Preliminary in Vitro Investigations on The Inhibitory Activity of The Original Dietary Supplement Oxidal® on Pathogenic Bacterial Strains

The antimicrobial action of the dietary supplement Oxidal® was tested using the classic Bauer and Kirby agar-gel diffusion method. Clinical and reference strains of Staphylococcus aureus and Escherichia coli were used in the studies. The tested dietary supplement showed a well-pronounced inhibitory effect against the microbial strains commensurable with that of the broad-spectrum chemotherapeutic agent Enrofloxacin and showed even higher activity than the broad spectrum antibiotic Thiamphenicol. The proven inhibitory effect of the tested dietary supplement against the examined pathogenic bacteria is in accordance with the established clinical effectiveness standards for antimicrobial agents.

Biofilms play a key role in infectious diseases, as they may form on the surface and persist after treatment with various antimicrobial agents. The Staphylococcus aureus, Klebsiella pneumoniae, S. typhimurium, P. aeruginosa, and Escherichia coli most frequently associated with medical devices. Chitosan sulphate from marine litter (SCH-MW) was extracted and the mineral components were determined using atomic absorption spectroscopy (AAS). The degree of deacetylation (DA) of SCH was predicted 50% and 33.3% in crab and shrimp waste respectively. The elucidation of the structure of the SCH-MW was portrayed using FT-IR and 1H-NMR spectroscopy. The molecular mass of SCH-MW was determined with Matrix-Assisted Laser Desorption/Ionization-Time of Flight (MALDI-TOF). The teratogenicity of SCH-MW was characterized by the zebrafish embryo (ZFE) model. Antimicrobial activity of SCH-MW was tested with the agar well diffusion method; the inhibitory effect of SCH-MW on biofilm formation was assessed in 96 flat well polystyrene plates. The result revealed that a low concentration of crab-sulfated chitosan inhibited bacterial growth and significantly reduced the anti-biofilm activity of gram-negative and gram-positive bacteria relatively to shrimp. It is potentially against the biofilm formation of pathogenic bacteria.

Two series of sulfonamides were synthesized from 4-hydrazinylbenzenesulfonamide as the key starting material. 1,3,5-Triarylpyrazoline sulfonamides (2a?i) were obtained by cyclocondensation of various chalcones in 53? ?64 % yields, while 4-thiazolidinone derivatives (4a?e) were synthesized by cyclocondensation between mercaptoacetic acid and different phenylhydrazones in 43?62 % yields. The synthesized compounds were characterized based on FTIR, 1H-NMR, 13C-NMR and HRMS data. The sulfonamides were evaluated for their in vitro antimicrobial activities against four bacterial strains (E. coli, P. aeruginosa, B. subtillis and S aureus), two filamentous fungal strains (A. niger and F. oxysporum) and two yeast strains (C. albicans and S. cerevisiae). Seven pyrazolines, 2a?c and 2e?h, exhibited significant inhibition of different microbial strains. Among them, compound 2b displayed good antifungal activity against A. niger (MIC value at 12.5 ?g mL-1) over the reference drug.

A straightforward rapid and efficient protocol for the synthesis of 2-azetidinone (D1-10) and 5-benzylidine-4-oxo-thiazolidine (F1-10) has been designed and synthesized in order to find newer antimicrobial compounds. The structure of entitle compounds have been evaluated on the basis of various spectroscopic techniques FTIR, 1H-NMR, 13C-NMR as well as elemental microanalysis. The title compounds were screened for their preliminary in vitro antibacterial activity against a panel of selected pathogenic bacterial strains, Staphylococcus aureus (MTCC 96), Escherichia coli (MTCC 443), Proteus vulgaris (MTCC 426) and Pseudomonas aeruginosa (MTCC 424) using cup-plate agar diffusion method at 40 μg/ml concentration. Out of synthesized compounds, compound nos. D4, D5, D7, D8, D9 and D10 have shown outstanding inhibitory effect against all pathogens and consider as the best bioactive desired antibacterial analogue of the series as compare to standard drugs ampicilline and chloramphenicol.

Background and Objective: With the significant increase in the prevalence of infectious diseases and the development of drug resistance by human pathogenic bacteria, there is a continuous need to discover new antimicrobial compounds from plants. Methods: Four extracts of wild Myrtus communis L. berries (myrtle berries) were prepared with the addition of Cinnamomum verum and Eugenia caryophyllata. The extracts were screened in vitro for their antimicrobial activities using agar-well diffusion method against Escherichia coli, Staphylococcus aureus, Enterobacter cloacae, Listeria monocytogenes, Pseudomonas aeruginosa and Proteus mirabilis cultures. Results: The inhibition zones ranged from 12 to 22 mm. The MICs values of extracts lies between the ranges of 30 to 100 mg/ml. Of the extracts studied, the most active ones were those obtained from the myrtle berries:cloves, myrtle berries:cinnamon:cloves with the highest inhibition zones 22 mm and 17mm against S. aureus and L. monocytogenes at 50 mg/ml and 80 mg/ml, respectively. None of the extracts was active against E. coli and P. mirabilis. Conclusion: The present investigations have exposed that the myrtle berries:cloves, myrtle berries: cinnamon:cloves extracts could be used in traditional medicine as natural antimicrobial agents in treatment the bacterial infections.

The principle aim of this work is to evaluate the antimicrobial and antiochratoxic power of the essential oil of Mentha spicata (EOM). This oil was obtained by hydrodistillation with a yield of 0.3%. EOM was screened for its possible antibacterial activity in vitro against five strains of pathogenic bacteria, using the solid disc diffusion method and the microdilution method. The extract reacted positively to the bacterial strains tested. The results of the antifungal activity show an inhibition of mycelial growth; it is total on Aspergillus ochraceus, Aspergillus flavus, Aspergillus niger, Penicillium viridicatum, and on Rhizopus stolonifer and Alternaria alternate at a concentration of 12.5 μl/ml. Tests carried out on yeasts mainly referenced as Candida albicans IP444 and Candida albicans 2679 showed minimal inhibition (MIC) of the order of 7.81 μl/ml and 0.97 μl/ml, respectively and theMICis of the order of 0.48 μl/ml for Candida albicans 10 231. In addition, the EOM antiochratoxigenic test for ochratoxin A production capacity following a series of high performance liquid chromatography separation and ultraviolet detection (HPLC–UV) analyses detected an inhibitory effect proportional to the EOM concentration. In general, the essential oil showed an effective antimicrobial action on the growth of the tested pathogens and an antiochratoxigenic.

The results of the comparative tests of the «Agar Muller-Hinton II - Obolensk» nutrient medium developed in SRCAMB, Obolensk, and the control nutrient medium imported «Mueller Hinton II Agar» are presented in the study. The susceptibility of bacterial clinical strains to antimicrobial agents (AMP) was determined by the disc diffusion method and the method of gradient diffusion (E-test). The carbapenemase activity of the strains carrying the carbapenemase genes was determined by CIM-test. Total 173 characterized bacterial strains of species Klebsiella pneumoniae, Pseudomonas aeruginosa, Acinetobacter baumannii, Proteus mirabilis, Serratia marcescens, Enterobacter aerogenes, Escherichia coli; Photorhabdus spp., Staphylococcus aureus, Enterococcus spp. were used in the study, including producers of OXA- and NDM-types carbapenemases for gram negative bacteria. A high degree of coincidence of the results obtained on both nutrient media was shown. The consistency index of the strain sensitivity categories to AMPs (S, I, and R) was 98.2% for the disc diffusion method, and 94.4-100% - for E-test and CIM-test methods. Thus, within the framework of the Import Substitution Program, the domestic nutrient medium «MHA II-Obolensk» has been successfully developed. The nutrient medium meets the requirements of GOST R ISO 20776-2-2010 «Clinical laboratory testing and in vitro diagnostic test systems - Susceptibility testing of infectious agents and evaluation of performance of antimicrobial susceptibility test devices».

The leaf galls of Terminalia chebula is used widely as Karkatasringi in south Indian markets. Karkatashringi is an important crude drug employed in various indigenous systems of medicine against several diseases and the drug has diverse medicinal properties. The present study was carried out to understand the antimicrobial activity of various extracts. The antibacterial activity of T. chebula (leaf gall) was evaluated against ten bacterial strains including Gram-positive and Gram-negative bacteria using the agar-well diffusion method. Among the two extracts tested, the ethanol extract presented the best results against all the bacteria while aqueous extract showed moderate inhibition of the microbial growth. Each extract is unique against different microorganisms; Staphylococcus aureus was more susceptible to both extract among the tested organisms, whereas Serratia marcescens and Proteus mirabilis were less susceptible for ethanol and aqueous extract respectively. ABSTRACT The increasing problem of antibiotic resistance among pathogenic bacteria requires development of new antimicrobial agents. One line of investigation is the synthesis of antimicrobial hybrid peptides. The aim of the present investigation was to determine the in vitro activities of 16 cecropin-melittin hybrid peptides (CAMEL analogues) against 60 anaerobic bacterial strains, to compare their activities with those of seven clinically used antimicrobial agents, and to compare different methods for anaerobic susceptibility testing of these peptides. The stability of one of the peptides, temporin B, with different stereoisomeric configurations was investigated in a fecal milieu. The CAMEL analogues showed antimicrobial activity against the anaerobic bacteria, with MICs ranging from 0.125 to 32 μg/ml. The overall activities (the MICs at which 90% of isolates are inhibited) of the CAMEL analogues against anaerobic bacteria were mainly inferior to those of imipenem, clindamycin, and piperacillin but were equal to or superior to those of metronidazole, cefoxitin, ciprofloxacin, and chloramphenicol. The agarose dilution method was found to be an accurate method for the testing of large numbers of bacterial strains. The d isomer of temporin B was inactivated more slowly in feces than the l isomer. This study shows that the CAMEL analogues are potential agents for the treatment of anaerobic infections.

In the current investigation, screening of ethanolic extract obtained from Ficus lyrata leaves against pathogenic bacteria has been done in order to assess the antimicrobial activity aimed at detecting new sources of antimicrobial agents. The antimicrobial activity of the extract was determined using agar disc diffusion method. The antibacterial activity of leaf extract of F. lyrata was tested against human pathogenic bacteria — both Gram-positive (Staphylococcus aureus, methicillin-resistant S. aureus and Streptococcus pneumoniae) and Gram-negative strains (Klebsiella pneumoniae, Pseudomonas aeruginosa, and Escherichia coli). The results of this study provide evidence that the ethanolic extract of F. lyrata leaves has a mild antimicrobial activities, apparently, attributed to the presence of various secondary metabolites, which confirm the traditional use of this plant for the treatment of diseases caused by pathogens. These data allow us to suggest that the extracts of F. lyrata can be used to discover antibacterial substances for developing new pharmaceuticals to control clinically important pathogens responsible for severe disorders.

 The activity of essential oils from Eucalyptus globulus, Pinus silvestris, Lavandula angustifolia, Juniperus virginiana, Rosmarinus officinalis and Citrus paradise against the soft-rot pathogens Pectobacterium carotovorum subsp. carotovorum, Pectobacterium atrosepticum, Pectobacterium parmentieri and Dickeya solani was determined in vitro. The antibacterial activity of the essential oils will be evaluated using the disk-diffusion method by Kirby-Bauer [Bauer et al. 1966]. It was found that all the presented essential oils varied in antimicrobial activity against the four bacterial strains. No differences in the influence of streptomycin on inhibition of growth of the four bacterial strains were observed. Among six tested plants, essential oils from P. sylvestris had the strongest inhibitory effect on the growth of soft rot bacteria from Pectobacterium genus. This paper constitute the first report on the activity of the essential oils obtained from J. virginiana against soft rot bacteria. They are also the first report on the activity of the essential oils obtained from E. globulus, P. silvestris, L. angustifolia and C. paradisi against P. atrosepticum, P. parmentieri and D. solani as well as on the activity of the R. officinalis essential oils against P. atrosepticum and P. parmentieri.
