Background: Norhraman is an alkaloid that can damage the nervous system. Objective: This experimental study aimed to investigate the effects of prenatal norharman exposure on seizure severity in adult rats. Methods: A total of 36 Wistar rats were randomly divided into 3 groups as follows: Control, sham (solvent), and norharman (1000 ug/kg). The sham and norharman groups were treated once daily for 8-18 of the fetal period. 2 months after birth, kindling was performed by the daily injection of Pentylenetetrazol (PTZ) (30 mg/kg); then, the seizure behavior was recorded for approximately 30 minutes. The obtained data were analyzed using Analysis of Variance (ANOVA) and Tukey’s posthoc test. Results: The present study data indicated that administrating norharman in the fetal period increased seizure responses to PTZ, i.e., significant in the examined female rats (P<0.05). Conclusion: According to the current research findings, the severity of seizures increases in subjects who have been exposed to norharman during the prenatal period.

To study the regulation of the multiple forms of erythromycin N-demethylase, we determined the influence of sex and inducer treatment on this mixed-function oxidase activity in adult Wistar rats injected intraperitoneally once daily for 4 days with dexamethasone, pregnenolone-16α-carbonitrile, phenobarbital, or 2% Tween 80 (control). Based on the results from a computer curve-fitting procedure (ENZFITTER) as well as Eadie–Hofstee and Lineweaver–Burk plots, at least two forms of erythromycin N-demethylase were present in control, dexamethasone, pregnenolone-16α-carbonitrile, and phenobarbital-treated male rats and in control and dexamethasone-treated female rats. Only a high-affinity form was apparent in pregneno-lone-16α-carbonitrile and phenobarbital-treated female rats. Therefore, more than one form of erythromycin N-demethylase exists in hepatic microsomes from adult rats, depending on sex and inducer treatment. As well, at a substrate concentration commonly used in the erythromycin N-demethylase assay, the relative contribution of the high- and low-affinity forms to the enzyme activity varies with sex and inducer treatment.Key words: erythromycin N-demethylase, dexamethasone, pregnenolone-16α-carbonitrile, phenobarbital, mixed-function oxidase.

In women, hyperprolactinemia is often due to a prolactin (PRL)-secreting adenoma or PRL cell hyperplasia. RRL excess stimulates the mammary glands and causes proliferation of the alveolar epithelium. Bromocriptine, a dopamine agonist, inhibits PRL secretion and is given to women to treat nonpuerperal galactorrhea. Old female rats have been reported to have PRL cell hyperplasia or adenoma leading to PRL hypersecretion and breast stimulation. Herein, we describe the effect of bromocriptine and consequently the reduction in serum PRL levels on the ultrastructure of rat mammary glands.Female Long-Evans rats, 23 months of age, were divided into control and bromocriptine-treated groups. The control animals were injected subcutaneously once daily with a 10% ethanol vehicle and were later divided into a normoprolactinemic control group with serum PRL levels under 30 ng/ml and a hyperprolactinemic control group with serum PRL levels above 30 ng/ml.

ABSTRACT Male and female rats of varying ages were placad on a low iodine diet (LID) plus KClO4 or 6-propyl-2-thiouracil (PTU) or on the same diet supplemented with I (control rats). Goitrogenesis was also induced with LID plus PTU in gonadectomized animals of both sexes. The weight of the control and goitrogen treated animals, and the weight and iodine content of their thyroids were determined, as well as the plasma PBI, TSH, insulin and glucose levels. The pituitary GH-like protein content was assessed by disc electrophoresis on polyacrylamide gels. If goitrogenesis was induced in young rats of both sexes starting with rats of the same age, body weight (B.W.) and pituitary growth hormone (GH) content, it was found that both the males and females developed goitres of the same size. On the contrary, when goitrogenesis was induced in adult animals, it was found that male rats, that had larger B.W. and pituitary GH content than age-paired females, developed larger goitres. However, both male and female rats were in a hypothyroid condition of comparable degree as judged by the thyroidal iodine content and the plasma PBI and TSH levels. When all the data on the PTU or KClO4-treated male and female rats of varying age and B.W. were considered together, it was observed that the weights of the thyroids increased proportionally to B.W. However, a difference in the slope of the regression of the thyroid weight over B.W. was found between male and female rats, due to the fact that adult male rats develop larger goitres than female animals. In addition, in the male rats treated with PTU, gonadectomy decreased the B.W., pituitary content of GH-like protein and, concomitantly, the size of the goitre decreased; an opposite effect was induced by ovariectomy on the female animals. However, when goitrogenesis was induced in weight-paired adult rats of both sexes, the male animals still developed larger goitres than the females. Among all the parameters studied here, the only ones which appeared to bear a consistent relationship with the size of the goitres in rats of different sexes, treated with a given goitrogen, were the rate of body growth and the amount of a pituitary GH-like protein found before the onset of the goitrogen treatment. Moreover, though the pituitary content of the GH-like protein decreased as a consequence of goitrogen treatment, it was still somewhat higher in male that in female animals. The present results suggest that GH may somehow be involved in the mechanism by which male and female rats on goitrogens develop goitres of different sizes, despite equally high plasma TSH levels.

ABSTRACT The influence of early androgen treatment, light deprivation (by blinding), pinealectomy and superior cervical ganglionectomy on the reproductive system of female rats was tested. Early postnatal treatment of rats with testosterone propionate caused adult rats to exhibit the characteristic signs of androgen sterilization; these included polyfollicular ovaries, normal-sized uteri and persistent vaginal cornification. If early androgentreated rats also were blinded the ovaries were smaller in size and contained fewer follicles, the uteri were greatly reduced in size and the incidence of vaginal oestrus was decreased by approximately 50% If in addition to blinding, androgen-sterilized animals were subjected to either removal of the pineal gland or superior cervical ganglia, the reproductive organs and the vaginal smears were indistinguishable from those of testosterone-treated rats with eyes. These data indicate that the inhibitory influence of blinding on the pituitary-ovarian axis was mediated through the sympathetic nervous system and the pineal gland. The restraining influence of light deprivation on the growth of the reproductive organs was not permanent as illustrated by the fact that if these animals were kept to 120 days of age the ovaries and uteri grew to the same level as those of pinealectomized control rats.

The basic nature of guanethidine and some of its effects suggested a possible action of this drug on histamine metabolism. A single intraperitoneal injection of guanethidine (10 mg/kg) in male rats was found to double the daily urinary excretion of free histamine; daily injection for three weeks caused a 10-fold increase. In male rats, guanethidine increased the number of mast cells in the peritoneal fluid and, in both peritoneal fluid and mesentery, caused a significant degranulation of these cells; this action was not observed in female rats. This finding may indicate that guanethidine blocks methylation of histamine by inhibiting imidazole methyl transferase since this enzyme is found in male but not in female rats. Bethanidine and reserpine had no effect on histamine excretion. Imidazole was found to be even more potent than guanethidine in causing an increase in urinary histamine. Guanethidine and imidazole neither potentiated nor mimicked the action of histamine on the isolated ileum.

ABSTRACT In vitro experiments carried out with uterus preparations of ovariectomized adult rats indicate the presence in this tissue of a 20β-hydroxysteroid-oxidoreductase which catalyzes the conversion of 20β-hydroxy-4-pregnen-3-one to progesterone. Since a hepatic 20β-hydroxysteroid-oxidoreductase is absent in adult female rats, the myometrial enzyme can be responsible for the biological activity of 20β-hydroxy-4-pregnen-3-one in these animals. Besides progesterone five metabolites were isolated and identified after incubation of [4-14C]20β-hydroxy-4-pregnen-3-one with uterine tissue: 20β-hydroxy-5α-pregnan-3-one, 20β-hydroxy-5β-pregnan-3-one, 5α-pregnane-3α,20β-diol, 4-pregnene-3α,20β-diol and 4-pregnene-3β,20β-diol. The conversion of 20β-hydroxy-4-pregnen-3-one to progesterone permits us to regard all five steroids isolated as progesterone metabolites in the rat uterus. 20β-hydroxy-5β-pregnan-3-one is the first C21-metabolite with a 5β(H)-configuration isolated in the rat uterus, which indicates the presence of 5β-reductase in this tissue.

We previously reported that maternal separation (MS), a model of early life stress, does not modify baseline blood pressure in adult rats, but increases sensitivity to hypertensive stimuli. Under baseline conditions, adult male rats exposed to MS have significantly reduced glomerular filtration rate (GFR). Acute phenylephrine-induced reductions in renal blood flow is significantly attenuated in rats exposed to MS compared to control rats. Furthermore, norephinephrine (NE) content was increased in renal cortex of MS rats compared to control rats (p<0.05). These data indicate that MS induces increased renal sympathetic outflow. Thus, we hypothesized that renal denervation will normalize GFR in rats exposed to MS. Male WKY rat pups were separated from their mothers for 3 hrs/day during the morning hours from day 2 to 14 of life. Male non-separated littermates served as control rats. Experiments were performed in 300-320 g adult rats. Denervation (DnX) was performed mechanically stripping all visible renal nerves followed by topical phenol (10%) on the renal artery. Control-sham, MS-sham, control-DnX, and MS-DnX rats were instrumented with catheters in the femoral vein and abdominal aorta. Rats were placed in metabolic cages, connected to swivels, and allowed to recover for 4-5 days. Sodium intake was clamped at 2.8 mEq/day in both groups by combining sodium deficient diet and 24 hr/day 0.9% iv saline infusion (20 ml/day). GFR was determined by plasma clearance of [125I]iothalamate in the conscious state. During baseline conditions, MAP was not different between control-sham and MS-sham rats (99±4 vs 97±2 mmHg, respectively). MAP was reduced in both control-DnX and MS-DnX rats (91±2 mmHg and 83±3 mmHg, p<0.05, respectively) compared with the respective sham group. The reduction in MAP tended to be greater in MS than in control rats (-9±1 and -14±2 mmHg, p=0.074). DnX did not modify GFR in control rats (sham: 3.1±0.1 ml/min vs DnX: 3.5±0.4 ml/min). However, DnX significantly increased GFR in rats exposed to MS (sham: 2.4±0.2 ml/min vs DnX: 3.8±0.4 ml/min, p<0.05). These data support our hypothesis that MS induces increased renal sympathetic tone to reduce GFR in MS male rats, and may contribute to the exacerbated response to hypertensive stimuli observed in MS rats.

Background: Puberty is a developmental transition in which an estrogenic surge occurs, mediating the release of xenoestrogens, like aluminium. Aluminium’s effect on anxiety in rodents at the different developmental stages is inconsistent. Aims: This study aimed at investigating the effect of the metalloestrogenic property of aluminium on anxiety-like behavioral changes in prepubertal and young adult female rats. Objective: Considering this aim, our objective was to evaluate the anxiety-like behavior by the elevated plus maze in prepubertal and young adult female rats with or without acute exposure to aluminium. Methods: To address this property of aluminium, 5mg/Kg body weight (Al-5) and 10 mg/Kg body weight (Al-10) of aluminium was administered intraperitoneally to female rats at two developmental stages, prepubertal (PP; n = 8 for each dose) and young adult (YA; n = 6 for each dose) for two weeks. Post-treatment, three days behavioral assessment of the rats was done employing elevated plus maze. Results: Reduced escape latency was seen in Al-5, Al-10 pre-pubertal rats, and Al-5 young-adult rats on day 3. A significant reduction in open arm time was seen in the Al-5 young-adult rats. Aluminium treatment in the pre-pubertal rats reduced their head dipping and grooming. Reduced sniffing, head dipping, and stretch-attended posture in the treated young-adult female rats showed that they had impaired risk-taking tendency. Conclusion: Differential effect on the anxiety-like behavior in the pre-pubertal and young-adult female rats might be due to the metalloestrogenic property of aluminium, acting differently on the two age groups.

Glutamine synthetase (GS) is a glucocorticoid-inducible enzyme that has a key role for glutamine synthesis in muscle. We hypothesized that the glucocorticoid induction of GS could be altered in aged rats, because alterations in the responsiveness of some genes to glucocorticoids were reported in aging. We compared the glucocorticoid-induced GS in fast-twitch and slow-twitch skeletal muscles (tibialis anterior and soleus, respectively) and heart from adult (age 6-8 mo) and aged (age 22 mo) female rats. All animals received dexamethasone (Dex) in their drinking water (0.77 +/- 0.10 and 0.80 +/- 0.08 mg/day per adult and aged rat, respectively) for 5 days. Dex caused an increase in both GS activity and GS mRNA in fast-twitch and slow-twitch skeletal muscles from adult and aged rats. In contrast, Dex increased GS activity in heart of adult rats, without any concomitant change in GS mRNA levels. Furthermore, Dex did not affect GS activity in aged heart. Thus the responsiveness of GS to an excess of glucocorticoids is preserved in skeletal muscle but not in heart from aged animals.

The purpose of this study is to find and describe (1) the use of Mandar ethnic values in the Passayang-Sayang songs of the 1960-1970 period, (2) the use of the Mandar ethnic values in the Passayang-Sayang songs period of 1970-1980, (3) the use of the Mandar ethnic values in the Passayang-Sayang song period of 1980-1990, (4) the use of Mandar ethnic values in the Passayang-Sayang song period of 1990-2000, (5) the use of the Mandar ethnic value in the Passayang-Sayang song period of 2000-2010, and (6 ) the use of the Mandar ethnic value in the song of the Passayang-Sayang period 2010-Present. The theory used in this research is the theory of literary geography put forward by Herder, Stael, and Ricardson. This research is a qualitative descriptive study. The research data are in the form of Passayang-Sayang Mandar poetry texts, informant statements, and facts in the field. Data collection activities carried out by observation and observation techniques, interviews, recording, recording, and documentation. In this study the researcher acted as a key instrument guided by data collection guidelines. Data analysis procedures use the spiral model, while data analysis uses objective hermeneutics. To avoid research bias, triangulation was carried out for each stage of the study, research tools, and clarification of research findings to the informants as poet owners, Focus Group Discussions, and external audits. The findings of this study are the development of ethnic values in the Passayang-Sayang Mandar poem in the form of (16) ethnic values found in the period 1960-1970, (5) ethnic values found in the period 1970-1980, (7) ethnic values found in the 1980 period -1990, (6) ethnic values were found in the period 1990-2000, (7) ethnic values were found in the period 2000-2010, (8) ethnic values were found in the period 2010-Now.
