Alterations in Ruminal Fluid and Serum Biochemical Constituents in Goats Affected with Ruminal Acidosis

Ruminal acidosis is an important clinical emergency in small ruminants. In this study, eight healthy farm goats and 24 goats presented at TVCC of the college of Veterinary Sciences and A.H., Anand with clinical signs of ruminal acidosis and having rumen liquor pH below 6 were examined for alterations in the ruminal fluid and serum biochemical parameters. Among various rumen fluid parameters evaluated, the mean values of rumen fluid pH decreased significantly (4.71±0.11 vs. 6.90±0.10), while sediment activity time (46.67±1.20 vs. 24.50±0.78 min) and methylene blue reduction time (29.50±0.73 vs. 10.03±0.27 min) increased significantly in acidotic goats. The normal greenish, aromatic viscous color, odour and consistency of rumen fluid of healthy goats also changed to milky grey/creamy, sour/pungent watery in acidotic goats. The rumen protozoal activity decreased to nil in acidotic goats as compared to the healthy goats. Among various serum biochemical constituents, the mean values of glucose (92.43±1.37 vs. 74.13±1.83 mg/dl), BUN (26.49±0.47 vs. 22.63±1.19 mg/dl), serum creatinine (01.01±0.02 vs. 00.83±0.02 mg/dl) and albumin (03.22±0.03 vs. 03.05±0.05 g/dl), ALT (56.75±1.55 vs. 27.88±1.14 IU/L) and AST (93.25±1.82 vs. 54.00±1.75 IU/L), increased significantly, while there was significant decrease in serum calcium (09.09±0.14 vs. 10.29±0.08 mg/dl) in acidotic goats. The mean values of alkaline phosphatase (IU/L) in acidotic goats increased non-significantly from the base values of healthy goats.

Ruminal acidosis is one of the most important clinical emergencies in sheep and goats resulting into high mortality rate. In the present study, eight healthy farm goats and 24 goats presented to the TVCC of the college with clinical signs of ruminal acidosis like anorexia, tympany, increased pulse and respiratory rate, reduced body temperature, doughy rumen, enteritis, oliguria, grinding of teeth, purulent nasal discharge, muscle twitching, arched back, dehydration and recumbency with rumen liquor pH below 6 were examined for haematological alterations using autohaematoanalyzer. Among various haematological parameters evaluated from acidotic goats, the mean values of Hb (12.21±0.17 vs. 10.86±0.15 g/dl), TEC (14.28±0.16 vs. 12.04±0.36 ×106/ μl), TLC (13.43±0.11 vs. 11.11±0.27 ×103/μl), PCV (36.91±0.53 vs. 29.88±0.55%), neutrophils (64.54±0.93 vs. 28.13±0.92%), MCV (23.38±0.37 vs. 19.38±1.34 fl) and MCH (7.03±0.08 vs. 6.31±0.25 pg) were found significantly increased, while the mean values of lymphocytes (28.00±0.82 vs. 65.38±0.80%) and MCHC (24.55 ±0.26 vs. 34.88±0.97 g/dl) were decreased significantly from the base values of healthy goats. It was concluded that ruminal acidosis induced due to accidental heavy ingestion of readily fermentable carbohydrate rich grains and food waste significantly altered the haematological profile concurrent with clinical manifestations in goats, and hence can be used to assess the severity of the disease.

Cow faeces have been investigated as alternative inoculum to replace rumen fluid to determine neutral detergent fibre (NDF) digestibility (NDFD). Aims of this study were to estimate: (1) the NDFD (48 h) of feed ingredients using a rumen inoculum in comparison with faecal inocula from cows fed diets with different forage basis; (2) the undigestible NDF (uNDF) at 240 and 360 h with ruminal fluid and faecal inocula from lactating cows fed two different diets. At 48 h incubation, the NDFD was affected both by feed and type of inoculum (p < 0.01) and by their interaction (p = 0.03). Overall, the mean NDFD was higher for rumen inoculum than for faecal inocula (585 vs. 389 g/kg NDF, p < 0.05), and faecal inoculum obtained from cows fed hay-based diets gave lower NDFD than those from cows fed maize silage (367 vs. 440 g/kg, p < 0.05). At long incubation times, the average uNDF was affected by substrate, inoculum and incubation time (p < 0.01), but not by their interactions. For each inoculum, significantly lower values were obtained at 360 than at 240 h. Regressions between uNDF with rumen and with the tested faecal inocula resulted in r2 ≥ 0.98. Despite the differences at 48 h, the uNDF showed that faecal inoculum could replace rumen fluid at longer incubation times.

Background: No outbreak has been reported on Trypanosoma evansi infection in Malaysia ponies since 1983, and little is known about the interaction between T. evansi and ponies in the country. Therefore, an experimental study was designed to evaluate the pathogenicity of a local strain of T. evansi in the local ponies. Method: For this purpose, four healthy local ponies were inoculated with 102 live trypanosomes/kg body weight, whereas two ponies served as negative control. Blood samples and rectal temperature were collected on alternate days from both groups for 54 days. Physical examination comprised visible mucous membrane and any appearance of clinical signs were observed daily. The number of trypanosomes was estimated using the Neubauer haemocytometer method. Complete haemogram measurements were performed immediately using an automated blood cell counter and the data obtained was evaluated using the general linear model as linear regression. All infected ponies were salvaged treated with 7 mg/kg of diminazene diaceturate. Results: The four infected ponies developed parasitaemia on the 4th day post-infection (DPI), whereas the first high mean of parasites count was recorded on the 8th DPI. Parasitaemia was detected at a level that fluctuated throughout the infection period (30 days) in all infected ponies with a mean of 13.5x106 trypanosomes/ml blood on the 30th DPI. Successive peaks of pyrexia were accompanied by the peaks of parasitaemia and the highest temperature (39.4°C) was observed on the 20th DPI. Excessive weakness and a reduction of appetite fluctuated in the infected ponies during the infection and one animal died unexpectedly on the 23rd DPI. The mean values for RBC, PCV, Hb and thrombocyte count were significantly lower in the infected ponies than the control groups. Neutrophil and eosinophil were significantly declined after the onset of parasitaemia, whereas monocyte increased significantly in the infected group. Conclusion: The appearance of clinical signs and changes in haematological parameters suggests that Malaysian local ponies are susceptible to T. evansi infection. Treatment of the infected ponies with the recommended dosage of diminazene diaceturate was successful in the surviving ponies.

This study aimed to evaluate the inclusion of 0.0%, 0.4%, 0.8%, and 1.2% slow release urea (SRU) in sheep feed on the intake and total digestibility coefficient (DC) of nutrients and ruminal parameters of sheep living in tropical regions. Four sheep with a body weight (BW) average of 30.8 ± 1.7 kg were assigned to a Latin square design. The animals were placed in metabolic cages and received two meals a day. Intake data and DC of nutrients of the sheep were subjected to analysis of variance and differences were tested using a regression equation to 5% probability. The inclusion of 0.0%, 0.4%, 0.8%, and 1.2% SRU in sheep feed did not change (p &gt; 0.05) the intake of dry matter (DM), organic matter (OM), crude protein (CP), ether extract (EE), neutral or acid detergent fiber (NDF, ADF), total carbohydrates (TC), and non-fibrous carbohydrate (NFC) expressed in g animal-1 day-1, g (kg0.75)-1, and % BW. The mean values of 3.04%, 2.82%, 0.41%, 0.07%, 1.36%, 0.87%, 2.31%, and 1.00% of BW were obtained for the intake of DM, OM, CP, EE, ANF, ADF, TC, and NFC, respectively. The inclusion of 0.0%, 0.4%, 0.8%, and 1.2% of SRU in sheep rations had a quadratic manner (p &lt; 0.05) the DC of DM and CP, with maximum values of 69.67% and 63.59%, respectively, for 0.51% and 0.66% levels of SRU inclusion in the experimental diets. The pH of the rumen fluid did not vary (p &gt; 0.05) for different levels of inclusion of SRU in the feed. However, the pH of the rumen fluid did differ (p &lt; 0.05) in relation to time (T) after feeding for all experimental diets. The inclusion levels of SRU in sheep feed did not affect (p &gt; 0.05) the concentration of ammonia nitrogen (NH3-N) of the rumen fluid, but there was a quadratic effect (p &lt; 0.05) with relation to T for all experimental diets. Thus, we conclude that the inclusion of 0.66% SRU provides the best digestibility value of CP and is the appropriate value for the digestion of DM. The inclusion of up to 1.2% of SRU in sheep rations did not change the intake of nutrients and ruminal parameters.

Canine visceral leishmaniasis (CVL) is a zoonotic disease that presents variable clinical and laboratory aspects. The aims of this study were to identify the main biochemical/hematological status of dogs naturally infected with Leishmania (Leishmania) infantum and to associate theses parameters with clinical forms of CVL. Blood samples were analyzed from 51 dogs, 15 uninfected (control group) and 36 infected, which were classified clinically in three groups: asymptomatic (n=12), oligosymptomatic (n=12) and symptomatic (n=12). All the infected dogs showed lower albumin/globulin ratios (A-G ratio) than the limit of reference. The mean values of total protein, urea, α-globulin 2, globulin and A-G ratio of infected dogs were outside the reference interval and differed significantly from those of the controls. Anemia was detected only in groups that showed clinical signs of the disease, and a statistical analysis indicated a significantly higher frequency of lower eritrogram in these groups than in the asymptomatic group. In addition, a significant association was observed between anemia and the presence of the symptoms, with dogs displaying higher erythrogram values showing better clinical conditions. These results provide additional evidence that the clinical forms of CVL may reflect on the erythrogram status.

The aim of this study was to determine the biochemical and haematological profiles of Kalahari Red goats reared in the tropical environment. The Kalahari Red goats were imported into Nigeria from South Africa by the Federal University of Agriculture Abeokuta, Nigeria. The study was carried out on fifty animals of both sexes. The animals were managed under semi-intensive system where they were allowed to go out and graze on paddocks during some part of the day and brought back to the pens in the evening. The goats were fed with concentrate and grazed on sown pastures of Chloris gayana and Stylosanthes amata. Water Rwas given ad-libitum. The serum biochemical analysis was determined using Randox analysis kits. The sex of each animal was noted and recorded. The packed cell volume, haemoglobin concentration, red blood cell count, white blood cell count, neutrophils, lymphocytes, monocytes, eosinophils and basophils were determined as haematological parameters while total protein, albumin, globulin, sodium, potassium, calcium and phosphorus were determined in serum analysis. The results showed that there was no significant (p>0.05) difference in all the haematological parameters determined across sex. There were significant (p<0.05) differences in total protein and albumin across sex of which the mean values were higher in the female goats. The value of the mean total protein in the female goats was 5.93±0.88 while the value was 5.31±0.23 in the male goats. It can be concluded that sex did not have any influence on the haematology but had association on only the protein component of the serum biochemistry. This result obtained can be used as a reference value for Kalahari Red goats reared in the tropics and under similar nutrition.

Subacute ruminal acidosis (SARA) has been considered a major pathology in high producing dairy herds for years. These findings were corroborated by several studies in Europe. However, different feeding practices and herds’ production levels are found in Southern Belgium. This study aimed to ascertain whether dairy cows of several herds from the south of Belgium (Wallonia) with a suspicion of SARA really did present too low ruminal pH values. Twenty-four herds were visited and 172 cows were sampled using an oropharyngeal device to collect ruminal fluid, i.e. Geishauser probe. On the samples, three tests were performed: pH measurement, methylene blue reduction test and microscopic evaluation of protozoa vitality. Based on these analyses, no cows demonstrated pH values lower than 5.5 and, only ten cows could be considered at risk for SARA. By contrast, in eightteen cows, pH values higher than 7.0 were measured and ruminal inactivity was suspected. In this study, ruminal alkalosis appeared to be more frequently observed than SARA.

Abstract A cross-sectional study was conducted to determine prevalence and risk factors of ruminal disorders with identification of pathophysiological changes of rumen fluid and blood in sheep from January to June, 2019 in and around Gondar town, Northwest Ethiopia. One hundred seventy two sheep were selected by systematic random sampling technique and 21 apparently healthy sheep were selected by purposive sampling technique. Complete case history and clinical signs were recorded. Samples (rumen fluid and blood) were collected and analysed in the laboratory. The data was analysed using SPSS version 20. The prevalence of ruminal disorders in sheep was 55.2% and identified major types of ruminal disorders were simple indigestion (25.6%), ruminal acidosis (20.35%), ruminal tympany (5%) and ruminal alkalosis (4.1%). The most prominent clinical signs recorded in sheep with ruminal disorders were anorexia and inappetence followed by diarrhoea, dullness, dehydration, bloat and abdominal pain. Ruminal motility and protozoal motility were significantly (p<0.05) reduced where as percentage of PCV was significantly increased (p<0.05) in sheep’s with ruminal disorders. Sedimentation activity time was significantly reduced in sheep with ruminal acidosis however, it was significantly (p<0.05) increased in ruminal alkalosis (p<0.05). Color, consistency and odour were affected during ruminal disorders. In multivariable logistic regression analysis showed that Male sex; (Adjusted Odds Ratio [AOR] = 6.044 ; 95% CI: 1.894-19.281 ), extensive farming system (AOR=6.358; 95% CI: 1.335-30.285), Crop residue and concentrate feed (AOR=23.01; 95% CI: 8.364-08.201), sudden change of feed (AOR=32.911, 95% CI: 7.959-136.092) and moldy feed (AOR=20.807, 95%CI: 6.245-69.325) were significantly (p<0.05) associated with occurrence of ruminal disorders in sheep. ABSTRACT: The objective of this study was to describe for the first time the clinical-epidemiological, laboratory and pathological aspects of different forms of enzootic ataxia in goats and lambs in the state of Bahia, as well as to propose an emergency oral treatment with copper sulfate in an individualized way for neonates. Three outbreaks of enzootic ataxia were studied. The history, clinical signs and epidemiological data were obtained from the owners and checked during the visits. The first outbreak (FO) occurred in 2013 in a property located in the municipality of Itaberaba and affected a herd composed of 90 goats and 130 sheep. The second outbreak (SO) occurred in 2014 in the municipality of Santa Luz and affected a herd of 90 goats and 110 sheep. The third outbreak (TO) occurred in 2018, in a property located in the same municipality of the SO, being the herd constituted by 80 sheep. Samples of blood were collected from all goats and sheep treated for serum copper dosage. Nine animals (five goats and four sheep) that presented a more severe clinical picture and unfavorable prognosis were necropsied for diagnostic confirmation. During the necropsy of the FO, SO and TO animals, liver samples were collected for copper dosage as well as fragments of several organs for histopathology. At the site where the SO occurred, samples of soil and the main forages where the animals were kept were collected to determine copper, iron, molybdenum, sulfur and zinc contents. The clinical and histopathological findings in the three outbreaks studied were characteristic of enzootic ataxia. In the outbreaks studied, the frequency of enzootic ataxia was higher in goats (52) than in lambs (39). In the goats the mean values of serum copper (0.05mg/kg) and hepatic (2.48mg/kg) of the FO and SO were well below their respective reference values, 12 times lower than serum levels and eight times lower in relation to liver contents. On the other hand, TO sheep presented mean values of serum copper (0.015mg/kg) reduced by 40 times. The levels of iron and sulfur were high in the fodder of the property where the SO occurred, already in the soil, iron levels were high and copper levels were reduced. The treatment tested in neonates and lambs with two doses of copper sulphate solution orally at a dose of 20mg/kg for sheep and 35mg/kg for goats during the first and second week of life was effective in prevention of enzootic ataxia in neonates and can be used for emergency control of the disease. The occurrence of enzootic ataxia (congenital and late form) in goats and sheep was first reported in Bahia. It is also worth noting that this disease caused by copper deficiency has caused great damage to the breeders of small ruminants, mainly due to the high mortality of the animals affected.

High speed cinefluorography was used to track articulatory movements preceding and following full-mouth tooth extraction and alveoloplasty in two subjects. Films also were made of a control subject on two separate days. The purpose of the study was to determine the effects of dramatically altering the structural dimensions of the oral cavity on the kinematic parameters of speech. The results showed that the experimental subjects performed differently pre and postoperatively though the changes were in different directions for the two subjects. Differences in both means and variabilities of kinematic parameters were larger between days for the experimental (operated) subjects than for the control subject. The results for the Control subject also showed significant differences in the mean values of kinematic variables between days though these day-to-day differences could not account for the effects found pre- and postoperatively. The results of the kinematic analysis, particularly the finding that transition time was most stable over the experimental conditions for the operated subjects, are used to speculate about the coordination of normal speech.
