Abstract Cellulolytic bacteria is one of the beneficial bacteria that can found in mangrove ecosystem. The purposes of study were to analyse the cellulolytic index, and to analyse the cellulase activity of bacteria isolated from soil mangrove. Qualitatively, assessment of cellulase activity were carried out at the Microbiology laboratory of Fish Quarantine Station, Quality Control and Safety of Fishery Products (SKIPM) Aceh, while quantitatively was observed in microbiology laboratory, Biology Department, IPB. Assessment of qualitative cellulase activity is performed by growing the selected pure isolate on 1% CMC medium then spilled 1% congo red to test its cellulolytic potential. Cellulolytic potential was determined by clear zone performed around the colony after congo red flooded. The quantitative cellulase enzyme activity test carried out by DNS method tested on one selective isolate. There were 21 from 39 isolates showed a clear zone isolated from mangrove soil. The cellulolytic index (CI) obtained ranged from 0.07 to 0.80 classified as low cellulolytic index criteria. The cellulolytic index was higher in bacteria isolated from mangrove rehabilitated than mangrove unrehabilitated. The highest cellulase activity and specific cellulase activity of BTMD32 was at 48 hours with the value were 0.0012 U/ml, 0.077 U/mg, respectively. The result concluded that the bacteria cellulolytic isolated from mangrove soil had low cellulolytic index, low cellulase activity, and low specific cellulase activity.

The purpose of this reasearch was to determine the effect of storage period goat’s milk kefir (GMK) on total LAB, total mineral levels included Ca and Mg and antimicrobial activity to pathogenic microbes. The materials used was GMK. This research was conducted using Completely Randomized Design with 4 treatments and 4 replications. The treatments were storage period that consist of D0 (0 day), D1 (7 days), D2 (14 days), and D3 (21 days). Total calculation of LAB used TPC (Total Plate Count). Samples were diluted from 10-1 to 10-8, then incoculated PCA medium by using pour plate method. Petri dish was incubated at 37ºC for 48 hours. Calcium and magnesium levels were calculed base AAS method. The result of this study was analyzed using analysis of variance (ANOVA), then followed by Duncan’s Multiple Range Test (DMRT). The storage time treatment has a significant effect on the total microbial count, mineral levels (Ca and Mg) and inhibition zone of Staphylococcus aureus, Escherichia coli and Salmonella enterica serovar typhi on goat's milk kefir. Average clear zone Staphylococcus aureus 2.60 – 3.18 mm. Average inhibition zone Escherichia coli 2.09 – 3.32 mm. Average clear zone Salmonella enterica serovar typhi 1.97 – 2.72 mm.

Characteristic features of district cloud-free zones and their day to day evolution along Indian coasts as observed in INSAT cloud imageries during southwest monsoon months  have been analysed and discussed along with sea surface and surface air temperatures and monsoon condition situation. It was a weak or break monsoon condition. Appearance of early morning clear zone just off shore along Indian Peninsula coasts is attributed to the gradual lowering of sea surface temperate due to upwelling caused by persistent favorable surface wind and slow-setting of air above colder water. With the advance of the day, wide extension of clear area over water where it ends abruptly and propagation of front-like zone inland manifest as a typical sea breeze. It is postulated that this is the effect of sea breeze circulation and shrinking of air above colder water. It is inferred that time-to-time appearance of such phenomenon may be an Indication of longer weak or break monsoon over the Peninsula.

Lipase is widely applied in various fields of industries elevating its economic value. The demand for lipase keeps increasing open up working opportunities. Isolates showing lipolytic activity obtained previously are tested for their activity on different pH mediums: 7, 8, 9, and 10; and at different temperatures. The fungi are grown on SDA medium supplemented with Olive Oil, emulsifier, Tween 80, and Rhodamine B. The experiment was made in 3 replicates and is incubated at room temperature of 30oC. Lipase activity was calculated based on the clear zone around colonies observed on day 2 and day 3 after inoculation. The result shows that those isolates are highly active on various pH at RT, and the activity slightly reduces at 30oC. This result suggests that a wide range of their applications are at room temperature when pH is a limiting factor for the applications

AbstractBitter gourd as a traditional medicine has a chemical content in its leaves that are useful as antimicrobials and as antioxidants. According to research by Tessa Undap, 2017 shows that bitter melon leaves have an inhibitory power against the growth of Staphylococcus aureus bacteria by inhibiting protein synthesis. The purpose of this study was to determine the antibacterial activity of bitter melon leaf extract (Momordica charantia L.). The research method used in this study is the disc diffusion method. Data was collected by measuring the diameter of the clear zone formed in each treatment. The antibacterial results showed that the 9% extract concentration had the highest inhibitory power.Keywords: Antibacterial, Pare Leaf Extract, Granules, Staphylococcus aureus. AbstrakTanaman pare sebagai obat tradisional memiliki kandungan kimia dalam daunnya yang bermanfaat sebagai antimikroba dan sebagai antioksidan. Menurut penelitian oleh Tessa Undap, 2017 menunjukkan bahwa daun pare mempunyai daya hambat terhadap pertumbuhan bakteri Staphylococcus aureus dengan cara menghambat sintesis protein. Adanya kandungan flavonoid dalam daun pare dapat menghambat pertumbuhan bakteri Staphylococcus aureus yang dapat mengkoagulasi protein pada sel bakteri. Tujuan dari penelitian ini yaitu untuk mengetahui aktivitas antibakteri yang terkandung dalam ekstrak daun pare (Momordica charantia L.). Metode penelitian yang digunakan dalam penelitian ini yaitu metode difusi cakram disk. Pengumpulan data dilakukan dengan cara mengukur diameter zona bening yang terbentuk pada setiap perlakuan. Hasil antibakteri yaitu menunjukkan bahwa konsentrasi ekstrak 9% mempunyai daya hambat paling tinggi.Kata kunci: Antibakteri, Ekstrak Daun Pare, Granul, Staphylococcus aureus.

AbstractChemical constituents contained in the beluntas plant, including alkaloids, essential oils, flavonoids, phenols and tannins have the ability to inhibit the growth of Staphylococcus aureus bacteria. This research uses maceration extraction method. Thick extracts of flowers and leaves were used with a series of extract concentrations. The purpose of this study was to determine differences in the effectiveness of the ethanol extract of the leaves and flowers of beluntas against the growth of Staphylococcus aureus bacteria. The research method used in this study is the disc diffusion method. Data was collected by measuring the diameter of the clear zone formed in each treatment. Extract concentrations used 5%, 7% and 9%. The extracts of flowers and leaves of beluntas (Pluchea indica (L.) In order to improve the safety design of roadside areas and reduce the loss of roadside accidents, this paper uses PC-Crash software to perform an accident simulation analysis. By recording the track of the vehicle after entering the roadside, the recommended widths of the roadside clear zone for different operating speeds and horizontal curve radii in straight and curved sections are given. According to our previous research data, the conditions for setting the roadside clear zone are proposed. Finally, based on a cost-benefit ratio analysis, a comprehensive risk index method is adopted to evaluate the social stability risk of the project and conduct research on the design method of the roadside clear zone. The results show that the width of the roadside clear zone has an exponential relation with the departure speed and a power relation with the horizontal curve radius. The research results realize the accurate calculation of the roadside clear zone width and fill in the gaps of the relevant specifications and guidelines in the setting conditions of the roadside clear zone.

Background: Dadiah is a traditional dish from West Sumatra made from buffalo milk, which is fermented in bamboo tubes and left at room temperature for ±2 days. Dadiah is included in the staple food category because it contains Lactic Acid Bacteria (LAB) which has the potential to be a probiotic. This study aims to determine the identification and characterization of LAB from Dadiah from Halaban, Kab. Fifty Cities, West Sumatra. Design and Methods: A survey method was used in this research with a descriptive analysis, Antimicrobial activity testing was done with bacteria Escherichia coli O157, Staphylococcus aureus, Listeria monocytogenes, and Listeria innocua. Molecular identification was done using the 16S rRNA gene. Results: Probiotic candidate test with the best results in testing for resistance to stomach acid at pH3 with the viability of 65.98%, bile salt resistance 0.3%, viability of 54.90% from 2DA isolates. Antimicrobial activity with the best clear zone area results was obtained in 2DA isolates with Escherichia coli O157 test bacteria of 21.16 mm, Staphylococcus aureus with a clear zone area of 23.17 mm, Listeria innocua of 19.24 mm and Listeria monocytogenes with a clear zone area 18.23 mm in 4DA isolate, LAB identification using 16S sRNA gene, results of running PCR base length 1419bp. Conclusions: Phylogenetic analysis shows that Dadiah of Limapuluh Kota Regency is a kin to Lactobacillus plantarum. The superiority of identification technology by using 16S rRNA gene only can be conducted if the nucleotide sequence information of the targeted bacteria is known beforehand.

The purpose of this research is to obtain hydrocarbon degrading bacteria that work synergistically in a consortium. Consortium microorganisms is mixture of microbial populations in the form of communities that have mutualistic relationships and doesn’t inhibition the growth of other microbes. In this study, isolates were obtained from the rhizosphere of soil contaminated with petroleum. The isolates obtained were tested for synergism to determine the relationship between bacterial isolates. Synergism testing was carried out using the spread plate method on agar media. The results of this study showed that isolate number one showed antagonistic properties to other bacterial isolates by forming a clear zone around the disc paper. A total of eight bacterial isolates showed the greatest percentage of synergism, namely ≥ 80% so that the eight rhizosphere bacterial isolates could be used as materials for mixed culture.

<p><strong>Plant Root Growth Promoting Bacteria or PGPR are bacteria that colonize plant rooting areas or the rhizosphere to increase the quality or quantity of plant growth. PGPR can provide dissolved phosphate for plants to be absorbed by the plant root system since 95-99% of phosphate in nature is present in insoluble form. Soil samples from different irrigation systems from Akar-akar village were isolated through serial dilution techniques and grown on NA media shown that the higher the dilution, the fewer colonies that grew on the media. Macroscopic and microscopic observations were made to see the criteria for bacteria, shown that the bacteria in the three soil samples were gram negative and was known that bacillus and cocobacillus bacteria present in the three soil samples. Coccus bacteria was found in soil without irrigation and drip-surface irrigated soil, while streptococcus bacteria was found in drip-surface and drip-subsurface irrigated soil. As many as 22 bacterial isolates were isolated and grown on Pikovskaya media, only one bacterial colony was phosphate solvent through a clear zone that grew around the bacterial colony. The bacterial colony has a phosphate dissolving index (IPF) of 250, the ratio between colony diameter and clear zone diameter was 1: 1.5.</strong></p><p><strong>Keywords – </strong><em>PGPR, Drip-surface irrigated soil, Drip-subsurface irrigated soil</em></p>
