Phytochemical analysis, total phenolic, total flavonoid contents and ferric reducing power of extracts from roots and leaves of Searsia burchellii

Searsia burchellii has therapeutic applications in traditional medicine. The aim of the present study was to analyse the phytochemical profiles, total phenolic contents (TPC), total flavonoid contents (TFC) and ferric reducing power of various extracts from roots and leaves of S. burchellii collected from the kingdom of Lesotho. A combination of maceration, hot solvent extraction and solvent-solvent partition techniques were used to obtain methanolic extracts, fractions such as hexane, chloroform, ethyl acetate and methanol/water from methanolic extracts and water extracts separately from the roots and leaves of S. burchellii. The presence of alkaloids, steroids, terpenoids, phenolics, tannins, flavonoids, coumarins, saponins, glycosides, carbohydrates, proteins and phlobatannins were identified from these extracts/fractions. The TPC of these extracts/fractions was found to be in the range of 3.22±0.37 to 247.46±4.33 mg TAE/g DW, respectively. The TFC of these extracts/fractions was found to be in the range of 34.70±4.31 to 278.20±2.01 mg QE/g DW, respectively. Additionally, these extracts/fractions showed a moderate to strong ferric reducing power compared to ascorbic acid. The IC50 values were found to be in the range of 5.35-143.12 and 65.64-348.28 µg/mL for extracts/fractions from the roots and leaves, respectively. Ascorbic acid showed an IC50 value of 20.93 µg/mL. Therefore, the extracts/fractions from S. burchellii have the potential to replace the existing synthetic antioxidants

Chloroform, ethyl acetate and methanolic extracts from leaves and stem bark of Buddleja salviifolia were prepared. The phytochemical profiles, total flavonoid contents (TFC), total phenolic contents (TPC) and ferric reducing power of these extracts were studied. The presence of alkaloids, tannins, phlobatannins, saponins, sterols, flavonoids, coumarins, phenolics, terpenoids, quinones and reducing sugars were identified from these extracts. The TFC of these extracts was found to be in the range of 11.36±0.98 to 30.76±1.15 mg QE/g DW, respectively. The TPC of these extracts was found to be in the range of 175.00±0.41 to 465.27±2.65 mg GAE/g DW, respectively. Additionally, these extracts showed a weak to moderate ferric reducing power compared to ascorbic acid. B. salviifolia finds therapeutic applications in traditional medicine. Further studies are required to commercialize products from this plant.

Sideritis niveotomentosa Hub. -Mor. is a local endemic species belonging to the Lamiaceae family. In this study, GC/MS analysis, total antioxidant capacity and anticancer effects of different extracts obtained from S. niveotomentosa were investigated comparatively. Total phenolic contents of extracts were determined by the Folin–Ciocalteu method, total flavonoid contents by aluminum chloride method, and also the free radical scavenging activities of the extracts by DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) assay. The cytotoxic effect of the extracts was studied via MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide) assay on DLD1, HL60 and ARH77 cell lines. Pro-apoptotic gene expression levels were also tested in the most sensitive cell line ARH77 by Real-Time PCR. The expression levels of 4 pro-apoptotic genes, APAF, BAX, CASP3, and HRK were found to be upregulated in ARH77 cells that were treated extracts. Results showed that methanolic extracts contain more phenolic content than acetone extracts, consistent with DPPH results. As a result, Sideritis niveotomentosa extracts, especially methanolic extracts, are rich in phenolic content and have a strong radical scavenging effect. In addition, the extracts showed selective effects on cell lines. This study is pioneering in terms of future studies, and the findings provide hope for future experimentation.

Modern civilization is facing more than hundreds of disorders associated with free radicals and natural antioxidants from non-edible plants are gaining importance to fight these disorders. The intention of this report is to evaluate a well known medicinal weed Ageratum conyzoides stems for its antioxidant and cytotoxic Effects. Antioxidant potentiality of the crude methanolic extract of the Ageratum conyzoides (AC) stems was investigated on DPPH scavenging activity, reducing ability, total antioxidant capacity as well as total phenolic contents. Cytotoxic study was done by brine shrimp lethality bioassay and vincristin sulphate was used as standard. The total phenols and total antioxidant capacity of AC was found to be 38.125 ± 2.01mg/g equivalent of gallic acid and 333.37 ± 4.22mg/gm equivalent of ascorbic acid, respectively. The percentage (%) scavenging of DPPH free radical of the extract was found to be concentration dependent with IC50 value 46.01 ± 2.23µg/ml while IC50 value of standard ascorbic acid was found to be 29.56 ± 0.11?g/ml. The reducing power of AC was found to be concentration dependent. The cytotoxicity exhibited by AC was found promising with LC50 value 1.32?g/ml, comparing with the LC50 (0.689?g/ml) values of vincristin sulphate. The Ablmoschus manihot (L.) Medic, an edible hibiscus of the Malvaceae family, is abundant with total flavonoid. The content of total flavonoid from A. manihot flowers (TFA) was extracted by 70% ethanol (ethanol:water, 70:30). The scavenging effects of TFA on superoxide anions and hydrogen radicals were related to the total flavonoid concentrations with the IC50 values of 63.90 ±2.21 μg/mL and 266.88±28.32 μg/mL, respectively. However, the IC50 values of standard ascorbic acid were 436.52 ±14.36 μg/mL and 439.58±21.41 μg/mL, respectively. The DPPH radical scavenging activity increased with increasing of TFA concentrations and the highest inhibition was 94.63 ±3.01% at 50 μg/mL, as compared to ascorbic acid 34.94 ±1.50%. The extract also showed good reducing power. The data from this study suggest A. manihot flower extract has significant potential to use as health supplements and nutraceuticals.

Searsia burchellii finds therapeutic applications in traditional medicine. Methanolic extracts, hexane, chloroform, ethyl acetate and methanol/water fractions of methanolic extracts and water extracts were obtained separately from the roots and leaves of Searsia burchellii by the combination of maceration, hot solvent extraction and solvent-solvent partition techniques. These extracts were evaluated for their antioxidant activity using 2,2- diphenyl-1-picrylhydrazil radical scavenging assay (DPPH). The extracts from roots and their fractions showed radical scavenging activity ranging from 6.60±4.50 to 63.27±1.93% at various concentrations. Similarly, the extracts from leaves and their fractions showed radical scavenging activity ranging from 3.32±0.95 to 64.91±0.15% at various concentrations. Ascorbic acid served as positive control which showed radical scavenging activity ranging from 53.62±2.80 to 60.82±0.62% at various concentrations. The IC50 values of these extracts and fractions were found to be < 200 to > 3000 µg/mL. The IC50 value of ascorbic acid was found to be <200 µg/mL. From this study, we concluded that extracts and their fractions from S. burchellii showed promising radical scavenging activity.

Abstract Background: Nymphaea capensis is an aquatic flowering plant which is included in the family of Nymphaeaceae. Literature review on the plants of Nymphaeaceae family exhibited significant medicinal activities. Therefore, the objective of the present study is to evaluate possible anti-oxidant activity of crude methanol extract of N. capensis leaf.Method: In antioxidant study, methanolic plant extract was evaluated for 1,1-diphenyl,2-picrylhydrazyl (DPPH) and reducing power capacity. Moreover, total phenolic and total flavonoid content of plant extracts were determined and expressed in mg of gallic acid equivalent per gram of dry sample (mg GAE/g dry weight).Result: In the DPPH free radical scavenging assay, methanol extract showed concentration dependent inhibition of the free radicals. IC50 of Ascorbic acid was 14.84 µg/ml whereas N. capensis was 130.94 µg/ml. In case of reducing capacity, at conc. 62.5, 125, 250, 500, 1000 ug/ml, the absorbances of Ascorbic acid were 0.65, 1.12, 1.45, 1.78 and 1.89 respectively. In case of N. capensis, the absorbances were 0.46, 0.75, 1.04, 1.27 and 1.50 respectively. The extract displayed a concentration dependent increase in reducing power. In the case of total phenolic content, the methanol extract of N. capensis contained a considerable amount of phenolic contents which was 215±7 of GAE mg/g. In the case of total flavonoid content, methanol extract of N. capensis contained significant amount of flavonoid contents which was 184.75±6.78 of GAE mg/g.Conclusion: These results suggested that the methanol extract of N. capensis possess considerable anti-oxidant activity.

Objective: Plant phenols are extensively studied, thanks to their many prophylactic (anti-tumoural, ulcer, inflammatory) and therapeutic effects. Their many antioxidants may protect against diabetes, cancer, autoimmune diseases. Our objective was to evaluate antioxidant and antimicrobial activities of our samples: Cocos nucifera (shell), Punica granatum (peel), Citrus limonum (rind), and Ocimum sanctum (stem); in conjugation with phytochemical analysis. Methods: The hydro-methanolic extracts of the selected plant parts were assessed for polyphenols. Antioxidant (ferric reducing power assay) and antibacterial (Kirby Bauer disc diffusion) assays were run for varying concentrations against-Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Salmonella typhi. Results: The results showed that the samples all contained polyphenols with high total phenolic activity in pomegranate and high antibacterial activity in coconut. A more comprehensive study could lead to their emphatic incorporation into mainstream medicine and pharmaceuticals–furnishing natural alternatives to their chemical counterparts. Conclusion: The present study concludes that hydro-methanolic extracts of the samples contain phytochemicals in high concentrations, conferring upon them promising antioxidant and antimicrobial activity.

Objective: This study was aimed to analyze the methanol or ethanol extract of Camellia mistletoe (Korthalsella japonica) growing in Jeju Island, Korea, for their composition of phenolics, flavonoids, and their antioxidant activity.Methods: Total flavonoid, total carotenoid and L-ascorbic acid contents were determined by using colorimetric assays, and the antioxidant attributes of the methanol and Camellia mistletoe extracts were assayed using in vitro models, including the following: Free radical (1,1-diphenyl-2-picrylhydrazyl, O2-• and NO•)scavenging, reducing power, and chelating ferrous ions.Results: Total flavonoid, total carotenoid and L-ascorbic acid contents were 20.3 and 14.9 mg RE/100 g, 4.4 and 1.7 mg/100 g, and 0.4 and 1.4 mg AA/100 g in the methanol and ethanol extracts of Camellia mistletoe, respectively. Based on IC50 values, both extracts were also found to possess potent antioxidant activities.Conclusions: Camellia mistletoe extracts could be the good antioxidant source and holds promise as a natural ingredient in functional food and pharmaceutical supplement.

Arbutus unedo L. is a plant widely distributed in the Mediterranean basin and North Africa, frequently used in traditional folk medicine to treat diabetes and arterial hypertension. This study aims to evaluate the phenolic composition and antioxidant activity of ethyl acetate (EA.E) and n-butanolic (But.E) extracts prepared from leaves of Arbutus unedo. Total phenolic and flavonoids contents were determined spectrophotometrically. The antioxidant activity was evaluated using DPPH•, OH•, H2O2, O•-2, ß-carotene bleaching and AAPH-induced erythrocyte oxidative hemolysis assays. The phytochemical analysis showed the presence of polyphenols and flavonoids in both extracts. The high amount was observed in EA.E which exerted the stronger antioxidant effect, with IC50 values of 3.43 μg/mL, 323.45 µg/mL, 38.40 µg/mL and 11.56 µg/mL, in DPPH•, OH•, O•-2 and H2O2 tests, respectively. Both extracts inhibited β-carotene bleaching, but EA.E is always more potent (92%) than But.E (85%). Furthermore, the EA.E showed the highest protective effect on erythrocyte hemolysis induced by AAPH, with half time hemolysis (HT50) of 122.02 min at 40 µg/mL. Taken together, this study showed that Arbutus unedo leaf extracts possess strong antioxidant potential, which may be attributed to the presence of a high amount of polyphenolic constituents. So, this plant might be exploited as a potential source of natural antioxidant agents for pharmaceutical and food applications. In the present study, methanol and aqueous extracts ofParkinsonia aculeataL. leaves were prepared and analyzed for phytochemical analysis and antioxidant potential in differentin vitroassays. Antioxidant activity was studied using DPPH, CUPRAC, reducing power assay, deoxyribose degradation (site and nonsite specific), ferric reducing antioxidant potential (FRAP), ferric thiocyanate (FTC), thiobarbituric acid (TBA), and molybdate ion reduction, respectively. The total phenolic contents of the methanol and aqueous leaf extract were 39 mg GAE/g and 38 mg GAE/g, whereas flavonoid contents of these extracts were found to be 0.013 mg RE/g and 0.006 mg RE/g, respectively. From the two extracts, the methanol extract shows maximum inhibition (%) of 57.82%, 71.23%, 48.26%, 69.85%, and 52.78% in DPPH, nonsite- and site-specific, FTC, and TBA assays and absorbance of 0.669 and 0.241 in reducing power and CUPRAC assays at the highest concentration tested. UPLC analysis was done to determine the presence of various types of polyphenols present in plant extracts.
