  Objectives: The purpose of the study is to characterize antimycobacterial phytoconstituent from ethyl acetate extract of dried bulbs of Allium sativum Linn. (Alliaceae) and elucidating the probable mode of action of the bioactive molecule.Methods: Serial extraction, Mycobacterium tuberculosis assay by agar well diffusion method, minimal inhibitory concentration by microplate alamar blue assay, Fourier transform infrared (FT-IR), nuclear magnetic resonance (NMR) spectroscopy, liquid chromatography (LC)-electrospray ionization (ESI)-mass spectrometry (MS)/MS, cell leakage assay, scanning electron microscopy (SEM), inhibition property of linear alkylbenzene sulfonate (LAS) in the presence of rifampicin on M. tuberculosis were performed.Results: Ethyl acetate extract displayed significant inhibition properties against M. tuberculosis H37Ra (MTCC 300). Subsequently, the bioactivity-guided fractionation was employed to purify the phytochemical. Analysis of FT-IR, LC-MS (ESI), 1H, and13C-NMR spectrum revealed that the bioactive phytochemicals are the variants of LAS, with C12-alkyl being predominant, and the minimum inhibitory concentration was found to be 5.56 μg/ml. Morphological examination by SEM and cell leakage assay indicated that these molecules change the membrane fluidity.Conclusion: The results thus suggest the possibility of using low concentrations of LAS to effect changes in membrane fluidity, thereby enhancing the efficacy of antibiotic treatment.

Aim: To investigate the antimicrobial efficacy of various solvents leaf extracts of Aegle marmelos, Plumbago zeylanica and Rhinacanthus nasutus against human pathogenic organisms such as Acinetobacter baumannii, Salmonella enteriditis, Bacillus cereus, Streptococcus pneumonia, Mycobacterium tuberculosis and Aspergillus fumigatous by agar well diffusion method. Background: Also, this is the first time report on MIC and MBC/MFC activities and identify the phytochemicals and functional groups using HPLC, GC-MS and FT-IR analysis. Objective: Minimum inhibitory concentration (MIC), minimum bactericidal (MBC/MFC) values were also carried out by the micro dilution method. Methods: Ethyl acetate extract of P. zeylanica showed better antimicrobial activity against the tested pathogens, especially for S. pneumoniae (40 mm) followed by ethyl acetate extract of R. nasutus (36 mm) against S. pneumoniae. While least inhibition was observed for aqueous extract of P.zeylanica against S. enteritis (10 mm). Results: The MIC ranged from 3.75 µg/ml to 60µg/ml and MBC/MFC 7.5 µg/ml to 60 µg/ml. Phytochemical analysis exhibited the presence of alkaloids, steroids, saponins, flavonoids, tannins, terpenes, phenolics and cardiac glycoside in all samples of selected plant extracts. Tannins, phenolics and glycoside were found only in the ethyl acetate extract of P.zeylanica. HPLC and GC-MS analysis of ethyl acetate leaf extract of P. zeylanica exposed ten major peaks and eleven compounds respectively. Chemical nature of the leaf extract was analysed by FT-IR spectrum. Conclusion: Ethyl acetate leaf extract of P. zeylanica showed better antimicrobial activity than other extracts as well as standard drugs. Therefore, this plant could be studied in detail for the identification of active biomolecules for antimicrobial agents.

Objective: Aim of the present study was to evaluate the antimicrobial activity of ripened and unripened fruits of Anthocephalus cadamba and Scirpus kysoor Roxb. tuber against food pathogens.Methods: Different solvent extracts of Anthocephalus cadamba fruits and Scirpus kysoor Roxb. tubers were screened for its antimicrobial activity against Gram negative and Gram positive bacterial cultures by agar well diffusion method as well as by minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). MIC was carried out by broth microdilution method.Results: All the plant materials exhibit antimicrobial activity. Among all plant materials, macerated ethyl acetate extract of Scirpus kysoor Roxb. tuber showed lowest MIC against Bacillus Cereus 0.312 mg/ml. In the case of ripened Anthocephalus cadamba fruits, macerated ethanol extract was found to be more potent whereas in the case of unripened Anthocephalus cadamba fruit macerated ethyl acetate extract was found to be more active on the basis of MIC.Conclusion: This study revealed that all the plant materials possess significant antimicrobial activity against food pathogens hence may be used as a source of safe herbal antimicrobial agent.

Background: The only remedy for up surging problem of antibiotic resistance is the discovery of antibacterial agents of natural origin. Objective: The present study was aimed at finding antibacterial potential of crude and solvent extracts of mature leaves of Plumeria pudica. Methods: Antibacterial activity of three different solvent extracts were evaluated in four human and four fish pathogenic bacteria by measuring the zone of inhibition and determining Minimum Inhibitory Concentration and Minimum Bactericidal Concentration values. Standard antibiotics were used as positive control. Preliminary phytochemical screening of most effective extract i.e., ethyl acetate extract, Fourier Transform Infra Red analysis and GC-MS analysis of the Thin Layer Chromatographic (TLC) fraction of ethyl acetate extract were done meticulously. All experiments were done thrice and analyzed statistically. Results: Crude leaf extracts and solvent extracts caused good inhibition of bacterial growth in all selected bacteria. Ethyl acetate extract showed highest inhibition zones in all tested strains with maximum inhibition (19.50±0.29 mm) in Escherichia coli (MTCC 739). MBC/MIC of the extracts indicated that all three solvent extracts were bactericidal. Preliminary phytochemical tests revealed the presence of tannins, steroids and alkaloids and FT-IR analysis revealed presence of many functional groups namely alcoholic, amide, amine salt and aldehyde groups. From the GC-MS analysis of TLC fraction of ethyl acetate extract five different bioactive compounds e.g., 2,4-ditert –butylphenyl 5-hydroxypentanoate, Oxalic acid; allyl nonyl ester, 7,9-Ditert-butyl-1-oxaspiro(4,5)deca-6,9-diene-2,8-dione, Dibutyl phthalate and 2,3,5,8-tetramethyl-decane were identified. Conclusion: Leaf extracts of P. pudica contain bioactive compounds that can be used as broad spectrum bactericidal agent.

Objective: The objective of this study was to investigate the antimicrobial activity of Phellinus linteus against skin infectious pathogens, Staphylococcus epidermidis ATCC12228 and Propionibacterium acnes DMST 14916.Methods: Fungal fruiting bodies were extracted with 95% ethanol and ethyl acetate, and then, vaporized. The antimicrobial activities were determined by the disc diffusion method against Propionibacterium acnes DMST 14916 and Staphylococcus epidermidis ATCC12228 skin infectious pathogens. A minimum inhibitory concentration (MIC) and a minimum bactericidal concentration (MBC) for those crude extracts were determined. Finally, the chemical profile of crude extract was determined by using thin layer chromatography and GC-MS.Results: The result demonstrated that the ethanolic extraction had more active fractions with an MIC of 0.5 mg/ml against the growth of Propionibacterium acnes DMST 14916 and Staphylococcus epidermidis ATCC12228 and also showed a minimum inhibitory concentration (MBC) at a concentration of 1.0 mg/ml, while ethyl acetate-based solvents failed to develop on TLC according to Retention factor (Rf) values of 0.71-0.76. The GC-MS was applied to investigate the chemical profile of crude extract of Phellinus linteus, revealing a component of hexadecanoic acid and 9, 12-octadecadienoic acid.Conclusion: Phellinus linteus fruiting body extracts have great potential as antimicrobial compounds against Propionibacterium acnes DMST 14916 and Staphylococcus epidermidis ATCC12228. Thus, they can be used in the treatment of infectious diseases caused by bacterial pathogens.

Typhoid is a problematic fever, caused by Salmonella typhi. Present investigation is based upon studies of antibacterial effects of different solvent extracted samples of O. sanctum stem on growth of S. typhi. The crude extracts of stem of O. sanctum were prepared from three different solvents such as methanol, ethyl acetate and hot water. Four different concentrations (25%, 50%, 75% and100%) were prepared from each one of the solvent extracted sample. Agar well diffusion method was used to test the antibacterial activity of different solvent extracted samples of O. sanctum stem. Analysis of data indicates that a metabolic extract of O. sanctum stem shows maximum zone of inhibition and it was of 26mm at 100% concentration and minimum zone of inhibition was of 19mm at 50% concentration. Ethyl acetate extract of O. sanctum stem shows maximum zone of inhibition and it was of 29mm at 100% concentration and minimum was of 14mm at 25% concentration. Hot water extract of O. sanctum stem shows maximum zone of inhibition and it was of 28mm at 100% concentration and minimum zone of inhibition was of 12mm at 25% concentration. This study indicates that ethyl acetate extract of O. sanctum stem is more effective against S. typhi as compared to methanol and hot water extracted solvent. Contrary to the antibiotics or synthetic medicine associated with many side effects, this plant part can be used as a herbal medicines against Salmonella typhi infection.

Endophytic fungi are one of the untapped resources of therapeutic compounds for various diseases. The present study focused on the antimicrobial as well as larvicidal activity of ethyl acetate extract of endophytic fungi isolated from Tragia involucrata. The ethyl acetate extract of Penicillium citrinum CGJ-C1 (GenBank No.KT780618), Penicillium citrinum CGJ-C2 (KP739821), Cladosporium sp. (KP739822), and Cryptendoxyla hypophloia CGJ-D2 (KT780619) was subjected to antimicrobial activity against a panel of microorganisms by disc diffusion method, larvicidal activity against Culex quinquefasciatus. All the extracts showed significant antimicrobial activity against the tested organisms ranging from 8±0.32 to 13±2.11. The extent of activity was comparable to the standard drugs. The larvicidal potential of the endophytes was superior to T.involucrata extract. The larvicidal activity was found to be dose and time dependent with LC50 value ranging from 4.25- 158.06ppm after 24hrs of treatment. This is the first report on the bioactivity of the endophytes isolated from T.involucrata. Further studies on the bio-guided isolation of lead compound will benefit the people suffering from microbe’s related diseases.

Penelitian uji daya hambat dan analisis KLT bioautografi hasil partisi ekstrak etanol herba bandotan (Ageratum conyzoides L.) terhadap Shigella dysentriae  telah dilakukan. Penelitian ini bertujuan untuk mengetahui kemampuan ekstrak tersebut dalam menghambat pertumbuhan S.dysentriae dan membandingkan daya hambat ekstrak dengan tingkat kepolaran yang berbeda yaitu pada ekstrak n-heksana, larut etil asetat, dan tidak larut etil asetat berdasarkan  pengukurandiameter hambatan yang terbentuk. Herba tersebut diekstraksi dengan etanol menggunakan metode maserasi, lalu dipartisi dengan pelarut n-heksana dan etil asetat. Pengukuran dilakukan dengan menggunakan metode difusi pada medium Miller Hinton Agar (MHA) dengan waktu inkubasi 24 jam pada suhu 37oC dan memberikan diameter daerah hambatan terbesar pada ekstrak tidak larut etil asetat herba bandotan yaitu 9,3 dan 10,3 mm. Pemisahan secara KLT pada ekstrak n-heksana, ekstrak etil asetat, dan ekstrak tidak larut etil asetat dengan cairan pengelusi berturut-turut, n-heksana:etil asetat (3:1), n-heksana:etil asetat (1:1), dan etil asetat:etanol (10:1) dengan jumlah bercak noda berturut-turut 4, 5, dan 2. Nilai Rf pada ekstrak n-heksana 0.25, 0.41, 0.52, dan 0.71 sedangkan pada ekstrak etil asetat 0.34, 0.53, 0.65, 0.81, dan 0.92 pada ekstrak tidak larut etil asetat 0.33 dan 0.64. Hasil KLT bioautografi diperoleh komponen antibakteri yang diidentifikasi ekstrak n-heksana adalah golongan steroid dan pada tidak larut etil asetat golongan polifenol ABSTRACTInhibition test research and TLC bioautographic bioassay method of the partition results of the ethanol extract of bandotan (Ageratum conyzoides L.) herb against Shigella dysentriae have been conducted. This study aims to determine the ability of these extracts to inhibit S.dysentriae growth and to compare the inhibition of extracts with different polarity levels, n-hexane soluble, ethyl acetate soluble, and ethyl acetate insoluble extracts based on the diameter measurement of the formed resistance. The herbs were extracted with ethanol using the maceration method, then partitioned with n-hexane and ethyl acetate solvents. Measurements were carried out using the diffusion method on Miller Hinton Agar (MHA) medium with an incubation time of 24 hours at 37oC and gave the largest diameter area of resistance to the ethyl acetate insoluble extract of bandotan herb, value 9.3 and 10.3 mm. Separation by TLC on n-hexane extract, ethyl acetate extract, and ethyl acetate insoluble extract with elusive liquid respectively, n-hexane: ethyl acetate (3: 1), n-hexane: ethyl acetate (1: 1), and ethyl acetate: ethanol (10: 1) with the number of stains 4, 5, and 2, respectively. 0.81, and 0.92 in ethyl acetate insoluble extracts 0.33 and 0.64. The results of the bioautography TLC bioassay method showed that the antibacterial component identified in the n-hexane extract was a steroid compound and ethyl acetate insoluble was a polyphenol compound.

The study investigated the antimicrobial properties and the chemical composition of ethyl acetate extract of Dennettia tripetala (pepper fruit) seeds. Crude extract obtained by maceration of pulverized seeds in ethyl acetate was evaluated for antimicrobial activity against Escherichia coli, Staphylococcus aureus, Klebsiella aerogenes, Pseudomonas aeruginosa, Bacillus subtilis, Candida albicans and Aspergillus niger using standard agar-well diffusion method. GC-MS method was used to determine the chemical constituents of the extract. The extract was oily, yellowish-brown with a yield of 1.66 % and had activity against most of the test microorganisms, with inhibition zone diameters ranging between 10 to 25 mm. About 41 chemical constituents were present in the extract with formic acid methyl esters and fatty acids accounting for 57.23 and 18.49 % respectively. Ethyl acetate extract of Dennettia tripetala seeds possessed antimicrobial activity against bacteria but not fungi. The observed activity may be due to the presence of formic and fatty acid esters in the seed. The study further established a scientific proof for the traditional use of Dennettia tripetala seed extracts in treating microbial infections.

Two compounds, ergosterol (1) and 4-hydroxy-hexadec-6-enoic acid methyl ester (2) were isolated from the ethyl acetate extract of the endophytic fungal strain labeled as MI-3, isolated from the leave of Magnifera indica L. The structures of the isolated compounds were elucidated by 1H NMR studies and comparing with published data. The crude ethyl acetate extract, three column fractions and ergosterol were tested for antimicrobial activity against five Gram-positive and eight Gram-negative bacteria and three fungi by disc diffusion method. The general toxicity and antioxidant activity of the parent extract, column fractions and ergosterol were also evaluated by using brine shrimp lethality assay and free radical scavenging assay, respectively. Hexane, ethyl acetate, ethanol and methanol extracts of Psidium guajava, Terminalia chebula, Mimusops elengi and Achyranthes aspera were tested against the dental caries causing bacteria Streptococcus mutans and fungus Candida albicans isolated from caries infected patients. All the four extracts of P. guajava showed activity against both S. mutans and C. albicans. Maximum zone of inhibition was observed in ethyl acetate of P. guajava. The four extracts of T. chebula and M. elengi showed antibacterial activity against S. mutans. M. elengi extracts and ethanol extract of T. chebula did not show any antifungal activity against C. albicans. Except for the hexane extract of A. aspera, the other three extracts showed activity against the tested microbes. The ethyl acetate P. guajava leaf extract showed the minimum inhibitory concentration (MIC) against S. mutans to be <0.076 mg/mL in both MHB and BHI. The P. guajava ethyl acetate extract was subjected to GC-MS.
