Coreopsis species (tickseed) can be regenerated from leaf segments allowing the possibility to exploit somaclonal variation as a means to develop novel phenotypes. We used true leaf explants from in vitro seedlings of perennial C. grandiflora (A. Gray) Sherff `Domino' and `Sunray' grown on Murashige and Skoog (MS) basal medium. Two of ten seedlings of `Domino' regenerated freely and others were generally recalcitrant. From these two seedlings, designated E2 and H2, shoots were regenerated and acclimatized to the greenhouse. About 175 plants were established and vernalized from which somaclones were selected based on distinct differences in flower orientation and appearance. The selected somaclones were propagated by division and transplanted to the field in August 2001 in a randomized complete block design with three-plant plots and three replications to determine whether novel characteristics persisted through an additional propagation cycle. In the field, plant height, leaf dimension, flowering, and flower dimensions were scored in June and July 2003. Differences were found between somaclones and similarly propagated E2 and H2 for desirable (more petals per flower, greater flowering, shorter plants), undesirable (less flowering, smaller flowers), and neutral (narrower leaves, taller plants) traits. Open-pollinated (OP) seed was collected and germinated and the seedlings from somaclones that differed significantly from E2 and H2 were evaluated. These maternally selected seedlings were overwintered then planted in the field in May 2004. Most traits that differentiated somaclones from E2 and H2 did not persist in the OP seedling population; however variation that was likely introduced through outcrossing resulted in desirable phenotypes with potential for new cultivar development.

Coreopsis grandiflora (tickseed) regenerates readily from leaf segments allowing the possibility to exploit somaclonal variation as a means to develop novel phenotypes. We used true leaves from in vitro seedlings of Coreopsis grandiflora `Domino' grown on MS basal medium as explants in a series of experiments to evaluate the effect of media, leaf explant orientation, and genotype on shoot regeneration. Genotype accounted for most of the variation with two particular seedlings regenerating freely and eight others generally recalcitrant. From these two seedlings, designated E2 and H2, shoots were regenerated and acclimated to the greenhouse over a period of weeks. Once the plants had established (≈6 weeks after acclimatization) they were vernalized by moving them to a lighted bench (12-h photoperiod) in a walk-in cooler at 4 °C .On transfer back into the greenhouse, the plants flowered within a few weeks; 15 of 175 somaclones were selected based on distinct differences in flower orientation and appearance. The selected somaclones were propagated by division and transplanted to the field in May 2002 in a randomized complete-block design with three-plant plots and three replications, to determine if the novel characteristics persisted through an additional propagation cycle. In the field, plant height, leaf dimension, flowering, and flower dimensions were scored in June-July 2003. Objective: The objective of this study was to establish the method of propagation of Oryganum vulgare and Lippia graveolens employing a plant tissue culture technique that decreased the phenolization percentages and increased the multiplication coefficients. Design/ methodology/ approach: The in vitro germination percentage was evaluated in both MS and MS medium + activated carbon. Microcuttings (small shoots) of both species were established in base medium added with different antioxidant agents to decrease the phenolization of explants; the treatments were arranged in a completely randomized block  design. For the propagation phase, a completely randomized factorial design was used, where the auxin/cytokinin phytoregulators, type of explants (axillary buds and leaves), and the species (Lippia graveolens and Oryganum vulgare)  were considered as factors. Results: maximum germination (63.3% ±12.5) was obtained on day 15 ​​in both culture media for L. graveolens and O. vulgare. The use of antioxidant agents mainly activated carbon, increased the in vitro establishment and activation of vegetative buds in both species by up to 90%. There were significant differences in the variables evaluated regarding the treatments, the explant, and the species in the multiplication phase. The combination 1.0/ 0.5 mg L-1 BA/AIB induces callus formation for both species. When used as leaf explants, callus formation was potentiated. Study Limitations / Implications: The results presented are advances from a long-term experiment. Findings/conclusions: The germination of L. graveolens seeds can be achieved in MS medium after 15 days. Microcuttings of both L. graveolens and O. vulgare were successfully established in MS basal medium enriched with 1 g L-1 charcoal that showed low oxidation percentages and induced up to 90% the production of shoots in the explants. The mixture of 1.0/0.5 mg L-1 BA/AIB induces callus formation for both species; when this medium is in contact with leaves as an explant, its formation is potentiated, achieving diameters up to 15 mm. In order to achieve the induction of shoots and roots, buds should be established in MS medium enriched with 0.5 mg L-1 IBA for both species; this mixture encreased the multiplication coefficients

Rudbeckia spp. are adaptable and valuable ornamental wildflowers. Development of new varieties of Rudbeckia spp., with improved commercial characteristics, would be highly desirable. Interspecific hybridization and induced polyploidy may be avenues for improvement within the genus. The objective of this study was to evaluate fertility, morphology, phenology of flowering, and perennialness (overwintering survival) for lines of diploid and induced allotetraploids of R. subtomentosa × hirta and diploid and autotetraploids of R. subtomentosa ‘Henry Eilers’. Polyploid lines were developed and propagated in vitro and then grown ex vitro in a randomized complete block design with 12 replications. Compared with their diploid counterparts, autotetraploid lines of R. subtomentosa ‘Henry Eilers’ had similar internode lengths, plant heights, number of stems, flowering times (date at first anthesis), and fall and spring survival (100%); reduced number of inflorescences and male and female fertility; and increased inflorescence diameters. Compared with their diploid counterparts, allotetraploids of R. subtomentosa × hirta had similar internode lengths, reduced number of inflorescences, delayed flowering times, and increased pollen staining. Allotetraploids had limited male and female fertility compared with no detectable fertility in their diploid counterparts. Plant height and number of stems either decreased or showed no change with induced allotetraploidy. Spring survival of diploid hybrid genotypes ranged from 0% to 82% and was not improved in the allotetraploid hybrids. For a given genotype, some polyploidy lines varied significantly in certain morphological traits (e.g., plant height) indicating somaclonal variation may have developed in vitro or there were variable genomic or epigenetic changes associated with induced polyploidy.

Abstract This study evaluated the effects of preservatives on the in vitro fermentation measures of wet brewer’s grain (WBG) silage at different stages of storage. Treatments (TRT) were sodium lignosulfonate at 1% (NaL1) and 2% (NaL2; w/w of fresh WBG), propionic acid (PRP; 0.5% w/w of fresh WBG), a combination inoculant (INO; Lactococcus lactis and Lactobacillus buchneri each at 4.9 log cfu/fresh WBG g), and untreated WBG (CON). WBG (Fresh) were packed into 8.8 L mini-silos and stored for 60 d at 21°C (Ensiled), then they were opened and aerobically exposed for 10d (AES). Samples from each stage of storage (STG; Fresh, Ensiled and AES) were analyzed for in vitro ruminal digestibility (24 h).Gas kinetics were recorded using the Ankom RF Gas Production System. Data were analyzed as a randomized complete block design (5 blocks) with a 5 (TRT) × 3 (STG) factorial arrangement. Apparent in vitro DM digestibility (DMD) decreased across STG, (51.5, 47.2 and 40.9 for Fresh, Ensiled and AES, respectively) and increased for NaL1, NaL2 and PRP (~47.8) vs. CON (43.0 ± 2.12%). PRP increased apparent in vitro OM digestibility (OMD) when Ensiled (54.5) and NaL2 increased it for AES (47.1) vs CON (46.3 and 39.9 ± 1.73%, respectively). The asymptotic maximal (M) and rate (k) of gas production decreased across STG (214.6, 181.5, 155.1 and 14.6, 12.6, and 9.8, for Fresh, Ensiled and AES, respectively). PRP increased (200.0) and NaL1 decreased (169.3) M vs. CON (183.9± 7.81ml/incubated DM g), while NaL1 and NaL2 (~11.4) decreased k vs. CON (13.4 ± 0.85%/h). Methane concentration and yield were higher in Fresh vs. other STG (0.94 vs. ~0.84 ± 0.07mM and 0.27 vs. ~0.23 ± 0.03mmol/g fermented OM). Spoilage of WBG decreases fermentability and methane production while PRP and NaL improve digestibility with the former also increasing M and k.

The reduction in herbicide use is one of the greatest interests for modern agriculture and several alternatives are being investigated with this objective, including the adoption of cultivars that suppress weeds. The objective of this study was to verify if maize cultivars develop differently, in competition with weeds, to produce green ears and grain. Randomized complete block design was used, with split-plots and five replications. Cultivars DKB 390, DKB 466, DKB 350, AG 7000, AG 7575 and Master, were evaluated in the plots, without weeding and two weedings (at 22 and 41 days after sowing) in sub plots. Twenty-one species were identified in the experimental area, the most frequent being Gramineae (Poaceae), Euphorbiaceae, Leguminosae (Fabaceae) and Convolvulaceae species. There was no difference in the dry biomass above-ground part of the weeds in the plots of the evaluated cultivars. The cultivars behaved similarly in treatments with or without hoeing, except for plant height and ear height evaluations. Without hoeing, plant height increased in cultivar DKB 390, while plant height and ear height decreased in cultivar AG 7575. In the other cultivars, these traits did not change under weed control. The presence of weeds decreased the values of all traits employed to assess green corn yield, with the exception of the total number of green ears and grain yield.

Today, soybean crops of the Tamba Kurodaisu cultivar are sown in beds prior to transplantation to the field. This planting system has caused crop failure due to damage to the root system. An experiment to test different sowing alternatives to obtain plantlets for cropping of food type big seeded soybean was set up with the following treatments: sowing in beds; sowing in 130 cm³ newspaper cups; sowing in test tubes of volumes of 30 cm³, 60 cm³ and 70 cm³; sowing in 70 cm³ disposable plastic cups; sowing on 90 cm³ styrofoam trays. A randomized complete block design was used, and the following traits were assessed: germination percentage; number of days to flowering; plant height at flowering; number of days to maturity; plant height at maturity; number of seeds per plant; individual plant yield; weight of a hundred seeds. Results should that three methods could be used to set up Tamba Kurodaisu cultivar crops: sowing in disposable plastic cups, sowing in beds with later transplant, or direct sowing in the field.

This trial aimed to evaluate the effect of sequential applications of different plant regulators over growth and flower rachis emission of 'Meyer' zoysiagrass (Zoysia japonica). The study was conducted on 15-month old green turfgrass under a randomized complete block design with four replications. The following plant regulator and doses were tested: trinexapac-ethyl (113+113, 226+113, 226+226, 452+113, 452+226, 452+452, 678+339 e 904+452 g a.i./ha-1), prohexadione-calcium (100+100 e 200+200 g a.i. ha-1) and bispyribac-sodium (40+40 e 60+60 g a.i. ha-1), as well as an untreated control. The turfgrass was mowed again at 3.0 cm aboveground and the second plant regulator was applied when 'Meyer' zoysiagrass was between 5.0 and 6.0 cm high. The effect of the treatments was visually rated for visual injury, plant height, height and number of flower rachis, and total dry mass production of clippings. Only bispyribac-sodium had visual symptoms of injury on 'Meyer' zoysiagrass, and no intoxication was observed at 28 days after the second application (DAAB). The sequential applications of trinexapac-ethyl, prohexadione-calcium and bispyribac-sodium reduced by more than 80% the total clipping dry mass produced by 'Meyer' zoysiagrass. All the plant regulators tested also showed promising results in reducing the height and emission of rachis, especially when trinexapac-ethyl was applied at the doses 452+452, 678+339 and 904+452 g a.i. ha-1. 'Meyer' zoysiagrass turfgrass can be handled with the sequential application of a plant regulator, which reduces the need for mowing over a period up to 110 days after the application of the second plant regulator, and it also avoids deleterious visual effects over turfgrass.

<p>New wetlands field generally has low productivity, so a technology is needed for improve productivity. New Superior Variety (NSV) is a technology that is considered easy to be adopted by farmers, so knowing a NSV that is able to adapt to the characteristics and conditions of new wetlands field is a must. The study of adaptive NSV on new wetlands field in Boven Digoel Regency is still limited so that the NSV recommendation for farmers is also still limited. The purpose of this study was to examine the growth and production of new superior varieties on the new wetlands fields in Boven Digoel Regency. The study was conducted on new wetlands in Asiki Village, Jair District during April-August 2019. Randomized Complete Block Design was employed with 3 replications using Inpari 36, Inpari 37, Inpago Rindang 1, Inpago Rindang 2 dan Inpara 8. The results showed different varieties perform different agronomy characters observed (plant height, number of tillers, panicle length, number of grains per panicle, filled grain, per panicle, empty grain per panicle and 1000 grain weight). Inpari 37 produced 5.3 ton/ha Dried Unhulled Rice, the highest production compared to other varieties, so that it could potentially be developed in Boven Digoel’s new wetlands field. Productivity may still be increased by improving soil fertility conditions.</p>

<p>Phenotypic Performance of Yield and Yield Component of Rice Promising Lines at Tidal Swamp Area in Karang Agung, South Sumatra. An experiment was conducted in Karang Agung, in April until September 2007. The experiment aimed at evaluating phenotypic performance of yield and yield components of promising lines in tidal swamp rice field. The experiment was arranged in a Randomized Complete Block Design with 16 treatments and four replications. The promising lines tested were B9858-KA-55, B9833C-KA-14, B9852E-KA-66, B5524G-SM-61-2-1, B7003D-MR-24-3-1, KAL9414F-MR-2-KN-0, KAL9418F-MR-2-KN-0, B10214FTB- 7-2-3, IR70213-9-CPA-12-UBN-2-1-3-1, IR70215-2- CPA-2-1-B-1-2. The check varieties used were IR42, Batanghari and Lembu Sawah. The results of this experiment showed that IR61242-3B-B-2 had better performance among varieties and promising lines tested, as well as giving the highest yield potential. Meanwhile, the line B10214F-TB-7-2- 3 had better performance of number of filled grain, and 1.000 grain weight. It also showed higher yield potential compared to IR42 and Batanghari. The line KAL9418F-MR-2-KN-0 had better performance of plant height, 1.000 weight grain and harvest age, but showed lower yield potential compared to those other check varieties.</p><p> </p><p><strong>Abstrak</strong></p><p>Percobaan dilaksanakan di KP Karang Agung, Sumatera Selatan pada bulan April sampai September 2007. Penelitian bertujuan untuk mendapatkan informasi tentang penampilan fenotipik karakter hasil dan komponen hasil dari galur-galur harapan padi rawa di lahan pasang surut Karang Agung. Percobaan ditata dalam rancangan acak kelompok dengan 16 perlakuan dan empat ulangan. Galur-galur yang diuji di antaranya B9858-KA-55, B9833C-KA-14, B9852E-KA-66, B5524GSM- 61-2-1, B7003D-MR-24-3-1, KAL9414F-MR-2-KN-0, KAL9418F-MR-2-KN-0, B10214F-TB-7-2-3, IR70213-9- CPA-12-UBN-2-1-3-1, dan IR70215-2-CPA-2-1-B-1-2. Varietas pembanding digunakan IR42, Batanghari, dan Lembu Sawah. Hasil penelitian menunjukkan bahwa galur IR61242- 3B-B-2 memiliki penampilan lebih baik dari varietas pembanding untuk karakter tinggi tanaman, jumlah gabah isi, bobot 1.000 butir gabah, dan potensi hasil. Galur B10214F-TB-7-2-3 memberikan penampilan lebih baik untuk karakter jumlah gabah isi, bobot 1.000 butir, dan potensi hasil lebih tinggi daripada pembanding IR42 dan Batanghari. Galur KAL9418F-MR- 2-KN-0 memiliki penampilan lebih baik untuk karakter bobot 1.000 butir, tinggi tanaman, dan umur panen, tetapi potensi hasilnya lebih rendah dari ketiga varietas pembanding.</p>

Abstract The objective of this study was to compare and explain the variations in the production and nutritive value of canarana erecta lisa grass (Echinocloa pyramidalis Lam.) at different harvest intervals. The treatments corresponded to the cut intervals of 20, 27, 34, 41, 48, 55, 62, and 69 days. The experimental design was a randomized complete block design with three replications. Each experimental unit had 6m2 (3m ×2m). The variables studied included forage production, canopy height, and nutritive value o harvested forage. Forage production varied from 11517 to 12324 kg ha-1; Plant heights from 48.8 to 234 cm; Neutral detergent fiber (NDF) from 68.8% to 71.13%; Acid detergent fiber (ADF) from 36.20% to 44.15%; Crude protein (CP) from 11.43% to 5.85% and in “in vitro” dry matter digestibility (IVDMD) from 63.30% to 47.42%. These values indicated that the nutritive value of canarana erecta lisa was reduced at longer harvest intervals. It is recommended to harvest the grass at 35 days of regrowth favoring productivity, nutritive value and forage digestibility.
