In this study, methanol extract from the root bark of Calotropis gigantea L. and its petroleum ether (40°C-60°C), chloroform and ethyl acetate soluble fractions were tested for their cytotoxic activity against brine shrimp nauplii (Artemia salina, Leach) and for antifungal activity against Aspergillus flavus, Aspergillus niger, Penicillium sp and Trichoderma harzianum. Thin layer chromatography (TLC) screening showed that methanol extract and its different fractions contained different type compounds such as steroid, terpene, glycoside, heterocyclic and flavonoid. In brine shrimp lethality bioassay, it was found that chloroform fraction was highly cytotoxic (LD50 14.72 μg/ml) among the tested samples. Though methanol extract and ethyl acetate fraction have no activity against all the tested fungi but petroleum ether and chloroform fractions showed potent activity against Aspergillus niger, Penicillium sp, Trichoderma harzianum and Aspergillus niger, Trichoderma harzianum, respectively, in antifungal activity test.  Key words: Calotropis gigantea; Methanol extract; Antifungal activity; Cytotoxicty.DOI: 10.3329/sjps.v2i2.2187Stamford Journal of Pharmaceutical Sciences Vol.2(2) 2009: 38-41

The antibacterial activity of methanol extract from the root bark of Akanda (Calotropis gigantea L.) and its petroleum ether, chloroform and ethyl acetate fractions were investigated. Both of methanol extract and its chloroform fraction showed activity against Sarcina lutea, Bacillus megaterium and Pseudomonas aeruginosa. Petroleum ether fraction showed activity against Bacillus subtilis and Shigella sonnei whereas ethyl acetate fraction showed activity against Pseudomonas aeruginosa and Escherichia coli at 20ìg/disc, 30ìg/disc and 40ìg/disc doses. Among the tested materials, methanol extract and its chloroform fraction showed comparatively better results. Minimum inhibitory concentration (MIC) for methanol extract and each fraction were also determined by serial dilution technique. The petroleum ether, ethyl acetate and methanol extracts of the stem bark of Callistemon citrinus were subjected to screenings for antimicrobial and antioxidant activities and brine shrimp lethality. The ethyl acetate  extract and its column fractions XVIII and XIV exhibited moderate antimicrobial activity, while the methanol extract  revealed significant antioxidant activity having IC50 of 3.84 ?g/ml. The present study was undertaken to evaluate in vitro antimicrobial activity of the successive leaf extracts of Cynodon dactylon in petroleum ether, ethyl acetate, acetone, ethanol, methanol and water, against various gram positive & gram negative bacterial strains using zone of inhibition. Both Agar well diffusion method & Agar disc diffusion method were used to evaluate the antibacterial efficacy. The microorganisms used in the test were – Escherichia coli, Proteus vulgaris, Staphylococcus aureus, Klebsiella pneumoniae, Pseudomonas aeruginosa, Shigella flexneri, Salmonella typhi, Salmonella paratyphi A, Salmonella paratyphi B, Bacillus subtilis, Streptococcus pyogenes, Vibrio cholerac and Enterobacter aerogenes. Two fungal strains - Candida albicans and Saccharomyces cerevisiae were also tested to evaluate the antifungal potential of the said plant extract. The reference antibiotics chloramphenicol & ampicillin (antibacterial); nystatin & clotrimazole and (antifungal) were also tested against these standard microorganisms used in the assay and the results were compared with that of the plant extracts.The results of the study revealed that all the seven successive extracts of the leaf powder of Cynodon dactylon ( L.) Pers. exhibited prominent antimicrobial and antifungal activity against all microorganisms used in the study. The nonpolar extracts i.e. petroleum ether, ethyl acetate and acetone showed the most significant antibacterial and antifungal activity against all tested organisms. The petroleum ether and ethyl acetate extracts showing maximum inhibition in the range of 8 mm - 15 mm. This was closely followed by, acetone extract, which showed the inhibition in the range of 9 mm - 13 mm.

Four essential oils from the leaf (P23) and rhizomes (P19, P22, P24) of Acorus calamus L., collected from various parts of Nepal, were obtained by hydrodistillation and analyzed by GC-MS. From a total of 61 peaks, 57 compounds were identified among the four essential oils accounting for 94.3%, 96.2%, 97.6%, and 94.1% of the oils, respectively. All of the essential oils were dominated by ( Z)-asarone (78.1%–86.9%). The essential oils also contained ( E)-asarone (1.9%–9.9%) and small amounts of γ-asarone (2.0–2.3%), ( Z)-methyl isoeugenol (1.5–2.0%), and linalool (0.2–4.3%). Allelopathic testing of the rhizome oil showed inhibition of seed germination of Lactuca sativa and Lolium perenne with IC50 values of 450 and 737 μg/mL, respectively. The rhizome essential oil demonstrated stronger seedling growth inhibition of L. perenne than of L. sativa, however. The rhizome oil also showed notable brine shrimp lethality ( LC50 = 9.48 μg/mL), cytotoxic activity (92.2% kill on MCF-7 cells at 100 μg/mL), and antifungal activity against Aspergillus niger (MIC = 19.5 μg/mL).

This study was carried out to evaluate the antioxidant, brine shrimp lethality, bioassay and antitumor effects of nhexane, petroleum ether, methanol and chloroform extracts of Annona reticulata leaves. All extracts demonstrated good cupric reducing activity with increasing concentrations keeping methanol extract at the top and n-hexane extract at the second position. The methanol extract was found to have the highest antioxidant capacity 28±26.88 followed by n-hexane 16.95±11.39 and chloroform 12.85±10.32 in the form of mg of ascorbic acid equivalent per gram of crude extract. The methanol extract also showed the highest values of 2.41±0.963 mg of gallic acid equivalent phenolic content and 6.54±4.08 mg of quercetin equivalent flavonoid content per g of crude extract. In the brine shrimp lethality bioassay, LC50 values of methanol, chloroform, petroleum ether, and n-hexane extracts were found to be 1.084?g/ml, 1.096?g/ml, 1.995?g/ml and 3.126 ?g/ml, respectively. These findings suggest that A. reticulata leaves can be a good source of antioxidants and possess significant cytotoxic effect. In a narrow spectrum investigation on antitumor activity of the extracts, we found that the methanol extract inhibited the tumor growing process (control group, chloroform group and petroleum ether group showed significant rise in body weight; i.e., 8% - 14% of initial weight compared to that of methanol group showing a rise of 0.45% - 0.94% of initial weight) and improved ILS (Percentage Increase in Life Span) with increasing doses (Doses of 50mg/kg, 100mg/kg and 200mg/kg showed ILS values of 13.70%, 19.20% and 21.92%, respectively) to some extent when compared with control group and groups receiving other extracts.Bangladesh J Microbiol, Volume 29, Number 2, Dec 2012, pp 70-74

AbstractThe phytochemical and antimicrobial activity of the petroleum ether and crude methanol extracts, chloroform and ethyl acetate fractions of the leaves of Lannea kerstingii were investigated. Phytochemical screening revealed the presence of steroids and triterpenes in the petroleum ether extract, steroid, triterpene, flavonoids and tannins in both crude methanol extract and chloroform fraction while the ethyl acetate fraction contained only flavonoids and tannins. The extracts exhibited antimicrobial activities with zones of inhibition ranging from 17.00 to 21.03, 20.10 to 25.24, 25.32 to 34.02 and 22.28 to 27.20 mm for petroleum ether extract, methanol extract, chloroform and ethyl acetate fractions respectively. The minimum inhibitory concentration was between 5 and 10mg/ml, 5mg/ml for the petroleum ether and methanol extract respectively, and between 2.5 and 5 mg/ml, 5mg/ml for the acetate fractions. The minimum bactericidal concentration for all the extracts was 40mg/ml respectively except for chloroform fraction which ranged from 20 to 40mg/ml. The minimum fungicidal concentration for all the extracts was found to be 40mg/ml respectively. This result indicates the broad spectrum antimicrobial potential of L. Kerstingii and justifies the use of this plant in traditional medicine.

Songi (Dillenia serrata) is a tree endemic to Southeast Sulawesi and its stem bark been used in folk medicine. Nonetheless, only a handful scientific knowledge regarding chemistry and biological activities has been investigated on the plant. The present study aimed to investigate the antioxidant and xanthine oxidase (XO) inhibitory activity of the stem bark of the plant. Methanol extract and organic fractions (petroleum ether, ethyl acetate, and methanol) of the dried powdered stem bark of songi were evaluated for phytochemical screening, total phenolic (TPC), total flavonoid contents (TFC), DPPH, FRAP, and XO in vitro assays. Flavonoids, tannins, terpenoids, steroids, and saponins present in the extract. TPC and TFC in extract were 59.2 mg GAE/g and 23.4 mg QE/g. The contents in organic fractions were in solvent-dependent manner (methanol>ethyl acetate>petroleum ether). Extract and fractions scavenged DPPH radicals (48.2–59.7%) at 100 μg/mL compared to ascorbic acid, trolox, and gallic acid (90.3–93.8%). FRAP values varied from 0.8–3.4 μg/μg equivalent trolox amount (quercetin and gallic acid were 25.7 dan 32.4 μg/μg, respectively). They also inhibited xanthine oxidase (15.3–50.3%) at 100 μg/mL (allopurinol, 98.2%). The study concluded the potential of methanol extract and organic fraction of the stem bark of songi, thus highlighted the prospect of songi to be used in herbal and drugs development from nature.

AbstrakToksisitas akut merupakan kemampuan suatu bahan kimia (obat/bahan obat) dalam menimbulkan kerusakan  pada suatu organisme dalam waktu yang relatif singkat. Tujuan penelitian ini adalah untuk mengetahui potensi ketoksikan akut dari ekstrak metanol, fraksi etil asetat dan fraksi air kulit batang ketapang laut (Terminalia catappa L.). Pengujian toksisitas akut dilakukan dengan menggunakan metode BSLT (Brine Shrimp Lethality Test) dan menggunakan analisis program untuk mengetahui nilai LC50 dari ekstrak dan fraksi. Hasil uji toksisitas akut menunjukan nilai LC50  dari ekstrak metanol, fraksi etil asetat dan fraksi air kulit batang ketapang laut (Terminalia catappa L. )  berturut-turut 247,997 ppm; 400,666 ppm dan 618,046 ppm. Hal ini menunjukan bahwa ekstrak metanol batang ketapang laut (Terminalia catappa L.) bersifat paling toksik dibandingkan fraksi etil asetat dan fraksi air.Kata kunci: Ekstrak, Fraksi, Terminalia catappa L., BSLT, Artemia salina LeachAbstractAcute toxicity is the ability of chemical (drug/drug ingredient) to cause damage to organisms in a relatively short time. The purpose of this study was to determine the potential for acute toxicity of methanol extract, ethyl acetate and water fractions of sea ketapang stem skin (Terminalia catappa L.). Acute toxicity testing was performed using the BSLT (Brine Shrimp Lethality Test) method and program analysis to determine the LC50 values of extracts and fractions. The results of the acute toxicity test showed the LC50 values of methanol extract, ethyl acetate and water fractions of the plant stem skin consecutively were 247,997 ppm; 400,666 ppm and 618,046 ppm. This shows that the methanol extract of sea ketapang stems (Terminalia catappa L.) is the most toxic compared to the ethyl acetate and the water fraction.Keywords: Extract, Fraction, Terminalia catappa L., BSLT, Artemia salina Leach

We evaluated the bioactivities of compounds from Cicuta virosa L. var. latisecta Celak. against red imported fire ants (Solenopsis invicta Buren) under field and laboratory conditions. The compounds were as follows: methanol extract; petroleum ether, chloroform, and ethyl acetate fractions from the methanol extract; and the active compound isoimperatorin, which was isolated from the chloroform-fraction. The 7 d LC50 values of the methanol extract, petroleum ether, chloroform, and ethyl acetate fractions and isoimperatorin toward micrergates were 111.20, 214.45, 40.90, 569.67, and 25.73 mg/kg, res­pectively. The corresponding LC50 values toward macrergates were 155.78, 308.38, 75.01, 776.75, and 42.77 mg/kg, res­pectively. Under field conditions, baits containing 0.2% methanol extract, 0.1% chloroform fraction, and 0.05% isoimperatorin efficiently controlled S. invicta, with effectiveness percentages of 95.56%, 97.78%, and 95.56%, respectively on the 30th day after bait application. Such effectiveness percentages were not significantly different from that obtained using the positive control fipronil. The present study showed that C. virosa L. var. latisecta has potential as a natural control agent for the red imported fire ants.

This work was aimed at evaluating the probable toxicity of the methanol extract of the leaf, stem bark and root of Sacrocephalus latifolius on Artemia nauplii and fronds of Lemna minor. The powder samples of the three morphological parts were independently extracted with soxhlet extractor apparatus using 95% methanol and were dried with aid of ritory evaporator at 40ºC. The extracts were subjected to biological activities involving brine shrimp cytotoxicity and Lemna minor test for phytotoxicity at 10-100 μg /mL respectively. The experiment was done in replicates of three. Concentration-dependent cytotoxic and phytotoxic activities were recorded for the extracts. The results revealed that extract of the leaves demonstrated significant cytotoxic and phytotoxic activities over the other parts of the plant. At 10μg/mL, the three extracts recorded little or no cytotoxic activity. However, at the maximum concentration of 1000 μg /mL, cytotoxicities of 66.67 and 3.33% were recorded for extracts of the leaf and root bark respectively, while the extract of the stem bark showed no activity. However, the LC50 of the leaf was 467.74μg /mL, and that of the other extracts were observed to be ˃1000μg /mL. The results for phytotoxicity followed a similar trend with the extracts of the leaf and root bark recording 52.96 and 23.33 % phytotoxicities respectively at 1000 μg /mL. Having shown a higher activity over the other extracts, the leaf extract of S. latifolius could serves as a natural alternative pesticide and weedicide.
