This study was conducted in Al-Alam region, which is located in Salah Al-Din Governorate, on first month pregnant Iraqi local cows (n-10). The follow up extended from the first month of pregency up to the end of the first month post-parturituion during that some clinical and biochemical parameters were measured in the serum. Blood samples from the jugular vein of cows were collected monthly for the whole period of experiment and divided into four stages: early pregnancy, mid pregnancy, late pregnancy and early lactation immediately after birth. It was observed that the temperature, respiration rate, and heart rate increased gradually and significantly (P ≤ 0.05) with the progress of pregnancy reaching to its highest value in the last period of pregnancy and decreased after birth. The last trimester of pregnancy and the early lactation were also showed a significant decrease (P ≤ 0.05) of the phosphorus, calcium, iron and copper concentrations compared to the first and second trimesters of the pregnancy howecer, significant increasees in other biochemical values (P ≤ 0.05) were observed in urea and creatinine concentrations in the last trimester of pregnancy and early lactation. In addition to a significant decrease (P ≤ 0.05) of the ratios of concentrations of glucose, total protein, albumin and glubulin in the last period of pregnancy compared to other periods. The decrease in the value of glubulin continued until the early birth period while the study did not showed any significant difference in the concentration of bilirubin between durations. Finally, values of the ALT, AST and ALP enzymes showed significant increases (P ≤ 0.05) in thier concentrations in the last period of pregnancy. It is concluded that possible changes in the biochemical parameters of the local Iraqi cows blood during pregnancy and early lactation are existed

Background: Dairy goats are highly susceptible to subclinical hyperketonemia (SCHK) during the transition period. This study aimed to compare the variation in metabolic parameters and surrogate indexes of insulin resistance (sIR) between goats with SCHK and clinically healthy (HEAL) goats during the transition period. Methods: Twenty Guanzhong dairy goats were assorted to HEAL (n = 10) and SCHK (n = 10) groups according to the blood β-hydroxybutyrate (BHBA) concentrations. The blood samples were taken from the jugular vein of each goat at −3, −2, −1, 0 (partum), +1, +2, and +3 weeks relative to kidding to analyses GLU and INS. The sIR was calculated from blood metabolic parameters. Results: Compared with the HEAL goats, the insulin concentrations were significantly higher in SCHK goats during the first three weeks postpartum. The QUICKI, revised QUICKI (RQUICKI), and RQUICKIBHBA were significantly lower in goats with SCHK at 1 week postpartum, while the homeostasis model assessment-IR (HOMA-IR) was significantly higher. Conclusion: Goats with SCHK made more efforts through elevated insulin levels at early lactation than HEAL goats, thereby maintaining the normal glucose concentrations.

SummaryThe aim of this study is to investigate the influence of hemolysis degrees on the values of hematological and biochemical parameters in the blood samples of cows in early lactation. The study included a total of 45 blood samples of cows in early lactation. Out of the total number of samples analyzed, 15 indicated no signs of hemolysis, whereas 15 indicated moderate hemolysis and 15 severe hemolysis. The influence of hemolysis was examined by determining the bias (%) and by means of the Wilcoxon signed-rank test. The results showed that the hemolyzed samples had decreased values of the HCT, RBC, MCV, and increased values of the MCH, MCHC, and PLT. The total number of LEU showed a declining trend with increasing degrees of hemolysis. The changes in the HGB values were not detected despite the visual identification. The hemolysis degrees showed a significant influence on the values of biochemical parameters. The variations in biochemical parameters were less significant in the samples with moderate hemolysis compared to the samples with severe hemolysis. The most hemolysis-sensitive biochemical parameters were the AST, TBIL, TGC, and NEFA as great variations of their values were found even in moderate hemolysis. A slight deviation was noted in the BHB and total protein values. With respect to all the parameters examined, the Wilcoxon test showed significant differences in the samples with moderate and severe hemolysis in comparison with the control group. In conclusion, hemolysis significantly influenced the values of hematological and biochemical parameters in the blood samples analyzed.

This study was conducted to determine the effect of bone marrow stromal cells (BMSCs) on progesterone level during pregnancy in ewes. Flurogestone Acetate Sponges 40 mg, followed by 400 i.u. Equine chorionic gonadotropin (eCG) were used to synchronize estrus and ovulation. The animals were divided into three equal groups (5 animals for each group). The 1st and 2nd group injected intravenously after eCG injection with 1x108 and 2x108 respectively with BMSCs while the 3rd group was injected with normal saline which serve as a control group. Blood samples were collected during pregnancy, at day 10, 21, 85 from the jugular vein. At day 10, 21 the results showed that there was a significant difference (p≤0.05) in the level of progesterone between treated groups as compared with the control group. While there was no significant difference between different groups at day 85. It was concluded from this study that BMSCs have a beneficial effect in ewe’s reproductive system, by increasing the level of progesterone at early pregnancy.

S-100B is used in melanoma follow-up. This serum biomarker is also present in adipocytes; therefore, subcutaneous adipocytes trapped in the needle before performing a venipuncture could contaminate the serum. The aim was to study the influence of adipocyte contamination on blood samples used for S-100B analysis, possibly resulting in falsely elevated S-100B values. A total of 294 serum samples were collected from 147 American Joint Committee on Cancer staging stage III melanoma patients. The mean difference between the first (dummy) and second tubes was 0.003 μg/l (p = 0.077), with a decrease in the second tube. Compared with the second tube, the S-100B level was higher in the first tube in 33.3% of the samples, equal in 36.8% of the samples and lower in 29.9% of the samples. No significant difference between the two consecutively drawn tubes was found. There seems to be no necessity of implementing a dummy tube system for accurate S-100B determination in melanoma patients.

Abstract Introduction: Acromegaly is a rare disease due to growth hormone (GH)-secreting pituitary adenoma. GH and IGF-1 levels are usually congruent, indicating either remission or active disease, however a discrepancy between GH and IGF-1 may occur. We aimed to evaluate the outcome of acromegalic comorbidities in patients with congruent GH and/or IGF-1 levels vs discordant biochemical parameters. Methods: Retrospective analysis of the data of 3173 patients from the Liège Acromegaly Survey (LAS) allowed to include 190 patients from 8 tertiary referral centers across Europe, treated by surgery, with available data concerning diabetes mellitus (DM) and hypertension (HT) both at diagnosis and at last follow-up. We recorded for all the patients the number of antihypertensive and antidiabetic drugs used at the first evaluation and at last follow-up. Results: Ninety-nine patients belonged to the REM group (Concordant parameters), sixty-five patients were considered as GHdis and 26 patients were considered as IGF-1dis. At diagnosis, 63 patients (33.1%) had HT and 54 patients had DM (28.4%). There was no statistically significant difference in terms of number of anti-HT and anti-diabetic drugs at diagnosis versus last follow-up (mean duration=7.3+/-4.5years) between all 3 groups. Discussion: The results highlight that the long-term outcome of acromegaly does not tend to be more severe in patients with biochemical discordance in comparison with patients considered as in remission on the basis of concordant biological parameters, suggesting that patients with biochemical discordance do not require a closer follow-up.

Objectives The aim of this study was to evaluate the effect of two differently sized butterfly catheter needles and the effect of venepuncture difficulty on thromboelastography (TEG) results in healthy cats. Methods Twenty-four healthy cats were included. Blood samples were collected from the jugular vein by syringe aspiration via direct venepuncture with 21 G and 22 G butterfly needles. The venepuncture difficulty score was classified into four categories. The first 1.5 ml blood drawn from each subject was discarded before collecting a sample for TEG analysis. TEG analyses were performed on citrated whole blood samples from 17 clinically healthy cats, using assays with kaolin as activators. Among the TEG parameters, reaction time (R), clot formation time (κ), alpha angle (α), maximum amplitude (MA) and global clot strength (G) were recorded from each tracing. Results Seven cats were excluded from the study; results were obtained for the remaining 17 cats. There were no statistically significant differences between the use of two different needles for R ( P = 0.72), κ ( P = 0.74), α ( P = 0.99), MA ( P = 0.08) and G ( P = 0.09). Samples with difficulty scores ⩾1 were not significantly different from samples with difficulty scores of 0 for R ( P = 0.24), κ ( P = 0.65), α ( P = 0.65), MA ( P = 0.72) and G ( P = 0.77). Conclusions and relevance The results of TEG in clinically healthy cats do not differ significantly when using two different gauge needles. There was no significant difference in the TEG results according to venepuncture difficulty scoring.

The object of this study was to determine the effect of prepartum supplementation of vitamin E with or without injective vitamin E and selenium (Se) on productive and reproductive performances and immune function in dairy cows. Sixty multiparous Holstein dairy cows were divided randomly into three groups at the end of gestation. Cows in each group received one of three treatments: (1) a single intramuscular (im) injection of vit. E + selenium 3 weeks prepartum; (2) daily supplementation of oral vit. E given from 3 weeks prepartum to parturition; (3) injective vit. E + Se with daily supplementation of oral vit. E. Blood samples were collected from cows at calving and from calves at 0 and 7 days of age. Concentration of IgG in serum of cows and calves as well as in colostrum was determined. No significant differences among treatments occurred in the concentrations of IgG, animal, and calf production and reproduction performance. Due to the lack of significant difference between injection and oral supplementation, it is recommended to replace the injection with oral supplementation.

Introduction. Cerebral infarction is more and more frequently present by massive participation and high percentage of mortality even in young population. It is notified as very hard, vitally endangered disease. Objective. To prove whether there is a difference in the gas analyses of the arterial and cerebral venous blood between the patients with the cerebral infarction of the left and right hemisphere and why it is significant for the treatment of those patients. Material and methods. Sixty-five (65) patients of both sexes who were diagnosed by CT as: cerebral infarction, cerebral multiinfarctions and cerebral haemorrhagic infarctions were investigated. Under the same conditions their radial artery and left and right internal jugular vein were tapped - on the fourth, eighth and tenth day of their disease. Gas analysis, calculation of oxygen content and statistical comparison of testing results were performed from the samples of arterial and venous blood. Results. By continuous monitoring of gas analysis parameters of arterial and cerebral venous blood from internal jugular vein, we can follow up the phases of disease, the velocity of changes of gas analysis parameters and adequacy of brain circulation and to correct therapy adequately for the purpose of preventing basic disease complications and to estimate the results of treatment. Conclusion. There is a significant difference between the results of gas analysis of arteries and the internal jugular vein, in patients with infarctions of the left and right hemisphere of the brain.

     The present study was aimed to check the amount of variation in some hematologic and biochemical parameters accompanied with natural acquired canine dirofilariasis. Blood samples were collected from sixty five stray dogs (5-10) years old belong to local breed dog in the villages of Al-Hindya area/ Karbala Governorate, The affected animal showed differences in hematological and biochemical values as compared with reference ranges. In conclusion, the disease showed no significant clinical signs although many pathological changes in some blood constituent with serum biochemical parameters were observed, before euthanasia for dogs.

Objective: To determine the circulatory miRNA expression levels in patients with Hashimoto thyroiditis (HT) at the time of diagnosis and follow-up period compared with healthy controls. Methods: We collected blood samples from 34 patients with HT (4 males and 30 females) at the time of first diagnosis (Group P) and euthyroid period (Group E). Thirty-three healthy controls (Group H) blood samples were also included in the study. Expression levels of five different circulating miRNAs (miR-22, miR-141, miR-155, miR-375, miR-451) were evaluated using real-time polymerase chain reaction. Results: There was a significant difference in miR-375 levels between the P group and the H. Also, for miR-451, there was a significant difference between the P and E groups. Finally, there was a moderate positive correlation between thyroidstimulating hormone values and miR-22 expression levels for the P group. Conclusion: miRNAs have important roles at all stages of the diseases. More studies must be performed in all thyroid diseases and autoimmune diseases, including HT.
