Flaky graphene oxide (GO) nanoparticles (NPs) were synthesized using Hummer’s method and then capped with polyethylene glycol (PEG) by an esterification reaction, then loaded with Nigella sativa (N. sativa) seed extract. Aiming to investigate their potential use as a smart drug delivery system against Staphylococcus aureus and Escherichia coli, the spectral and structural characteristics of GO-PEG NPs were comprehensively analyzed by XRD, AFM, TEM, FTIR, and UV- Vis. XRD patterns revealed that GO-PEG had different crystalline structures and defects, as well as a higher interlayer spacing. AFM results showed GONPs with the main grain size of 24.41 nm, while GONPs–PEG revealed graphene oxide aggregation with the main grain size of 287.04 nm after loading N. sativa seed extract, which was verified by TEM examination. A strong OH bond appeared in FTIR spectra. Furthermore, UV- Vis absorbance peaks at (275, 284, 324, and 327) nm seemed to be correlated with GONPs, GO–PEG, N. sativa seed extract, and GO –PEG- N. sativa extract. The drug delivery system was observed to destroy the bacteria by permeating the bacterial nucleic acid and cytoplasmic membrane, resulting in the loss of cell wall integrity, nucleic acid damage, and increased cell-wall permeability.

The aim of this study was to determine the protection effect of SNEDDS piroxicam ulcerogenic agent against malondialdehyde (MDA) level and protein expression of caspase-3, COX-1, COX-2. The research was conducted using the test animals as much as 30 male white Sprague dawley (SD) rats aged 1-2 months with a weight of 100-200 grams divided into 5 groups. Treatment was given for 28 days orally. On the 29th day blood samples were also taken for the determination of MDA (Malondialdehid) levels by Thiobarbituric Acid Reactive Substance (TBARs) method using a visible spectrophotometer. Rats were sacrificed, then gastric organs were taken for immunohistochemical testing of caspase-3 and COX-1 expression, COX-2. The statistical analysis showed that the piroxicam SNEDDS group and the piroxicam suspension group decreased expression of the caspase-3 protein, increased COX-1 expression, decreased COX-2 and significantly decreased MDA levels. The piroxicam-containing SNEDDS (Self-Nanoemulsifying Drug Delivery System) form has protection against ulcogenic piroxicam.

Background: Functional and nutraceutical foods are gaining immense popularity among the masses. Plants and their bioactive molecules are of prime importance. Although various plants from different geographical areas have been tested in the past, many horizons still need to be addressed. Black cumin (Nigella sativa L.) is one such example that is quite popular in South Asia and the Middle East.Context and purpose: The present research study was designed to expedite the role of black cumin seed in reducing the risk of hypercholesterolemia. For the purpose, thirty Sprague dawley rats were procured from the National Institute of Health (NIH) in Islamabad, Pakistan, and further split up into three groups, (10 rats each). Experimental diets were prepared using powdered black cumin (PBC) at 1% and 2%, and compared with the placebo. Results: The results revealed that PBC was effective in reducing the serum cholesterol, triglycerides, and low-density lipoproteins (LDL). Additionally, the experimental diets resulted in a non-significant increase in high-density lipoprotein (HDL). Overall, powdered black cumin at 1% and 2% reduced cholesterol level by 6.73, and 4.48%, LDL by 24.79, and 24.32% respectively. However, the supplementation of PBC at 2% resulted in marked variations as increasing tendency, which was recorded for cholesterol and triglycerides contents after 28 days of study. Conclusion: Present research investigation brightened the prospects of using powdered black cumin seed in diet based therapies to improve the lipid profile. Further studies are still required to assess the phytochemistry of the plants and indeed the functional ingredients responsible for such health benefits. Such studies would bring meticulousness for utilization of black cumin seeds as a functional food.Keywords: Functional foods, black cumin, lipid profile, cholesterol, triglycerides

Aims: The aims of this research were to develop and characterize liquid and solid micro emulsifying drug delivery system (L-SMEDDS and S-SMEDDS) containing black seed oil.  Study Design: Experimental Research Design (laboratory). Place and Duration of Study: The study was conducted at research laboratory of pharmacy department UNISBA, between August 2018- August 2019. Methodology: The optimization of L-SMEDDS was carried out using various comparison of oil, surfactant, and cosurfactant. All formulations were evaluated for percent transmittance, emulsification time, dispersibility, robustness, and thermodynamic stability. The best formula of L-SMEDSS was evaluated for globule size distribution and converted to S-SMEDDS by spray drying method using aerosil 200 as adsorbent. S-SMEDDS were evaluated for organoleptic, flowability, compressibility, emulsification time, dispersibility, robustness and surface morphology. Results: The best formula of L-SMEDDS contains tween 80 as a surfactant and PEG 400 as cosurfactant (2:1) with a ratio of oil and Smix (2:8). The L-SMEDDS preparation meets the requirement of percent transmittance (95.77%), emulsification time (37.67 seconds), grade A of dispersibility, stable of robustness and thermodynamics study with the average of globule size was 231 nm. S-SMEDDS preparation meets the requirement of the moisture content, flowability, emulsification time, and stable on robustness testing with a spherical shape. Conclusion: L-SMEDDS and S-SMEDDS of black seed oil have been developed and have good physical characteristics and stability.

<p>Jinten hitam (<em>Nigella sativa</em>) mengandung senyawa timokuinon yang berefek sebagai imunostimulan. Ekstrak jinten hitam dikembangkan menjadi SNEDDS (<em>Self-nanoemulsifying Drug Delivery System</em>) karena masalah kelarutan. Penelitian dilakukan untuk mengetahui karakteristik SNEDDS ekstrak jinten hitam yang meliputi viskositas, ukuran tetesan nanoemulsi, <em>extract loading</em>, dan stabilitas. Pengujian aktivitas imunostimulan SNEDDS meliputi rasio sel makrofag dan indeks fagositosis.</p><p>SNEDDS ekstrak jinten hitam dioptimasi dengan metode <em>Simplex Lattice Design</em> menggunakan <em>Design Expert 7.1.5., </em>selanjutnya SNEDDS optimal diuji ukuran tetesan nanoemulsi dan zeta potensial, viskositas, serta uji stabilitas. Uji aktivitas imunostimulan dilakukan dengan metode <em>biolatex assay</em> terhadap tikus <em>Sprague Dawley</em> sebanyak 5 tikus/kelompok selama 15 hari dengan pemberian satu kali sehari yaitu kontrol positif (ekstrak meniran 7,2 mg/tikus), kelompok perlakuan yaitu ekstrak jinten hitam dengan dosis 200 mg/kgBB serta SNEDDS ekstrak jinten hitam (200 mg/kgBB), kelompok plasebo berupa formula SNEDDS tanpa ekstrak jinten hitam, dan kontrol normal, selanjutnya dihitung rasio dan indeks fagositosis makrofag.</p>SNEDDS ekstrak jinten hitam optimal mengandung 15% minyak ikan hiu cucut botol, 67,344% surfaktan (10,102% croduret 50 ss dan 57,242% tween 80), 17,656% PEG 400 sebagai ko-surfaktan dengan hasil ukuran tetesan nanoemulsi 16,3 nm, PI sebesar 0,202, zeta potensial -43,5 mV, dan viskositas antara 234,69 – 255,71 cP. Hasil <em>extract loading</em> sistem SNEDDS mencapai 600 mg ekstrak/g sistem. SNEDDS stabil setelah penyimpanan selama 90 hari pada suhu kamar dan uji <em>freeze-thawing</em>. SNEDDS ekstrak jinten hitam dengan dosis 200 mg/kgBB dapat meningkatkan rasio sel makrofag dan indeks fagositosis dibandingkan dengan ekstrak jinten tanpa formulasi (P&lt;0,05).
