People living with diabetes mellitus across the world are expected to rise to 592 million by 2035 and the pandemic disease is worst in the developing countries like Nigeria, where the cost of treating/managing diabetes mellitus is relatively out of the reach of many diabetic patients. The study investigated the effect of Anogeissus leiocarpus Guill and Perr ethanolic leaf extract on blood glucose and other biochemical indices in alloxan-induced diabetic adult male Wistar albino rats. Diabetes mellitus was induced in overnight-fasted rats by a single intraperitoneal injection of alloxan monohydrate (150 mg/kg body weight). Diabetic rats were then treated with the extract for 14 days. Blood glucose, lipid profile and liver glycogen levels were determined. After the treatment with the extract, the fasting blood glucose levels significantly reduced (p < 0.05) in treated diabetic rats (62.2 ± 18.4 mg/dl) compared to untreated diabetic rats (73.8± 8.2 mg/dl). There was also significant (p < 0.05) reduction in the hyperlipidemic parameters. The results revealed that A. leiocarpus ethanolic extract has antidiabetic and hypolipidemic effects in alloxan-induced diabetic rats.Dhaka Univ. J. Pharm. Sci. 17(1): 65-72, 2018 (June)

Background: Diabetes mellitus is a chronic metabolic disorder of glucose metabolism characterised by hyperglycaemia. Long standing diabetes mellitus leads to various complications affecting multiple organ systems. Management of diabetes mellitus includes lifestyle modification and pharmacotherapy. Pharmacotherapy of diabetes mellitus includes a wide variety of drugs that help in achieving adequate glycaemic control. Anti-diabetic medications are however associated with several adverse effects. Phytochemicals are being used extensively for the treatment of various diseases. Use of phytochemicals would minimize adverse effects due to various anti-diabetic drugs and improve patient compliance. In the present study, authors studied the effect of turmeric on alloxan induced diabetes mellitus in albino rats.Methods: Albino Wistar rats of either sex weighing 180 - 250grams were utilized for the present study. Diabetes mellitus was induced by intraperitoneal administration of alloxan. Ethanolic extract of turmeric was administered to diabetic rats daily orally for duration of 28 days. Blood glucose levels were monitored using glucometer before and after intervention with turmeric.Results: Statistically significant reduction in mean blood glucose levels (p value <0.05) was seen after intervention with turmeric in diabetic rats. There was a significant reduction in mean blood glucose levels.Conclusions: Ethanolic extract of turmeric showed antihyperglycemic effect in diabetic rats.

 Objective: Lawsonia inermis (Linn.) leaves are one of the alternative medicines to treat diabetes mellitus in Indonesia. We investigated the blood glucose level (BGL) of the L. inermis (Linn.) leaves ethanolic extract (LLEE) leaves and evaluated the histopathological alterations in diabetic rats.Methods: This study was an experimental study with posttest - only control group design. Alloxan (120 mg/kg, intraperitoneally)-induced diabetic rats. 35 of Wistar rats (Rattus norvegicus) were divided randomly into five groups, i.e. K: Normal control, P1: Diabetic control, and P2, P3, and P4 (200 mg/kg body weight [BW], 400 mg/kg BW, and 600 mg/kg BW of LLEE, orally) for 28 days. At the end of the treatment, the rats were sacrificed to obtain the kidney for histopathological evaluation using hematoxylin and eosin technique. BGLs were conducted using a glucose meter (GlucoDR).Results: One-way ANOVA test showed that dose 400 mg/kg BW of the LLEE was related to BGL of alloxan-induced diabetic rats (p=0.000). The histopathological of kidney showed glomerular inflammation (GI), epithelial membrane lining degeneration, vascular congestion, and interstitial tubule hemorrhage at diabetic control (P1). Meanwhile, treated with 600 mg/kg BW of LLEE (P4) showed increase cellular regeneration as normal architecture of the kidney.Conclusion: The LLEE at dose 400 mg/kg BW effective decreased BGL and was able to restore the kidney destruction of alloxan-induced diabetic rats at dose 600 mg/kg BW.

Objective. Mallotus oppositifolius(Geiseler) Müll. Arg. (Euphorbiaceae) is folklorically used to “treat” diabetic conditions in some parts of Nigeria therefore the study, to investigate the extract of the leaves for activities on hyperglycaemia, lipid peroxidation, and increased cholesterol levelsin vivoin alloxan diabetic rats as well as its potential antioxidant activityin vitro.Methods. Albino rats (240–280 g) were given an injection of 120 mg/kg body weight, i.p. of alloxan monohydrate. After 8 days, diabetic animals with elevated fasting blood glucose levels (>9 mmol/L) were considered and selected for the study.Results. Oral treatment with the extract administered every 12 h by gavage at doses of 100, 200, and 400 mg/kg of the extract to the test rats, for 14 days, resulted in a significant dose-dependent decrease in blood glucose levels from 12.82 ± 1.02 mmol/dL to 4.92 ± 2.01 mmol/dL at the highest dose of 400 mg/kg compared to the control drug and glibenclamide as well as attendant significant decline in diabetic rats employed in the study.Conclusion. The extract also showedin vitroconcentration-dependent antioxidant activity following the 1,1-diphenyl-2-picryl-hydrazyl (DPPH) and ferric reducing assays. Findings further suggest the presence of active antidiabetic and antioxidant principles inM. oppositifoliusleaves.

Objective: The present study was aimed to evaluate the antidiabetic activity of ethanolic extract of the whole aerial plant of Portulaca grandiflora Hook on streptozotocin (STZ)-induced diabetic rats. Methods: Experimental diabetes was induced by a single dose of intraperitoneal injection of STZ (150 mg/kg). Adult male Wister albino rats were divided into five groups; normal control, diabetic control, diabetic glibenclamide (5 mg/kg), diabetic P. grandiflora H. extract (200 mg/kg), and diabetic P. grandiflora H. extract (400 mg/kg) for 21 days and analyzed for body weight (BW) and blood glucose. Results: The STZ-treated diabetic control rats showed a significant increase in blood glucose with a concomitant decrease in BW. Oral administration of P. grandiflora H. extract (200 and 400 mg/kg) and glibenclamide (5 mg/kg) for 21 days showed a significant reduction in blood glucose levels and elevation in the bodyweight studies as compared to control and glibenclamide-treated rats. Conclusion: The results of the present study showed that a potent antidiabetic activity was present in the aerial part of plant P. grandiflora H. extract.

Background: Over the recent year there has been a startling rise in the number of people suffering from diabetes mellitus. Estimation of blood glucose levels has been an essential laboratory investigation for screening and monitoring of diabetes. Saliva is one of the secretions in human body whose collection is easy and non-invasive. Salivary glucose hence could serve as an easy and non-invasive tool.Methods: Institutional review committee of NHRC gave us permission to carry out this study. 105 subjects with Type 2 diabetes mellitus who attended the OPD at Star hospital, Sanepa, Lalitpur, Nepal and 106 healthy individuals were consented to participate in this study. Glucose was measured by the GOD-POD (Glucose oxidase peroxidase) methods using the semi-autoanalyser and salivary glucose was compared with corresponding blood glucose levels.Results: A significant positive correlation of fasting salivary glucose level and fasting blood glucose level was observed in healthy (r=0.241, p=0.001) and in diabetic patients (r=0.202, p=0.001).Conclusions: The study implies a potential for saliva in monitoring and screening of diabetes mellitus.

Diabetes mellitus has high global prevalence and occurrence and is considered to bean endocrinological and/or metabolic disorder. Conventional drug treatment is costly and has toxic side effects, although it is successful in treating diabetes mellitus. If effective and less toxic, herbal medicine will thus include alternative therapy. This research has been designed to investigate the role of Grewia asiatica extract in the control of diabetes in male albino rats with Streptozotocin mediated type 2 diabetes. Grewia asiatica fruit extract at a dose of 200mg/kg was given to Streptozotocin mediated type II DM Rats. A known anti-diabetic drug, Glibenclamide has been used as a standard drug. The method of the research was to monitor the effect of Grewia asiatica on the blood glucose level of Rats. In this study, Rats were split into four categories i.e. Control, Streptozotocin treated, Streptozotocin + Glibenclamide treated and Streptozotocin +Grewia asiatica extract-treated group.  Grewia asiatica fruit extract significantly improve the blood glucose levels as compared to the standard drug Glibenclamide in Streptozotocin mediated diabetic group. Conclusion: It was concluded that Grewia asiatica may be used in the treatment of diabetes or decreasing the elevated level of blood sugar.

Background: Rheum emodi been used traditionally to treat diabetes in India. This study was designed to elucidate the action of R. emodi (Rhizome) ethanolic extract (75%) and its isolated compounds like emodin and chrysophanol on alloxan-induced diabetic rats to check the anti-diabetic efficacy. To access acute toxicological and histopathological study to evaluate the anti-diabetic efficacy of R. emodi. Method: Experimental rats were grouped into seven and each group consisting of 6 rats. 75% ethanolic extract and its compounds emodin and chrysophanol were given for 30 days. Results: The experimental rats were sacrificed on 30 days by cervical dislocation. Parameters like renal profile and lipid parameters were determined. As well as histopathological changes in diabetic rat’s liver, kidney and pancreas were studied. The 75% ethanolic extract of R. emodi (EE-ReR) fed orally to diabetic rats, resultant in a decline in the fasting blood glucose, total cholesterol, free fatty acids, creatinine, urea, and a rise in the insulin levels were observed almost normal range in the rats fed with the extract. Histopathological studies of pancreas, kidney, and liver in diabetic rats revealed that the treated group of rats showed regeneration of islets cells to almost as normal. Acute toxicological studies revealed that the extract is safe up to 2000mg/kg bw of extract feed orally. Conclusion: These findings are suggestive of a possible protective and prevent damage to the internal organs played by the R. emodi and its compounds like emodin and chrysophanol compounds and elevate insulin production during high blood glucose levels without any acute toxicologically effect.

Introduction: A number of recent literatures suggest a potential role of H2S and H2S modifying agents in the etiology and management of type-2 diabetes mellitus. Objective: The current study was aimed to evaluate the plasma levels of H2S in the patients with type 2 Diabetes mellitus and to find out if there is any relationship of H2S concentrations with the fasting blood glucose levels. Methods: Plasma H2S levels were measured in sixty two recently diagnosed type 2 diabetic patients and compared with similar number of healthy volunteers as controls. Results: The plasma H2S level in the patients ( 81.17 ± 16.40 micromol/l ) is significantly higher (P< 0.001) than the healthy controls (50.69 ± 8.69 micromol/l) and the H2S levels in plasma have significant positive correlation (r= 0.359, P=0.004) with fasting blood glucose levels. Conclusion: The present study has elucidated that the patients with type-2 diabetes mellitus are associated with elevated plasma H2S levels which are well correlated with glucose levels. This reveals a potential the role of H2S modulators towards the management of this non-communicable epidemic disorder. Alhagi maurorum(camel thorn plant) is a promising medicinal plant due to the presence of flavonoids and phenolic compounds as major contents of its constituents. No previous study has been conducted before onA. maurorum extractsas an antioxidative stress and/or antidiabetic herb in STZ-induced DM in rats. Therefore, four groups of rats were allocated as control (C), STZ-induced DM (D), and STZ-induced DM supplemented with 300 mg/kg BW of either aqueous extract (WE) or ethanolic extract (EE) ofA. maurorum. The plasma levels of glucose, TG, TC, LDL-C and VLDL-C, MDA, and bilirubin and the activities of transaminases and GR were significantly increased in the diabetic group. Also, diabetic rats showed severe glucose intolerance and histopathological changes in their livers. In addition, levels of insulin, total proteins, GSH, and HDL-C and the activities of SOD, GPx, and GST were significantly decreased in the diabetic rats compared to those of the control group. The ingestion ofA. maurorumextracts lowered the blood glucose levels during the OGTT compared to the diabetic rats and restored all tested parameters to their normal levels with the exception of insulin level that could not be restored. It is concluded thatA. maurorumextracts decreased elevated blood glucose levels and hyperlipidemia and suppressed oxidative stress caused by diabetes mellitus in rats.

Antidiabetic effects of ethanolic extract of Pleurotus ostreatus (mushroom) on alloxan-induced diabetic rats was studied. The median lethal dose (LD50) of the extract was determined to be 3,472.14 mgkg-1 and a single dose of 380.0, 760.0 and 1140.0 body weight of the extract were intraperitoneally administered as the treatment dose and the blood glucose levels (BGL) examined for 7 hours and 15 hours (prolonged) at 2 and 4 hours intervals respectively. The extract exhibited significant (p 0.05 and p 0.01) reduction in the blood glucose levels of the albino rats. The extract compared favourably with the standard reference drug (metformin) which all gave their maximum BGL reduction at 5 hours duration. The confirmation of antidiabetic potentials of the Pleurotus ostreatus tuber has been justified in this study as claimed by traditional medicine practitionersin Akwa Ibom State.
