Two compounds, ergosterol (1) and 4-hydroxy-hexadec-6-enoic acid methyl ester (2) were isolated from the ethyl acetate extract of the endophytic fungal strain labeled as MI-3, isolated from the leave of Magnifera indica L. The structures of the isolated compounds were elucidated by 1H NMR studies and comparing with published data. The crude ethyl acetate extract, three column fractions and ergosterol were tested for antimicrobial activity against five Gram-positive and eight Gram-negative bacteria and three fungi by disc diffusion method. The general toxicity and antioxidant activity of the parent extract, column fractions and ergosterol were also evaluated by using brine shrimp lethality assay and free radical scavenging assay, respectively. The objective of this study was to evaluate the diversity of endophytic fungi of different parts of Ligusticum chuanxiong Hort (CX) and further characterize their biological activities and identify chemical compounds produced by these endophytic fungi. A total of 21 endophytic fungi were isolated and identified from CX. Penicillium oxalicum, Simplicillium sp., and Colletotrichum sp. were identified as promising strains by the color reaction. Comparing different organic extracts of the three strains, it was observed that the ethyl acetate extract of Penicillium oxalicum and Simplicillium sp. and the n-butanol extract of Colletotrichum sp. showed significant antioxidant and antibacterial activities. The ethyl acetate extracts of Penicillium oxalicum had outstanding antioxidant and antibacterial effects, and its radical scavenging rates for ABTS and DPPH were 98.43 ± 0.006% and 90.11 ± 0.032%, respectively. At the same time, their IC50 values were only 0.18 ± 0.02 mg/mL and 0.04 ± 0.003 mg/mL. The ethyl acetate extract of Penicillium oxalicum showed MIC value of only 0.5 mg/mL against Escherichia coli and Staphylococcus aureus. By liquid chromatography-mass spectrometry (LC-MS), we found that Penicillium oxalicum could produce many high-value polyphenols, such as hesperidin (36.06 μmol/g), ferulic acid (1.17 μmol/g), and alternariol (12.64 μmol/g), which can be a potential resource for the pharmaceutical industry. In conclusion, these results increase the diversity of CX endophytic fungi and the antioxidant and antibacterial activities of their secondary metabolites.

Background: Although the chemical constituents and biological activities of a large number of plants in the Croton genus have been studied, there are still recently discovered plants to be investigated. Objective: 1. To investigate the anti-bacterial, anti-HIV1-RT, and cytotoxicity activities of crude extracts from these plants. 2. To investigate the chemical constituents of Croton fluviatilis, Croton acutifolius, and Croton thorelii. Method: The anti-bacterial, anti-HIV1-RT, and cytotoxicity of the three plants were evaluated by standard techniques. Extraction, separation, and purification of extracts from the three plants were undertaken. Results: The ethyl acetate extract of C. fluviatilis showed low anti-bacterial activity against E. aerogenes, E. coli 0157: H7, and P. mirabilis, together with the ethyl acetate extract of C. acutifolius displayed low anti-bacterial activity against E. aerogenes, while all the crude extracts of C. thorelii were inactive. The ethyl acetate extracts of C. thorelii, and C. fluviatilis showed strong inhibited HIV1-RT, whereas the ethyl acetate extract of C. acutifolius, and the hexane extract of C. fluviatilis displayed moderate inhibited HIV1-RT. Cytotoxic properties of three Croton plants were specific to KKU-M213, MDA-MB-231, A-549, and MMNK-1. Especially, the ethyl acetate extract of C. acutifolius exhibited strong cytotoxic activities against MDA-MB-231, A-549, and MMNK-1. Furthermore, the ethyl acetate extract of C. thorelii showed high cytotoxic activities against KKU-M213, and MDA-MB-231. Compounds 1, and 4 were found in C. fluviatilis. Compounds 2 and 4 were also found in C. acutifolius. Moreover, compound 3 was only found in C. thorelii. Conclusion: The present study revealed that the three Croton species are good sources of flavonoid compounds and further investigation of the chemical constituents from these plants may prove to be fruitful to discover more active compounds to be tested as potential medicines.

The study investigated the antimicrobial properties and the chemical composition of ethyl acetate extract of Dennettia tripetala (pepper fruit) seeds. Crude extract obtained by maceration of pulverized seeds in ethyl acetate was evaluated for antimicrobial activity against Escherichia coli, Staphylococcus aureus, Klebsiella aerogenes, Pseudomonas aeruginosa, Bacillus subtilis, Candida albicans and Aspergillus niger using standard agar-well diffusion method. GC-MS method was used to determine the chemical constituents of the extract. The extract was oily, yellowish-brown with a yield of 1.66 % and had activity against most of the test microorganisms, with inhibition zone diameters ranging between 10 to 25 mm. About 41 chemical constituents were present in the extract with formic acid methyl esters and fatty acids accounting for 57.23 and 18.49 % respectively. Ethyl acetate extract of Dennettia tripetala seeds possessed antimicrobial activity against bacteria but not fungi. The observed activity may be due to the presence of formic and fatty acid esters in the seed. The study further established a scientific proof for the traditional use of Dennettia tripetala seed extracts in treating microbial infections.

Objective: The current investigation was conducted to evaluate phenol and flavonoid contents, antibacterial, antioxidant and antihypertensive potentials of three extracts of Margotia gummifera aerial parts. Methods: The antibacterial effect was assessed using wells agar diffusion method against 11 strains. The antioxidant effect was evaluated using different methods, 2, 2-diphenyl-1-picrylhydrazyl, total antioxidant capacity, ferric reducing ability power, cupric reducing antioxidant capacity and β-carotene/linoleic acid bleaching assay. The antihypertensive activity was performed using hippuryl-histidyl-leucine substrates. Results: The highest yield was obtained by hydro-methanolic extract, 19.801 %, however ethyl acetate extract seemed to be rich on phenolic and flavonoid compounds comparing to other extracts, 822.14±69.10 mg GAE g-1 ext. and 117.28±1.80 mg RE g-1 ext., respectively. The high free radical scavenging activity was mentioned in ethyl acetate extract with IC50 of 48.820±1.25 µg/ml. The same extract showed better antioxidant activity in all tested procedures. In antibacterial activity, the hydro-methanolic extract exhibited moderate effect against all tested bacteria except Salmonella enterica and Enterococcus faecalis which gave 18±2.0 and 16±1.0 mm of inhibition, whereas, it seemed to be inactive towards two Gram-negative bacteria. Concerning antihypertensive activity, all extracts can inhibit the Angiotensin-I Converting Enzyme, but the potential effect was given by hydro-methanolic and aqueous extracts, 97.75 % and 96.65 %, respectively. Conclusion: It can be stated that bioactive compounds of Margotia gummifera extracts had different biological activities which confirm their therapeutic uses in traditional medicine.

We investigated the effect of different solvents (ethyl acetate, methanol, acetone, and chloroform) on the extraction of phytoconstituents fromLantana camaraleaves and their antioxidant and antibacterial activities. Further, GC-MS analysis was carried out to identify the bioactive chemical constituents occurring in the active extract. The results revealed the presence of various phytocompounds in the extracts. The methanol solvent recovered higher extractable compounds (14.4% of yield) and contained the highest phenolic (92.8 mg GAE/g) and flavonoid (26.5 mg RE/g) content. DPPH radical scavenging assay showed the IC50value of 165, 200, 245, and 440 μg/mL for methanol, ethyl acetate, acetone, and chloroform extracts, respectively. The hydroxyl scavenging activity test showed the IC50value of 110, 240, 300, and 510 μg/mL for methanol, ethyl acetate, acetone, and chloroform extracts, respectively. Gram negative bacterial pathogens (E. coliandK. pneumoniae) were more susceptible to all extracts compared to Gram positive bacteria (M. luteus,B. subtilis, andS. aureus). Methanol extract had the highest inhibition activity against all the tested microbes. Moreover, methanolic extract ofL. camaracontained 32 bioactive components as revealed by GC-MS study. The identified major compounds included hexadecanoic acid (5.197%), phytol (4.528%), caryophyllene oxide (4.605%), and 9,12,15-octadecatrienoic acid, methyl ester, (Z,Z,Z)- (3.751%).

Euphorbia hirta Linn. (co sua la lon in Vietnamese) belongs to Euphorbiaceae family, is a group of small prostrate herbaceous annual weed in Vietnam. It is abundant in waste places and open grasslands and distributes in most Asian countries. E. hirta is traditionally used in the treatment of gastrointestinal disorders, bronchial and respiratory. The aqueous extract exhibits anxiolytic, analgesic, antipyretic, and anti-inflammatory activities. Strong anti-diabetic activity of Euphorbiaceae family in general and E. hirta in particular was reported in the past investigations. E. hirta has been studied by various investigations and several active constituents have been isolated and identified successfully. Most of those compounds have strong biological activities. At the first step in the processing of the isolation of bioactive compounds from the ethyl acetate extract, we isolated four purified compounds, including methyl gallate (1), quercetin (2), myrecitin (3), and quercitrin (4). The chemical structures of those compounds were elucidated by spectroscopic methods and compared with published data in the literature.

The in vitro biological activities of the ethyl acetate extract of the culture filtrate from Streptomyces achromogenes TCH4 (TCH4 extract) were evaluated. The ethyl acetate extract of TCH4 produced a bacteriostatic effect against Enterobacter cloacae, Staphylococcus aureus, Staphylococcus saprophyticus, Bacillus subtilis, methicillin-resistant S. aureus (MRSA) and Klebsiella pneumoniae. The extract had bactericidal activity against S. aureus, S. saprophyticus, S. aureus (MRSA) and K. pneumoniae with minimum bactericidal concentration (MBC) values in the range of 500-1000 ?g/mL. The total phenolic and flavonoid contents in TCH4 were 107.20?2.57 mg gallic acid equivalent (GAE)/g and 44.91?0.84 mg quercetin equivalent (QE)/g of dry extract. Antioxidant activity was determined by DPPH radical (IC50 299.64?6.83 ?g/mL) and ABTS radical scavenging (IC50 65.53?0.95 ?g/mL), and the ferric-reducing antioxidant power (FRAP) (822.76?9.12 mM FeSO4.7H2O/g dry extract) assays. TCH4 exhibited cytotoxic activity in the DU-145 cell line (IC50 9.36?0.37 ?g/mL). Analysis of extract constituents by GC-MS revealed pyrrolo[1,2-a]pyrazine-1,4-dione, hexahydro-3-(2-methylpropyl) (36.85%), benzeneacetamide (23.76%), and deferoxamine (12.85%) as the major compounds, which have been reported to possess pharmaceutical properties. S. achromogenes TCH4 could be a potential source of bioactive metabolites with antibacterial, antioxidant and anticancer activities for pharmaceutical applications.

The Panchthar, a hilly district of Eastern Nepal, has several medicinal plants which are used by indegenous people for the treatments of different diseases. In the present work, phytochemical and biological activities of hexane, ethyl acetate, acetone, and methanol extracts of Litsea monopetala leaves from the Pachthar district were analyzed. The phytochemical screening demonstrated the presence of major secondary metabolites like flavonoids, alkaloids, terpenoids, polyphenols, glycosides, and proteins. The antibacterial activity was estimated by the disc diffusion method against Bacillus substilus and Escherichia coli. The result showed that ethyl acetate extract exhibit the highest zone of inhibition (ZOI) value of 18 mm/disc. Acetone extract also showed significant antibacterial activity at the concentration of 100 mg/mL. Antioxidant activity from DPPH radical scavenging assay displayed the inhibitory concentration (IC50) values of dried methanol and acetone extracts were 30.74 μg/mL and 202.86 μg/mL, respectively. The methanol extract has the highest total phenolic content of 360.14 mg GAE/g extract and the highest total flavonoid content of 27.13 mg QE/g extract. The present study revealed that Litsea monopetala plant from the Panchthar district is a source of several bioactive substances and can be used in several drugs formulation.

Introduction: Triphala, a herbal medicine which is a product from Chae Son, Lampang, Thailand, is an unequal-proportional mixture of fruits of three herbs, Phyllanthus emblica, Terminalia chebula, and Terminalia bellirica (2:1:1). Materials and Methods: The contemporary study concentrated on extracts detection and comparative analysis between various annual year (A.D.2017/2018) activities in hexane, ethyl acetate, and methanolic extracts of Triphala. The determination of antioxidant was using by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP). The broth dilution and agar well diffusion assay were determined antibacterial. Results: Results exposed the valuable biological activity extracts from ethyl acetate and methanol solvents of T. chebula and T. bellirica, which strength is accountable for biological activities. Extracts displayed acceptable radical-scavenging activity analogous with ascorbic acid. The methanol extract of T. bellirica (0.0013 mg/mL), ethyl acetate extract of P. emblica (0.0009 mg/mL), in A.D. 2017 and the ethyl acetate extract of P. emblica (0.0090 mg/mL), the methanol extract of Triphala (0.0176 mg/mL) in 2018, were exhibited higher antioxidant activity. Also, extracts indicated hopeful antibacterial potential against tested strain comparable to chloramphenicol. Conclusion: The results in A.D. 2017 showed that inhibition zone of P. emblica in ethyl acetate extract; S. aureas (8.67 mm), E. aerogenes (8.67 mm), E. coli (EPEC) (7.67 mm), P. mirabilis (7.73 mm) , S. typhimurium (8.33 mm), S. flexneri (8.33 mm) and V. cholera (7.33 mm) together with P. emblica methanol extract; S. aureas (6.67 mm), E. aerogenes (7.76 mm.)  and V. cholera (4.0 mm). Therefore, it could be concluded that Triphala may be a talented candidate in pharmaceutical and future medicine.

The ethyl acetate extract of bark of Aporosa wallichii Hook.f. was extensively investigated for the chemical characterization of secondary metabolites and for biological activities. After repeated chromatographic separation and purification, two compounds were elucidated as a phenolic compound ferulic acid (1) and an uncommon triterpene glut-5(6)-en-3β-ol (2) from the ethyl acetate extract of barks of A. wallichii by using high field NMR analyses. This is the first report of isolation of both compounds from this plant species. No significant antimicrobial activity was observed for any fraction after Kupchan partitioning of the extract. During the antioxidant activity assay, the ethyl acetate soluble fraction of A. wallichii demonstrated significant DPPH radical scavenging capacity with an IC50 value of 1.25 μg/mL. The other fractions viz. petroleum ether, aqueous and chloroform soluble fractions exhibited moderate, mild, and weak activity respectively. The petroleum ether soluble fraction demonstrated maximum thrombolytic property (51.33%) which was similar to that of standard streptokinase (66.81%). Dhaka Univ. J. Pharm.
