Background: This study aimed to evaluate the growth ability of cassava mini stem cuttings with different node number and a variety of stem cutting shapes and their correlation with starch content in the stems at initial growth stages. Methods. In this study, the viability of cassava stem cuttings was identified in two type experiments i.e. mini-stem cuttings consisting 1 and 2 nodes and shape variation of single node mini-stem cutting. Parameters observed were shoots emergence period, number of sprouting cuttings, shoots number  of individual stem cuttings, shoots height and number of leaves. In addition, starch histochemical test was  also carried out on stems of young shoots and initial stem cuttings using Lugol’s solution. Results. Both cassava stem cuttings consisting of 1 and 2 buds indicated the same survival rate of 100%. 1 bud stem cuttings with different shapes showed different survival rate, i.e. 60-80% for semicircular and fully circular cuttings and 30-40% for box shape cuttings. The difference in survival rate with different stem size is probably related to the availability of the amount of starch to support shoots growth. Observations at week 3 after planting generally showed that the stem cuttings with 2 buds were higher than those of stem with 1 bud. Conclusion: There were differences in the scores on the starch content test qualitatively with Lugol staining, in various parts of the plant originating from 1 bud and 2 bud cuttings which may indicate a breakdown of starch during shoot development.

Katuk (Sauropus  androgynus (L.) Merr.) has potential as a vagetable and medicinal plant.  This study aimed  to determine effect of node  position and number of stem cuttings  on the  growth  of  katuk. The experiment   used  Completely Randomized Design with two treatments arranged in split  plots. The node  position of stem cuttings as the main plot consists of top stem cutting (T, top – 8 nodes under shoot tip),  middle stem cutting (M, 8 – 11 nodes under shoot tip) and bottom stem cutting (B, 16-20 nodes under shoot tip). The node number of stem cuttings as a subplot consists of 2 nodes  (J1), 3 nodes (J2), 4 nodes (J3), and 5 nodes (J4). Each treatment  combination was replicated  three times.The results revealed that the top stem cutting  resulted in the  highest  root, total plant dry weight and leaf area, as well stem cutting with 4 nodes produced  the highest total plant dry weight and leaf area.

Abstract Background Cassava mosaic disease (CMD) is one of the most devastating viral diseases for cassava production in Africa and Asia. Accurate yet affordable diagnostics are one of the fundamental tools supporting successful CMD management, especially in developing countries. This study aimed to develop an antibody-based immunoassay for the detection of Sri Lankan cassava mosaic virus (SLCMV), the only cassava mosaic begomovirus currently causing CMD outbreaks in Southeast Asia (SEA). Methods Monoclonal antibodies (MAbs) against the recombinant coat protein of SLCMV were generated using hybridoma technology. MAbs were characterized and used to develop a triple antibody sandwich enzyme-linked immunosorbent assay (TAS-ELISA) for SLCMV detection in cassava leaves and stems. Assay specificity, sensitivity and efficiency for SLCMV detection was investigated and compared to those of a commercial ELISA test kit and PCR, the gold standard. Results A TAS-ELISA for SLCMV detection was successfully developed using the newly established MAb 29B3 and an in-house polyclonal antibody (PAb) against begomoviruses, PAb PK. The assay was able to detect SLCMV in leaves, green bark from cassava stem tips, and young leaf sprouts from stem cuttings of SLCMV-infected cassava plants without cross-reactivity to those derived from healthy cassava controls. Sensitivity comparison using serial dilutions of SLCMV-infected cassava sap extracts revealed that the assay was 256-fold more sensitive than a commercial TAS-ELISA kit and 64-fold less sensitive than PCR using previously published SLCMV-specific primers. In terms of DNA content, our assay demonstrated a limit of detection of 2.21 to 4.08 × 106 virus copies as determined by quantitative real-time PCR (qPCR). When applied to field samples (n = 490), the TAS-ELISA showed high accuracy (99.6%), specificity (100%), and sensitivity (98.2%) relative to the results obtained by the reference PCR. SLCMV infecting chaya (Cnidoscolus aconitifolius) and coral plant (Jatropha multifida) was also reported for the first time in SEA. Conclusions Our findings suggest that the TAS-ELISA for SLCMV detection developed in this study can serve as an attractive tool for efficient, inexpensive and high-throughput detection of SLCMV and can be applied to CMD screening of cassava stem cuttings, large-scale surveillance, and screening for resistance.

The legumes (Fabaceae family) are the second most important agricultural crop, both in terms of harvested area and total production. They are an important source of vegetable proteins and oils for human consumption. Non-thermal plasma (NTP) treatment is a new and effective method in surface microbial inactivation and seed stimulation useable in the agricultural and food industries. This review summarizes current information about characteristics of legume seeds and adult plants after NTP treatment in relation to the seed germination and seedling initial growth, surface microbial decontamination, seed wettability and metabolic activity in different plant growth stages. The information about 19 plant species in relation to the NTP treatment is summarized. Some important plant species as soybean (Glycine max), bean (Phaseolus vulgaris), mung bean (Vigna radiata), black gram (V. mungo), pea (Pisum sativum), lentil (Lens culinaris), peanut (Arachis hypogaea), alfalfa (Medicago sativa), and chickpea (Cicer aruetinum) are discussed. Likevise, some less common plant species i.g. blue lupine (Lupinus angustifolius), Egyptian clover (Trifolium alexandrinum), fenugreek (Trigonella foenum-graecum), and mimosa (Mimosa pudica, M. caesalpiniafolia) are mentioned too. Possible promising trends in the use of plasma as a seed pre-packaging technique, a reduction in phytotoxic diseases transmitted by seeds and the effect on reducing dormancy of hard seeds are also pointed out.

The crystallographic orientations of echinoderm skeletal elements can supplement standard morphological comparisons in the exploration of echinoderm evolution. At a coarse scale, many echinoderms share a crystallographic pattern in whichcaxes radiate away from the axis of pentaradial symmetry. Within this common pattern, however,caxes of different taxa can differ dramatically in their degree of variability, angles of inclination, and relationships to the external morphology of skeletal elements. Crystallographic data reflect a variety of taxon-specific influences and therefore reveal different information in different taxa. In echinoids, orientations ofcaxes in coronal plates correlate well with high-level taxonomic groupings, whilecaxes of apical plates record modes of larval development. In blastoids,caxes of radial plates have a structural interpretation, with thecaxis oriented parallel to the orientation of the surface of the radial plate during its initial growth stages. In crinoids,caxes do not correlate with taxonomic group, plate morphology, or developmental sequence, but instead correlate with relative positions of skeletal elements on the calyx. Although their full potential has yet to be explored, the varied crystallographic patterns in echinoderms have been used to clarify skeletal structure, characterize developmental anomalies, and infer homologies of skeletal plates both within specimens and between groups. A axes are less constrained in their orientations thancaxes and offer less promise of revealing novel paleobiological information.

Development of precision machines to handle some of the field activities involved in the cultivation of cassava is a highly welcomed project among young enterprising farmers who are keen in resolving the posterity of their wellbeing as a big time farmer. Mechanization of cassava in the tropics is grossly hampered by lack of extensive knowlegde on the behaviour, physical properties of cassava stems and parameters that have great influence on the handling of the stems during cultivation, hence the limita-tion of advancing the present semi-automatic planter to fully automated one is still a mirage in the tropics. This paper presents the flow properties of cassava cuttings that can lend it to proper handling in a metering machine. A variable inclined wood surface rig with an ejection outlet was used to study the properties of the stem. Measured cassava stem predictive parameters; diamater, lentgh,weight; angle variation on the rig; response parameters; percentage ejected, percentage sliding through, forces acting on stems were obtained. The results from the study could be used to predict acurately the exact dynamics of a given stem isolated from the bulk of stem cuttings stacked in the metering hopper. Models to predict the flow properties of the stems were obtained; further simulation of the dynamics was embarked upon using MATLAB software. A major transition zone of stem characteris-tics per pecentage sliding to ejection occurred within the range of angles of 150 -300. This might be a favourable angular zone for isolating the stems from the bulk mass while the effectively isolated stems can be metered appropriately by another mechanism in the process of developing a metering device for cassava stem cuttings.

The increasing interest in<em> Arundo donax,</em> a perennial lignocellulosic species only reproducing by propagation, requires the setup of cheap, simple and reliable techniques. Considering these targets, stem cutting offers considerable advantages. The present investigation aimed to compare: i) plants obtained by different propagation methods (by rhizome and micropropagation mother plants); ii) plants obtained by stem cuttings from basal, central and apical parts of the stem; iii) different planting periods (spring, summer, autumn). The obtained results showed that the number of new shoots from stem buds was: i) higher in the spring and lower in the summer planting period; ii) higher from cuttings obtained by micropropagated than rhizome mother plants, both in spring and summer plantings; iii) decreasing passing from the basal to the apical stem portion; iv) partly unexpressed in the autumn planting period; v) lower from one-year stem cuttings as compared to two-year stem cuttings.

ABSTRACTEffects of surface morphology of buffer layers on ZnO/sapphire heteroepitaxial growth have been investigated by means of “nitrogen mediated crystallization (NMC) method”, where the crystal nucleation and growth are controlled by absorbed nitrogen atoms. We found a strong correlation between the height distribution profile of NMC-ZnO buffer layers and the crystal quality of ZnO films. On the buffer layer with a sharp peak in height distribution, a single-crystalline ZnO film with atomically-flat surface was grown. Our results indicate that homogeneous and high-density nucleation at the initial growth stages is critical in heteroepitaxy of ZnO on lattice mismatched substrates.

Goal. The effect of fungicides on the dynamics of the activity of peroxidase, catalase (CAT) and chlorophyll content in lupine plants during seed dressing has been established. Methods. Laboratory and vegetation researches were conducted in the laboratory of analytical chemistry of pesticides of the Institute of Plant Protection. Yellow lupine (Lupinus luteus L.), variety Obriy has been grown. The objects of research were fungicides triticonazol (40 g/t) and its combination with prochloraz (120 g/t). Determination of the content of fungicides in plants was carried out using chromatographic methods according to officially approved methods and me­thods developed in the laboratory of analytical chemistry of pesticides. Chlorophyll content and peroxidase activity were measured by colorimetric method, catalase activity — by titrimetric method. Results. According to the research results, the varying sensitivity of the enzymatic system of antioxidant defense (catalase, peroxidase) in response to seed dressing by fungicides was recorded. It showed that on the 10th day after sowing, content of triticonazol in plants was 0.8 mg/kg, and the peroxidase activity was similar to that in untreated plants. Subsequently, against the background of a decrease in the content of the active substance, a gradual activation of the enzyme was observed. Catalase activity also gradually increased beginning from the 14th day, and on the 30th day it exceeded the corresponding control indicator by 40%. When using a combination of triticonazol with prochloraz, the disturbance in the balance of peroxidase catalase was more significant. However, by the phase of 7—8 leaves, with a minimal total content of fungicides (0.38 mg/kg), the enzyme activity approached the control level, which is associated with the restoration of plant homeostasis and the formation of its adaptive potential under stress conditions. The stimulating effect of these fungicides on chlorophyll content at the initial growth stages of lupine was established. The chlorophyll concentration in fungicides-treated plants exceeded the control indicator by 11—29%. Conclusions. The use of systemic triazole fungicides to protect seedlings, improves the photosynthetic activity of plants and at the same time acts as a stress factor that activates protecting enzymes (catalase, peroxidase), which trigger the development of protective adaptive reactions of plants.
