Protection of the liver by oxymethyluracil and derivatives of succinic acid at influence of tetrachlormethane in the experiment

Цель. Сравнительная оценка гепатопротекторных свойств комплексного соединения «оксиметилурацил + натрия сукцинат» и отдельных его составляющих на экспериментальной модели острого токсического поражения печени, индуцированного тетрахлорметаном (ТХМ). Результаты. Исследуемое комплексное соединение в дозе 50 мг/кг оказывало максимальный гепатозащитный эффект, ограничивая выраженность некроза и сохраняя метаболизм печени за счет положительного влияния на активность ферментов (уроканиназы, АсАТ, ЩФ), показатели уровня билирубина и триглицеридов, а также на состояние перекисно-антиперекисной системы - ПОЛ-АОС (ДК, ТК, СОД, КАТ), восстановленного глутатиона, уровня SH-групп. В группах крыс, получавших раздельно оксиметилурацил и натрия сукцинат по 25 мг/кг по лечебно-профилактической схеме, выявлена нормализация функционирования ПОЛ-АОС, в меньшей степени было выражено влияние на функциональную активность печени и ее морфологическую структуру. Заключение. Целесообразность одновременного назначения оксиметилурацил и натрия сукцината подтверждалась более высокой выживаемостью крыс при введении комплексного соединения «оксиметилурацил + натрия сукцинат» после введения летальной дозы ТХМ. The effect of a new complex compound «oxymethyluracil + sodium succinate» on the morphological state and AOS of the liver damaged by tetrachloride has been studied using 50 experimental white male rats,. Purpose. Comparative assessment of hepatoprotective properties of the «oxymethyluracil + sodium succinate» compound with OMU and sodium succinate using the experimental model of acute toxic damage of the liver induced by TCR. Results. The complex compound studied has been shown to produce maximal hepatoprotective effects at a dose of 50 mg/kg through limiting TCR necrosogenic effect, preserving metabolic activity of the liver due to beneficial effects of enzymes (urokaninase, AcAT), levels of bilirubin and triglycerides as well as the POL-AOS system (DK, TK, SOD, KAT), reduced glutathione, SH-group levels. In the rat groups, with oxymethyluracil and sodium succinate administered separately at a dose of 25 mg/kg for treatment and prevention, POL-AOS functioning has also been normalized, while the liver functional activity and its morphological structure - to a lesser degree. Conclusion. The usefulness of simultaneous administration of oxymethyluracil and sodium succinate is confirmed by a higher survival rate of the rats which received the complex compound «oxymethyluracil + sodium succinate» in case of poisoning with TCR at a lethal dose.

An important element in ensuring the epizootic well-being of the poultry industry is disinfection. Modern poultry farming requires a large number of effective disinfectants. It is known that the resistance of microorganisms to the effects of disinfectants is based on a genotypic mechanism. The nature of the formation of resistance to disinfectants and antiseptics is different than antibiotics. With regard to disinfectants, resistance is formed more slowly and the proportion of resistant strains in the population of microorganisms may not be high for a long time. This is due to different mechanisms of formation of resistance to antibiotics and disinfectants, in the first case – plasmid mechanism, in the second – chromosomal. However, increasing the resistance to the active substance in disinfectants can be widespread, so it is necessary to periodically rotate disinfectants. The goal of the work – to investigate the parameters of acute toxicity of the disinfectant biocide “Zodizin”. The studies were conducted in the laboratory of Veterinary Pharmacy and the Vivarium of Sumy National Agrarian University. The drug “Zodizine” contains: polyhexamethyleneguanidine hydrochloride – 21.0 %, alkylldimethylbenzylammonium chloride – 3.0 %. For toxicological examination of the disinfectant, healthy white male rats and white female rats weighing 200 ± 10 g 1.5 years of age were used. In the study of acute toxicity of animals observed daily, noted the general condition of the animals, features of their behavior. Studies have found that the toxic effect of the disinfectant “Zodizin” clinically manifested almost equally in both males and females. The average lethal dose for the rat female was 1000.0 ± 35.0 mg/kg body weight, males 1033.0 ± 34.3 mg/kg. Therefore, according to the classification of substances by toxicity, the drug by intragastric administration can be attributed to low-toxic substances. Observations on animals revealed that 1–3 hours after oral administration of the drug in a subtoxic dose in laboratory animals, shortness of breath and inhibition of the central nervous system were noted. Most of them died during the first day. Subsequent observations of the surviving animals indicated that their motor response was suppressed over the next 24–72 hours. Conclusions and prospects for further research: 1. It was found that the average lethal dose of the drug “Zodizin” with oral administration to rats-females was 1000.0 ± 35.0 mg/kg body weight, males – 1033.0 ± 34.3 mg/kg. 2. Experimental studies have proved that the disinfectant “Zodizin” according to GOST 12.1.007-76, belongs to the IV class of danger, that is, to the low-dangerous compounds, and according to GOST 12.1.07 – to the III class of hazard of substances and can be used for disinfection premises where animals and poultry are kept. Further, the sporoсide and corrosion properties of the “Zoodizin” biocide will be studied.

The aim of the paper is to study thymus variability in white rats, which were exposed to formaldehyde, and to compare these data with the indicators in control animals. Materials and Methods. The trial enrolled 72 white male rats, initial body weight 40–50 g. The animals were divided into 2 groups (36 rats in each). The first group consisted of control rats. Animals of the second group were exposed to formaldehyde inhalation, 2.766 mg/m3. To characterize the variability of the organ size, centroids were determined. The superposition of landmark configurations was performed using the generalized Procrustes analysis method, MorphoJ 1.06d program. The principal component analysis and canonical analysis of the obtained data were carried out. Results. One-Way ANOVA revealed a high level of intergroup differences in Procrust distance (F=1.34; p<0.0001). The significant effect of the duration of formaldehyde exposure on centroid size was established. The Kruskal-Wallis criterion was 19.778 (p=0.0014). The analysis of the principal components indicated that each of the first 10 components stands for more than 1 % of Procrustes coordinate variance. In this case, the first 7 components compatibly explain 91.398 % of thymus variability. The proportion of the first main component to the total variance of the Procrustes coordinates is 40.236 %. PC1 (-) shows changes in the thymus shape, mostly affecting the tops of its lobes, the middle part of the right boundary and the entire left thymus boundary. The scattering ellipses of the thymus ordinates in rats exposed to formaldehyde, in the first two canonical variables are located higher than those in the control animals. Conclusion. Formaldehyde inhalation leads to thymus changes in white rat. The most significant differences with control data are determined along the second canonical variable. Изучение изменчивости формы тимуса белых крыс, находившихся в условиях влияния формальдегида, и сравнение этих данных с показателями, полученными у контрольных животных. Материалы и методы. Работа выполнена на 72 белых крысах-самцах с начальной массой тела 40–50 г. Животные были разделены на 2 серии (по 36 крыс). Первую серию составляли контрольные крысы. Животные второй серии подвергались ингаляционному воздействию формальдегида (ФА) в концентрации 2,766 мг/м3. Для характеристики изменчивости размеров органов определяли размер их центроидов. Процедуру суперимпозиции конфигураций ландмарок выполняли методом генерализованного прокрустова анализа с использованием программы MorphoJ 1.06d. Проводили анализ главных компонент и канонический анализ полученных данных. Результаты. Однофакторный дисперсионный анализ выявил высокий уровень межгрупповых различий по показателю прокрустовых расстояний (F=1,34; р<0,0001). Установлено значительное влияние продолжительности нахождения животных в условиях воздействия ФА на размер центроида. Критерий Краскела–Уоллиса составил 19,778 (р=0,0014). Анализ главных компонент указывал на то, что каждая из первых 10 компонент объясняет более 1 % дисперсии прокрустовых координат. При этом первые 7 компонент совместно объясняют 91,398 % изменчивости формы тимуса. Вклад первой главной компоненты в общую дисперсию прокрустовых координат составляет 40,236 %. РС1 (–) показывает изменения формы тимуса, в большей степени затрагивающие верхушки его долей, среднюю часть правого контура и весь левый контур тимуса. Эллипсы рассеивания ординат тимусов, принадлежащих крысам, подвергавшимся влиянию ФА, в пространстве первых двух канонических переменных расположены выше по отношению к таковым контрольных животных. Заключение. Ингаляционное воздействие формальдегида приводит к изменению формы тимуса белых крыс. Наибольшие различия с контрольными данными определяются вдоль второй канонической переменной. Incorporation of sodium nitrite in the diet of cats and white rats on the basis of an average sized man consuming 1 lb. (454 g.) of fish containing 0.2 per cent (908 mg.) of this salt daily for six days each week does not appear to affect their growth rate nor the development (weight) of their thyroid, heart, lungs, spleen, liver, kidneys or adrenals. The fecundity of white rats as judged by their ability to breed and raise normal litters is apparently not affected thereby. The lethal dose of sodium nitrite by oral route is about 1.1 to 2.0 g./kg. for healthy male rats, 0.46 to 1.2 g./kg. for healthy female rats and 0.073 g./kg. for cats (one animal). The lethal dose by subcutaneous route is about 0.19 to 0.20 g./kg. for healthy male rats and 0.057 to 0.13 g./kg. for healthy female rats.

Medicinal plants are widely used as supplements for the treatment of various diseases because of their few side-effects. Here, we examined the anti-obesity effects of a mixture extract of Cornus officinalis and Ribes fasciculatum (CR) in high-fat diet (HFD)-induced obese male mice. Four week old male C57BL/6J mice were fed a normal diet (ND) or 60% high-fat diet (HFD) with different concentrations of CR extracts (75, 150, and 300 mg/kg/day) by oral administration for 12 weeks. CR extract administration prevented HFD-induced weight gain, hepatic steatosis, and adipocyte enlargement through the downregulation of adipogenesis-associated genes in obese male mice. In addition, CR administration improved the impaired glucose metabolism, insulin action, biochemical obesity parameters, and metabolic profiles in HFD-induced male mice. Consequently, the CR extract exhibited beneficial effects on HFD-induced systemic metabolic challenges. Taken together, our findings suggest that CR extract may be a potent therapeutic supplement for the treatment and prevention of obesity.

The sufferers of Diabetes Mellitus (DM) in Indonesia is the highest with fourth number in the world, with a mortality rate of 1.2 million in 2012. Because of it, DM research using antihyperglycemic green tea is needed. This study aims to determine the effectiveness of green tea extracts on decreasing fasting blood glucose (KGDP) levels in alloxan-induced white male Wistar rats. Samples were 30 male rats, aged 8-12 weeks, weight ± 200 grams. Rats were grouped into 6 groups with the first group (K1) as a control, the second group (K2) induced alloxan, the third group (K3) induced alloxan with glibenclamide, the fourth group (K4), the fifth group (K5), and the sixth group (K6) induced alloxan with green tea extract as the treatment with dose of green tea extract, 200 mg/kg, 400 mg/kg, and 800 mg/kg. The first, FBG level examination is done 3 days after induction of alloxan using a glucometer. Giving green tea extract was given for 16 days orally, then FBG level re-examined from the tail of rats. One Way ANOVA Test Results, there is the effectiveness of green tea extract on reducing FBG level (p = 0,000). The post hoc Bonferroni test showed that giving 800 mg/kg of green tea extract was the best dose in reducing KGDP close to normal (p = 0,000).

Introduction. The toxicity of indomethacin was studied for its hygienic regulation. Material and methods. The toxic properties of indomethacin in the experiments on out-bred and linear mice, rats, Guinea pigs and rabbits contained in standard vivarium conditions and quarantined have been studied. In the experiments, various modes (single, repeated, chronic) and ways of exposure (intragastric, inhalation, epicutaneous) were modeled. The average lethal dose (LD50) of Indomethacin and the threshold of a single acute action (Limac) were determined; irritant effect on the skin and mucous membranes, cumulative and allergenic activity were revealed. In subacute and chronic intake to the body, the main target organs were determined on the based of the results of biochemical and hematological studies. Results. DL50 for male rats, females and male mice, when introduced into the stomach, were have been established to be 20, 15 and 25.6 mg/kg respectively. It refers to the substances of hazard class 2. DL50 in the intraperitoneal introduction for the rats accounted for 13.8 mg/kg, for Guinea pigs - 500 mg/kg. The clinical picture of acute poisoning in mice and rats was characterized by low mobility, decreasing breathing, ataxia, muscle relaxation, anorexia, diarrhea, ulceration with the perforation of the intestines, and the death on the 2-4th days after the poisoning. In the experiments on Guinea pigs, the ulcerogenic effect was not detected. Local irritant effect on the skin and mucous membranes of the eyes was not revealed. It has a marked skin-resorptive action causing ulcerogenic effect and the death of the animals after 6 applications. The introduction of verospiron to the rats in a dose of 25 mg/kg simultaneously with the application of indomethacin ointment on the skin prevented the ulcer development in the gastrointestinal tract and the death of the animals. No sensitizing effect was detected. It has an average cumulative ability: the cumulation coefficient amounted to 2.6. In a subacute experiment, there was a decrease in the body temperature and summation-threshold index, an increase in the vertical motor activity and a threshold of pain sensitivity. During the study of blood serum an increase in AcAt activity, a rise of chlorides in the blood serum and their decrease in the urine, and an increase in the number of erythrocytes and hemoglobin in peripheral blood were revealed. In the pathomorphological study, there was an increase in the coefficients of liver mass and ulceration of the stomach and intestines. The threshold of acute inhalation action accounted for 0.52 mg/m3 (by the reduction of the summation-threshold index and the content of sodium and chlorides in the urine). Conclusion. The maximum permissible concentration of indomethacin in the air of the working area was of 0.05 mg/m3 with the mark “special protection of the skin and eyes”, hazard class 1, aerosol.

The rational organization and implementation of effective disinfection activities plays an important role in the complex of measures for the prevention of infections. The development and introduction of new disinfectants into production is an urgent issue of modern poultry farming. When developing a disinfectant, it is important to determine the cumulative effect of the drug. The purpose of the study was to determine the cumulative and skin-resorptive action of the disinfectant “Zoоdizin”. For the toxicological study of the drug used healthy white male rats and white female rats weighing 200 ± 10 g 1.5 years of age. To study the toxicity of the drug “Zoоdizin” when applied to the skin used the method of immersion of the tails of rats in a test tube with the test substance. The tail was injected 2/3 into a regular tube with a 5 % solution of the drug “Zoоdizin”. The tube was closed with a cork ring whose diameter was slightly larger than the tail diameter. For 15 days, the tubes were placed daily in a water bath at 28–30 °C for 2 hours. Control animals tails were immersed in distilled water. To establish the local action of the drug “Zoodizin” on the mucous membranes of the study drug was introduced into the conjunctival sac of the right eye of the rabbit at a dose of 50 mg, and in the left eye was buried saline in a volume of 0,05 cm3. When studying the cumulative effect of Zoоdizin, no significant changes in the biochemical parameters in the serum of rats were observed. In the study of possible irritant or damaging effect on the skin and the development of contact non-allergic dermatitis found that a single application of disinfectant “Zoоdizin” on the unaffected skin of the back of white rats in the maximum significant recommended concentration of working solutions (2 %) did not cause signs. The single effect of the drug on the intact areas of the skin did not cause skin irritation, but it can be stated that prolonged daily epicutaneous exposure of high concentration (5 %) of the solution of the drug “Zoоdizin”, which is 2.5 times higher than the maximum recommended concentration, caused a general resorption. When assessing the cumulative properties, it was taken into account that the total dose administered to rats was Zodizin 42000 mg/kg body weight and did not result in animal death. It did not allow to calculate the cumulative coefficients for the “lethal effect”. A single effect of the product on the intact areas of the skin did not cause skin irritation, but it can be stated that prolonged daily epicutaneous exposure of a high concentration (5 %) of the Zodizin solution, which is 2.5 times the maximum recommended concentration, caused a general resorption. In the future, it is planned to study the virulidal properties of the biocide “Zoоdizin”.

Guava (Syzygium aqueum), a typical Indonesian crop plant, has diuretic properties. Plants used as diuretic medicines can also be valuable sources for endophytic fungi containing diuretic drugs. The aims of this study were to isolate and identify endophytic fungi from S. aqueum stems, to isolate their secondary metabolites, and to conduct in vivo tests of the diuretic activity of an endophytic fungus extract on white male rats of the Wistar strain. Endophytic fungi were isolated by a direct plating method, and fungal isolates were identified molecularly. A phenolic compound was isolated by chromatography, and the chemical structure was identified spectroscopically. Doses of 100, 200, and 300 mg/kg BW were administered to white male rats divided into six groups (normal control, negative control, positive control, and the three treatment groups). Diuretic tests included urine volume; sodium, chloride, and calcium electrolyte levels; and pH. Phylogenetic analysis identified the endophytic fungal isolate as Trichoderma ghanense (isolation code SA1). The secondary metabolite isolated from the T. ghanense extract was a phenolic compound. Diuretic tests using the endophytic fungus extract showed the highest urine volume with a dose of 300 mg/kg BW. The diuretic test results showed an increase in urine volume and levels of sodium, potassium, and chloride ions in the urine. The extract of T. ghanense isolated from the stem of S. aqueum has the potential as initial therapy for hypertension because it contains phenolic secondary metabolites that show diuretic activity.

Aim: to study morphometrically the peculiarities of remodeling of chondrocytes of the articular surfaces of the temporomandibular joint in hyperglycemia.Materials and methods: Quantitative morphological methods were used to study diameters of chondrocytes and their nuclei, nuclear-cytoplasmic relationship in these cells, relative volume of damaged chondrocytes, intercellular-chondrocytic relationship in mature chondrocytes of the articular surfaces of the temporomandibular joint of 45 laboratory mature white male rats with simulated diabetes.Results: A comprehensive analysis of the morphometric parameters presented in the article showed that they did not differ in the articular surfaces of the temporal fossa and mandibular head of the intact temporomandibular joint. The expressed structural changes of chondrocytes were revealed at a monthly and a two-month diabetic arthropathy of this joint.Conclusions: Proceeded studies and obtained results suggest that prolonged hyperglycemia leads to diabetic arthropathy of the TMJ, which is characterized by atrophy of chondrocytes, violation of nuclear-cytoplasmic relationships, an increase in the relative volume of damaged chondrocytes and volume of intercellular substance. The detected changes in quantitative morphological parameters depended on the duration of hyperglycemia and dominated in the cartilaginous tissue of the articular surface of the mandibular head.

Aluminum is recognized as a public health concern because of its potential toxic effects on human health. Therefore, the present experiment was undertaken to determine the effectiveness of curcumin (CUR) in reducing the hepatotoxicity induced by aluminum chloride (AlCl3) in animals. In this experimental study, forty male rats were allocated to five groups (N = 8), viz. no treatment (control), solvent (DMSO or distilled water), CUR (10 mg/kg B.W.), AlCl3 (10 mg/kg B.W.), and CUR+AlCl3 (each with 10 mg/kg B.W.). Treatments were performed by intra-peritoneal injections for 28 days. On the final day, animals were sacrificed, and liver function markers in blood plasma, hepatic antioxidants, and lipid peroxidation index in liver homogenate were estimated. AlCl3 treatment resulted in a significant increase in plasma AST, ALT, ALP and LDH activities with decreased total protein compared to control. AlCl3 significantly reduced superoxide dismutase (SOD), catalase (CAT) and glutathione (GSH) levels but increased malondialdehyde (MDA) level in the liver compared to control. AlCl3 also caused various histopathological changes in the livers of rats. Curcumin could normalize nearly all these parameters. CUR improved levels of changes in different parameters when was combined with AlCl3. It is concluded that CUR has beneficial effects being able to antagonize AlCl3 toxicity.
