HUMORAL AND CELLULAR RESPONSE TO SARS-CoV-2 IN COVID-19 PATIENTS

The study of the immune response to SARS-CoV-2 is crucial for the prognosis and control of coronavirus infection, diagnosis and formation of individual and population immunity, the development of indications and evaluation of the effectiveness of vaccinations, and, ultimately, the scientific prediction of the course of a pandemic. One year after the infection debuted in numerous immunological studies in COVID-19 patients, kinetics, duration and evolution of immune memory in humans due to infection are not well predictable, as data obtained represent the initial effector phase of the immune response, and the responses after recovery from infection cannot be used for long-term prediction. The paper presents an analysis of the results of studies of immune response and immune memory to SARS-CoV-2, including all three branches of adaptive immunity: immunoglobulins, memory B-cells, CD8+ and CD4+ T-cells in sick and cured patients in the dynamic period of 6–8 months after the onset of the disease.

Few, if any, of children's behavioural or cognitive characteristics assessed in the first years of life demonstrate stability until later childhood; early characteristics have so far failed to show an association with future psychopathology. This longitudinal study, from 4–8 months to 4.7 years old, focused on stability and change of extreme temperamental traits in groups of infants subselected from a large birth cohort. Persistent extreme temperament at four and eight months old did not increase stability of temperament to four years of age, relative to other children in the whole population. Sizeable change occurred, and the environmental parameters associated with negative temperamental change did not seem to be the same as those related to positive change. Boys with extreme scores were more stable, while girls appeared more prone to positive change. It is hypothesised that the direction of temperamental change in the first years could be more meaningful for long-term prediction of disorders than any one assessment of temperament taken at any one year.

Many biological responses may develop over long periods of time, and annual community variation should therefore be controlled in ecological research. We sampled carabid beetles over ten years in Norway spruce dominated forests in Southern Finland, harvested using replicated logging treatments of different intensities. We collected carabids in 1995 (prior to logging) and during four post-harvest seasons, 1996-98 and in 2006. The treatments were clear-cutting (no retained trees), modified clear-cutting (retention of three groups of 20-30 trees within one-hectare core) and gap cutting (three 0.16-ha openings within a one-hectare core), and control (mature unharvested forest). Carabids showed remarkable annual and regional variation at assemblage, ecological-group and species levels, such that was independent of treatments. The total species richness, and that of open-habitat carabids, were higher in cleared sites of all treatments than in control stands in 1997-1998 but not in 2006, suggesting that the logging response was ephemeral by many species. The abundances of forest and generalist carabids were little affected by logging. Open-habitat carabids were more abundant in clear-cuts and modified clear-cuts than in gap cuts, which was still detectable in 2006, suggesting a long-term effect. Open-habitat carabids were less abundant in retention sites of modified clear-cuts and gap cuts than in cleared sites, suggesting that retention attenuates assemblage change. Carabid assemblages of logged stands did not differ from control stands in 1996 but they did in 1997-1998, suggesting a one-year delay in logging response. Evaluation of the short-term and long-term immunological responses in a preclinical model that simulates the targeted age population with a relevant vaccination schedule is essential for human vaccine development. A Göttingen minipig model was assessed, using pertussis vaccines, to demonstrate that vaccine antigen-specific humoral and cellular responses, including IgG titers, functional antibodies, Th polarization and memory B cells can be assessed in a longitudinal study. A vaccination schedule of priming with a whole cell (DTwP) or an acellular (DTaP) pertussis vaccine was applied in neonatal and infant minipigs followed by boosting with a Tdap acellular vaccine. Single cell RNAsequencing was used to explore the long-term maintenance of immune memory cells and their functionality for the first time in this animal model. DTaP but not DTwP vaccination induced pertussis toxin (PT) neutralizing antibodies. The cellular immune response was also characterized by a distinct Th polarization, with a Th-2-biased response for DTaP and a Th-1/Th-17-biased response for DTwP. No difference in the maintenance of pertussis-specific memory B cells was observed in DTaP- or DTwP-primed animals 6 months post Tdap boost. However, an increase in pertussis-specific T cells was still observed in DTaP primed minipigs, together with up-regulation of genes involved in antigen presentation and interferon pathways. Overall, the minipig model reproduced the humoral and cellular immune responses induced in humans by DTwP vs. DTaP priming, followed by Tdap boosting. Our data suggest that the Göttingen minipig is an attractive preclinical model to predict the long-term immunogenicity of human vaccines against Bordetella pertussis and potentially also vaccines against other pathogens.

Background. The mechanism of the immune response in patients with COVID-19 is still poorly understood and differs from other respiratory infections. Immunodeficiencies and age-related characteristics of immunity can be regarded as factors influencing reinfection and a predisposition to prolonged persistence of SARS-CoV-2. Most of the studies report about post-infection immunity to SARS-CoV-2 for adults and less for children. However, understanding the dynamics of antibody production to SARS-CoV-2 among children as well as other unique features of immune response is extremely important, because this provides to avoid possible complications and long-term outcomes of COVID-19, and predict the epidemic spread of the new coronavirus in the organized groups of children such as kindergartens, schools, study groups and clubs.Aim: to evaluate the humoral and cellular immunity after SARS-CoV-2 infection in children.Materials and methods. We reported data of the first phase prospective cohort study of immunity among 60 children living in Irkutsk, Russia within a month after SARS-CoV-2 infection, which took place in October and November 2020.Results. Immunity of children one month after SARS-Cov-2 infection was characterized by suppressed cell-mediated and humoral immunity, and phagocytosis dysfunction. Reduced phagocytosis by neutrophils was noted for 61.6 % of individuals.Conclusions. Children after COVID-19 had impaired immunity to SARS-CoV-2, which may be a predictor of chronic infection and other long-term outcomes of COVID-19.

Purpose Geographical indications (GIs) have been implemented across the EU for agricultural and food products for many years and consumers know them well. However, developing countries and/or transitioning economies do not have sufficient experience to apply GIs. The purpose of this paper is to demonstrate ways to implement GI in domestic markets in countries in which there are no common logo and control/tracking systems to help GI holders manage the process. Design/methodology/approach This study uses a qualitative case study to describe a process to implement GI registered food products in domestic markets. The system was developed by the author and applied by the Gemlik Commodity Exchange (GTB). Findings A registration system that allows inspection of producers in GI limits should be established. For good practice and to ensure correct registration, an efficient, established control system, promotion, and conservancy of GI-holding organizations are essential. Practical implications GTB implemented two projects, for which a GI logo and labels with quick-response (QR) codes were created. A regional, GI promotion project was conducted, and after one year, there was no increase in consumer demand, but entering markets became easier, and traders of PDO products began to experience increases in orders and/or shortening of intervals. Long-term implications of the system could not be measured since one year had passed. Originality/value This study develops and demonstrates a QR tracking system for implementation of GIs.

Disrupted immune response is an important feature of many neurodegenerative conditions, including sepsis-associated cognitive impairment. Accumulating evidence has demonstrated that immune memory occurs in microglia, which has a significant impact on pathological hallmarks of neurological diseases. However, it remains unclear whether immune memory can cause subsequent alterations in the brain immune response and affect neurobehavioral outcomes in sepsis survivors. In the present study, mice received daily intraperitoneal injection of low-dose lipopolysaccharide (LPS, 0.1 mg/kg) for three consecutive days to induce immune memory (immune tolerance) and then were subjected to sham operation or cecal ligation and puncture (CLP) 9 months later, followed by a battery of neurobehavioral and biochemical studies. Here, we showed that repeated low-dose LPS injection-induced immune memory protected mice from sepsis-induced cognitive and affective impairments, which were accompanied by significantly decreased brain proinflammatory cytokines and immune response. In conclusion, our study suggests that modulation of brain immune responses by repeated LPS injections confers neuroprotective effects by preventing overactivated immune response in response to subsequent septic insult.

Chicken meat is often a major component of a modern diet. Allergy to chicken meat is relatively rare and occurs independently or in subjects allergic to ovalbumin (OVA). We examined the effect of adoptive transfer of OVA-CD4+ T cells on the immune response to OVA in mice fed chicken meat. Donor mice were injected intraperitoneally with 100 µg of OVA with Freund’s adjuvant two times over a week, and CD4+ T cells were isolated from them and transferred to naïve mice (CD4+/OVA/ChM group), which were then provoked with OVA with FA and fed freeze-dried chicken meat for 14 days. The mice injected with OVA and fed chicken meat (OVA/ChM group), and sensitized (OVA group) and healthy (PBS group) mice served as controls. Humoral and cellular response to OVA was monitored over the study. The CD4+/OVA/ChM group had lowered levels of anti-OVA IgG and IgA, and total IgE. There were significant differences in CD4+, CD4+CD25+, and CD4+CD25+Foxp3+ T cells between groups. OVA stimulation decreased the splenocyte proliferation index and IFN-γ secretion in the CD4+/OVA/ChM group compared to the OVA group. IL-4 was increased in the OVA/ChM mice, which confirms allergenic potential of the egg–meat protein combination. Transfer of OVA-experienced CD4+ T cells ameliorated the negative immune response to OVA.
