An experiment was conducted to know the effect of supplemental phosphorus on biomass growth and composition of a floating aquatic fern, Azolla pinnata, cultured in a pit system for 21 days with 4 levels of phosphorus (0, 5, 10, and 15 ppm) supplementation. Water quality parameters of the pits were within a suitable range for A. pinnata culture. It was observed that fresh and dry weights of A. pinnata increased with phosphorus supplementation up to 10 ppm. Doubling time was the fastest when the culture medium was supplemented with 10 ppm of phosphorus. Phosphorus content of A. pinnata was proportional to the phosphorus supplementation in the culture medium. Supplementation of phosphorus also improved the protein and lipid contents of A. pinnata. It was concluded that supplementation of 10 ppm phosphorus to water used for culturing A. pinnata is optimum under outdoor conditions. SAARC J. Agric., 19(1): 177-184 (2021)

The aim of this study was to find out the effect of different organic fertilizers on the population growth of Chydorus sp. This research was conducted at Laboratorium of Nutrition and Food Technology, Faculty of Fishery and Marine Sciences, Sam Ratulangi University, from December, 2012 to January, 2013. The experiment was run in the Completely Randomized Design with 3 treatments and 3 replication. Chydorus sp. was cultured in 9 glass jars with a volume of 1 L each. Three treatments applied in this research included A: medium culture composed of 500ml of water and 50 grams of soil; B: medium culture composed of 500ml of water, 50 grams of soil and 10g of cow dung; C: medium culture composed of 500ml of water, 50 grams of soil and 10g of horse dung. The density of Cydorus sp. in each media was 10 individuals /500 ml water. Observation on the population growth, development of live preys, and water quality parameters was conducted for 20 days period. Data were statistically analized with Analysis of variance. The results showed the highest density of Chydorus sp. was reached by Chydorus cultured in medium with horse manure (2169 individual/500 ml), followed by medium culture with cow manure (1715 individual/500 ml), and the lowest in medium culture with soil which was 1065,33 individual/500 ml. However, Analysis of variances showed that Chydorus population growth were not significantly affected by different culture medium. Keywords: manure, liquid fertilizer, growth, Chydorus sp.

In a laboratory experiment, liming resulted in an average decline of 3.6 per cent in the total organic phosphorus content of incubated surface samples of seven acid soils from eastern Canada. Increases of 2.6 and 5.1 per cent in 1N H2SO4- and 4N HCl-soluble inorganic phosphorus, respectively, and a decrease of 46.4 per cent in NaHCO3-soluble organic phosphorus (pH 8.5) provided further evidence of mineralization of organic phosphorus following liming. There was some evidence, however, that the differences in NaHCO3-soluble organic phosphorus following liming were due only in part to mineralization, since Ca(OH)2 added to a soil just prior to extraction with NaHCO3 had a repressive effect on the solubility of the organic phosphorus compounds.Some mineralization of organic phosphorus occurred when unlimed samples were incubated in the laboratory for 9 months.Marked increases in microbiological activity, as indicated by increased numbers of microorganisms, and increased CO2 and NO3-nitrogen production, were associated with lower values for extractable organic phosphorus following liming. Partial sterilization of samples with toluene lowered biological activity in the unlimed and limed samples. Toluene was found, however, to have a positive effect on release of phosphorus from organic form.

AbstractThe objective of the paper was to analyse the use of the designed photobioreactor for freshwater microalgae cultivation in the controlled laboratory conditions. The work covered the design and construction of photobioreactors (PBR) and setting up comparative cultivations of freshwater microalgae chlorelli vulgaris along with determination of the biomass growth intensity for a varied amount of supplied culture medium. It was found out that the constructed PBR may be used for microalgae cultivation in the controlled conditions. The impact of the culture medium amount on the growth of chlorelli vulgaris was proved. As a result of the increase of culture medium concentration to 30.1-120.4 ml·l−1 of water, dry mass in photobioreactorsincreased respectively from 1.33 g·dm−3 to 4.68 g·dm−3.

Dengue is vector-borne diseases with 390 million infections per year extending over 120 countries of the world. Aedes aegypti (L.) (Diptera: Culicidae) is a primary vector for dengue viral infections for humans. Current focus on application of natural product against mosquito vectors has been the main priority for research due to its eco-safety. The extensive use of chemical insecticides has led to severe health problems, environmental pollution, toxic hazards to human and nontarget species, and development of insecticide resistance on mosquitoes. Azolla pinnata is an aquatic fern and predominantly used as feed in poultry industry and as fertilizer in agricultural field for enhancing the fertility of rice paddy soil. The present study was conducted to explore the larvicidal efficacy of A. pinnata using fresh and powdered form against late third-stage larvae (6 days, 5 mm in larvae body length) of Ae. aegypti (L.) (Diptera: Culicidae). The larvicidal bioassays were performed using World Health Organization standard larval susceptibility test method for different concentration for powdered and fresh A. pinnata. Powdered A. pinnata concentration used during larvicidal bioassay ranges from 500ppm to 2000ppm; meanwhile, fresh A. pinnata ranges from 500ppm to 9,000,000 ppm. The highest mortality was at 1853 ppm for powdered A. pinnata compared with fresh A. pinnata at 2,521,535 ppm, while the LC50 for both powdered and fresh A. pinnata recorded at 1262 ppm and 1853 ppm, respectively. Finally, the analysis of variance (ANOVA) showed significant difference on Ae. aegypti larval mortality (F=30.439, df=1, p≤0.001) and concentration (F=20.002, df=1, p≤0.001) compared to powdered and fresh A. pinnata at 24-hour bioassay test. In conclusion, the powdered A. pinnata serves as a good larvicidal agent against Ae. aegypti (L.) (Diptera: Culicidae) and this study provided information on the lethal concentration that may have potential for a more eco-friendly Aedes mosquito control program.

The aim of this paper is the evaluation of the trophic status of three lakes riparian to the Colentina River, namely Mogosoaia, Herastrau, Pantelimon, as Part III of an extended study on evaluation of the freshwater resources quality in Bucharest and surrounding areas [1, 2], supplementing the general quality assessment based on indices and indexes presented in Part I with a more in depth analysis of nutrient speciation and partitioning between water and upper sedimentary layers. Water and sediment samples were taken from the inlet and outlet areas of each lake in two sampling campaigns in July 2016 and September 2016. Some of the nutrient related water quality parameters presented in Part I of this series have been used here, together with the analysis of total nitrogen and total phosphorus content in sediments, to assess the general trophic state of the lakes and to highlight the relationships between the nitrogen and the phosphorus content from water and sediments.The studied lakes were found to be eutrophic, Mogosoaia and Pantelimon being in the most advanced state. A significant amount of nitrogen and phosphorus was detected in all sediments analysed, constituting a high accumulation of nutrients from anthropogenic activities. Actions to improve local sanitation infrastructure and to prevent anaerobic leakage of nutrients from sedimentary deposits are recommended.

Serum in embryo culture medium may be a potential cause of abnormal accumulation of lipid droplets, which is correlated to a higher sensitivity to cryopreservation. Moreover, serum may introduce pathogens. With the aim of developing a serum-free culture medium, we first (Experiment 1) investigated the effect of adding ITS (5 μg/mL insulin, 5 μg/mL transferrin, 5 ng/mL selenium) as a serum substitute in SOF medium on embryos cultured in large groups (20 embryos per culture drop of 20 μL) and we then (Experiment 2) analyzed the effect of adding BSA. In this second experiment, our serum-free culture media were also tested on embryos cultured in small numbers (5 embryos per drop of 20 μL) in order to mimic ovum pickup (OPU) conditions. Embryos were obtained from slaughterhouse oocytes, matured in vitro for 24 h in a serum-free enriched 199 medium (Donnay et al. 2004 Reprod. Fertil. Dev. 16, 274) containing ITS, and fertilized for 18 h. In experiment 1, embryos were cultured in SOF (Holm et al. 1999 Theriogenology 52, 683–700) supplemented with 0.1 mg/mL polyvinylpyrrolidane (PVP) without (SOF) or with ITS (SOF-ITS), or with 5% FCS (SOF-FCS). Cavitation occurred earlier in presence of serum (Table). Adding ITS to SOF increased blastocyst rates at Day 7 and Day 8 post-insemination (p.i.) and also the hatching rate. In experiment 2, embryos were cultured in SOF-FCS, SOF-ITS, or SOF-ITS supplemented with 4 mg/mL fatty acid free BSA (SOF-ITS-BSA). Within each condition, no differences were observed for blastocyst and hatching rates between embryos cultured in large or in small groups. Adding BSA to SOF-ITS increased blastocyst rate at Day 6 p.i. and also the hatching rate. At Days 7 and 8 p.i., blastocyst rates were higher in SOF-FCS than in SOF-ITS and tended to be higher than in SOF-ITS-BSA, especially for embryos cultured in small groups. Cell numbers of the resulting embryos were unaffected. These results indicate that: (1) ITS as supplement to SOF medium promotes embryo development in vitro. (2) BSA as protein supplement to SOF-ITS medium accelerates blastulation and improves hatching rate. (3) SOF-ITS and SOF-ITS-BSA are two serum-free culture media that can sustain development of embryos, also when cultured in small number, even though SOF-FCS tended to afford better rates of development. Further studies will include evaluation of other quality parameters including resistance to cryopreservation. Taman Nasional Sembilang merupakan salah satu taman nasional yang terletak di Sumatera Selatan dan menjadi kawasan mangrove terbesar di barat Indonesia. Sebagian besar wilayah mangrovenya telah mengalami alih fungsi menjadi tambak pasang surut. Hal ini menyebabkan timbulnya kekhawatiran kerusakan mangrove di taman nasional. Salah satu upaya untuk mempertahankan wilayah mangrove adalah dengan melakukan sistem silvofishery, yaitu penanaman mangrove dan budidaya bandeng dilakukan secara bersamaan di tambak. Penelitian ini bertujuan untuk menganalisis kualitas air dan kondisi nutrien di tambak di daerah restorasi silvofishery di Taman Nasional Sembilang. Penelitian dilakukan dengan metode purposive sampling di tambak silvofishery, tambak non-silvofishery, dan badan air sungai di seluruh wilayah taman nasional. Pengukuran dilakukan terhadap parameter kualitas air dan kandungan nutrien khususnya nitrat dan fosfat. Hasil penelitian menunjukkan bahwa kehadiran tanaman mangrove di daerah tambak mampu memperbaiki kandungan oksigen dan pH air tambak. Mangrove juga menunjukkan kemampuan dalam mengikat nitrat yang secara tidak langsung akan mencegah air tambak dari polusi. Namun, kandungan fosfat tinggi menunjukkan bahwa tanaman mangrove muda di wilayah restorasi belum mampu mengikat nutrien ini secara signifikan. Hasil penelitian ini menunjukkan bahwa hutan mangrove sangat bermanfaat bagi kegiatan budidaya tambak karena kemampuannya dalam menyerap polutan nutrient. Kata kunci: silvofishery; kolam; nitrat; fosfat; Taman Nasional Sembilang Sembilang National Park is one of the national parks which is located in South Sumatera and became the largest mangrove area in western Indonesia. Most of the mangroves area in this national park has been experiencing conversion to be tidal ponds. This has resulted in concerns on mangrove forest destruction in the park. One of the efforts to maintain mangroves area is by applying silvofishery system, which is mangrove planting and cultivation of milkfish in ponds performed simultaneously. This study aims to investigate the water quality and nutrient condition in the ponds in the area of silvofishery restoration at Sembilang National Park. The study was conducted by purposive sampling method in the silvofishery ponds, non-silvofishery ponds, and river water bodies throughout the park. Measurements conducted on water quality parameters and nutrient content, especially nitrate and phosphate. The results showed that the presence of mangrove in the ponds area is able to improve the oxygen content and pH of pond water. Mangroves also demonstrated the ability to bind nitrates which indirectly will prevent the pond water from pollution. However, the high phosphorus content showed that young mangrove plants in the restoration area have not been able to significantly bind the of this nutrient. The results of this study showed that mangrove forests are very beneficial for aquaculture activities because of its ability to absorb nutrient pollutants. ABSTRACT The effect of surface electrochemical polarization on the growth of cells of Pseudomonas fluorescens (ATCC 17552) on gold electrodes has been examined. Potentials positive or negative to the potential of zero charge (PZC) of gold were applied, and these resulted in changes in cell morphology, size at cell division, time to division, and biofilm structure. At −0.2 V (Ag/AgCl-3 M NaCl), cells elongated at a rate of up to 0.19 μm min−1, rendering daughter cells that reached up to 3.8 μm immediately after division. The doubling time for the entire population, estimated from the increment in the fraction of surface covered by bacteria, was 82 ± 7 min. Eight-hour-old biofilms at −0.2 V were composed of large cells distributed in expanded mushroom-like microcolonies that protruded several micrometers in the solution. A different behavior was observed under positive polarization. At an applied potential of 0.5 V, the doubling time of the population was 103 ± 8 min, cells elongated at a lower rate (up to 0.08 μm min−1), rendering shorter daughters (2.5 ± 0.5 μm) after division, although the duplication times were virtually the same at all potentials. Biofilms grown under this positive potential were composed of short cells distributed in a large number of compact microcolonies. These were flatter than those grown at −0.2 V or at the PZC and were pyramidal in shape. Polarization effects on cell growth and biofilm structure resembled those previously reported as produced by changes in the nutritional level of the culture medium.
