Anthocyanins, the naturally occurring pigments responsible for most red to blue colours of flowers, fruits and vegetables, have also attracted interests because of their potential health effects. With the aim of contributing to major insights into their structure-activity relationship (SAR), we have evaluated the radical scavenging and biological activities of selected purified anthocyanin samples (PASs) from various anthocyanin-rich plant materials: two fruits (mahaleb cherry and blackcurrant) and two vegetables (black carrot and ‘Sun Black’ tomato). PASs from the above-mentioned plant material have been evaluated for their antioxidant capacity, using TEAC and ORAC assays. In human endothelial cells, we analysed the biological activity of different PASs by measuring their effects on the expression of endothelial inflammatory markers, including endothelial adhesion molecules VCAM-1 and ICAM-1. We demonstrated that all the different PASs showed biological activity. They exhibited antioxidant capacity of different magnitude, higher for samples containing non-acylated anthocyanins (typical for fruits) compared to samples containing more complex anthocyanins acylated with cinnamic acid derivatives (typical for vegetables), even though this order was slightly reversed when ORAC assay values were expressed on molar basis. Concordantly, PASs containing non-acylated anthocyanins reduced the expression of endothelial inflammatory antigens more than samples with aromatic acylated anthocyanins, suggesting the potential beneficial effect of structurally diverse anthocyanins in cardiovascular protection.

AbstractLuteolin is a flavonoid found in a wide range of plant materials, including commonly eaten fruits and vegetables. It displays a wide range of biological activities but is known to have poor bioavailability. In this study, ten different mono-acyl (nine 5-O-acyl and one 7-O-acyl) derivatives of luteolin were synthesised for the purpose of improving bioactivity and bioavailability, and therefore enhance their therapeutic potential. The antiproliferative activity of these derivatives was assessed against the HCT116 colon cancer and MDA-MB-231 breast cancer cell lines using a 3[H] thymidine incorporation assay. The radical scavenging activity of these derivatives against 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical cation and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical using Trolox as a standard, was also assessed. Some of these derivatives were found to have improved antiproliferative activity with comparable radical scavenging activity compared to luteolin. Increased lipophilicity has been shown to increase the bioavailability of flavonoids implying these analogues will also have increased bioavailability.

Quercetin is a flavonoid that is found in many plant materials, including commonly eaten fruits and vegetables. The compound is well known for its wide range of biological activities. In this study, 5-O-acyl derivatives of quercetin were synthesised and assessed for their antiproliferative activity against the HCT116 colon cancer and MDA-MB-231 breast cancer cell lines; and their radical scavenging activity against the 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical cation and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical species. Four derivatives were found to have improved the antiproliferative activity compared to quercetin whilst retaining radical scavenging activity.

Herbal medicines has been the most cost-effective and valuable medical practice to cure diseases and emphasize modern health care treatment. The present research was conducted to assess the biological activities of 10 fractions obtained from methanolic extract which was derived from dried seeds of Sesamum indicum. Antioxidant activity was assessed using DPPH radical scavenging, total antioxidant capacity and total reducing power assays. Highest free radical scavenging activity (80.3 ± 1.36%), total antioxidant capacity (104.7 ± 4.04 μg AAE/mg) and ferric reducing power activity (238.76 ± 1.23 μg AAE/mg) was shown by fraction SE. Fraction SE showed the highest phenolic contents (63.72 ± 1.5 μg GAE/mg) while fraction SG sample showed highest flavonoid contents (54.62 ± 2.61 μg QE QE/mg). Antibacterial activity was performed against four selected bacterial strains including Staphylococcus aureus, Escherichia coli, Enterobacter aerogens and Bacillus subtilis. Highest inhibition was shown by fraction SD (11 ± 1.04 mm) against Staphylococcus aureus, fraction SH against Bacillus subtilis (11 ± 1.06 mm) and fraction SB against Escherichia coli. All fractions were found inactive against the selected fungal strains. While performing antileishmanial activity, fraction SC showed highest percent mortality (78%) of Leishmania tropica. In brine shrimp lethality bioassay, fraction SG showed significant LD50 value (23.48 μg/mL).

Casein hydrolysates (CH) were prepared using papain and modified by the plastein reaction (CH-P) in the presence of extrinsic phenylalanine (CH-P-Phe) or tryptophan (CH-P-Trp). The in vitro protective activity of CH and its modified products against ethanol-induced damage in HHL-5 cells was investigated. The results showed that the modification by the plastein reaction reduced the amino group content of CH. However, the modification by the plastein reaction in the presence of extrinsic amino acids could enhance the antioxidant, proliferative, cell cycle arresting, and anti-apoptosis activity of CH. Biological activities of CH and its modified products in the HHL-5 cells varied depending on the hydrolysate concentration (1, 2, and 3 mg/mL) and treatment time (24, 48, and 72 h). Generally, higher biological activities were found after cell treatment with CH or its modified products at concentration of 2 mg/mL for 48 h compared to other treatments. In addition, CH modified in the presence of tryptophan (CH-P-Trp) showed higher biological activity than that modified in the presence of phenylalanine (CH-P-Phe). Based on the obtained results, it can be concluded that casein hydrolysates with enhanced biological activity and potential health benefits can be produced by papain and the plastein reaction with the incorporation of extrinsic amino acids.

Different parts of Bellis perennis were subjected to hydrodistillation and the products were subsequently analyzed by GC and GC/MS. Aqueous and methanol extracts were prepared from the aerial parts. Antioxidant [1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging, reducing activity, total antioxidant] and antimicrobial activity of the plant materials were studied. The aqueous extracts showed higher DPPH scavenging activity (85.8% at 102.5 μg/mL) than the methanol extract. Reducing power was also observed for both tested extracts, where the formation of linoleic acid peroxides was more for the aqueous extract than the methanol extract. Antimicrobial activities of all plant materials were evaluated at varying concentrations against a panel of pathogens, but without pronounced inhibitory effect.

Recently, the use of bioactive compounds in improving human health has received more attention. The aim of the present study was to hydrolyze orange seed proteins using pepsin enzyme to obtain bioactive peptides as well as to study the stability of such activity after simulated gastrointestinal digestion conditions. The method was optimized using different enzyme concentrations from 1% to 3%, hydrolysis times between 2 and 5 h, and an optimal temperature of 33 °C. Biological activities including α-glucosidase inhibition, α-amylase inhibition, Angiotensin I-Converting Enzyme (ACEI) inhibition, ferric reducing antioxidant power, and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity were evaluated. According to the results, a significant higher value of the biological activity (p < 0.05) was observed using an enzyme ratio of 0.03 E/S and hydrolysis time of 3.5 h. After size-exclusion chromatography separation, fractions 45–49 and 50–54 showed the highest biological roles such as antioxidant, ACEI inhibitory, and hypoglycemic. Fractions with the highest biological activity were purified using RP-HPLC and analyzed using nano-liquid chromatography and mass spectrometry. The results obtained after simulated gastrointestinal digestion indicated that peptide fractions obtained after chromatographic separation significantly maintain their activity.

AbstractA blueberry anthocyanin extract was obtained from Brightwell blueberry fruits cultivated in eastern China and the extraction and purification conditions were optimized. The components of the anthocyanin extract were identified using ultra-performance liquid chromatography-electrospray ionization interface-mass spectrometer. The antioxidant and antibacterial activities of the blueberry fruit supernatant (BFS), blueberry anthocyanin crude extract (BCE), and blueberry anthocyanin rich extract (BRE) were evaluated. The extraction yield was 1.79 ± 0.0014 mg/g under the following optimal conditions: 1:20 solid-to-liquid ratio (v/w), 24 h, 34 °C, and 90% ethanol containing 0.21% (v/v) hydrochloric acid. With regard to purification, anthocyanin purity increased 19.1-fold. Nine fractions were identified as the glycosides of delphinidin, cyanidin, petunidin, and malvidin. The biological activities of the blueberry anthocyanin extract were improved through extraction and purification. Compared with BFS and BCE, BRE had a higher DPPH radical scavenging activity (EC50 = 0.51 mg/mL), ABTS antioxidant capacity (EC50 = 0.32 mg/mL), and oxygen radical absorbance capacity (0.43 mmol Trolox/g). Furthermore, BRE (2 mg/mL) showed a maximum of 84.64 ± 0.35% reduction in the biofilm biomass of Listeria monocytogenes and the inhibition zone given by BRE against Escherichia coli was 16.04 ± 0.38 mm. BRE showed the highest antioxidant capacities and obvious antibacterial effects against food-related microorganisms than the other samples. Therefore, BRE can be used as a natural antioxidant and antibacterial agent and has potential health advantages and food industry applications.

Consumption of fruits and vegetables has been associated with reduced risk of disease, such as neurodegenerative disorders and certain forms of cancer, and aging. Antioxidants prevent the damage to macromolecules and cells by interfering with the free radicals. Several natural compounds that posses antioxidant activity have been reported from plant sources and are commercially promoted as nutraceuticals. Citrus fruits contain certain bioactive compounds such as phenolics, flavonoids, limonoids, carotenoids, and ascorbic acid. In this context, navel oranges were freeze-dried and extracted with five different solvents, such as hexane, ethyl acetate, acetone, methanol, and 8 methanol: 2 water. The extracts were dried under vacuum and screened for their radical scavenging activity using the 1,1-diphenyl-2-picrylhydrazyl method at 250, 500, and 1000 ppm. The methanol: water and methanol extracts of navel orange were found to be maximum (92.9%) and minimum (63.89%) radical scavenging activity at 1000 ppm. Furthermore, the antioxidant capacity of all extracts was assayed through the phosphomolybdenum method and expressed as equivalent to ascorbic acid (μmol/g of the extract). The order of antioxidant capacity for navel orange extracts was found to be ethyl acetate > acetone > methanol: water > methanol > hexane. It seems that the antioxidant capacity of the extracts is in accordance with the amount of phenolics/lycopene present in each fraction and may provide a good source of antioxidants. There has been increasing interest in the research on flavonoids from plant sources because of their versatile health benefits reported in various epidemiological studies. Since flavonoids are directly associated with human dietary ingredients and health, there is need to evaluate structure and function relationship. The bioavailability, metabolism, and biological activity of flavonoids depend upon the configuration, total number of hydroxyl groups, and substitution of functional groups about their nuclear structure. Fruits and vegetables are the main dietary sources of flavonoids for humans, along with tea and wine. Most recent researches have focused on the health aspects of flavonoids for humans. Many flavonoids are shown to have antioxidative activity, free radical scavenging capacity, coronary heart disease prevention, hepatoprotective, anti-inflammatory, and anticancer activities, while some flavonoids exhibit potential antiviral activities. In plant systems, flavonoids help in combating oxidative stress and act as growth regulators. For pharmaceutical purposes cost-effective bulk production of different types of flavonoids has been made possible with the help of microbial biotechnology. This review highlights the structural features of flavonoids, their beneficial roles in human health, and significance in plants as well as their microbial production.

Two species of Agaricus mushroom grown in Mongolia were analyzed for their element content. Biological activity and chemical components study of Agaricus, grown in the Mongolian flora has been investigated for the first time. The ethanol extracts of dried Agaricus sp. mushrooms were analyzed for antioxidant activity on 1,1-diphenyl-2- picrylhydrazyl (DPPH) radicals and interferon-like activity. The ethanol extracts from Agaricus arvensis showed the most potent radical scavenging activity. The IC50 of A. silvaticus and A. arvensis were 216 and 17.75 g/ml respectively. Among the twenty three mushroom extracts, the extracts from A. silvatisus and A. arvensis have shown the interferon-like activity.
