scholarly journals EFFECTS OF SILVER NANOPARTICLES ON PENTYLENETETRAZOL-INDUCED KINDLED SEIZURES IN RATS

Silver nanoparticles are used for delivering neurotropic agents to brain, increasing the bioavailability of insoluble drugs as well as for brain tumours treatment. Their effects upon brain excitability, especially under conditions of chronic epilepsy modelling are underinvestigated. The purpose of the study was to study the effects of silver nanoparticles upon pentylenetetrazol-induced kindled seizures in rats. Material and Methods. Experiments were performed on Wistar rats kindled during four weeks with pentylenetetrazol administrations in a dose of 30.0 mg/kg, i.p. Only rats, which demonstrated generalized clonic-tonic seizures in a response to each of three last epileptogen injections, were included into the study. Nanoparticles and ionized argentum were administered in a dosage of 0.2 mg/kg, i.p. Observation was performed in early and postponed period of kindling – in 24 h and three weeks from the moment of kindling induction, correspondently. Results and discussion. The calculation of the pentylenetetrazol dose, which effectively induced seizures in 50 % of kindled rats (ED50), was performed in kindled animals in 24 h from the moment of last epileptogen administration was 22.0 mg/kg. In postponed period of kindling (three weeks from the moment of last pentylenetetrazol administration in a dose of 30.0 mg/kg, i.p.), ED50 of pentylenetetrazol was 16.0 mg/kg. ED50 of pentylenetetrazol, which induced clonic seizures in 50 % animals at the early stage of kindling, was recalculated following the administration of ionized and nanoparticle forms of silver, and were less when compared with such ones determined in the control group by 13.5 % (P>0,05) and 26.0 % (P<0,05), respectively. In the postponed period of kindling ED50 of ionized and nanoparticle forms of silver were less when compared with control by 20.0 % (P<0.05) and by 42.0 %, respectively (P<0.05). Significant difference was noted between groups treated with ionized and nanoparticle silver (P<0.05). The net intensification of pentylenetetrazol seizure manifestations was seen in kindled rats treated with silver nanoparticles, when seizure discharges amplitude achieved 1.0-1.2 mV and frequency of 3/sec, which exceeded the ones in rats treated with ionized silver. Conclusion. Silver nanoparticles induce the increase in seizures severity in pentylenetetrazol-kindled rats, and this effect was more pronounced at the postponed stage of kindling development.

Poly methyl methacrylate (PMMA) is one of the most widely used materials in modern prosthodontics. It is widely known due to its simplicity in use and acceptable aesthetic. A new concept of polymerization fluid resin technique was instead of heat and gypsum material. Forty specimens were prepared from two brands of fluid acrylic resin. The samples were divided into two groups, the first one was the control while the other group was incorporated with silver nanoparticles(modified). The tests performed were impact strength, transverse strength, color stability and candida retention ability on the samples. 10 samples for each test were used. The results showed that modified group had significantly higher impact strength than the control group. There was non-significant difference between group of modified fluid acrylic and control group. Regarding color stability, there were highly significant color change after 10 days of immersion in the tea solution for the modified group the results of the biological test showed that the candida retention of the control group was significantly higher than modified group. Within the limitation of this study it can be concluded that addition of silver nanoparticles has resulted in significant difference between control and modified group for impact strength test. While non-significant difference was seen for transverse strength test. In regard to color stability, showed enhancement in color stability for both before and after placement in tea solution. Addition of silver nanoparticles also caused reduction in candida albicans retention in the added samples.

Insertional Achilles tendinopathy (IAT) is caused by traction force of the tendon. The effectiveness of the suture bridge technique in correcting it is unknown. We examined the moment arm in patients with IAT before and after surgery using the suture bridge technique, in comparison to that of healthy individuals. We hypothesized that the suture bridge method influences the moment arm length. An IAT group comprising 10 feet belonging to 8 patients requiring surgical treatment for IAT were followed up postoperatively and compared with a control group comprising 15 feet of 15 healthy individuals with no ankle complaints or history of trauma or surgery. The ratio of the moment arm (MA) length/foot length was found to be statistically significant between the control group, the IAT group preoperatively and the IAT group postoperatively (p < 0.01). Despite no significant difference in the force between the control and preoperative IAT groups, a significantly higher force to the Achilles tendon was observed in the IAT group postoperatively compared to the other groups (p < 0.05). This study demonstrates that a long moment arm may be one of the causes of IAT, and the suture bridge technique may reduce the Achilles tendon moment arm.

The regeneration of bone defects caused by periodontal disease or trauma is an important goal. Porous hydroxyapatite (HA) is an osteoconductive graft material. However, the hydrophobic properties of HA can be a disadvantage in the initial healing process. HA can be coated with TiO2 to improve its hydrophilicity, and ultraviolet irradiation (UV) can further increase the hydrophilicity by photofunctionalization. This study was designed to evaluate the effect of 5% TiO2-coated HA on rabbit calvarial defects and compare it with that of photofunctionalization on new bone in the early stage. The following four study groups were established, negative control, HA, TiO2-coated HA, and TiO2-coated HA with UV. The animals were sacrificed and the defects were assessed by radiography as well as histologic and histomorphometric analyses. At 2 and 8 weeks postoperatively, the TiO2-coated HA with UV group and TiO2-coated HA group showed significantly higher percentages of new bone than the control group (p < 0.05). UV irradiation increased the extent of new bone formation, and there was a significant difference between the TiO2-coated HA group and TiO2-coated HA with UV group. The combination of TiO2/HA and UV irradiation in bone regeneration appears to induce a favorable response.

BACKGROUND: Chronic kidney disease (CKD) is a chronic illness with complex disease which could lead to other underlying diseases such as diabetes mellitus (DM), hypertension, and dyslipidemia. Urban population must manage their illness due to their occupation. Coaching support is an advanced method to help individuals manage their illnesses, especially chronic illness. Symptoms and complaints in early-stage renal disorders tend to be mild, making it difficult to diagnose only by clinical examination. Impaired kidney function can lead to progressive kidney damage. AIM: This study aimed was to analyze the effect of coaching support in maintaining kidney function in patients with CKD. METHODS: This research used quasi-experiment with pre-test and post-test with control group design. Respondents in this study were 40 CKD patients which were taken by consecutive sampling technique and divided into two groups, namely, control group and treatment group. Data were collected using blood urea nitrogen and creatinine values observation sheet. Coaching support was divided into four steps of therapy, identify the disturbance, identify based on experience, use a family support system, and evaluating the results. Data were analyzed using paired t-test and independent t-test with a significance of p < 0.05. RESULTS: This study found that there was a significant difference in kidney function between the control group and the treatment group (p = 0.000). Coaching support interventions were effective on kidney function in patients with CKD. The implementation of coaching support went well because respondents and families were proactive. CONCLUSION: Coaching support should be applied by nurses as daily activity management of CKD patients at early stage to inhibit the kidney function damage progression.

A total of 144, day old as hatched Cobb-500 broilers were fed adlibitum basis on a diet with 0%, 0.5%, 1.0% and 1.5% of Black Cumin Seed Meal up to 35 days of age to compare the effect of BCSM as herbal growth promoters on growth performance, meat yield characteristics and profitability of broilers. The experimental diets consisted of broiler starter and broiler grower, which were fed from 0-16 and 17-35 days of age, respectively. Diets supplemented with 1.5% BCSM showed significant effect on live weight at 21, 28 and 35 days of old in broilers in various groups as compared to that of control group. Better feed conversion (2.02) also noticed at 1.5% level of BCSM at early stage (14 days of old) of broilers. There was no significant difference (P>0.05) among the average live weight, dressing yield, thigh meat, wing meat, heart, gizzard liver, at 0%, 0.5%, 1.0% and 1.5% level of BCSM diet. But difference found within breast meat, drumstick meat, abdominal fat and skin of broilers at 1.5% level of BCSM diet. It may be concluded that supplementation of BCSM at 1.5% had the highest significant effect (P<0.01) on feed cost (Tk kg-1 b), production cost (Tk kg-1 b) and profit (Tk kg-1 b) and lower mortality. So that 1.5% BCSM in diets can be used for economic and efficient broiler production than 0.5% and 1.0 % level.SAARC J. Agri., 13(2): 151-160 (2015)

Abstract BackgroundNAFLD is a common metabolic disorders disease which influenced 20~30% people. NAFLD can progress to cirrhosis, liver fibrosis and even liver cancer. Liver puncture is the gold standard. However, due to its trauma and possible complications, its clinical use is currently limited. Therefore, it is of great clinical significance and value to find a noninvasive biochemical index that can diagnose NAFLD early.ObjectiveOur aim was to identify the potential biomarkers in NAFLD people in early stage via untargeted metabolomics study.MethodsIn our research, From January to October 2019, 224 patients aged 18-55 were selected from the outpatient department and ward of gastroenterology department of Putuo Hospital in Shanghai.According to the NAFLD diagnostic criteria of the guidelines for diagnosis and treatment of nonalcoholic fatty liver disease (2018) formulated by the National workship on Fatty liver and alcoholic liver disease and Chinese Society on Hepatology, they were divided into the healthy control group and the experimental group.Besides,on the same day, the height, weight, waist, BMI, blood pressure and heart rate of patients were measured, and fasting blood was taken to obtain blood glucose,ALT, AST, TB, DB, TP, ALB, Che, ALP, γ - GT, TG, TC, HDL-C, LDL-C and other serum data.Serum samples were analyzed using LC/MS and data was processed by SIEVE software and simca-P to validate the potential biomarkers. The altered metabolites were identified by variable importance in projection value (VIP > 1) and ANOVA (p<0.01). The pathway analysis was performed by using MetaboAnalyst 4.0. In addition, our project has passed the review of ethics committee of Putuo Hospital Affiliated to Shanghai University of traditional Chinese medicine, and its ethics approval number is ptec-a-2018-49-1.ResultsThe serum biochemical indicators of early NAFLD patients showed no significant difference with NC (p>0.05). While there was significant difference of blood lipids indicators between NAFLD and NC (p<0.001). Finally, 55 metabolites were identified and the AUC of ROC curve results showed that new identified biomarkers owned high predictability and reliability. ConclusionIt is found that 15 metabolites in serum were of great diagnostic value in early NAFLD patients. AUC of these biomarkers (>0.9) were much higher than clinical indicators (0.770). This may be worth further research in the clinic.

TNF-α, HGF and TGF-β1 are Involved in Liver Regeneration Following Partial Hepatectomy Using Portal Vein ArterializationsExperiments on liver regeneration after partial hepatectomy have shown that TNF-α, HGF and TGF-β1 and other cytokines play important roles in the different stages of liver regeneration, however, the effect of portal vein arterialization (PVA) on the expressions of these cytokines during liver regeneration is not clear. Sprague Dawley rats were randomly divided into the PVA group and control groups, and blood was collected for the detection of ALT using an automatic biochemical analyzer. The expressions of TNF-α, HGF and TGF-β1 in liver tissues were detected by quantitative RT-PCR. The ALT levels in both groups in the early period after surgery were significantly higher than those before operation, and gradually returned to normal at 7 days after surgery. At 12 h and 24 h after operation, the TNF-α expression in the PVA group was significantly higher than that in the control group (P<0.05), but no significant difference at 7 days after surgery was observed between the two groups. At 12 h, the HGF expression in the PVA group was similar to that in the control group, but significantly higher than in the control group at 24 h (P<0.05). At 24 h, the TGF-β1 expression in the PVA group was significantly lower than that in the control group (P <0.05), but no significant difference was found at 48 h after surgery between the two groups. The promotive effects on the portal vein arterialization at the early stage of liver regeneration were associated with the changes in the expressions of TNF-α, HGF and TGF-β1.

Graft cell repopulation and tendon-bone tunnel healing are important after allograft anterior cruciate ligament reconstruction (ACLR). Freshly isolated bone marrow mononuclear cells (BMMNCs) have the advantage of short isolation time during surgery and may enhance tissue regeneration. Thus, we hypothesized that the effect of intra-articular BMMNCs in post-allograft ACLR treatment is comparable to that of cultured bone marrow stromal cells (BMSCs). A rabbit model of hamstring allograft ACLR was used in this study. Animals were randomly assigned to the BMMNC, BMSC, and control groups. Fresh BMMNCs isolated from the iliac crest during surgery and cultured BMSCs at passage four were used in this study. A total of 1 × 107 BMMNCs or BMSCs in 100 µL phosphate-buffered saline were injected into the knee joint immediately after ACLR. The control group was not injected with cells. At two and six weeks post operation, we assessed graft cell repopulation with histological and cell tracking staining (PKH26), and tendon-bone healing with histological micro-computed tomography and immunohistochemical analyses for collagen I and monocyte chemoattractant protein-1 (MCP1). At two weeks post operation, there was no significant difference in the total cell population within the allograft among the three groups. However, the control group showed significantly higher cell population within the allograft than that of BM cell groups at six weeks. Histological examination of proximal tibia revealed that the intra-articular delivered cells infiltrated into the tendon-bone interface. Compared to the control group, the BM cell groups showed broader gaps with interfacial fibrocartilage healing, similar collagen I level, and higher MCP1 expression in the early stage. Micro-CT did not reveal any significant difference among the three groups. BMMNCs and BMSCs had comparable effects on cell repopulation and interfacial allograft-bone healing. Intra-articular BM cells delivery had limited benefits on graft cell repopulation and caused higher inflammation than that in the control group in the early stage, with fibrocartilage formation in the tendon-bone interface after allograft ACLR.

ObjectivesStandard treatment in early-stage cervical cancer is a radical hysterectomy (RH) with pelvic lymphadenectomy. In women who wish to preserve fertility radical vaginal trachelectomy has been proposed; however, this is not feasible in larger tumors, and nerve-sparing surgery is not possible. Nerve-sparing radical abdominal trachelectomy (NSRAT) overcomes these disadvantages.MethodsCase-control study of women with early-stage cervical cancer (International Federation of Gynecology and Obstetrics IA2-IB) submitted to NSRAT from 2000 until 2011. Women submitted to nerve-sparing RH with early-stage cervical cancer were included as control subjects.ResultsTwenty-eight patients and 77 control subjects were included. Neoadjuvant chemotherapy was administered in 3 women before NSRAT because the linear extension was or exceeded 40 mm. Local recurrence rate was 3.6% (95% confidence interval [CI], 0.00–10.6) in the NSRAT group compared with 7.8% (95% CI, 1.7–13.9) in the control group (P = 0.44). No significant difference was found between both groups regarding disease-free survival and survival. The overall pregnancy rate was 52.9% (95% CI, 28.7%–77.2%). The mean follow-up was 47.3 months (range, 6–122 months) for NSRAT and 51.8 months (11–129.6 months) for nerve-sparing RH.ConclusionsNerve-sparing radical abdominal trachelectomy seems safe and effective in women with early-stage cervical cancer who wish to preserve fertility. Respective women should be informed about this treatment option, especially if the tumor is too large for radical vaginal trachelectomy.

Empiric broad-spectrum antimicrobials therapy is suggested to be started immediately for sepsis patients. Empiric antimicrobial therapy should be narrowed once pathogen identification and sensitivities are established. However, the detail mechanisms of de-escalation strategy are still unclear. Here we hypothesized neutrophil extracellular trap (NETs) played an essential role and de-escalation strategy might alleviate organs injury through regulation of NETs formation in sepsis. We evaluated the effect of imipenem and ceftriaxone on NETs formation in vitro and examined the role of reactive oxygen species (ROS). Next, we designed de-escalation and escalation strategy based on their effects on NETs formation in CLP model. Organ injury, inflammatory cytokines, NETs levels were compared and evaluated. The in vitro study showed that imipenem and ceftriaxone had opposite effects on NETs formation in activated neutrophils. De-escalation therapy resulted in an evaluated MPO-DNA during early stage and decreased MPO-DNA during late stage, which exerted the reverse effects in escalation therapy sepsis animal model. Inflammatory response and organ injury exacerbated when eliminated NETs with DNAseI during early stage of sepsis (p<0.01). Histopathological analysis showed decreased injury in lung, liver and intestine in de-escalation therapy compared with escalation therapy (p<0.01). De-escalation therapy results in the highest 6-day survival rate compared with the control group (p<0.01), however, no significant difference was found between de-escalation and escalation group (p=0.051). We demonstrate that de-escalation, not escalation, therapy reduces organ injury, decreases inflammatory response by promoting NETs formation in the early stage and inhibiting NETs formation in the late stage of sepsis.
