coli, honey

Anogeissus leiocarpus (DC.) Guill. & Perr. (Combretaceae) et Combretum collinum Fresen. (Combretaceae) sont deux plantes de la flore ivoirienne couramment utilisées pour traiter plusieurs pathologies telles que les affections cutanées, le paludisme et la fatigue générale. Ce travail s’inscrit dans le cadre de la valorisation de cette flore. Il vise donc à évaluer l’activité antibactérienne des extraits méthanoliques bruts des feuilles de Combretum collinum et des écorces de racines de Anogeissus leiocarpus sur la croissance "in vitro" des souches de Staphylococcus aureus résistantes à la méticilline. La méthode de diffusion en milieu gélosé (solide) a été utilisée pour évaluer la sensibilité des souches bactériennes aux extraits et à déterminer les concentrations minimales inhibitrices (CMI), Quant au screening phytochimique, il a été réalisé par une chromatographie sur couche mince (CCM). La concentration minimale inhibitrice est comprise entre 0,62 mg/ml et 1,25 mg/ml pour Anogeissus leiocarpus et est de 0,325 mg/ml pour Combretum collinum. Le screening phytochimique a révélé la présence de flavonoïdes et de tanins dans l’extrait méthanolique de Anogeissus leiocarpus alors que dans l’extrait méthanolique de Combretum collinum, ce sont des acides phénoliques qui ont été mis en évidence. Il ressort donc que ces deux plantes sont dotées d’un important pouvoir antibactérien et contiennent plusieurs composés chimiques. Elles pourraient donc constituer des voies de prospection pour la recherche de nouvelles molécules antibactériennes en réalisant une étude bio-guidée des extraits bruts et en évaluant leur cytotoxicité sur des cellules hépatiques saines.Mots clés: Antibactérienne, plantes médicinales, flore ivoirienne, extraits methanoliques English Title: Phytochemical screening of methanolic extracts from leaves of Combretum collinum and roots of Anogeisus leiocarpus and in vitro antibacterial effect on multiresistant strains of Staphylococcus aureus Both Anogeissus leiocarpus (DC.) The current study consists of the valorization of this flora, and aims therefore at revealing antibacterial activity of the methanol crude extracts obtained from stem bark and leaves of the respective plants, against Methicillin-resistant Staphylococcus aureus strains. Agar gel diffusion method was used to assess bacteria susceptibility, and broth dilution method allowed determination of minimum inhibition concentrations (MICs). These parameters varied from 0.62 mg/ml to 1.25 mg/ml for Anogeissus leiocarpus, while they reached 0.325 mg/ml for Combretum collinum. In addition, phytochemical screening brought about flavonoids and tanins for the plant, and phenolic acids for the other one. In conclusion, these named plants contain several chemical compounds with antibacterial properties, and could be of great interest in the search of new molecular compounds provided with antibacterial activity through bio-guided experiments. Furthermore, they could best hold value by testing those methanol extracts over hepatic sane cells for cytotoxicity assessment. Nymphaea pubescens Willd telah diketahui mempunyai efek antibakteri, terutama pada biji dan daunnya. Tetapi sampai saat ini bagian bunga dari tanaman tersebut belum dilakukan pengujian aktivitas antibakteri. Tujuan dari penelitian ini untuk melakukan skrining fitokimia, uji aktivitas antibakteri dan penentuan konsentrasi hambat minimum (KHM) dari ekstrak metanol bunga N. pubescens terhadap S. aureus dan E. coli. Proses ekstraksi bunga N. pubescens dimaserasi menggunakan pelarut metanol dengan perbandingan 1: 4 b/v. Metode pengujian yang digunakan ada dua yaitu metode difusi untuk pengujian aktivitas antibakteri dan metode dilusi untuk pengukuran konsentrasi hambat minimum (KHM). Hasil skrining fitokimia didapatkan bahwa ekstrak metanol bunga N. pubescens mengandung senyawa golongan fenolik, saponin dan flavonoid. Hasil pengujian aktivitas antibakteri menunjukkan bahwa ekstrak metanol bunga N.pubescens mampu menghambat S. aureus (diameter hambat 10 ± 0,29 mm) dan E.coli (diameter hambat 10,2 ± 0,50 mm). Konsentrasi hambat minimum dari ekstrak metanol bunga N. pubescens terhadap S.aureus sebesar 12,5% dan terhadap E.coli sebesar 25%. Dapat disimpulkan bahwa ekstrak metanol bunga N. pubescens memiliki aktivitas sebagai antibakteri. Nymphaea pubescens Willd has known to have antibacterial effects, especially on the seeds and leaves. However, until now the flower of the plant has not been tested for antibacterial activity. The purpose of this study was to perform phytochemical screening, antibacterial activity test and determine the minimum inhibitory concentration (MIC) of the methanol extract of N. pubescens flowers against S. aureus and E. coli. The process of extracting N. pubescens flowers is macerated using methanol as a solvent with a ratio of 1: 4 w / v. There are two test methods used, namely the diffusion method for testing antibacterial activity and the dilution method for measuring the minimum inhibitory concentration (MIC). The results of phytochemical screening showed that the methanol extract of N. pubescens flowers contained phenolic compounds, saponins, and flavonoids. The results of the antibacterial activity test showed that the methanol extract of N.pubescens flowers was able to inhibit S. aureus (inhibition diameter 10 ± 0.29 mm) and E. coli (inhibitory diameter 10.2 ± 0.50 mm). The minimum inhibitory concentration of the methanol extract of N. pubescens flowers against S.aureus was 12.5% and against E. coli was 25%.            The purpose of this research was to identified the compounds in ethanol extract of soursop flower and determined the antibacterial activity of the soursop flower extract on Salmonella enteritidis. The phytochemistry screening was done to identify the secondary metabolite compounds of this extract. The concentrations used of ethanol extract of soursop flower were 10%, 20%, 30%, 40%, 50%, with ampicillin 10 μg/disc as the positive control and 10% Dimethyl Sulfoxide (DMSO) as the negative control. The antibacterial activity test of soursop flower ethanol extract against Salmonella enteritidis was held in vitro using the paper disc diffusion method by measuring the diameter of the inhibition zone. The phytochemistry screening showed that the ethanol extract of soursop flower had secondary metabolites such as alkaloids, phenolic, and flavonoid. The result of the antibacterial activity test showed that there was no inhibition zone (bright zone) at various concentrations. So, it can be concluded that soursop flower ethanol extract (Annona muricata L.) contained the secondary metabolites such as alkaloids, phenolic, and flavonoid, and also this extract had no antibacterial activity against Salmonella enteritidis

Background: View to its interesting role in the peptidoglycan biosynthesis pathway the enzyme UDP-N- acetylglucosamine enolpyruvyl transferase is an attractive target to develop new antibacterial agents, it catalyzes the first key step of this pathway and its inhibition leads to the bacterial cell death. Fosfomycin is known as the natural inhibitor of MurA. Objective: Call new inhibitors of MurA by virtual screening of different chemical compounds libraries, and test the best scored “virtual hits” against three pathogenic bacteria: Escherichia coli, Bacillus subtilis, and Staphylococcus aureus. Methods: A Virtual screening of the structural analogues of fosfomycin downloaded from PubChem database was performed on one side and of the French National Chemical Library as well as using ZINC database to identify new structures different from fosfomycin on the other, FlexX was the software used for this study. The antibacterial testing was divided into methods: disk diffusion and broth dilution. Results: A set of virtual hits was found with better energy score than that of fosfomycin, seven between them were tested in vitro. Therefore, disk diffusion method explored four compounds exhibited antibacterial activity: CID-21680357 (fosfomycin analogue), AB-00005001, ZINC04658565, and ZINC901335. The testing was continued by broth dilution method for both compounds CID-21680357 and ZINC901335 to determine their minimum inhibitory concentrations, and ZINC901335 had the best value with 457µg/ml against Staphylococcus aureus. Conclusion: Four compounds were found and proven in silico and in vitro to have antibacterial activity: CID-21680357, AB-00005001, ZINC04658565, and ZINC901335.
