Chemical screening and biological activities of extracts from Cistus munbyi Pomel

Background: Cistus munbyi Pomel is a well-known medicinal plant in Western Algeria for its several applications in medicine and food. This endemic species to North Africa is poorly studied among the genus of Cistus, which gives the opportunity to carry out original pharmacological studies to value this medicinal plant. Objective: To investigate the antioxidant and the antimicrobial activities of Cistus munbyi, an endemic medicinal plant to Algeria. Methods: The antioxidant properties of C. munbyi crudes extracts were evaluated using the DPPH radical-scavenging activity tests. While their antimicrobial activity was studied by the agar diffusion method and MICs determination against eighteen bacterial and fungal strains, some of which were clinical. Results: The obtained results showed that aqueous and ethanol extracts possess a strong scavenging activity against DPPH radicals with EC50 values estimated at 1.12 and 8.31 µg/mL, respectively. On the other hand, extracts from C. munbyi revealed homogeneous and good antimicrobial activity, in which the inhibition zone diameters observed were ranging from 08 to 19 mm, and the MICs values were included between 0.312 and 5.000 mg/mL. Aqueous extract was the less active except against Proteus mirabilis and Staphylococcus aureus. Conclusion: The findings of this study indicate that C. munbyi possess a strong antioxidant activity and an interesting antimicrobial potential, which allow justifying its uses by the local population in several applications. Therefore, C. munbyi could be as a good source of natural antioxidants and antimicrobial agents.

Aerva javanica (Burm. f) Juss. ex Schult. (Family: Amaranthaceae) family is one of the traditional medicinal plant growing in the United Arab Emirates. Apart from studies related to some medicinal properties, phytochemical, GC MS compound characterization and biological activities still to be investigated. An experiment was conducted to determine the possible bioactive components with their chemical structures and elucidation of phytochemicals from the aerial parts of the plant. The macro and micro-mineral constituents and antioxidant activities were also evaluated. Aerial parts of A. javanica were extracted sequentially with hexane, chloroform, ethyl acetate, acetone, methanol by cold percolation method. Free radical scavenging and antioxidant properties of methanolic extract were evaluated by using in vitro antioxidant assays such as hydroxyl radical scavenging activity, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, superoxide radical scavenging activity and ABTS radical scavenging activity. Primary phytochemical and micro-macro element was tested using standard protocol. The chemical characterization was done with the help of Gas Chromatography-Mass Spectrometry (GC–MS), and the mass spectra of the total compounds in the extract were matched with the National Institute of Standard and Technology (NIST) library. Mineral constituents were identified and estimated by ICP-OES. Ninety-nine metabolites were obtained by GC-MS anslysis; indole was found to be major components followed by 2-Chlorallyl diethyldithiocarbamate (CDEC), Carbaril, Bis(2-ethylhexyl) phthalate, Quinoline, 4H-Cyclopenta[def]phenanthrene,2-[Bis(2-chloroethylamino)]-tetrahydro-2H-1,3,2-oxazaphosphorine-2-oxide, Phenobarbital, 1H-Indole, 2-methyl-, 2,3,7,8-Tetrachlorodibenzo-p-dioxin Disulfide, diphenyl. The presence of various bioactive compounds in the extract validates the traditional medicinal uses of this plant.

Apart from being the primary source of food to other living things, plants also have medicinal value to treat various kinds of diseases. In recent years, it has been proposed that the extract from plants may be used as natural antioxidants which can help to prevent the generation of carcinogens in human body. In addition, plants also have antimicrobial agents to inhibit the growth of pathogenic microbes. This study was intended to investigate the antioxidant properties and antimicrobial activity of methanolic extract of Tinospora crispa stems extracted using soxhlet extraction method. The antimicrobial properties of T. crispa stems extract were tested using disc diffusion method against Staphylococcus aureus ATCC 25923, Bacillus cereus ATCC 11778, Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853, Candida albicans IMR C S23/11 A and Saccharomyces cerevisiae IMR S 617/068. The antioxidant properties of the extract were investigated by using Total Phenolics Content (TPC), Total Flavonoids Content (TFC), DPPH free radical scavenging and b–carotene bleaching assays.  The TPC value was 6.12 g GAE/100 g of dried extract while the TFC value was 55.58 g QE/100 g of dried extract. The IC50 of DPPH scavenging assay for the extract and ascorbic acid were 0.21 and 0.04 mg/mL, respectively. The average percentage of b–carotene bleaching assay was 38.3 % as compared to BHT, which was 45.1%.  The disc diffusion method showed no inhibition zone against all the strains of microorganisms at all concentrations of the extracts (0.5, 1.0, 2.5 and 5.0 mg/disc).

Introduction: The role of medicinal plants in meeting the healthcare needs of the populace, particularly in developing countries cannot be overemphasized. They provide holistic treatment and wellbeing due to a plethora of phytochemicals found in them. Among such is Tetrapleura tetraptera (Schum. & Thonn.) Taub., a multipurpose tree plant with varied biological activities. The current study was aimed at assessing the basic phytochemical constituents, free radical scavenging activity as well as the antimicrobial effect on various solvent extracts of leaves, fruit and stem bark of T. tetraptera of Ghanaian origin. Materials and Methods: Aqueous, 50% hydroethanolic and methanolic extracts were made from the leaves, fruits, and stem bark of T. tetraptera. They were assessed for the qualitative and quantitative phytochemical constituents, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, antimicrobial activity as well as the spectroscopic properties (Ultraviolet – visible and Fourier-transform infrared spectroscopy). Compounds in the methanolic extracts were evaluated using the gas chromatography–mass spectrometry. Results: Tannins, reducing sugars, coumarins and flavonoids were present in all extracts while hydroethanolic and methanolic extracts were rich in phenols, tannins and flavonoids, especially for the stem bark. Leaf extracts had better antimicrobial activity against Pseudomonas aeruginosa, Staphylococcus aureus, Escherichia coli, Candida albicans and Proteus mirabilis. Conclusion: Generally, leaves and stem bark extracts of T. tetraptera are rich in biologically active phytochemicals, supporting their use in ethnomedicine and could be exploited further for other biological activities.

Propolis is the mixture of resins that honeybees collect from plant buds, leaves and exudates. Antimicrobial and antioxidant properties of propolis have been widely studied, however, there have few reports on Thai propolis. To increase the efficient extraction of organic propolis compounds, extraction methods have been developed. We applied the ultrasound technology to reduce extraction time and increase extraction yields of propolis. The propolis was extracted using ultrasound technology for 15-60 minutes and the propolis extracts were examined to compare their biological activities. Antimicrobial activities of the propolis were investigated using the agar well diffusion method. The free radical-scavenging and antioxidant activities of propolis extracts were measured using the DPPH and FolinCiocalteau colorimetric methods. The propolis extracted using ultrasound for 15 and 30 minutes showed inhibitory effects on tested fungi and bacteria as well as antioxidant activities.

Several biological activities have now been associated with egg protein- derived peptides, including antihypertensive, antimicrobial, immunomodulatory, anticancer and antioxidant activities, highlighting the importance of these biopeptides in human health, and disease prevention and treatment. Special attention has been given to peptides with antioxidant and antimicrobial activities as a new source of natural preservatives in food industry. In this study, the antioxidant properties of the egg-yolk protein by-product (YP) hydrolysates were evaluated based on their radical scavenging capacity (DPPH), Fe2+chelating effect and ferric reducing power (FRAP). Furthermore, antimicrobial properties of obtained hydrolysates against Bacillus species were studied. The degrees (DHs) of hydrolysis for 4h hydrolysates were: 19.1%, 13.5% and 13.0%, for pepsin, chymotrypsin and trypsin, respectively. Pepsin was the most effective in producing the free amino groups (1410.3 μmolGly/g). The RP-HPLC profiles of the protein hydrolysates showed differences in the hydrophobicity of the generated peptides. Trypsin hydrolysate obtained after 4h reaction demonstrated the strongest DPPH free radical scavenging activity (0.85 µmol Troloxeq/mg). Trypsin and chymotrypsin hydrolysates obtained after 4h reaction exhibited 4 times higher ferric reducing capacity than those treated by pepsin. The hydrolysis products obtained from YP exhibited significant chelating activity. The 4h trypsin hydrolysate exhibited weak antimicrobial activity against B. subtilis B3; B. cereus B512; B. cereus B 3p and B. laterosporum B6. Objective: The present work deals with the synthesis and characterization of biologically active new indole derivatives viz., 2-((1H-benzo[d]imidazol-2-yl)thio)-N-(4-(3-(1H-indol-3-yl)acryloyl)phenyl)acetamide 6a-d, N-(4-(3-(1H-indol-3-yl)acryloyl)phenyl)-2-((5-methoxy-1H-benzo[d]imidazol-2-yl)thio)acetamide 7a-d and N-(4-(3-(1H-indol-3-yl)acryloyl)phenyl)-2-(benzo[d]thiazol-2-ylthio)acetamide 8a-d.Methods: All these newly synthesized compounds were screened for their in vitro antimicrobial activity by an agar plate diffusion method, antioxidant activities: like 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity (RSA), ferric ions (Fe3+) reducing antioxidant power (FRAP), ferrous (Fe2+) metal ion chelating activity.Results: The structures of all the newly synthesized compounds were characterized by their IR, 1HNMR, mass spectral studies and elemental analysis. Compounds 6b, 7a, 7b, 8a and 8b exhibited good radical scavenging activity (RSA) at a concentration 100 µg/ml, compounds 7b, 7c, 7d and 8b displayed good ferric ions (Fe3+) reducing antioxidant power (FRAP) at a concentration 100 µg/ml, compounds 7b, 7d, 8b and 8d showed good ferrous (Fe2+) ion metal chelating activity. Compounds 7d and 8d exhibited good activity against all the screened bacteria and fungi.Conclusion: The synthesised compounds 7d having methoxy group at 5 positions of benzimidazoles ring, bromo group at 5 positions of indole ring, and 8d having bromo group at position 5 of indole ring, both the compounds have exhibited potent antimicrobial activity. Some compounds have shown very good antioxidant activity.

Teas made from 18 Hungarian medicinal plants were investigated for their total phenolic content and antioxidant effect as radical scavenging activity using the Folin-Ciocaltau and DPPH assays. Antimicrobial effect of the herbal teas was measured by agar diffusion method. In total, the results showed that the smallflower hairy willowherb (Epilobium parviflorum) had the highest antioxidant effect among the plants studied. Teas made from fennel (Foeniculum vulgare) and bean pods (Phaseoli legumen) have the lowest activity. Only four herbal teas showed some antimicrobial activity: the smallflower hairy willowherb (Epilobium parviflorum), common agrimony (Agrimonia eupatoria), spearmint (Mentha crispa) and bean pods. At smaller concentrations the relationship found between total phenolic content and radical scavenging activity was linear but with increasing phenol content the antioxidant activity remained the same.

Objectives: The present work is to evaluate the presence of phenol and flavonoid contents, 2, 2-Diphenyl-1-picrylhydrazyl (DPPH) activity and antimicrobial activity of Corbichonia decumbens leaf extracts. Methods: C. decumbens leaves were collected, shaded, dried, powdered, and were subjected to Soxhlet apparatus for extraction using ethanol and petroleum ether. The ethanol and petroleum ether extract of the plant was tested for phenol, flavonoid, H2O2 contents, and DPPH scavenging activity using standard procedures. It is also subjected to antimicrobial activity against pathogenic bacteria and fungus using the well diffusion method. Results: The result revealed that the leaf extracts contain phenol, flavonoid, and H2O2 contents. The values of DPPH radical scavenging activity recorded for ethanol and petroleum ether extract were 55.90±0.10 and 41.23±0.20, respectively. Antimicrobial activities of ethanolic leaf extracts showed lesser values compared to standard values. Conclusion: The result of this study showed the presence of phenol and flavonoid content, evidenced the potential antioxidant and antimicrobial activity against all tested micro-organisms.

Emergence of antimicrobial resistance to standard commercial drugs has become a critical public health concern worldwide. Hence, novel antimicrobials with improved biological activities are urgently needed. In this regard, a series of quinoline-stilbene derivatives were synthesized from substituted quinoline and benzyltriphenylphosphonium chloride using Wittig reaction. Furthermore, a novel pinacol of quinoline was synthesized by pinacolinazation of 2-methoxyquinoline-3-carbaldehyde which was achieved by aluminum powder-potassium hydroxide reagent combination at ambient temperature in methanol. The structures of the synthesized compounds were established based on their spectral data. The antibacterial activities of the synthesized compounds were evaluated in vitro by the paper disc diffusion method against two Gram-positive bacteria (Staphylococcus aureus and Bacillus subtilis) and two Gram-negative bacteria (Escherichia coli and Salmonella typhimurium). The best activity was displayed by compound 19 against E. coli with an inhibition zone of 16.0 ± 0.82 mm and 14.67 ± 0.94 mm at 500 and 250 μg/mL, respectively. This is close to ciprofloxacin which is used as a positive control. The results of in silico molecular docking evaluation of the compounds against E. coli DNA gyraseB were in good agreement with the in vitro antibacterial analysis. Compounds 19 (−6.9 kcal/mol) and 24 (−7.1 kcal/mol) showed the maximum binding affinity close to ciprofloxacin (−7.3 kcal/mol) used as positive control. Therefore, the antibacterial activity displayed by these compounds is encouraging for further investigation to improve the activities of quinoline-stilbenes by incorporating various bioisosteric groups in one or more positions of the phenyl nuclei for their potential pharmacological use. Findings of the DPPH radical scavenging assay indicated that some of the quinolone stilbenes and pinacol possess moderate antioxidant properties compared to ascorbic acid used as a natural antioxidant.

Quercetin is a flavonoid that is found in many plant materials, including commonly eaten fruits and vegetables. The compound is well known for its wide range of biological activities. In this study, 5-O-acyl derivatives of quercetin were synthesised and assessed for their antiproliferative activity against the HCT116 colon cancer and MDA-MB-231 breast cancer cell lines; and their radical scavenging activity against the 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical cation and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical species. Four derivatives were found to have improved the antiproliferative activity compared to quercetin whilst retaining radical scavenging activity.
