Background: Diabetes mellitus (DM) is a heterogeneous group of metabolic disorders with micro and macrovascular complications which are the major causes of morbidity and mortality in diabetic patients. Peanut due to its anti-oxidant property can reduce blood glucose level and may reduce the risk of diabetes. Objective: To observe the anti-diabetic effects of peanut (Arachis hypogaea L.) in alloxaninduced diabetic male rats. Method: This experimental study was carried out in the Department of Physiology, Sir Salimullah Medical College (SSMC), Dhaka in 2013. For this purpose, 15 wistar albino male rats, aged 90-120 days, weighing 160-200 g (initial body weight) were included in the experimental group (DG-A-P). Age and weight matched 20 wistar albino rats without peanut supplementation was taken as control and divided into (BC and DC-A) . All groups of animals received basal diet for 21 consecutive days. In addition to basal diet, animals of DC-A received alloxan intraperitoneally only on the first day of the study. Moreover, animals of DG-A-P also received peanut extract for 21 consecutive days. After 16 hours of fasting blood samples were collected from tail vein of all rats on day-1 and day- 3. After taking final body weight all the rats were sacrificed on 22nd day. Their pancreas was removed and weighed. To observe glycemic control serum blood glucose and blood HbA1c levels were measured by usual laboratory technique. ANOVA, independent t-test and paired t-test were used for statistical analysis. Result: The % change of body weight was significantly (p<0.001) lower in DC-A in comparison to that of BC. The weight of pancreas was significantly lower in DC-A (p<0.001) and DG-A-P (p<0.05) when compared to that BC, whereas this level was significantly higher (p<0.001) in DG-A-P than that of DC-A. The mean fasting blood glucose level on day-3 was significantly (p<0.001) higher in DC-A and DG-A-P in comparison to that of BC. Then this level was significantly (p<0.001) lower in DG-A-P in comparison to that of DC-A on day-22. Moreover, significantly higher levels of HbA1c were observed in DC-A (p<0.001) and DG-A-P (p<0.05) in comparison to those of BC. Again, significantly (p<0.001) lower level of blood HbA1c were observed in DG-A-P than those of DC-A. Conclusion: From this study, it can be concluded that peanut (Arachis hypogaea L.) has anti-diabetic effect by reducing fasting blood glucose, HbA1c levels. Background and Aims: Small dense  low-density lipoproteins (sd-LDL) particles are smaller and heavier than typical LDL ones. They can penetrate into the endothelium of coronary arteries more easily because of their small size. Diabetes mellitus is accompanied by dyslipidemia such as increasing concentration of plasma very low density lipoprotein and sd-LDL. Peroxisome proliferator activated receptor γ (PPARγ ) can decrease the level of sd-LDL in plasma. Biochanin A (BCA), a natural compound, is a PPARγ agonist. The present study was designed to investigate the effect of BCA on sd-LDL-Clolesterol level in diabetic animals. Materials and Methods: Adult male rats (Wistar strain) were used as the animal models in this study. Animals were made diabetic by single intraperitoneal injection of Streptozotocin- Nicotinamide and then treated by 1 and 5 mg/kg of BCA for 28 days. Body weight and fasting blood glucose were also tested before and at the end of treatment. Furthermore, the size of LDL particles were measured by nondenaturing polyacrylamide gradient gel electrophoresis assay. Results: Results of the present study indicated that BCA administration at dose of 5mg/kg decreased fasting blood glucose level and increased body weight and diameter of LDL particles in diabetic animals significantly. Conclusions: BCA seems to be an appropriate agent in diabetes mellitus, because it improves the diabetic dyslipidemia, which is the most important complication in diabetic patients.

Abstract: Glucose is a main compound that has important role in preparation and development  of energy in human body. Blood glucose level is the amount of glucose in the blood. One of the factors that affects blood glucose level is body weight. Body weight is affected by several factors, such as aged, gender, and physical activities. Overweight may cause insulin resistance, results in increasing of the blood glucose level. This study aimed to describe the fasting blood glucose level at students Faculty of Medicine year 2011 Sam Ratulangi University with Body Mass Index (BMI) ≥23 kg/m2. Research method used for this study was cross sectional descriptive survey with purposive sampling method. From 26 respondents, 12 overweight (BMI ≥23 kg/m2) respondents (46,15%) had average fasting blood glucose level 85,4 mg/dL and 14 obese (BMI ≥25 kg/m2) respondents (53,85%) had average fasting blood glucose level 86 mg/dL. Conclusion: the description of fasting blood glucose level at students Faculty of Medicine year 2011 Sam Ratulangi University with Body Mass Index (BMI) ≥23 kg/m2 is at normal level (80-100 mg/dL). Kadar glukosa darah adalah jumlah kandungan glukosa dalam darah. Salah satu faktor yang mempengaruhi kadar glukosa darah adalah berat badan. Berat badan dipengaruhi oleh beberapa faktor, antara lain usia, jenis kelamin, dan aktivitas fisik. Kelebihan berat badan dapat menyebabkan resistensi insulin sehingga kadar glukosa dalam darah meningkat. Peningkatan kadar glukosa darah merupakan salah satu penanda sindroma metabolik. Penelitian ini bertujuan untuk mengetahui gambaran kadar glukosa darah puasa pada mahasiswa Fakultas Kedokteran Universitas Sam Ratulangi Angkatan 2011 dengan Indeks Massa Tubuh (IMT) ≥23 kg/m2. Jenis penelitian berupa penelitian survey deskriptif cross sectional dengan metode purposive sampling. Sampel penelitian sebanyak 26 orang responden. Hasil penelitian ini didapatkan 12 orang responden (46,15%) yang overweight (IMT ≥23 kg/m2) memiliki rata-rata kadar glukosa darah puasa 85,4 mg/dL dan 14 orang responden (53,85%) yang obesitas (IMT ≥25 kg/m2) memilki rata-rata kadar glukosa darah puasa 86 mg/dL. Simpulan: semua responden mahasiswa Fakultas Kedokteran Universitas Sam Ratulangi Angkatan 2011 memiliki kadar glukosa darah puasa normal (80-100 mg/dL). Combination therapy is considered as a new and effective therapeutic strategy for control of hyperglycemia. The present study aimed to determine the hypoglycemic effect of Afghan Hibiscus sabdariffa L. and Carum Carvi L. hydro-alcoholic extracts either alone or in combination on diabetic rats. Streptozotocin-induced diabetic rats were administered intraperitoneally with hydro-alcoholic extracts of H. sabdariffa (75, 150 and 300 mg/kg), C. carvi (150, 300 and 500 mg/kg) and their combinations for 21 days. The Fasting blood glucose and body weight of rats were determined on 0, 7, 14 and 21st days. The results showed a significant (P<0.0001) hypoglycemic effect of H. sabdariffa extract (75, 150 and 300 mg/kg) on 7, 14 and 21th days, C. carvi extract (500 mg/kg), and their effective doses combination (75:25), especially on 7th day. Only 75 mg/kg of H. sabdariffa had positive effect on body weight loss of diabetic rats (P<0.05). One can conclude that Afghan H. sabdariffa and C. carvi hydro-alcoholic extracts and their combination have a hypoglycemic effect. However, their combinations do not have any synergic effects, so the hypoglycemic effect of H. sabdariffa was much greater and even normalized the blood glucose level.

Objective: To find out the effects of ginger (Zingiber officinale) juice blood glucose in alloxan induced-diabetes mellitus in rats. Methods: This experimental study was done in the Department of Pharmacology & Therapeutics, Dhaka Medical College, Dhaka, in collaboration with the Departments of Pathology, Ibrahim Medical College, Dhaka between January and December 2009. This experimental animal study was divided into two parts, which were Experiment-1 and Experiment-2. Experiment 1 comprises of 12 rats and divided into 2 groups each group having 6 rats. Rats of group-A was non-diabetic normal control group and group-B was fed with ginger (Zingiber officinale) in a dose of 4ml/kg body weight orally through Ryle’s tube. Experiment-2 comprised of 12 rats divided into 2 groups each containing 6 rats labeled as group C, group D. Rats of groups C administered alloxan 150 mg/kg intraperitoneally on the 2nd day of the study. Rats of group D were administered alloxan 150mg /kg intraperitoneally and ginger (4ml/kg of body weight orally) on the 2nd day of the study. Results: The fasting blood glucose level at day 12 in the rats treated with ginger (Zingiber officinale) 4 ml /kg body weight orally daily for 12 days showed reduction in fasting blood glucose level as compared to control group, but not significant, which indicates that ginger has no effect in lowering blood glucose of normal rats. The fasting blood glucose levels at day 12 in the rats of group D (treated with ginger and alloxan) showed highly significant reduction in fasting blood glucose level as compared to diabetic control group (p<0.002). Conclusion: Consumption of ginger produced a significant antihyperglycemic effect in experimentally induced diabetic rats. Background: Diabetes Mellitus is one of the leading cause of morbidity and mortality globally. According to WHO there is a steady increase in the number of diabetic patients annually. Maintenance of good glycaemic control in type 2 diabetic patients typically becomes progressively more difficult as the duration of disease lengthens due to decline in the capacity of the pancreatic beta cells for glucose stimulated insulin release, in the presence of insulin resistance. This study is conducted to observe the effect of metformin and bromocriptine individually and sub-therapeutic doses of these drugs when used as a combination therapy. Objective: The objective of this study is to primarily investigate and then compare the antihyperglycemic effects of bromocriptine with metformin, also to see the combined effect of sub therapeutic doses of both these drugs. Methodology: Random allotment of 24 albino male rats was done in four groups. Group 1 was kept as control. Alloxan monohydrate was given to group 2, 3 and 4 and diabetes was induced. Group 2 and 3 were treated with metformin (1.5mg/kg body weight) and bromocriptine (3 mg/kg body weight) respectively while group 4 was treated with sub therapeutic doses of metformin (1 mg/kg body weight) and bromocriptine (1.5 mg/kg body weight)both. Serum glucose levels were estimated at 1, 10, 20 and 30 days. Results: Results showed that metformin reduces blood glucose level significantly where as bromocriptine also showed reduction of blood glucose level but not as significantly as metformin. However, the combination of metformin and bromocriptine showed much reduction in blood glucose level than metformin and bromocriptine used alone. In this study, catechin (CTN) isolated from Elaeagnus umbellata was evaluated for in vitro antioxidant potential and inhibition of carbohydrate digestive enzymes (α-amylase and α-glucosidase). The compound was also tested for its in vivo antidiabetic potential using Sprague-Dawley rats as experimental animals. The effects of various doses of catechin in STZ (Streptozotocin) induced diabetic rats on fasting blood glucose level, body weight, lipid parameters, hepatic enzymes, and renal functions were evaluated using the reported protocols. The CTN exhibited the highest percent antioxidant for free radical scavenging activity against DPPH and ABTS free radicals, and inhibited the activity of carbohydrate digestive enzymes (with percent inhibition values: 79 ± 1.5% α-amylase and 80 ± 1.1% α-glucosidase). Administration CTN and standard glibenclamide significantly decreased the fasting blood glucose level and increased the body weight in STZ-induced diabetic rats. CTN significantly decreased the different lipid parameters, hepatic, and renal function enzyme levels along with Hb1c level in diabetic rats, while significantly increasing the high-density lipoprotein (HDL) level with values comparable to the standard glibenclamide. Further, the altered levels of glutathione and lipid peroxides of liver and kidney tissues were restored (by CTN) to levels similar to the control group. CTN significantly increased the antioxidant enzyme activities, total content of reduced glutathione, and reduced the malondialdehyde (MDA) level in rat liver and kidney tissues homogenates, and also corrected the histopathological abnormalities, suggesting its antioxidant potential.

Abstract: Grass jelly leaf is a traditional Indonesian beverage that is believed to be beneficial for our health. This study was carried out to observe the increased number of pancreatic  cells and the decreased fasting blood glucose in male Wistar rats with diabetes induced by streptozotocin (STZ). This was a pure experimental study with a post-test only control group design. Subjects were 36 male rats divided into 2 groups: the control group (P0), given glibenclamide dose 0.09 mg /200 g rat body weight and 2 ml aquadest as placebo; and the treatment group (P1), given glibenclamide 0.09 mg/200mg rat body weight and grass jelly leaf extract 54 mg/200 mg rat body weight. Pancreatic  cell count and blood glucose examination were performed after 28 days of treatment. The results showed that the number of pancreatic  cells in the treatment group (P1) was statistically higher than of the control group (P0) (56.72±5.644 vs 29.11±2.698 cells (P ≤0.001). In addition, the fasting blood glucose level of the treatment group (P1) was statistically lower than of the control group (P0) (79.50±37.75mg/dl vs 191.00±123.15mg/dl) (P <0.001). Conclusion: The administration of grass jelly extract increased pancreatic β cells and decreased fasting blood glucose level in diabetic male rats. Penelitian ini dilakukan untuk mengetahui adanya peningkatan jumlah sel β pankreas dan penurunan kadar gula darah puasa pada tikus Wistar diabetes yang diinduksi streptozotocin (STZ). Jenis penelitian ialah eksperimental murni dengan post-test only control group design. Subyek penelitian ialah 36 ekor tikus putih jantan, dibagi menjadi 2 kelompok, yaitu: kelompok kontrol (P0) yang diberikan glibenklamid oral dosis 0,09 mg/200gr BB tikus dan 2 ml aquadest sebagai plasebo; dan kelompok perlakuan (P1) yang diberikan glibenklamid oral dosis 0,09 mg/200 gr BB tikus dan ekstrak daun cincau dosis 54 mg/200gr BB tikus. Penghitungan jumlah sel β pankreas dan pengukuran gula darah dilakukan setelah 28 hari perlakuan. Hasil penelitian menunjukkan bahwa kelompok perlakuan (P1) memiliki jumlah sel β pankreas yang secara statistik lebih tinggi dibandingkan kelompok kontrol (P0) (56,72±5,644 sel/lapang pandang vs 29,11±2,698 sel/lapang pandang) (P ≤0,001). Selain itu, kelompok perlakuan (P1) memiliki kadar gula darah puasa yang secara statistik lebih rendah dibandingkan kelompok kontrol (P0) (79,50±37,75 mg/dl vs 191,00±123,15 mg/dl) (P <0,001). Simpulan: Pemberian ekstrak daun cincau (Mesona palustris BL) meningkatkan jumlah sel β pankreas dan menurunkan kadar gula darah puasa pada tikus (Rattus novergicus) jantan galur Wistar diabetes.Kata kunci: ekstrak daun cincau, gula darah puasa, sel beta pankreas, diabetes melitus

Objective: To evaluate the anti-diabetic and anti-diarrheal activity of methanol extract of Flemingia stricta Roxb. (Fabaceae) leaf.Methods: In anti-diabetic study, the extract was administered to alloxan-induce diabetic mice at two concentrations (200mg/kg and 400mg/kg body weight) for acute (12 hours) and prolong treatments (15 days) and blood glucose levels (Blood glucose level) of diabetic mice were monitored at intervals of hours and days throughout the duration of treatment. Antidiarrheal test was conducted by castor oil induced diarrhea and enteropooling as well as intestinal motility in mice at three different concentration (100mg/kg, 200mg/kg and 400mg/kg body weight).Results: Treatment of alloxan induce diabetic mice with the extract caused a significant reduction in fasting blood glucose level of the diabetic mice both in acute (12 hours) and prolong treatment (15 days) and it was determined that the extract at both concentration (200mg/kg and 400mg/kg) showed the significant (P&lt;0.05) hypoglycemic effect in comparison to the standard drug Metformin (10mg/kg). In the case of castor oil induced diarrheal test, enteropooling test and gastrointestinal motility test, the extract of F. stricta at 100mg/kg, 200mg/kg and 400mg/kg has given significant effect (P&lt;0.05) in comparing to standard drug Loperamide (5mg/kg). Conclusion: These result suggested that the methanol extract of F. stricta Roxb. possess promising anti-diabetic effect on alloxan-induced mice and significant antidiarrheal effect on castor oil induced diarrheal mice.
