This research was conducted by conducting experiments in UPTD laboratory of Livestock and Livestock Feeding Agricultural Service of Sulawesi Barat Province. The purpose of this research is to compare the age of males during March to April 2013. The results of this research are expected to be used as a guideline for the use of Bali Cattle research as a semen producer based on the age of livestock that can be used as one of the criteria in the selection of prospective semen producing males. The research material is fresh semen of Bali Cattle with requirement of 70% individual motility. The research method used is observation method by observing the fresh semen quality of Bali Cattle research which has been classified into 3 groups based on age 3, 5, and 6 years. The data analysis used is the analysis of the variety and the design is Completely Randomized Design. The results showed that the quality of fresh semen in the three age groups (3, 5, and 6 years) where volume: 5.2 ± 1.2; 6.4 ± 1.1; And 6.4 ± 0.8 ml, color: milk, white, pH: 6,4 ± 0,1; 6.4 ± 0.1; And 6.3 ± 0.1, consistency: concentrated; Medium; Concentrated and concentrated, concentrations: 1909.6 ± 418.8; 1223.3 ± 220.8; And 1790.8 ± 240.5 million / ml, and 92.5 ± 2.9%, abnormality: 4.4 ± 1.2; 4.3 ± 1.2; And 3.3 ± 1.0%. The conclusions of this research indicate that the age of males gives a significant effect on the fresh semen volume, fresh semen concentration, gives a very significant effect on fresh semen concentration and does not have a significant effect on fresh semen sperm abnormalities. And a male of Bali Cattle age 3-6 years old in UPTD Livestock Feeding and Animal Feed Agricultural Service of Sulawesi Province has good semen quality for Artificial Insemination.

This study was conducted to find the rate of body weight ideal study Simental cows to produce production and the best quality fresh semen. The material used in this study were 9 males Simental cows were divided into 3 ranges of body weight as a treatment that is P1 = low body weight (822-878 kg), P2 = moderate weight (910-958 kg) and P3 = body weight high  (983-1041 kg). The number of cows in each group of body weight is considered as replicates. The method used was a survey method. The research is designed to completely randomized design (CRD) with 3 treatments and weight range 3 replications. Each stud cows housed cement 2 times per week for 12 weeks so that each cow cement accommodated as many as 24 times. Parameters observed in this study is that the volume of fresh cement production of cement per shelter and semen quality bulls include sperm motility and concentration of spermatozoa. Data were analyzed using analysis of variance, followed by Duncan test if there are significant levels of different body weights. The results showed that differences in body weight bulls simental not significant (P>0.05) for fresh semen volume, sperm motility and concentration of spermatozoa. Mean semen volume is best found in the high body weight group, whereas sperm motility and concentration of spermatozoa present in body weight groups were.

This study was conducted to examine the comparison the quality of spermatozoa in four genetic groups of poultry (Pelung chicken, Nunukan chicken, Sentul chicken, and Bangkok chicken). The parameters of this study were the concentration of spermatozoa, motility, viability, semen volume, color, and consistency. The study was designed using a Completely Randomized Design (CRD) with four local chicken nations and four replications. The results showed that the volume of Nunukan chicken semen was 0.32±0.01 mL/ejaculation, Bangkok chicken was 0.31±0.01 mL/ejaculate, Sentul chicken was 0.15±0.02 mL/ejaculate and Pelung chicken was 0.23±0.02 mL/ejaculation. the color of fresh semen is white (Pelung chicken, Nunukan chicken and Sentul chicken) except chicken Bangkok has the color of cream semen. Pelung chicken has a concentration of spermatozoa 5,043.33±51 million/mL, Nunukan chicken 3,250.22 ±45 million/mL, Sentul chicken 3,002.87±67 million/mL, and Bangkok chicken 3,002.87 ± 67 million/mL. Motility of Pelung chicken 84.,69 ± 1.12%, Bangkok chicken 82.35±1.85%, Nunukan chicken 77.74±1.57% and Sentul chicken 77.64±1.65%. Viability spermatozoa of Sentul chicken was 90.35 ± 1.21%, Bangkok chicken was 90.64 ± 1.16%, Pelung chicken was 89.17 ± 1.23% and Nunukan chicken was 86.29 ± 1.15%. It can be concluded that the motility spermatozoa of Pelung chicken and Bangkok chicken is better than the spermatozoa motility of Nunukan chicken and Sentul chicken. The viability of Sentul chicken and Bangkok chicken was higher compared to the viability semen of chicken Pelung and chicken Nunukan.

The purpose of this study was to observe the macroscopic and microscopic quality of Bronze turkey fresh semen on various ages. The observed toms were at the ages of 9 to 10 months (P1), 13 to 14 months (P2), and 17 to 18 months (P3), with each taken from 3 different toms. The collection was done by abdominal massage, and collected for 3 times a week in triplicate. This data was analyzed by analysis of variance (ANOVA) in a completely randomized design, followed by Duncan's New Multiple Range Test (DMRT) if there was any difference. The results showed that the average volume of turkey fresh semen were P1 (0.16 ± 0.04 ml), P2 (0.13 ± 0.02 ml), and P3 (0.10 ± 0.02 ml). The average pH of fresh semen were P1 (7.19 ± 0.05), P2 (7.12 ± 0.10), and P3 (7.06 ± 0.06). The average motility percentage of fresh semen were P1 (78.11 ± 1.38%), P2 (82.55 ± 1.17%), and P3 (74.55 ± 4.16%). The average viability percentage of fresh semen were P1 (78.63 ± 1.55%), P2 (83.26 ± 1.54%), and P3 (79.20 ± 1.06%). The average abnormal percentage of spermatozoa were P1 (3.01 ± 0.04%), P2 (3.04 ± 0.88%), and P3 (3.21 ± 0.44%). In conclusion, the best fresh semen quality is found on toms aged from 13 to14 months (P2). Furthermore, at the age 17 to18 months and more, the semen quality was decreased.

This study aimed to find out the quality of fresh semen from Sumba Ongole (SO) bulls which was diluted with Bestvile Thawing Solution (BTS) diluent modified with soybean milk (SKD). The study used a completely randomized design consisting of four treatments and 5 replications, P0 (100 % BTS), P1 (95 % BTS + 5 % SM), P2 (90 % BTS + 10 % SM), and P3 (85 % BTS + 15 % SM). The parameters observed were the colour, smell, consistency, pH, volume, motility, viability, and abnormalities of spermatozoa. The results showed that the characteristics were in the normal, creamy white semen colour, semen volume 3,6 ml, medium consistency, pH 6,3, typical SO bulls smell, sperm motility 85 %, mass activity +++, sperm concentration 1276 million/ml, live sperm 90,05 % and sperm abnormalities 6,15 % and quality there was no significant difference between the four treatments. The spermatozoa motility and viability in each treatment was able to which achieved on day 1. The characteristics of the spermatozoa of SO bulls were in the normal category. Meanwhile, BTS diluent with the addition of SKD was unable to maintain sperm quality.

ABSTRAK                       Tujuan penelitian ini adalah untuk mengetahui kualitas spermatozoa semen hasil sexing kambing Peranakan Etawa (PE) dengan metode sedimentasi putih telur menggunakan pengencer yang berbeda. Materi yang digunakan dalam penelitian ini adalah semen segar kambing PE.  Metode percobaan yang digunakan adalah percobaan laboratorium dengan menggunakan rancangan acak lengkap (RAL) Setiap perlakuan semen sexing kambing Peranakan Etawa dengan pengencer yang berbeda yaitu (P0) CEP, (P1) CEP+kuning telur+putih telur, (P2)CEP+putih telur (P3), CEP+kuning telur dan diulang sebanyak 10 kali. Variabel yang diukur pada penelitian ini adalah motilitas, viabilitas, dan abnormalitas spermatozoa. Data yang diperoleh dianalisis dengan mengunakan analisis varian. Apabila perlakuan memberikan pengaruh maka dilanjutkan dengan uji Beda Nyata Terkecil (BNT). Hasil penelitian menunjukkan bahwa kualitas spermatozoa kambing Peranakan Etawa (PE) dengan berbagai macam pengencer menunjukkan pengaruh yang sangat nyata (P<0,01). Motilitas dan Viabilitas menunjukkan perbedaan yang sangat nyata (P<0,01) pada berbagai pengencer Motilitas terbaik pada beberapa lapisan atas sebesar 65% dan CEP+PT lapisan atas 60% dilanjutkan oleh CEP+KT+PT lapisan bawah 55,9% dan CEP+PT lapisan bawah 55% dilanjutkan lagi oleh CEP+KT lapisan atas 50% dan  lapisan bawah 45% dan yang paling terkecil adalah CEP lapisan bawah 40%. Viabilitas spermatozoa dari yang tertinggi yaitu CEP+KT+PT lapisan atas sebesar 69,553% dan CEP+PT lapisan atas sebesar 69,519% dan dilanjutkan dengan CEP+PT lapisan bawah sebesar 65,504% dan CEP+KT+PT lapisan bawah 65,473%, dan dilanjutkan lagi dengan CEP+KT lapisan atas sebesar 60,269% dan lapisan bawah sebesar 53,476% dan yang paling terkecil yaitu CEP lapisan bawah sebesar 40,371%. Presentase Abnormalitas menunjukkan bahwa perlakuan tidak memberikan pengaruh nyata (P>0,05). Tetapi CEP+KT+PT lapisan atas menunjukkan persentase yang paling rendah yaitu sebesar 4,88% dibandingkan perlakuan lainnya. Dari hasil penelitian dapat disimpulkan bahwa kualitas spermatozoa semen sexing kambing PE dengan menggunakan pengencer CEP+ kuning telur + putih telur pada lapisan atas memberikan hasil terbaik terhadap kualitas spermatozoa ditinjau dari motilitas, viabilitas dan abnormalitas.   ABSTRACT                      The purpose of this study was to determine the sexing sperm quality of Etawa cross-bred goat (PE) with egg white sedimentation method using different diluents. The material used in this study was the fresh semen Etawa crossbreed goat (PE) of the center for Artificial Insemination (BBIB) Singosari Malang. The method is laboratory research using completely randomized design. Consist of CEP, CEP + KT, CEP + PT, CEP + KT + PT and repeated 10 times. The variables are motility, viability and abnormality of sperm. Data were analyzed by using variance analysis. If the treatment effect then continued by Least Significant Difference (LSD). The results showed that the quality of Etawa cross-bred goat sperm with various diluents showed a significant influence (P <0.01). Motility and viability showed differences (P <0.01) in various diluents. The best motility and viability on top layer of CEP+KT+PT diluent were 65% and 69%, 55%. Percentage abnormalities showed that the treatment was not significant effect (P> 0.05). but top layer CEP+KT+PT diluent shows the percentage of abnormality west 4.88% compared to the other treatments. From the results of this study concluded that the quality of sexing semen quality by using dilution CEP + yolk white egg on the top layer gives the best results on the quality of sperm in terms of motility, viability and abnormalities. Based on this study it is suggested that the use of sexing semen with egg white sedimentation method using diluent CEP + yolk + white egg.

Semen was collected from four rams, using artificial vagina and viability%, motility% and plasma membrane integrity% were measured. Fresh ejaculates (n = 32) were separated by modified swim-up separation using modified human tubal fluid medium. Four fractions of supernatant were collected at 15-minute intervals. The mean volume, mass activity, concentration, motility%, viability%, normal morphology and membrane integrity% (HOST +ve) of fresh semen were 1.0 ± 0.14, 4.1 ± 0.1 × 109 spermatozoa/ml, 85.0 ± 1.3, 89.4 ± 1.0, 85.5 ± 0.7, 84.7 ± 0.5 respectively. There was no significant (P>0.05) difference in fresh semen quality parameters between rams. The motility%, viability% and HOST +ve % of first, second, third and fourth fractions were 53.4 ± 0.5, 68.2 ± 0.3, 74.8 ± 0.3 and 65.5 ± 0.4; 55.5 ± 0.4, 66.2 ± 0.4, 74.5 ± 0.3 and 73.6 ± 0.3 and 66.7 ± 0.5, 66.8 ± 0.5, 65.2 ± 0.4 and 74.7 ± 0.5 respectively. The motility%, viability% and membrane integrity% of separated semen samples differed significantly (P<0.05) between four fractions. The mean motility% and viability% were significantly higher (P<0.05) in third fraction (74.8 ± 0.3%), whereas the mean HOST +ve% was significantly higher (P<0.05) in fourth fraction (74.7 ± 0.5). All quality parameters of separated spermatozoa were significantly (P<0.05) lower than that of fresh semen. Objectives: We explored the safety of Tadalafil once-daily treatment for 12 week and its effects on semen quality in a clinical subpopulation of men with psychogenic erectile dysfunction (ED). Patients and Methods: Twenty-seven men, between 19 and 35 years, unaware of their fertility status, suffering from psychogenic ED were enrolled. The examination of the seminal fluid was performed twice before administration of Tadalafil and twice after three months of continuous daily administration of Tadalafil 5 mg. The volume of the seminal fluid, the concentration of sperm, the concentration of sperm with type “a+b” motility, the concentration of sperm with type “a” motility, the concentration of normal sperm were taken into consideration. The results before treatment with Tadalafil (T0) and after 3 months of treatment (T3m) were compared. Results: The administration of once-daily Tadalafil 5mg, brings to an average increase of the total number of sperm cells, both total and fast motility (type a) and the percentage of nemasperms, and to an average increase of semen volume of only 0.41 ml. These quantitative and qualitative improvements of the seminal fluid resulted statistically significant as regard motility, nemasperm percentage and seminal fluid volume. No unespected safety findings were observed. Conclusions: Tadalafil administration improves the quality of sperm cells and seminal fluid: in particular motility, percentage of nemasperms and volume of seminal fluid. We emphasize the safety of the once-daily treatment with tadalafil 5mg and the positive effects on spermatogenesis.

The cryopreservation process might reduce the quality of spermatozoa due to an increase in the production of reactive oxygen species (ROS) compounds during cooling, freezing, and thawing. The quality of spermatozoa can be maintained by adding glutathione as an exogenous antioxidant into cryo-diluent. This study aimed to examine the effect of the addition of different doses of glutathione in cryopreservation of Aceh Local catfish (Clarias gariepinus) spermatozoa after freezing. The method used was a completely randomized design (CRD) with four treatments and four replications. Fresh semen was diluted in Ringer, 15% DMSO, and 20% Fetal Bovine Serum (FBS) and then added with glutathione antioxidants of 0.0 mgL-1, 0.5 mgL-1, 1.0 mgL-1, and 2.0 mgL-1. The parameters observed in this study were motility, integrity of the plasma membrane, fertility, and DNA integrity. The results showed that the concentration of glutathione had no effect on motility, integrity ofthe plasma membrane, or fertility, but had an effect on DNA integrity. The highest percentage of motility and plasma membrane integrity respectively was 40.50% (P3) and 70.87% (P2). Furthermore, the assessment of DNA integrity showed that there was no DNA fragmentation both treatments and fresh spermatozoa. This research is the first study regarding glutathione supplementation in cryo-diluent of Aceh Local catfish spermatozoa. Finally, the results obtained can provide information about the exact concentration of glutathione in the extender on the quality of spermatozoa of Aceh Local catfish (C. gariepinus) after the cryopreservation process. These results can also increase the success of fertility be used by the seed hall unit and the aquaculture industry to increase the productivity and supply high quality seeds.

Animal feed should ideally be inexpensive and not competing with humans. Ampas know is a waste of the manufacture of tofu that contains protein and high energy. Its use has problems due to the existence of antinutrient and cannot be kept long. Lactobacillus Plantarum is a lactic acid (Gram-positive) bacteria used in the fermentation process to improve the quality of feed. The goal of fermentation is to remove the anti-nutrients that exist in the tofu pulp and increase the content of Isoflavone (Daidzin, Glycitein, and Genistein) and Isoflavone is a source of antioxidants important to growth. The study uses a Completely Randomized Design with 5 treatment of microbial culture concentrations of 1%, 5%, 10%, 15%, and 20% (V/w), respectively repeated 3 times. The result of analyzes in the pulp of tofu fermentation suggests that fermentation by Lactobacillus Plantarum can increase the content of Daidzein, Glycitein, and genistein with the highest value resulting from the use of L. Plantarum 20% respectively 191.21 + 24.9 μg/g, 85.96 μg/g, and 120.82 μg/g. It can be concluded that the higher the use of the concentration of L. Plantarum in tofu pulp can increase the levels of isoflavone, so it is advisable to conduct research with the concentration of L. Plantarum higher in tofu pulp

Stallions affected by seminal vesiculitis present history of infertility or subfertility, ejaculatory disturbance, spread of sexually transmitted pathogens, and changes in semen characteristics, leading to reduced semen quality and longevity. The aim of this study was to evaluate the semen quality of stallions with seminal vesiculitis before and after local treatment. Five stallions with a mean age of 12.4 years diagnosed with seminal vesiculitis were used. The identification of the microorganism involved in the pathogenesis of seminal vesiculitis of each animal was performed by bacterial culture of the seminal vesicles flush with Ringer Lactate solution, performed in duplicate at 1-week intervals. After identification of bacteria was performed, there was susceptibility testing to antibiotic (antibiogram) and the appropriate antibiotic was chosen. The local treatment was performed by endoscopy for 10 consecutive days, and this consisted of flushing with Ringer Lactate solution, followed by infusion of the antibiotic selected. The semen analyses were performed before starting the local treatment for seminal vesiculitis (M0), after a week (M1), and after a month (M2) of therapy. Sperm kinetics were performed by computerized method – CASA for the following parameters: percentage of sperm with total motility, progressive motility, and rapid sperm. Analysis of plasma membrane integrity was performed by epi-fluorescence microscopy, using the combination of fluorescent probes carboxyfluorescein diacetate and propidium iodide. Percentage of leukocytes was assessed through evaluation in light optical microscopy of semen smears stained with DiffQuick. The content of nitric oxide (NO) was determined by colourimetric Griess reaction by a spectrophotometer through the concentrations of nitrate (NO3–) and nitrite (NO2–). To perform the count of colony forming units per millilitre (CFU mL–1), an aliquot of 0.1 mL of semen was diluted in 9.9 mL of saline. A 0.1-mL aliquot of this sample was plated on Mueller-Hinton agar. The seeded plates were incubated, and the bacterial colonies were counted after 24 h. According to the performed dilution, total colonies identified corresponds to ×10 000 CFU mL–1. The data were analysed by two-way ANOVA followed by Tukey's test (P < 0.05). The values (mean ± standard error) of seminal parameters on M0, M1, and M2 were the following, respectively: sperm kinetics (total motility: 46.5 ± 5.13a; 75.1 ± 3.42b; 42.8 ± 5.28a; progressive motility: 19.3 ± 3.86a; 33.4 ± 2.39b; 16.5 ± 2.40a; rapid sperm: 22.2 ± 1.82a; 52.2 ± 5.65b; 22.1 ± 2.62a); plasma membrane integrity (47.5 ± 4.65a; 62.9 ± 5.41b; 39.1 ± 4.32a); percentage of leukocytes (35.2 ± 2.36a; 15.1 ± 2.55b; 36.1 ± 4.04a); CFU (119 980 × 103 ± 19 528.0 × 103a; 5375 × 103 ± 2453.7 × 103b; 65 850 × 103 ± 19 701.0 × 103ab) on fresh semen; and NO content (0.645 ± 0.172a, 0.117 ± 0.023b, 0.364 ± 0.110ab) on seminal plasma. The results demonstrate that local treatment after a week leads to an improvement in sperm quality; however, this was not maintained after 1 month of therapy, since the seminal parameters at this time are similar to pretreatment, which can be justified by recurrent disease.
