The study was carried to investigate pathological effects of fimbrial antigen of proteus vulgaris 20 rabbit has been divided into (4) equal groups The first group immunized subcutaneous twicely with (01) ml of pv fimberial Ag (200 μ/ml) with 2 week interval the second group was treated as the 1st group but in a dose containing 100 μ/ml. Third and fourth group considered as positive and negative control groups respectively. After 45 days post immunization first second and third groups were challenged with 01ml of bacteria suspension contain (107x5cfu/ml) of virulent pvulgaris. All rabbits of the third group died during 24-48 hr post challenge with severe congestion in all internal organs associated with necrotic foci mainly in liver and kidney at days 3-20 post challenge immunized infected rabbits (died and survival) were sacrified histopathologically the third group showed focal necrosis and polymorphonuclear cells (PMNCs) infiltration in liver parenchyma pulmonary edema with severe congestion and depletion of spleen Mild pathological changes were revealed in the 1st and 2nd immunized groups characterized by kupffer cell proliferation dilation of sinusoids with mononuclear cells (MNCs) aggregation (mainly lymphocyte) around blood vessels lymphoid hyperplasia in spleen with hyperplasia of goblet cells and secretion of mucin in the intestine mainly in the first groupfimberial antigen considered as effective immunogen for protecting rabbits against p volgaris infection and it is synchronized with dose of this antigen.

The aim of this study to conduct the effects of fimbrial and lipopolysacchride (LPS) immunization is on the pathohistological changes in rabbits, Fifteen rabbits of both sexes (Weight 1500-2000 gm) divided into three groups (5 animals of each group). The first group was immunized by 1ml (200µg /animal) of fimbrial subcutaneously the second group gave 1 ml ( 200 µg /animal) LPS while the third group was left as negative control group that injected 1 ml phosphate buffer control subcutaneously. First and second groups recived the same dose after two weeks give as booster dose. All animals challenged after 5 weeks of immunization by5X107CFU/ml Proteus vulgaris intra peritoneally .After 7 days from challenge all the animals, sacrificed for histopathological examination . The results showed that the fimbrial group had a severe infiltrations of mononucleart cells in liver and kidney ,but there was no clear histopathological changes observed in the spleen compared with lipopolysaccharide and control group . Also this group showed a slight mononuclear cells infiltration in lamina properia of intestine ;while the lipopolysaccharide group showed hypertrophy of epithelial cells with a mild mononuclear cells infiltration in lamina properia as compared with the control group that showed presence of mononuclear cells aggregation in the lamina properia .Our conclusion That the defense of animals against Proteus vulgaris more efficient in fimbrial antigen than lipopolysaccharide antigen by decrease the pathological effects of this bacteria.

     The aim of this study is to describe the morphology of Cysticercus fasciolaris by using light microscopy, and the pathological changes in the liver of rats naturally infected. A total of 50 liver specimens of local rats (Wister rats) were collected for examination. The gross lesions showed the presence of single or multiple cysts. Microscopic findings revealed the presence of larvae within the cysts which represent the larvae Cysticercus fasciolaris of the adult parasite Taeniae taeniaeformis which inhabited the small intestine of the domestic cats surrounded by fibrous connective tissue infiltrated with inflammatory cells (mononuclear cells and plasma cells). These lesions cause pressure atrophy to the adjacent hepatic parenchyma. In advanced hepatic infection there is a tendency to undergo neoplastic changes (fibroma). Other pathological lesions seen in the liver parenchyma were necrosis, apoptosis with infiltration of chronic inflammatory cells in the portal area, in addition; to formation of early granulomas with congestion of blood vessels which contain neutrophiles in their lumina with extensive area of hemorrhages in liver parenchyma. In conclusion the C. fasciolaris infction induce hepatic neoplasia in rat livers (fibroma) in advance cases of heavy infection, which could be developed to fibrosarcoma in future.

The study was carried to investigate the pathological effect of lyophilized antigen of Trichophyton mentagrophytes in mice. Fifty mice were divided into three groups. The first group 20 mice were immunized subcutaneous (s/c) with 0.5 ml of T.mentagrophyte antigen 20 µgm/ml, by two doses, 14 day intervals, between them. The second group 20 mice and third group 10 mice considered as positive and negative control groups respectively. After 30 days post immunization first and second groups were challenged intradermal I/d. with 0.1 ml of fungal suspension contain (1×107 ml) of viable virulence T.mentagrophyte while the third group injected intraperitoneally I/P. with 0.5 ml of sterile phosphate buffer saline. All mice of the first and second groups were sacrificed at (5, 14, 30 and 60) days post challenge for gross and histopathological examination. Histopathologically the second group showed epidermal hyperkeratosis with appearance of crust lesions seen with abscess formation especially in early stage of lesion, while the main feature of advance cases were characterized by folliculitis with fungal hyphae invasion in all epidermal and dermal layers together with eosinophilic infiltration. Mild pathological changes were seen in the 1st immunized group characterized by infiltration of mononuclear cells mainly macrophages in dermal connective tissue with dense proliferation of collagenous fibers , appearance of young fibroblasts together with cellular hypodermal infiltration of eosinophil no evidence of clear follicular lesions were seen. Lyophilized antigen of T.mentagrophyte can be considered as an effective immunogen for protecting mice against T.mentagrophyte infection and it is synchronized with its dose.

The aim of the study was to make knowledge on the histopathological changes in some internal organs ( liver, kidney and spleen) of albino mice after treatment with therapeutic dose(16mg/kg BW/day) of pentoxifylline (PTX). Thirty albino mice which are approximately at same age (8week) and body weight were, randomly divided into three equal groups, group 1:Received tape water along the period of experiment and considered as a control group, Group 2:Treated with Pentoxifylline ( 16 mg /kgBW/day)for 30 days Group 3:Treated with Pentoxifylline ( 16 mg/kgBW/days)for 60 day. The histopathological findings of liver, kidney and spleen, showed infiltration of mononuclear cells within the liver parenchyma and portal areas and in the interstitial tissue of the kidney with perivascular lymphocytic cuffing and mild degenerative changes represented by acute cellular swelling of hepatocytes and epithelial cells lining the cortical renal tubules in addition to congestion of blood vessels Spleen showed lymphoid hyperplasia of white pulp with congestion and infiltration of lymphocytes in red pulp.

<p>Penelitian ini bertujuan untuk melihat pengaruh pemberian ekstrak etanol kulit petai (Parkia speciosa Hassk) terhadap penurunan kadar glukosa darah mencit jantan yang diinduksi aloksan. Hewan percobaan dibagi atas 5 kelompok diantaranya kelompok kontrol negatif, kelompok kontrol positif,dosis I (280 mg/kgBB mencit), dosis II (560 mg/kg BB mencit), dosis III (840 mg/kg BB mencit). Penelitian dilakukan selama 21 hari. Persentase penurunan kadar glukosa darah mencit jantan setelah diberikan ekstrak etanol kulit petai pada hari ke-21 adalah dosis I (77,52 %) lebih besar dibandingkan dengan dosis II (69,5 %) dan dosis III (73,37 %). Data yang diperoleh dianalisis dengan uji Two Way Anova dengan program SPSS 17. Hasil penelitian ini menunjukkan bahwa pemberian ekstrak etanol kulit petai untuk tiga variasi dosis menyatakan perbedaan yang bermakna secara statistik terhadap penurunan kadar glukosa darah mencit jantan.</p><p><em>Petai (Parkia speciosa Hassk) has a compound β-sitosterol and stigmasterol that have efficacy to decreased blood glucose levels. This study aimed to determine the effect of ethanol extract of petai peel for decrease blood glucose levels of male mice induced by alloxan. Experimental animals were divided into 5 groups including negative control group, positive control group, the first dose (280 mg/kg in mice), the second dose (560 mg/kg in mice), the third dose (840 mg/kg in mice). The study was conducted for 21 days. After 21 days, the result found that the percentage of blood glucose levels after the male mice given the ethanol extract of petai peel was, the first dose (77.52%) biger than the second dose (69.5%) and the third dose (73.37%). The data obtained were analyzed by Two Way ANOVA using SPSS 17. The results showed that have signicantly difference between three dose variation of ethanol extract of petai peel in blood glucose levels.</em></p>

(1) The aim of the present study was to describe the endoscopic and histopathological findings in the esophagus, stomach, and duodenum in patients with Crohn’s disease. (2) Methods: This was a cross-sectional study that included patients receiving treatment from the inflammatory bowel disease outpatient clinic. Esophagogastroduodenoscopies with biopsies of the stomach and proximal duodenum were performed. Presence of Helicobacter pylori bacteria was assessed by Giemsa staining. (3) Results: We included 58 patients. Erosive esophagitis was identified in 25 patients (43.1%), gastritis was diagnosed in 32 patients (55.2%) and erosive duodenitis was found in eight (13.8%). The most frequent histopathological finding in the H. pylori-positive group was increased inflammatory activity in the gastric body and antrum, with a predominance of mononuclear and polymorphonuclear cells. In turn, the most frequent finding in the H. pylori-negative group was chronic inflammation with predominance of mononuclear cells. Focally enhanced gastritis was identified in four patients (6.9%), all of whom were negative for H. pylori. Granulomas were not observed. H. pylori infection was present in 19 patients (32.8%). (4) Conclusions: Nonspecific endoscopic and histological findings were frequent in patients with Crohn’s disease. Focally enhanced gastritis was uncommon and observed only in H. pylori-negative patients. The time from the diagnosis, patient age, and therapy in use may have influenced the nondetection of epithelioid granuloma.

Purpose: By taking advantage of 18F-FDG PET imaging and tissue nuclear magnetic resonance (NMR) metabolomics, we examined the dynamic metabolic alterations induced by liver irradiation in a mouse model for hepatocellular carcinoma (HCC). Methods: After orthotopic implantation with the mouse liver cancer BNL cells in the right hepatic lobe, animals were divided into two experimental groups. The first received irradiation (RT) at 15 Gy, while the second (no-RT) did not. Intergroup comparisons over time were performed, in terms of 18F-FDG PET findings, NMR metabolomics results, and the expression of genes involved in inflammation and glucose metabolism. Results: As of day one post-irradiation, mice in the RT group showed an increased 18F-FDG uptake in the right liver parenchyma compared with the no-RT group. However, the difference reached statistical significance only on the third post-irradiation day. NMR metabolomics revealed that glucose concentrations peaked on day one post-irradiation both, in the right and left lobes—the latter reflecting a bystander effect. Increased pyruvate and glutamate levels were also evident in the right liver on the third post-irradiation day. The expression levels of the glucose-6-phosphatase (G6PC) and fructose-1, 6-bisphosphatase 1 (FBP1) genes were down-regulated on the first and third post-irradiation days, respectively. Therefore, liver irradiation was associated with a metabolic shift from an impaired gluconeogenesis to an enhanced glycolysis from the first to the third post-irradiation day. Conclusion: Radiation-induced metabolic alterations in the liver parenchyma occur as early as the first post-irradiation day and show dynamic changes over time.

Цель - выявление реактивных изменений гистологических элементов печени и почек у мышей в экспериментальных моделях сепсиса, вызванного штаммами 1840 и 1623 Pseudomonas aeruginosa (PsA1840, 1623). Материал и методы. Сепсис моделировали на двух группах половозрелых самцов мышей линии C57Bl/6 интраперитонеальным введением Pseudomonas aeruginosa. Животным 1-й группы (8 особей) вводили штамм 1840, животным 2-й группы (12 особей) - штамм 1623. Контрольная группа состояла из 3 животных. Перед началом опыта штаммы тестировали на присутствие генов экзотоксинов U, S, T, Y (ExoU, ExoS, ExoT, ExoY) с помощью полимеразной цепной реакции (ПЦР). Для визуализации продуктов ПЦР применяли электрофорез в горизонтальном 1,5 % агарозном геле. Животных вскрывали на терминальной стадии сепсиса. Серийные парафиновые срезы печени и почек толщиной 4 мкм окрашивали гематоксилином - эозином, анализировали особенности гистоструктуры органов и фотографировали в световом микроскопе «AxioPlus» (фирма Zeiss, Германия). Результаты. Штамм PsA 1840, имеющий ген exoU, вызывал выраженные деструктивные изменения пластинок гепатоцитов и замещение участков паренхимы печени гомогенным эозинофильным веществом. Присутствовали признаки стаза крови в синусоидных капиллярах, расширение и тромбоз центральных вен, немногочисленные скопления лейкоцитов. Морфологические изменения нефронов состояли в локальных деструктивных изменениях проксимальных канальцев на периферии коркового вещества почек. При введении PsA1623, имеющего ген exoS, возникали массовая гибель почечных телец и дегенерация канальцев нефронов. В печени дольковая гистоархитектура в основном сохранялась. Выводы. Предполагается связь выявленных различий в реактивных изменениях гистологических элементов печени и почек в подопытных группах с особенностями геномов штаммов PsA, использованных для моделирования сепсиса. Objective - to identify reactive changes of liver and kidney histological elements in experimental models of sepsis in mice caused by 1840 and 1623 Pseudomonas aeruginosa strains (PsA1840, 1623). Material and methods. Sepsis was modeled in two groups of mature male C57Bl/6 mice by intraperitoneal administration of Pseudomonas aeruginosa. Strain 1840 was administered to animals of the first group (n=8), animals of the second group (n=12) were administered strain 1623; the control group consisted of 3 animals. Before the experiment, the strains were tested for the presence of genes of exotoxins U, S, T, Y (ExoU, ExoS, ExoT, ExoY) using polymerase chain reaction (PCR). Electrophoresis in horizontal 1,5 % agarose gel was used to visualize PCR products. The animals were euthanized at the terminal stage of sepsis. The extracted liver and kidneys were fixed according to the generally accepted histological method, and embedded into paraffin blocks. Serial 4 μm thick sections of organs were stained with hematoxylin and eosin, analyzed and photographed using «AxioPlus» light microscope (Zeiss, Germany). Results. Strain PsA 1840, carrying the gene of exotoxin U (ExoU), caused severe destructive changes of hepatocytes plates and the replacement of the liver parenchyma with homogeneous eosinophilic substance. There were signs of blood stasis in sinusoidal capillaries, expansion and thrombosis of central veins, a few accumulations of leukocytes. Morphological changes of nephrons consisted of local destructive changes in the proximal tubules at the periphery of kidney cortical substance. After the introduction of PsA1623, carrying the gene of exotoxin S (ExoS), the massive death of renal corpuscles and degeneration of nephron tubules were registered. However, the lobular histoarchitecture in the liver remained mostly unaltered. Conclusions. It is supposed that there is a possible connection of the observed differences in reactive changes of liver and kidney histological elements in two experimental groups with genome features of PsA strains used for the sepsis modeling.

Abstract We have studied the coagulation status of eight patients with the Chediak-Higashi syndrome (CHS), both in the chronic and the accelerated phase of the disease. It has been shown that during the accelerated phase there are coagulation abnormalities. These abnormalities include a peripheral thrombocytopenia, minor alterations of liver clotting factors, and mainly a profound hypofibrinogenemia and hypoplasminogenemia, which cause life-threatening bleedings. These disorders are of complex origin, but a fibrinolytic process, possibly primary, appears to play a significant role, since the present evidence for intravascular coagulation is not definitive. The accelerated phase of the CHS is characterized by a visceral infiltration by macrophages and lymphocytes. Therefore, we have investigated the possible role of the macrophages in the fibrinolytic process. We have found an excessive plasminogen activator (PA) production by CHS mononuclear cells in the accelerated phase and to a lesser extent in the chronic phase, except in one patient in whom no anomaly was found. Single-cell studies revealed an increased number of PA-producing cells among the monocyte- macrophage lineage rather than a higher level of production per cell. Polymorphonuclear cells (PMN) from patients with CHS were also shown to contain more PA. Slight but significant abnormalities in PA production were observed in obligatory heterozygotes (five out of nine), indicating the inherited nature of the excessive PA production. Finally, an enhanced PA production was similarly demonstrated using beige mice macrophages. The exacerbated production of PA by macrophages in the accelerated phase of the CHS can account to some extent for the coagulation abnormalities that have been observed.
