Introduction. Smart food packaging that alerts consumers to spoilt food by changing color is based on affordable and biodegradable raw materials. The research objective was to develop films from anionic polysaccharides and anthocyanin pigment that can be used as a freshness indicator of minced fish. Study objects and methods. The study featured frozen black currant berries (Ríbes nígrum), polysaccharide-based anthocyanin films, and minced fish. Extracts of anthocyanin pigment and films based on agar, kappa-carrageenan, chitosan, starch, and anthocyanin pigments were analyzed by IR spectroscopy. Results and its discussion. Anionic polysaccharides, i.e. agar and kappa-carrageenan, demonstrated good film-forming properties. Films based on 1.5% agar and 2% kappa-carrageenan showed elasticity, resilience, plasticity, and sufficient resistance to mechanical deformation. Neutral polysaccharide starch and cationic polysaccharide chitosan appeared to have no such qualities. An IR spectral analysis revealed chemical interactions between polysaccharide and anthocyanin molecules. It indicated the electrostatic nature of the polyelectrolyte complexes of the anthocyanin pigment with anionic polysaccharides. A film based on 1.5% agar fortified with anthocyanin pigment was used as a test-system for analyzing the quality of fish. The minced fish samples were wrapped in the anthocyanin film and left for 2–7 min to register the color change of the film. When the anthocyanin film came in contact with fresh fish, the color of the film did not change even after prolonged contact. When the film came into contact with spoilt fish, the color of the film began to change after 2 min of contact. When the contact time reached 7 min, the film turned blue. Conclusion. The type of polysaccharide and the interaction between polysaccharides and anthocyanin pigment had a significant effect on film formation. Anionic polysaccharides demonstrated the best results. Electrostatic interactions between anionic polysaccharides and anthocyanin pigments produced stable polyelectrolyte complexes. The new smart films were able to determine the quality of minced fish.

The study aims to analyze about the low graduation and certification exam training participants of the procurement of goods / services of the government and its contributing factors, and formulate a strategy of education and training and skills certification exams procurement of goods / services of the government. Collecting data using the method of study documentation, interviews, and questionnaires. Is the official source of information on the structural and functional Regional Employment Board, as well as the participants of the training and skills certification exams procurement of goods / services of the government in Magelang regency government environment. Analysis using 4 quadrant SWOT analysis, to determine the issue or strategic factors in improving the quality of education and training and skills certification exams procurement of government goods / services within the Government of Magelang regency. The results show organizer position is in quadrant I, which is supporting the growth strategy, with 3 alternative formulation strategies that improve the quality of education and training and skills certification exams procurement of government goods / services, and conducts certification examination of the procurement of government goods / services with computer assisted test system (CAT). Based on the research recommendations formulated advice to the organizing committee, namely: of prospective participants of the training and skills certification exams procurement of goods / services the government should consider the motivation of civil servants, is examinees who have attended training in the same period of the year, the need for simulation procurement of goods / services significantly, an additional allocation of training time, giving sanction to civil servants who have not passed the exam, the provision of adequate classroom space with the number of participants of each class are proportional, as well as explore the evaluation of education and training and skills certification exams procurement of goods / services for Government of participants.

We formulated the principles of designing bioluminescent enzyme tests for assessing the quality of complex media which consist in providing the maximum sensitivity to potentially toxic chemicals at a minimal impact of uncontaminated complex media. The developed principles served as a basis for designing a new bioluminescent method for an integrated rapid assessment of chemical safety of fruits and vegetables which is based on using the luminescent bacterium enzymes (NAD(P)H:FMN oxidoreductase and luciferase) as a test system.

Investigation of the levels of organochlorine pesticides (OCPs) in fish samples was carried out to assess the contamination status of Niger River. Ten different brackish water species of fish (6 samples for each, making a total of 60) were purchased from landing sites at the Delta area of Niger River. These were Drapane africana, Mochokus niloticus, Chrysichthys nigrodigitatus, Pristipoma jubelini, Vomer septapinis, Pseudotolithus senegalensis, Mugil cephalus, Pseudotolithus elongatus, Sphyraena piscatorum, and Lutjanus goreensis. OCPs were determined using standard methods. Certified reference standards from Accustandard USA were used for the instrument calibration and quantification of OCPs. Twenty OCPs, namely, α-HCH, β-HCH, γ-HCH, δ-HCH, endrin, endrin aldehyde, endrin ketone, heptachlor, heptachlor epoxide, aldrin, dieldrin, endosulfan I, endosulfan II, endosulfan sulfate, methoxychlor, α-chlordane, γ-chlordane, DDE, DDT, and DDT, were identified/quantified using Gas Chromatography (GC) (Hewlett Packard GC 5890 series 11 with electron capture detector). Confirmation was done using Shimadzu GCMS QP2010. The highest concentration of ∑OCPs in the brackish fish samples of the Niger River, 4302±2066 µg/kg fresh weight, with a range of 2237-6368 µg/kg, was detected in Drapane africana, while the lowest concentration, 2320±876.4, with a range of 1006-3288 µg/kg, was found in Mochokus niloticus. The concentration of total OCP compounds varied markedly amongst the different fish species. The guideline value of 2000 µg/kg fresh weight by WHO/FAO was exceeded and therefore implied potential harmful effects on humans. Since contamination of the fish samples was an indication of contamination of the river, the quality of the water for public water supply should be of concern; and therefore further monitoring is suggested.

The study is to investigate the effectiveness of several anthocyanins on quality of papaya jam. It was conducted using a randomized block design, arranged with two factors. The first factor is the anthocyanin with different source with several levels that of control (without pigment), canna flower, grape and skins dragon fruit, factor II is sugar content which is 40%, 50%, and 60%. The results of the study, indicating that there is significant to increase quality products. The best treatment is papaya jam with canna flower anthocyanin pigment and 60% sugar.

The study was conducted to investigate the possibility of enhancing the quality of cultured snakehead fish (Channastriata) protein, especially used in the processing of sausage products, by treatment with protease from its viscera. The change in physicochemical properties (expressed by water holding capacity, emulsifying capacity of protein, texture characteristics and whiteness index) of snakehead fish paste related to various protease activity (0, 3, 6, 9, 12, 15 U/L) and washing time (20, 30 and 40 min) were determined. In addition, protease treatment conditions were fixed at ambient temperature (30 ± 2 °C) and the ratio of minced fish and protease solution was 1:1(w/v). The results showed that physicochemical properties, gel quality and sensory properties of sausage significantly improved from soaked minced fish in protease solution at 9 U/L for 30 minutes.

Aims: Caseins are among the main milk proteins that determine many of its properties. Bovine kappa-casein (CSN3) is associated with the qualitative composition of milk, as well as with the quality of cheese obtained from this milk. The rs43703016 single-nucleotide substitution (g.88532332A>C; Asp148Ala) in exon 4 of the bovine CSN3 gene plays an important role in the production of quality hard cheeses. Various methods for the DNA testing of this substitution have been developed in the last three decades. Emergent DNA technologies provide an opportunity to modernize methods of genotyping single-nucleotide polymorphisms. Results: We have developed and verified a method to differentiate A/C alleles of the rs43703016 substitution in the bovine CSN3 gene by real-time PCR using allele-specific fluorescent probes. Conclusion: Our new method allows fast genotyping of animals, and may be used for selection of cows carrying the CC genotype, which determines good cheese-making properties of milk.

Miana (Coleus scutellaroides) is an ornamental plant of single leaf a purple color. The purple color found from miana leaves is an indicator of the presence of anthocyanin pigments. Anthocyanin from miana leaves can be utilized as a natural pigment for various staining purposes especially in the food industry, anthocyanin harvesting on miana leaves can be done at any time and does not depend on the season due to its abundant availability in nature.The aims of this study were to isolate, identify, and microencapsulated anthocyanin pigments of miana leaves. The result, pigment extracted from miana leaves had characteristic of purple color and having maximum absorbance at wavelength 529 nm, suspected as anthocyanin derivative (cyanidin-3-routosida).The total microcapsulated anthocyanim was 0.149542 mg / g. Thermostability of encapsulated anthocyanin pigment from miana leaves stable with heat exposure temperature 100C up to 60 minutes. Based on those collected information, encapsulated miana leaves pigment could be used as an additional natural coloring in food industries.Keywords: anthocyanin, miana, microencapsulation, pigment, thermostability. ABSTRAKMiana (Coleus scutellaroides) termasuk ke dalam tanaman hias yang berdaun tunggal dan berwarna ungu. Warna ungu yang terdapat pada daun miana adalah indikator keberadaan pigmen antosianin.Pemanfaatan daun miana sebagai sumber antosianin dapat dimanfaatkan sebagai pigmen alami untuk berbagai keperluan pewarna terutama dalam industri pangan, pemanenan antosianin pada daun miana dapat dilakukan setiap saat dan tidak bergantung pada musim dikarenakan ketersediannya yang selalu melimpah di alam.Tujuan dari penelitian ini adalah untuk mengisolasi, mengidentifikasi, dan mikroenkapsulasi pigmen antosianin dari daun miana.Berdasarkan hasil analisis didapati bahwa ekstrak pigmen daun miana memiliki karakteristik warna nampak ungu dan serapan maksimum absorbansinya berada pada panjang gelombang 529 nm, pigmen diduga sebagai turunan antosianin yaitu sianidin-3-rutinosida.Total antosianim yang sudah dimikroenkapsulasi sebesar 0.149542 mg/g. Dari hasil uji thermostabilitas pigmen terenkapsulasi maltodekstrin, stabilitas pigmen antosianin dari daun miana cenderung stabil dengan adanya pemaparan panas suhu 100C hingga 60 menit.Berdasarkan hasil dan pengujian, pigmen daun miana terenkapsulasi dapat menjadi pertimbangan produk untuk digunakan sebagai bahan tambahan pewarna dalam industri pangan.Kata kunci: antosianin, miana, mikroenkapsulasi, pigmen, termostabilitas.

Karaginan merupakan senyawa hidrokoloid yang diekstraksi dari rumput laut merah jenis Kappaphycus alvarezii. Karaginan dapat digunakan untuk meningkatkan kestabilan bahan pangan baik yang berbentuk suspensi (dispersi padatan dalam cairan), emulsi (dispersi gas dalam cairan). Tujuan dari penelitian ini adalah mengetahui Kualitas ekstrak karaginan hasil ekstraksi rumput laut K. alvarezii dari dua lokasi sampling yang berbeda. Penelitian ini dilaksanakan pada bulan Agustus 2015 sampai bulan Januari 2016, di perairan Pantai Kartini, Jepara dan peraian Kemojan Karimunjawa, Jepara. Hasil penelitian menunjukkan analisis spektra FTIR produk telah memenuhi spesifikasi karaginan standar karena gugus-gugus fungsi yang terdapat pada spektrum sampel yang dihasilkan sama seperti gugus pada kappa karaginan yaitu gugus ester sulfat, OH, ikatan glikosidik. Hasil analisis sifat fisik-kimia karaginan (rendemen, kadar air, kadar abu, kadar sulfat, viskositas, dan kekuatan gel) yang paling baik yaitu karaginan dari perairan Kemojan Karimunjawa dibandingkan karaginan Pantai Kartini. Kadar sulfat Pantai Kartini dan Kemojan Karimunjawa masih dibawah kadar mutu yaitu 15-40 %, Sedangkan kekuatan gel kedua perairan yaitu Kemojan Karimunjawa 78,57 g.cm-2 dan Pantai kartini 61,86 g.cm-2 masih jauh di bawah baku mutu yaitu 685,50 g.cm2. Carrageenan is a hydrocolloid compounds extracted from red seaweed types Kappaphycus alvarezii. Carrageenan can be used to improve the stability of food in the form of suspension or emulsions. The purpose of this study was to determine the quality of the extract of carrageenan extracted from K. alvarezii at two different sampling locations. This research was conducted in August, 2015 and January, 2016. Samples of seaweed obtained from two different waters, namely Kartini Beach and Kemojan Karimunjawa Island, bolt at jepara regency. The results showed the FTIR spectra analysis of the product met the specifications for a standard carrageenan functional groups contained in the sample spectrum as expressed in clusters of kappa carrageenan namely sulphate ester group , OH , glycosidic bond. The results of analysis  of the physical-chemical properties of carrageenan ( yield, moisture content , ash content , sulphate content , viscosity and gel strength ) the best quality of carragenan was obtained from carrageenophyte taken from Kemojan Karimunjawa Island, although sulphate content from two different waters was under quality level at 15 – 40%, while the  gel strength kartini 61.86 g.cm-2 and Kemojan 78.57 g.cm-2 both were still far below the of carrageenophyte quality at 685.50 g.cm-2.

Abstract Background: Beer spoilage caused by wild yeast and bacteria is a major concern to both commercial and home brewers. Objective: To address this problem, Molecular Epidemiology Inc. (MEI, Seattle, WA) has developed a beer spoilage organism detection kit consisting of an enrichment media (BSE) and a multiplex PCR DNA dipstick that simultaneously detects these organisms within 2 h following enrichment. Methods: The kit was tested by using samples obtained from breweries located in the Greater Seattle area. Samples were spiked with the target microbes, when necessary, and used for assessing the performance characteristics of the DNA dipstick assay. Microbial enumerations were performed as per the standard microbiological plating methods. The suitability of the BSE medium to support the growth of beer spoilage microbes was compared with the industry-approved NBB-C medium (Dohler, Darmstadt, Germany). Results: Inclusivity (a panel of 50 isolates) and Exclusivity (a panel of 92 isolates) testing indicated that the dipstick assay can exclusively detect the indicated target beer spoilage microbes. When compared with the NBB-C medium (Dohler, Darmstadt, Germany) approved by the European Brewers Convention for beer spoilage organisms, the BSE medium supported faster growth of critical spoilage lactic acid bacteria such as Lactobacillus brevis, L. lindneri, and Pediococcus damnosus. Conclusions: The beer spoilage organism detection kit has a detection limit of 10 cells/mL. Highlights: The kit can be used at different stages of the brewing process, thus offering a convenient, cost effective, and faster test system for brewers interested in monitoring the quality of their product.

Latex products have long been recognized as a cause of latex protein allergy. The increased usage of latex gloves, with the consequent increased occurrence of latex allergies appears to have escalated with increasing awareness of the transmission of HIV–AIDS and other infections. The use of condoms as a means to prevent the transmission of STD's (sexually transmitted diseases) and HIV–AIDS has been widely promoted. Although extensive testing is done to evaluate the physical quality of condoms, no information is available regarding the biological safety of condoms. This study was undertaken to determine the effects of short‐term exposure to physiological levels of condom surface material on cell viability (MTT assay) and cell growth (crystal violet assay). A direct contact cell culture testing method (FDA test method F813‐83 used to evaluate the cytotoxic potential of medical materials and devices) was used. The modified test method was found to be a sensitive test system for the evaluation of the biological safety of condoms. This study reveals the importance of evaluating the biological safety of all condoms that are commercially available, because of the potential health risk that may be associated with prolonged use of certain types of condoms.
