Abstract One year of continuous observation of bacterial and viral pathogen concentrations in overlying water and sediment of three urban landscape ponds replenished with reclaimed wastewater (RW) ponds was carried out to establish the distribution of pathogens and investigate the effects of environmental factors on that in RW ponds. The pathogens were represented by Escherichia coli and three common viral pathogens (enterovirus, norovirus, and rotavirus). Results indicated that the peak concentrations of pathogens occur from August to October. Pathogens present in sediment should be paid much more attention than those in overlying water, as they mainly contribute to the favorable conditions for survival and regrowth of pathogens in sediments. Cluster and redundancy analyses revealed that the environmental factors of chlorophyll a (Chl-a), organic matter, and water transparency have key impacts on the occurrence of pathogens. This infers that the practical way to reduce pathogenic risks in RW ponds is to control the algae bloom and improve the transparency of water bodies. Furthermore, based on breakpoint regression analyses, the appropriate ranges of Chl-a and transparency are suggested to be less than 57 mg/m3 and greater than 68 cm, respectively, to reduce the concentration of pathogens in urban landscape ponds replenished with RW.

The relationship between different environmental factors and abundance of phytoplankton species was studied for one year in a pond of old Dhaka named Sikkatuli pond. The range of annual mean of different variables in the pond was air and water temperatures 20 - 31.75˚C, secchi depth 21 - 54 cm, pH 7.39 - 8.3, alkalinity 3.9 - 9.2 meq/l, conductivity 484.5 - 2273.34 μS/cm, DO 3.35 - 8.33 mg/l, TDS 224.67 - 380.5 mg/l, SRS 12.03 - 79.93 mg/l, NO3-N 0.18 - 0.435 mg/l, SRP 0.33 – 4.28 μg/l, chlorophyll a 196.08 - 362.76 μg/l and phaeopigment 30.51 - 212.2 μg/l. During the investigation Cryptomonas erosa var. reflexa, Rhodomonas lens, Cyclotella comensis, Merismopedia gluaca, Euglena acus, Lyngbya limnetica, Chlorella vulgaris, Arthrospira platensis, Trachelomonas volvocina, Oscillatoria agardhii, Synechocystis aquatilis, Pelonema aphane and Peridinium sp. were found to be dominant phytoplankton. Pearson correlation showed that only alkalinity correlated with phytoplankton at 5% significant level. RDA orientation showed that air temperature, water temperature, secchi depth, chl a and pheopigment concentration are the important environmental factors. RDA ordination plot also showed that phytoplankton species of Pelonema aphane and Peridinium sp. were negatively correlated with secchi depth. ABSTRACT Introduction Acute respiratory diseases account for a substantial number of outpatient visits and hospitalizations among U.S. military personnel, significantly affecting mission readiness and military operations. We conducted a retrospective analysis of respiratory viral pathogen (RVP) samples collected from U.S. military personnel stationed in Hawaii and tested at Tripler Army Medical Center from January 2014 to May 2019 in order to describe the etiology, distribution, and seasonality of RVP exposure in a military population. Materials and Methods Samples were analyzed by viral culture or multiplex PCR. Distribution of respiratory viruses over time was analyzed as well as subject demographic and encounter data. Presenting signs and symptoms were evaluated with each RVP. Results A total of 2,576 military personnel were tested, of which 726 (28.2%) were positive for one or more RVP. Among positive tests, the three most common viral pathogens detected were influenza A (43.0%), rhinovirus (24.5%), and parainfluenza (7.6%). Symptoms were generally mild and most frequently included cough, fever, and body aches. Conclusion Our study evaluated respiratory virus prevalence, seasonality, and association with clinical symptoms for military personnel in an urban tropical setting in Oahu, HI, over a 5-year period. We show that viral prevalence and seasonality in Hawaii are distinct from those of the CONUS. Results contribute to the broader understanding of seasonality, clinical manifestation, and demographics of RVP among active duty military personnel stationed in Hawaii.

In the New World, the leishmaniases are primarily transmitted to humans through the bites ofLeishmania-infectedLutzomyia(Diptera: Psychodidae) phlebotomine sand flies. Any or both of two basic clinical forms of these diseases are endemic to several cities in Brazil—the American cutaneous leishmaniasis (ACL) and the American visceral leishmaniasis (AVL). The present study was conducted in the urban area of a small-sized Brazilian municipality (Jaboticatubas), in which three cases of AVL and nine of ACL have been reported in the last five years. Jaboticatubas is an important tourism hub, as it includes a major part of the Serra do Cipó National Park. Currently, no local data is available on the entomological fauna or circulatingLeishmania. During the one-year period of this study, we captured 3,104 phlebotomine sand flies belonging to sixteenLutzomyiaspecies. In addition to identifying incriminated or suspected vectors of ACL with DNA of the etiological agent of AVL and vice versa, we also detectedLeishmaniaDNA in unexpectedLutzomyiaspecies. The expressive presence of vectors and naturalLeishmaniainfection indicates favorable conditions for the spreading of leishmaniases in the vicinity of the Serra do Cipó National Park.

Schistosomiasis has expanded to the coast of Pernambuco State, Brazil, where there are frequent reports of Biomphalaria glabrata snails and human cases of the disease. This study analyzes factors related to schistosomiasis transmission risk in Porto de Galinhas. A one-year malacological survey was conducted to identify biological, abiotic, and environmental factors related to the host snail breeding sites. Data analysis used Excel 2010, GTM Pro, and ArcGis 10. A total of 11,012 B. glabrata snails were captured in 36 breeding sites, and 11 schistosomiasis transmission foci were identified. A negative correlation was found between breeding site temperature and snail density and infection rate, and a positive correlation with pH and salinity. The rainy season showed a positive correlation with snail density and infection rate. The study emphasizes the factors involved in the maintenance of schistosomiasis breeding sites, in light of persistence of this disease in Porto de Galinhas for more than 10 years.

In the Mediterranean Sea, the fleshy red alga Phyllophora crispa forms dense mats of up to 15 cm thickness, mainly located on rocky substrates in water depths below 20 m. Because of the observed density of these mats and some first observations, we hypothesize that P. crispa is a yet undescribed ecosystem engineer that provides a multitude of ecological niches for associated organisms along small-scale environmental gradients. Therefore, we conducted an in-situ pilot study in the Western Mediterranean Sea to assess potential influence of the algae mats on the key environmental factors water movement, temperature and light intensity. We comparatively and simultaneously measured in P. crispa mats, in neighboring Posidonia oceanica seagrass meadows, on neighboring bare rocky substrates without algae mats, and in the directly overlying water column. We used several underwater logging sensors and gypsum clod cards. Findings revealed that P. crispa significantly reduced water movement by 41% compared to the overlying water column, whereas water movement was not affected by P. oceanica meadows and bare rocky substrates. Surprisingly, P. crispa increased the water temperature by 0.3°C relative to the water column, while the water temperature in P. oceanica and on bare rocky substrates was reduced by 0.5°C. Light intensity inside the red algae mats was reduced significantly by 69% compared to the water column. This was similar to measured light reduction of 77% by P. oceanica. These findings highlight the strong influence of the dense red algae mats on some key environmental factors. Their influence is obviously similar or even higher than for the well-known seagrass ecosystem engineer. This may be a factor that facilitates associated biodiversity similarly as described for P. oceanica.

Acute infectious gastroenteritis is an important illness worldwide, especially on children, with viruses accounting for approximately 70% of the acute cases. A high number of these cases have an unknown etiological agent and the rise of next generation sequencing technologies has opened new opportunities for viral pathogen detection and discovery. Viral metagenomics in routine clinical settings has the potential to identify unexpected or novel variants of viral pathogens that cause gastroenteritis. In this study, 124 samples from acute gastroenteritis patients from 2012–2014 previously tested negative for common gastroenteritis pathogens were pooled by age and analyzed by next generation sequencing (NGS) to elucidate unidentified viral infections. The most abundant sequences detected potentially associated to acute gastroenteritis were from Astroviridae and Caliciviridae families, with the detection of norovirus GIV and sapoviruses. Lower number of contigs associated to rotaviruses were detected. As expected, other viruses that may be associated to gastroenteritis but also produce persistent infections in the gut were identified including several Picornaviridae members (EV, parechoviruses, cardioviruses) and adenoviruses. According to the sequencing data, astroviruses, sapoviruses and NoV GIV should be added to the list of viral pathogens screened in routine clinical analysis.

Viral agents of human gastroenteritis affect people of all ages across the globe. As a mainly self-limiting disease, it is difficult to evaluate the real prevalence of etiological agents circulating in each region. Many of the analyzed outbreaks are caused by viruses of the family Caliciviridae, especially the genus Norovirus (NoV). Most studies have focused on other enteric viruses, leaving sapovirus (SaV) underestimated as an important emerging human threat. This one-year study analyzed clinical samples from hospital outpatients with acute gastroenteritis in Spain, with the aim of revealing the importance of human SaV as an emerging viral pathogen. A total of 2667 stools were tested using reverse transcription (RT)-qPCR to detect and quantify SaV. Sapovirus was detected in all age groups, especially in infants, children, and the elderly. The prevalence was 15.64% (417/2667), and was slightly higher in 0–2- and 3–5-year-olds (19.53% and 17.95%, respectively) and much lower in 13–18-year-olds (9.86%). Positive samples were detected throughout the year, with peaks of detection during autumn and the late winter to early spring months. The mean value for the quantified samples was 6.5 × 105 genome copies per gram of stool (GC/g) (range 2.4 × 103–6.6 × 1011 GC/g). RT-nested PCR and sequencing were used for further genotyping. Genetic characterization showed a predominance of genogroup I (GI), followed by GII and GIV. The detection of multiple genotypes suggests the circulation of different strains without any clear tendency. The results obtained suggest SaV as the second major gastroenteritis agent after NoV in the region.

One free-ranging Gray fox (Urocyon cinereoargenteus) underwent autopsy following neurologic disease, with findings including morbilliviral inclusions and associated lesions in numerous tissues, adenoviral intranuclear inclusions in bronchial epithelial cells, and septic pleuropneumonia, hepatitis, splenitis, and meningoencephalitis. Molecular diagnostics on fresh lung identified a strain within a distinct clade of canine distemper that is currently unique to wildlife in New England, as well as the emerging multi-host viral pathogen skunk adenovirus-1. Bacterial culture of fresh liver resulted in a pure growth of Listeria monocytogenes, with whole genome sequencing indicating that the isolate had a vast array of antimicrobial resistance and virulence-associated genes. One year later, a second fox was euthanized for inappropriate behavior in a residential area, and diagnostic workup revealed canine distemper and septic L. monocytogenes, with the former closely related to the distemper virus found in the previous fox and the latter divergent from the L. monocytogenes from the previous fox.
