The United Nations’ One Health initiative advocates the collaboration of multiple sectors within the global and local health authorities toward the goal of better public health management outcomes. The emerging global health threat posed by Aspergillus species is an example of a management challenge that would benefit from the One Health approach. In this paper, we explore the potential role of molecular epidemiology in Aspergillus threat management and strengthening of the One Health initiative. Effective management of Aspergillus at a public health level requires the development of rapid and accurate diagnostic tools to not only identify the infecting pathogen to species level, but also to the level of individual genotype, including drug susceptibility patterns. While a variety of molecular methods have been developed for Aspergillus diagnosis, their use at below-species level in clinical settings has been very limited, especially in resource-poor countries and regions. Here we provide a framework for Aspergillus threat management and describe how molecular epidemiology and experimental evolution methods could be used for predicting resistance through drug exposure. Our analyses highlight the need for standardization of loci and methods used for molecular diagnostics, and surveillance across Aspergillus species and geographic regions. Such standardization will enable comparisons at national and global levels and through the One Health approach, strengthen Aspergillus threat management efforts.

AbstractAround the world, human health and animal health are closely linked in terms of the One Health concept by ticks acting as vectors for zoonotic pathogens. Animals do not only maintain tick cycles but can either be clinically affected by the same tick-borne pathogens as humans and/or play a role as reservoirs or sentinel pathogen hosts. However, the relevance of different tick-borne diseases (TBDs) may vary in human vs. veterinary medicine, which is consequently reflected by the availability of human vs. veterinary diagnostic tests. Yet, as TBDs gain importance in both fields and rare zoonotic pathogens, such as Babesia spp., are increasingly identified as causes of human disease, a One Health approach regarding development of new diagnostic tools may lead to synergistic benefits. This review gives an overview on zoonotic protozoan, bacterial and viral tick-borne pathogens worldwide, discusses commonly used diagnostic techniques for TBDs, and compares commercial availability of diagnostic tests for humans vs. domestic animals, using Germany as an example, with the aim of highlighting existing gaps and opportunities for collaboration in a One Health framework.

Abstract Background Antimicrobial resistance (AMR) is a growing global problem to which the ongoing COVID-19 pandemic may further contribute. With resources deployed away from antimicrobial stewardship, evidence of substantial pre-emptive antibiotic use in COVID-19 patients and indirectly, with deteriorating economic conditions fuelling poverty potentially impacting on levels of resistance, AMR threat remains significant. Main body In this paper, main AMR countermeasures are revisited and priorities to tackle the issue are re-iterated. The need for collaboration is stressed, acknowledging the relationship between human health, animal health and environment (“One Health” approach). Among the stated priorities, the initiative by the European Medicines Regulatory Network to further strengthen the measures in combatting AMR is highlighted. Likewise, it is asserted that other emerging health threats require global collaboration with the One Health approach offering a valuable blueprint for action. Conclusion The authors stress the importance of an integrated preparedness strategy to tackle this public health peril.

The One Health concept offers an integrative approach to disease and health at the human-animal-environment interface. It has often been suggested to view the COVID-19 outbreak within this framework to better understand and mitigate this global crisis. Here, we discuss how the evolutionary ecology of host-pathogen systems can add a valuable additional perspective to the debate around SARS-CoV-2 and its implications for public health awareness and policy-making. In this context, it is especially important to highlight that changes in nature, such as zoonotic spillover events, are often irreversible, and that humans, while deeply embedded in ecosystems, are intricate ecosystems themselves. A better recognition of the complex biology and evolution of human-parasite interactions will assist our understanding of such zoonoses.

Public health belongs to the “One Health” umbrella. As military veterinary medicine evolved, it became embedded in national security. Many armed forces still have active veterinary services, both regular and reserve components. The military veterinarian can serve as an interface between civilians and civil organizations, can handle complex and interdisciplinary cases. Introducing the “One Health” concept both in practice and education has encountered many difficulties. Over time, “One Health” has been judged to be a “buzz word” in civilian areas; however, it is a weighty concept. Its importance is pronounced in military areas where practicing along One Health principles were present before the appearance of the term itself. Nevertheless, military “One Health” has not penetrated into the overwhelming “One Health” literature. Emphasizing the military aspects of One Health not only reveals an obscure corner but might help to regain the proper importance of the “One Health” concept.

In this chapter, the author aim to approach new ways to understand how emerging technologies can better be applied in organizational contexts. For this purpose, collaborative methodological approaches were addressed—multi, inter, and transdisciplinary paradigms—aiming to promote a better level both of comprehension and adoption of technologies, paying special attention to the healthcare sector and to the One Health initiative, just defined as an interdisciplinary front. As an overall goal for the chapter, the adoption of those methodological principles is advised to the reader, enabling a better understanding of those technologies and their way to be effectively implemented.

Coronaviruses (CoVs) are worldwide distributed RNA-viruses affecting several species, including humans, and causing a broad spectrum of diseases. Historically, they have not been considered a severe threat to public health until two outbreaks of COVs-related atypical human pneumonia derived from animal hosts appeared in 2002 and in 2012. The concern related to CoVs infection dramatically rose after the COVID-19 global outbreak, for which a spill-over from wild animals is also most likely. In light of this CoV zoonotic risk, and their ability to adapt to new species and dramatically spread, it appears pivotal to understand the pathophysiology and mechanisms of tissue injury of known CoVs within the “One-Health” concept. This review specifically describes all CoVs diseases in animals, schematically representing the tissue damage and summarizing the major lesions in an attempt to compare and put them in relation, also with human infections. Some information on pathogenesis and genetic diversity is also included. Investigating the lesions and distribution of CoVs can be crucial to understand and monitor the evolution of these viruses as well as of other pathogens and to further deepen the pathogenesis and transmission of this disease to help public health preventive measures and therapies.
