Sound microbiological quality of ice cream should be ensured for being a widely popular dairy food in the world. Present study was conducted to determine the microbiological quality of different ice cream samples available in Dhaka, Bangladesh. Total 3 ice cream samples were collected and processed to detect the microbiological quality as well as drug resistant trait of the isolates through several conventional Kirby Bauer method. All the samples were found to be contaminated with the total viable bacteria and fungi within the range of 1.2×104 cfu/ml to 4.3×106 cfu/ml. The presence of E. coli, Staphylococcus aureus was also observed up to 104 cfu/ml. However, the fecal contamination was totally absent in all the samples. Antibiotic profile of two isolates was measured against 8 commonly used antibiotics and both E.coli and Staphylococcus aureus were found to be resistant against more than one antibiotics. Novel antibiotic drugs are urgently needed because of the increase in drug-resistant bacteria. The use of antimicrobial peptides has been suggested to replace antibiotics as they have strong antimicrobial activity and can be extracted from living organisms such as insects, marine organisms, and mammals. HPA3NT3-A2 ([Ala1,8] HPA3NT3) is an antimicrobial peptide that is an analogue of the HP (2–20) peptide derived from Helicobacter pylori ribosomal protein L1. Although this peptide was shown to have strong antimicrobial activity against drug-resistant bacteria, it also showed lower toxicity against sheep red blood cells (RBCs) and HaCaT cells compared to HPA3NT3. The l-Lys residues of HPA3NT3-A2 was substituted with d-Lys residues (HPA3NT3-A2D; [d-Lys2,5,6,9,10,15] HPA3NT3-A2) to prevent the cleavage of peptide bonds by proteolytic enzymes under physiological conditions. This peptide showed an increased half-life and maintained its antimicrobial activity in the serum against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) (pathogen). Furthermore, the antimicrobial activity of HPA3NT3-A2D was not significantly affected in the presence of mono- or divalent ions (Na+, Mg2+, Ca2+). Finally, l- or d-HPA3NT3-A2 peptides exhibited the strongest antimicrobial activity against antibiotic-resistant bacteria and failed to induce resistance in Staphylococcus aureus after 12 passages.

Abstract The current global death rate has threatened humans due to increase in deadly unknown infections caused by pathogenic microorganisms. On the contrary, the emergence of multidrug-resistant bacteria is also increasing which is leading to elevated lethality rate worldwide. Development of drug-resistant bacteria has become one of the daunting global challenges due to failure in approaching to combat against them. Methicillin-resistant Staphylococcus aureus (MRSA) is one of those drug-resistant bacteria which has led to increase in global mortality rate causing various lethal infections. Polymer synthesis can be one of the significant approaches to combat MRSA by fabricating polymeric coatings to prevent the spread of infections. Introduction: The bacteria colonising the oral cavity and the dentures acquire drug resistance due to frequent usage of antibiotics systemically and application of mouth rinses and denture disinfectants locally. These multidrug resistant bacteria pose potential threat to the health of the patient as infections caused by them do not respond to conventional antibiotics. Aim: The present study aims at detecting the drug resistant bacteria in patients who wear complete dentures. Materials and Methods: The study is a descriptive study and follows laboratory invitro study design involving 30 complete denture patients. Swabs were collected from their oral cavity as well as complete denture surfaces. Antibiotic sensitivity tests were performed for the following bacteriae namely Viridans streptococci species, Staphylococcus aureus, Klebsiella pneumoniae and E.coli. Isolation of the bacteria were done by means of selective media and subjected to biochemical tests. The 16S rRNA sequencing was done to ascertain the microorganisms by which 20 isolates of each of the selective bacteria were obtained. The bacteria were classified as sensitive, intermediate sensitive and resistant based on antibiotic sensitivity tests. Those isolates which exhibited Multi-Drug Resistance (MDR) were visualised using SEM. Results:Viridans streptococci spp. (40%) and Staphylococcus aureus (25%) isolates were resistant to Amoxiclavulinic acid and Methicilin, whereas Klebsiella pneumoniae (30%) and (30%) E.coli isolates were most resistant to Cefotaxime and Doxicilin. Conclusion: Drug resistant bacteria have been identified from complete dentures and oral cavity in the present study. Antibiotic sensitivity tests, 16S rRNA sequencing and SEM are vital investigative tools to detect and to visualise drug resistant bacteria. Cell density, Extracellular Polymeric Substances (EPS) and capsule could be important factors for providing drug resistance.

The lack of antibiotics with a novel mode of action associated with the spread of drug resistant bacteria make the fight against infectious diseases particularly challenging. A quinoline core is found in several anti-infectious drugs, such as mefloquine and bedaquiline. Two main objectives were set in this work. Firstly, we evaluated the anti-mycobacterial properties of the previous quinolines 3, which have been identified as good candidates against ESKAPEE (Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, Enterobacter spp. and Escherichia coli) bacteria. Secondly, a new series 4 was designed and assessed against the same bacteria strains, taking the pair of enantiomers 3m/3n as the lead. More than twenty compounds 4 were prepared through a five-step asymmetric synthesis with good enantiomeric excesses (>90%). Interestingly, all compounds of series 3 were efficient on M. avium with MIC = 2–16 µg/mL, while series 4 was less active. Both series 3 and 4 were generally more active than mefloquine against the ESKAPEE bacteria. The quinolines 4 were either active against Gram-positive bacteria (MIC ≤ 4 µg/mL for 4c–4h and 4k/4l) or E. coli (MIC = 32–64 µg/mL for 4q–4v) according to the global lipophilicity of these compounds.

Freshly prepared and pasteurized fruit juices sold by vendors in local market of Rawalpindi city from 10 locations were analyzed for the microbiological quality. Total viable count (TVC), total coliform, faecal coliform, molds and the presence of pathogenic microorganisms such as E. coli, Pseudomonas, Staphylococcus aureus, Salmonella, and fungi like Aspergillus, Pencillum, Rhizopus were determined .In open fruit juices available in city were highly contaminated with bacteria and fungi. E. coli, Salmonella, Staphylococcus, and Pseudomonas were isolated with different frequency. Aspergillus, Pencillum and Rhizopus were also found in juices especially Aspergillus was with high percentages. The pasteurized juices have less contamination as compared to the fresh juices samples. The number and type of microorganisms recovered from the freshly squeezed fruit juices made them unsafe for drinking. The results of this study demonstrate the unhygienic quality of popular types of market vended freshly squeezed fruit juices and their risk to the consumers. Freshly squeezed or freshly prepared fruit juices sold by local market vendors in Dhaka city were analyzed for the microbiological quality. Total viable bacterial counts, fungal counts, total coliform, faecal coliform and the presence of pathogenic microorganisms such as E. coli, Bacillus cereus, Staphylococcus aureus, Salmonella, Streptococcus were analyzed by standard methods. The total viable count of samples ranged from 3.00×102 to 9.60×108 and fungal counts ranged from 1.00×102 to 8.05×104. Out of 114 freshly prepared fruit juices samples collected 113 samples (99%) showed the presence of coliform and E. coli. The other bacteria like B. cereus, Staphylococcus aureus, Salmonella, Streptococcus were found in 64.91%, 6.14%, 7.89% and (5.26%) of the tested samples. The number and type of microorganisms recovered from the freshly squeezed fruit juices made them unsafe for drinking. It was concluded that due to unhygienic fruit handling in the unsanitary environmental conditions under which the vendors operate the juices become contaminated with harmful bacteria. The results of this study demonstrate the unhygienic quality of popular types of market vended freshly squeezed fruit juices and their risk to the consumers. Vegetables are major source of vitamins and fibers but presence of pathogens in salad vegetables can cause outbreak of diseases. Several studies have been conducted on commonly consumed vegetables so far to detect the level of microbial contamination. However, few of them compare the microbial quality of local and super shop vegetables. The present study was conducted for microbiological assessment and comparing four types of vegetables collected from two different market conditions. In this study, Green chili (Capsicum frutescens L.), Tomato (Solanum lycopersicum), Coriander (Coriandrum sativum) and Lady’s finger (Abelmoschus esculentus) were analyzed to determine Total Viable Bacterial (TVB) Counts, Total Fungal (TF) Counts, Total Coliform Counts (TCC), Total Fecal Coliform (TFC) Counts and occurrence of Staphylococcus aureus, Pseudomonas spp., Listeria spp., Bacillus spp., Salmonella spp., and Vibrio spp. Among these microorganisms, Klebsiella spp. and Pseudomonas spp. were significantly present in coriander of both local market and super shop. All pathogenic bacteria and fungi were found in huge amounts in the vegetable samples of the local market. Although some pathogenic bacteria and fungi were also found in the same samples of the super shop, relatively lower than the local market’s vegetables. In conclusion, contamination in collected vegetable samples were not acceptable but comparatively higher load in local market samples indicated that the hygienic condition in these markets is not sufficiently maintained. ABSTRACT Homodimerization of histatin-derived peptides generally led to improved bactericidal activity against Staphylococcus aureus in vitro. In vivo, monomers and dimers were equally active in killing bacteria in mice with a soft tissue infection. Altogether, these peptides are promising compounds for the development of novel therapeutics against infections with drug-resistant bacteria.
